Electron transport in the cni-1 mutant of Neurospora crassa.

1. Mitochondria from the nuclear mutant cni-1 have no optically detectable cytochrome aa3 in early log phase growth. These mitochondria have a high level of respiration that is not inhibited by cyanide but is inhibited by salicylhydroxamic acid. They also show a substantial amount of cyanide-sensitive respiration. 2. As cultures of mutant cni-1 age, flux through the hydroxamate-sensitive pathway decreases markedly while flux through the cytochrome chain remains constant. 3. Growth studies with mutant cni-1 indicate that the cytochrome chain in this mutant is more important in supporting growth

Properties and reconstitution of a cytochrome oxidase deficient in subunit III.

Three different preparations of beef heart cytochrome oxidase (EC 1.9.3.1) were reconstituted into the membranes of artificial liposomes, and the electrical charge/electron ratios were determined for charge translocation coupled to enzymic activity. Our previously characterised subunit-III-deficient preparation, which apparently lacks H+ translocation capacity [Saraste et al. (1981) Eur. J. Biochem. 115, 261-268] has a decreased charge/electron ratio (0.9-1.0) as determined from the uptake of potassium in the presence of valinomycin, in contrast to the intact reconstituted cytochrome oxidase (

First Report of Cutaneous Lepidopterism by Hylesia invidiosa (Lepidoptera: Saturniidae): Outbreak Description and Review of the Literature.

Cutaneous lepidopterism associated with exposure to setae from Hylesia moths has been frequently reported in the Americas. Despite efforts to understand its pathophysiology, little is known about the biology of the implicated species, treatment approaches, and outbreak prevention and control. This affects the direction of clinical management and exacerbates the social and public health implications of lepidopterism. In March 2024, the Ministry of Health of Costa Rica was alerted about human cases of dermatitis in the Tucurrique District, Cartago Province, which residents attributed to moths. F

First Record of Cinara tujafilina (Hemiptera: Aphididae) in Myanmar Based on Morphological and Molecular Evidence.

The cypress pine aphid, Cinara (Cupressobium) tujafilina (Del Guercio, 1909), is a widely distributed aphid associated with Cupressaceae and known for forming dense colonies that promote sooty mold development and reduce host vigour. Here, we report the first confirmed occurrence of this species in Myanmar. Specimens collected from Platycladus orientalis were identified using morphological characters and mitochondrial COI sequences. Genetic analyses revealed low intraspecific divergence and a clear barcode gap relative to closely related species, supporting accurate species identification. Hap

Alteration in Cerebral Metabolism in a Rodent Model of Acute Sub-lethal Cyanide Poisoning.

INTRODUCTION: Cyanide exposure can occur in various settings such as industry and metallurgy. The primary mechanism of injury is cellular hypoxia from Complex IV (CIV) inhibition. This leads to decreased ATP production and increased reactive oxygen species production. The brain and the heart are the organs most affected due to their high metabolic demand. While the cardiac effects of cyanide are well known, the cerebral effects on cellular function are less well described. We investigated cerebral metabolism with a combination of brain respirometry, microdialysis, and western blotting using a

Inter-Specific Genetic Exchange Despite Strong Divergence in Deep-Sea Hydrothermal Vent Gastropods of the Genus Alviniconcha.

Deep hydrothermal vents are highly fragmented and unstable habitats at all temporal and spatial scales. Such environmental dynamics likely play a non-negligible role in speciation. Little is, however, known about the evolutionary processes that drive population-level differentiation and vent species isolation and, more specifically, how geography and habitat specialisation interplay in the species history of divergence. In this study, the species range and divergence of Alviniconcha snails that occupy active Western Pacific vent fields was assessed by using sequence variation data of the mitoc

Adaptive evolution and functional redesign of core metabolic proteins in snakes.

BACKGROUND: Adaptive evolutionary episodes in core metabolic proteins are uncommon, and are even more rarely linked to major macroevolutionary shifts. METHODOLOGY/PRINCIPAL FINDINGS: We conducted extensive molecular evolutionary analyses on snake mitochondrial proteins and discovered multiple lines of evidence suggesting that the proteins at the core of aerobic metabolism in snakes have undergone remarkably large episodic bursts of adaptive change. We show that snake mitochondrial proteins experienced unprecedented levels of positive selection, coevolution, convergence, and reversion at funct

New insights into the phylogeny of fig pollinators using Bayesian analyses.

The interaction between figs and fig pollinators is one of the most species-specific mutualisms. Recently, phylogenies of both partners based on molecular data provided insights into a wide spectrum of co-evolutionary questions. However, for the phylogeny of fig pollinators, there are some discrepancies between different studies and left some relationships unresolved, especially for deep nodes. The phylogenetic uncertainties of pollinators prohibit our further understanding of the history of the mutualism. Here, we present phylogenetic analyses of a larger COI sequence dataset that includes pr

Distribution of glycogen phosphorylase and cytochrome oxidase in the central nervous system of the turtle Trachemys dorbigni.

Glycogen phosphorylase (GP) and cytochrome oxidase (CO) activities were mapped histochemically in the brain of the turtle Trachemys dorbigni. In the telencephalon, both activities occurred in the olfactory bulb, in all cortical areas, in the dorsal ventricular ridge, striatum, primordium hippocampi and olfactory tubercle. In the diencephalon, they were identified in some areas of the hypothalamus, and in rotundus and geniculate nuclei. Both reactions were detected in the oculomotor, trochlear, mesencephalic trigeminal nuclei, the nucleus of the posterior commissure, torus semicircularis, subst

The reduced minus oxidized difference spectra of cytochromes a and a3.

We have re-investigated the reduced minus oxidized difference spectra of the two heme centers of cytochrome oxidase, cytochromes a and a3. In contrast to data obtained in an earlier study (Vanneste, W.H. (1966) Biochemistry 5, 838-848), we find that the spectrum for cytochrome a3 agrees with that found with a 5-coordinate high-spin heme A model compound. Small but significant additional differences are noted for both heme centers.

"All-or-none" cytochrome c oxidase positivity in mitochondria in chronic progressive external ophthalmoplegia: an ultrastructural--cytochemical study.

Teased single muscle fibers from 6 patients with chronic progressive external ophthalmoplegia (CPEO) showed a segmental defect in cytochrome c oxidase (COX) activity. On ultrastructural--cytochemical examination, the majority of mitochondria in COX-positive segments were COX-positive, whereas all mitochondria in COX-negative segments were COX-negative. This "all-or-none" COX positivity in mitochondria in CPEO with deleted mitochondrial DNA can be explained by the "threshold effect," which induces the tissue-specific involvement and clinical heterogeneity.

Cytochrome c oxidase and coenzyme Q in neuromuscular diseases: a histochemical study.

Cytochrome c oxidase (CCO) has been histochemically studied in 250 muscle biopsies from patients with different neuromuscular diseases. The results were compared with those obtained on serial sections stained with Gomori's trichrome and with the methods for NADH tetrazolium reductase, succinate dehydrogenase and lactate dehydrogenase. In 58 selected cases serial sections were also stained with a method demonstrating coenzyme Q (CoQ) activity. Demonstration of structural alterations was as good with CCO as with the methods for other oxidative enzymes: particularly evident were alterations of th

Detection of cryptic Taenia species from domestic and sylvatic transmission cycles in southern pudu deer (Pudu puda), Chile.

Anthropogenic landscape fragmentation promotes interactions between domestic animals and wildlife, facilitating cross-species parasite transmission. Through molecular identification, we report cryptic infection by three Taenia species (T. hydatigena, T. krabbei, and T. omissa) in the Southern pudu (Pudu puda), a threatened cervid endemic to temperate rainforests of South America. From necropsies of twelve pudus from rural areas of Southern Chile, we recovered 44 metacestode cysts from diverse organs. Molecular and phylogenetic analyses of the cytochrome c oxidase subunit I (COX-1) gene confirm

Genetic characterization and new records of snail species of the genus Galba (Lymnaeidae) in two environments of Salta province, Northwestern Argentina.

The development and transmission of Fasciola hepatica, a parasitic flatworm responsible for causing fasciolosis, depend on the presence of freshwater snails belonging to the family Lymnaeidae, mainly of the genus Galba, which are obligate intermediate hosts. Although accurate identification of these snails is critical to understand disease transmission, the morphological similarities between species make taxonomic identification difficult. This study provides the first genetic characterization of freshwater snail species of the genus Galba in Salta province, Argentina. Snails were collected fr

Mitochondrial Complex IV Deficiency Nuclear Type 11 Caused by a Novel Start-Lost Variant in the COX20 Gene.

Background: The COX20 gene encodes a critical assembly factor for cytochrome C oxidase (complex IV), and biallelic loss-of-function variants in this gene cause mitochondrial complex IV deficiency, typically presenting in infancy or childhood with hypotonia, ataxia, neuropathy, or dystonia. Methods: This study describes an adult male patient with a broad clinical spectrum of central and peripheral nervous system involvement. Different medical genetic tests were performed for the patient, and only whole-genome trio sequencing identified pathogenic variants in the COX20 gene. A review of previous

Identification of a Clinical Spirometra mansoni Plerocercoid Isolate Using Molecular and Morphological Data.

Sparganosis has been a neglected parasitic zoonosis for a long time. The accurate identification of Spirometra tapeworms in clinical practice is poorly understood. A case of breast sparganosis was reported in Henan Province of central China. One plerocercoid approximately 3.5 cm in length was collected from the patient. The clinical isolate was identified as Spirometra mansoni based on the barcoding sequences of mitochondrial cytochrome c oxidase subunit 1 (cox1). Finally, the epidemiology of sparganosis in central China was reviewed. Comprehensive public health education should be carried out

Methemoglobin-albumin clusters for cyanide detoxification.

Sodium nitrite (NaNO2) is a universal antidote for patients with cyanide poisoning. However, its use has serious drawbacks in terms of efficacy and safety. Herein, we present a promising antidote: methemoglobin (metHb)-albumin clusters. The metHb-albumin cluster is made by a metHb core wrapped by covalently bound human serum albumin. Spectral analyses proved that the metHb-albumin clusters possessed cyanide-binding properties similar to those of naked metHb. In vitro cell experiments showed that metHb-albumin clusters prevented the cyanide-induced inhibition of cytochrome c oxidase activity, r
