Repeated early-adolescent ethanol intoxication promotes riskier decision-making in adult male mice and increases drinking in adult female mice.

Exceeding binge-level alcohol drinking is common among adolescents and is associated with both short- and long-term adverse outcomes. This study evaluated the effects of repeated ethanol intoxication during early-adolescence on behavioral outcomes in early-adulthood. Male and female C57BL/6 mice (5 weeks old) received four intraperitoneal (i.p.) injections, over two weeks, of either saline (control) or 3.2 g/kg ethanol (intoxicated). A subset of intoxicated mice had blood collected on days 1 and 4, confirming heavy intoxication (∼289 mg/dL). At 9 weeks of age, animals were tested in the Light-

Exploratory activity, anxiety, and motor coordination in bigenic APPswe + PS1/DeltaE9 mice.

Bigenic APPswe + PS1/DeltaE9 mice with Abeta plaque formation in neocortex and hippocampus were evaluated in three tests measuring exploratory activity. By comparison to a non-transgenic group controlled for age and gender, 7-month-old APPswe + PS1/DeltaE9 mice spent more time in the open arms and had higher open/total entries and duration in the elevated plus-maze, indicative of disinhibitory tendencies. On the contrary, the groups did not differ in T-maze and open-field tests. Moreover, the motor coordination of the bigenic group was equivalent to that of controls in stationary beam, coat-ha

Differential long term effects of early diisopropylfluorophosphate exposure in Balb/C and C57Bl/J6 mice.

The long-term effect of postnatal administration of a sub-toxic dose of the irreversible acetylcholinesterase inhibitor diisopropylfluorophosphate (DFP) on depression and anxiety behavior was compared in two strains of inbred mice. C57BL/6J and Balb/C mice were injected for 7 consecutive days with either 1 mg/kg DFP or saline on postnatal days 14-20. Mice were tested at age 3-4 months for initial and learned anxiety using double-exposure elevated plus maze and to a novel enclosed environment. Depression was assayed using the sweet preference model of anhedonia and the forced swim test for desp

Illumination has no effect on rats' behavior in the elevated plus-maze.

Male Wistar rats Shoe:Wist(Shoe) were tested in the elevated plus-maze under three different illumination levels (30, 300, and 900 lx). It was found that illumination did not change percentage of time spent in closed arms, number of closed arm entries, or time spent on open arms, or total arm entry. This confirms earlier findings that rat's behavior in the elevated plus-maze is independent of light levels.

Gut microbiota composition is correlated to grid floor induced stress and behavior in the BALB/c mouse.

Stress has profound influence on the gastro-intestinal tract, the immune system and the behavior of the animal. In this study, the correlation between gut microbiota composition determined by Denaturing Grade Gel Electrophoresis (DGGE) and tag-encoded 16S rRNA gene amplicon pyrosequencing (454/FLX) and behavior in the Tripletest (Elevated Plus Maze, Light/Dark Box, and Open Field combined), the Tail Suspension Test, and Burrowing in 28 female BALB/c mice exposed to two weeks of grid floor induced stress was investigated. Cytokine and glucose levels were measured at baseline, during and after e

Potential anxiogenic effects of cannabinoid CB1 receptor antagonists/inverse agonists in rats: comparisons between AM4113, AM251, and the benzodiazepine inverse agonist FG-7142.

Cannabinoid CB1 inverse agonists suppress food-motivated behaviors, but may also induce psychiatric effects such as depression and anxiety. To evaluate behaviors potentially related to anxiety, the present experiments assessed the CB1 inverse agonist AM251 (2.0-8.0mg/kg), the CB1 antagonist AM4113 (3.0-12.0mg/kg), and the benzodiazepine inverse agonist FG-7142 (10.0-20.0mg/kg), using the open field test and the elevated plus maze. Although all three drugs affected open field behavior, these effects were largely due to actions on locomotion. In the elevated plus maze, FG-7142 and AM251 both pro

Individual differences in circadian locomotor parameters correlate with anxiety- and depression-like behavior.

Disrupted circadian rhythms are a core feature of mood and anxiety disorders. Circadian rhythms are coordinated by a light-entrainable master clock located in the suprachiasmatic nucleus. Animal models of mood and anxiety disorders often exhibit blunted rhythms in locomotor activity and clock gene expression. Interestingly, the changes in circadian rhythms correlate with mood-related behaviours. Although animal models of depression and anxiety exhibit aberrant circadian rhythms in physiology and behavior, it is possible that the methodology being used to induce the behavioral phenotype (e.g.,

Prenatal nicotine exposure increases anxiety and modifies sensorimotor integration behaviors in adult female mice.

Prenatal exposure to nicotine (PNE) has been associated with a myriad of physiological, cognitive, and behavioral effects in the developing offspring. In this study, CD-1 dams were given injections of nicotine or control vehicle throughout gestation and their offspring were raised to 6 months of age. Adult mice were administered a battery of behavioral tests (the Suok test, the elevated platform test, and the elevated plus maze test) to assess anxiety and sensorimotor integration. PNE resulted in a decreased likelihood of jumping during the elevated platform test and decreased directed explora

Adolescent nicotine treatment causes robust locomotor sensitization during adolescence but impedes the spontaneous acquisition of nicotine intake in adult female Wistar rats.

Very few people are able to quit smoking, and therefore it is essential to know which factors contribute to the development of compulsive nicotine use. These studies aimed to investigate if early-adolescent nicotine exposure causes locomotor sensitization and affects anxiety-like behavior and the spontaneous acquisition of intravenous nicotine self-administration. Early-adolescent male and female rats were treated with nicotine from postnatal (P) days 24 to 42, and anxiety-like behavior and locomotor activity were investigated one day after the cessation of nicotine treatment and in adulthood

The effects of perinatal fluoxetine exposure on emotionality behaviours and cortical and hippocampal glutamatergic receptors in female Sprague-Dawley and Wistar-Kyoto rats.

RATIONALE: There is increasing concern regarding the use of selective serotonin reuptake inhibitors (SSRIs) in pregnancy. Animal studies repeatedly show increased anxiety- and depressive-like behaviours in offspring exposed perinatally to SSRIs, however much of this research is in male offspring. OBJECTIVES: The primary aim of this study was to investigate the effects of perinatal SSRI exposure on emotionality-related behaviours in female offspring and associated glutamatergic markers, in Sprague-Dawley (SD) rats and in the Wistar-Kyoto (WKY) rat model of depression. Secondly, we sought to in

Anxioselective profile of glycineB receptor partial agonist, D-cycloserine, in plus-maze-naïve but not plus-maze-experienced mice.

Recent successes as a pharmacological adjunct to exposure therapy has focused attention on the therapeutic potential of the glycine(B) receptor partial agonist, D-cycloserine (DCS), in certain clinical anxiety disorders. Although widely believed to reflect a facilitation of extinction learning, previous research with DCS and other glycine(B) partial agonists suggests the additional possibility of intrinsic anxiolytic activity. In the present study, ethological methods were used to profile the behavioural effects of DCS (7.5-30.0mg/kg) and the positive control chlordiazepoxide (CDP, 15 mg/kg) i

The benzodiazepine receptor inverse agonist FG 7142 induces cholecystokinin gene expression in rat brain.

The effects of acute administration of the anxiogenic benzodiazepine receptor ligand, N-methyl-beta-carboline-3-carboxamide (FG 7142) and of a single exposure to the elevated plus-maze test of anxiety on preprocholecystokinin mRNA levels in rat brain were examined using the technique of in situ hybridisation. Administration of FG 7142 (10 mg/kg i.p.), but not elevated plus-maze exposure, increased cholecystokinin (CCK) mRNA levels in the basolateral amygdala and the CA3 pyramidal cell layer of the hippocampus. Neither stimulus produced changes in thalamic structures. These data suggest that dr

Increased anxiety behavior in OLETF rats without cholecystokinin-A receptor.

Cholecystokinin (CCK) may have a role in the mediation of human panic disorder and anxiogenic (anxiolytic)-like activity in an animal model of anxiety. Otsuka Long Evans Tokushima Fatty (OLETF) rats lacked CCK A receptors (CCKAR) because of a genetic abnormality. In order to elucidate the involvement of CCKAR in the regulation of anxiety, we investigated the exploratory behavior on elevated plus-maze test, the black and white box test, and open field test with OLETF rats in comparison with normal [Long-Evans Tokushima Otsuka (LETO)] rats. And OLETF rats increased the number of stretched attend

A unique polygenic mouse model of obesity exhibits a distinct immunological profile that may offer protection against systemic inflammation, diabetes, and behavioral impairments.

In both humans and mice, obesity is often associated with peripheral and central inflammation, which can lead to diabetes, dysregulation of the stress response, changes in affective behavior, and memory impairment. The DU6 polygenic mouse line was selected over more than 180 generations for a high body mass. Unlike other mouse lines, DU6 mice do not develop diabetes despite significant obesity. We performed a series of behavioral tests on male mice because obesity is often associated with cognitive and emotional impairment. DU6 mice showed no differences in spatial memory or anxiety compared t

Utility of ethological analysis to overcome locomotor confounds in elevated maze models of anxiety.

The elevated plus-maze is a commonly used model to identify putative anxiolytic and anxiogenic drugs. However, the validity of elevated plus-maze and other recently developed variants such as the elevated zero-maze has recently been questioned on the grounds that both the reference anxiolytic drug chlordiazepoxide and the psychostimulant d-amphetamine increase open arm exploration and stimulate locomotor activity. These findings suggest that measures of "anxiety" in the elevated maze cannot be adequately dissociated from simple changes in locomotor activity, which may confound the interpretati

Fear-like behavioral responses in mice in different odorant environments: Trigeminal versus olfactory mediation under low doses.

Odors can have repulsive effects on rodents based on two complementary adaptive behaviors: the avoidance of predator odors (potentially dangerous) and the avoidance of trigeminal stimulants (potentially noxious). The present study aimed to compare the behavioral effects on mice of odors according to their trigeminal properties and ecological significance. We used three different odors: 2,4,5-trimethylthiazoline (TMT: a fox feces odor frequently used to elicit fear-induced behaviors), toluene (a strong stimulant of the trigeminal system) and phenyl ethyl alcohol (PEA: a selective stimulant of t
