Assisted reproduction technology (ART) has advanced significantly over the past four decades, leading to improved pregnancy outcomes and a reduction in complications, particularly those associated with multiple pregnancies. These improvements largely stem from advances in understanding embryonic physiology, which has enabled better culture conditions. As a result, embryologists can now efficiently culture embryos to the blastocyst stage and successfully cryopreserve them for future use. However, while incubators aim to replicate the maternal environment of the oviduct and uterus, embryos in vi

Sheep somatic cell nuclear transfer (SCNT) is widely used in both agricultural and biomedical research, especially for generating animal models of human disease. Here, we present the most accessible method of sheep oocyte collection for use in subsequent SCNT, using commercial in vitro maturation and culture media to increase SCNT efficiency. We describe a detailed procedure of sheep SCNT and SCNT animal production, as well as outline two methods to determine embryo quality: in vitro culture until blastocyst formation and blastocyst inner cell mass and trophectoderm cell staining.

Spent human embryo culture media contains low-molecular-weight metabolites that can provide a non-invasive readout of preimplantation embryo physiology. However, nuclear magnetic resonance (NMR)-based metabolomic analysis of these samples is technically challenging because routine embryo culture conditions involve microliter volumes, protein supplementation, and paraffin oil overlays that generate background signals and compromise spectral quality. Here, we present an optimized NMR metabolomics workflow for spent embryo culture media incorporating controlled droplet collection to minimize oil

It is surprising that so little attention is currently given to in vitro culture of preimplantation rabbit embryos, even though the rabbit is the only laboratory animal in which there is very considerable embryo growth before implantation, resulting in a 300-fold increase in protein content of embryonic cells during the preimplantation period and the formation of more than a 100,000 cells in the blastocyst. This growth pattern explains why blastocyst formation in vitro has an absolute requirement for amino acids, and vitamins, particularly inositol, are esssential for blastocyst growth. A semi

BACKGROUND: The recent identification of embryonic cell-free DNA in spent blastocyst media has opened a new era of possibilities for noninvasive embryo aneuploidy testing in assisted reproductive technologies. Yet, previous studies assessing a limited number of embryos reported variable concordance between embryonic cell-free DNA and trophectoderm biopsies, thus questioning the validity of this approach.
OBJECTIVE: This study aimed to evaluate the concordance and reproducibility of testing embryonic cell-free DNA vs trophectoderm DNA obtained from the same embryo in a large sample of human bl

OBJECTIVE: To determine if a dynamic embryo culture system affects the reproductive potential of human embryos resulting from in vitro fertilization (IVF).
DESIGN: Paired randomized controlled trial (RCT).
SETTING: IVF center.
PATIENT(S): IVF patients with normal ovarian reserve eligible for two-embryo transfer.
INTERVENTION: IVF care was routine until fertilization was confirmed. Two-pronuclear embryos (2PNs) were then randomized: One-half of each patient's 2PNs were cultured in dynamic culture and one-half in static culture. Preimplantation genetic testing for embryonic aneuploidy was us

The development of the preimplantation embryo seems morphologically very simple, and embryologists previously assumed that an embryo that developed to the blastocyst stage was fully capable of normal development after transfer to the uterus of a recipient female. This complacency was disturbed by reports that exposure of early embryos to mutagens such as methylnitrosourea led to fetal abnormalities, decreased birth rates, and decreased life-span. Even more disturbing are recent reports that culture of early embryos in supposedly benign conditions can adversely affect their subsequent developme

Guinea pigs are a valuable animal model for studying various diseases, including reproductive diseases. However, techniques for generating embryos via embryo engineering in guinea pigs are limited; for instance, in vitro maturation (IVM) technique is preliminary for guinea pig oocytes. In this study, we aimed to establish and optimize an IVM method for guinea pig oocytes by investigating various factors, such as superovulation induced by different hormones, culture supplementation (e.g., amino acids, hormone, and inhibitors), culture conditions (e.g., oocyte type, culture medium type, and trea

Embryo transfer in cattle is an increasingly important technique for cattle production. Full attainment of the benefits of the technology will depend on overcoming hurdles to optimal performance using embryos produced in vitro. Given its importance, embryo technology research should become a global research priority for animal reproduction science. Among the goals of that research should be developing methods to increase the proportion of oocytes becoming embryos through optimization of in vitro oocyte maturation and in vitro fertilization, producing an embryo competent to establish and mainta

Advanced age is a risk factor undermining women's fertility. Hence, the optimization of assisted reproduction techniques is an interdisciplinary challenge that requires the improvement of in vitro culture systems. Here, we hypothesize that supplementation of embryo culture medium with extracellular vesicles from endometrial-derived mesenchymal stem cells (EV-endMSCs) may have a positive impact on the embryo competence of aged oocytes. In this work, 24 weeks old B6D2 female mice were used as egg donors and in vitro fertilization assays were performed using males from the same strain (8-12 weeks

In this study, the productivity and post-transfer pregnancy rates of bovine embryos in vitro-produced (IVP) by a two-step culture system were compared with those of in vivo-developed (IVD) embryos. Ovum pick-up was designated as Day 0, presumptive zygotes were cultured in modified synthetic oviduct fluid without glucose (mSOFaa), whereas late-stage embryos from Day 5 onward were cultured in TCM 199 supplemented with 5% fetal calf serum and 100 μM β-mercaptoethanol. An average of 5.9 ± 4.8 blastocysts was obtained from 26 donors. A total of 46 fresh and 18 vitrified IVP embryos were transferred

Preimplantation genetic testing for aneuploidies (PGTA) diagnoses chromosomal abnormalities and can be complemented by cumulus cell transcriptional analysis to assess egg and ovarian quality and development during embryo culture. In addition to selecting the best available embryo, intrauterine administration of platelet-rich plasma (PRP) and correction of insulin resistance (IR) through nutritional intervention can improve endometrial quality. This case report examines the potential feasibility of combining in vitro fertilization (IVF) with four techniques to achieve a viable pregnancy. A coup

In the early days of assisted reproductive technology (ART), the main target was achieving gestation. Success rates were low, and multiple embryo transfers became common practice, with multiple pregnancies being 20 times higher than in natural conception. Multiple pregnancy is associated with a higher risk of complications for the mother and the baby than a singleton pregnancy. Added to healthcare costs, multiple pregnancy also involves other costs and psychosocial risks, with a high social and health costs. At present, success rates of assisted human reproduction (AHR) have improved dramatica

This study investigated the potential of assessing human embryo viability through the analysis of the embryo culture medium used in assisted reproductive technology (ART) via Raman spectroscopy. Two types of culture media-one for early development (medium A) and one for recovery after vitrifying/warming (medium B)-were analyzed. Our results showed that protein concentrations were higher in medium A in which embryos reached the blastocyst stage compared to those in which embryos arrested before this stage. Moreover, the pH of medium A associated with higher-grade blastocysts tended to be more a

It can be assumed that the natural processes of selection and developmental condition in the animal provide the best prerequisites for embryogenesis resulting in pregnancy and subsequent birth of a healthy neonate. In contrast, circumventing the natural selection mechanisms and all developmental conditions in a healthy animal harbors the risk of counteracting, preventing or reducing the formation of embryos or substantially restricting their genesis. Considering these facts, it seems to be obvious that assisted reproductive techniques focusing on early embryonic stages serve an expanded and un

NMR offers the potential to holistically screen hundreds of metabolites and has already proved to be a powerful technique able to provide a global picture of metabolic changes in a wide range of biological systems underlying complex and multifactorial matrixes. This review covers the literature until May 2020 centered on the early prediction of the viability of in vitro developed embryos using several analytical techniques, including NMR. Nowadays, the predominant non-invasive technique for selecting viable embryos is based on morphology, where variables associated with the rate of cleavage an

We describe two complementary methods for the study of early thymus organogenesis in the mouse. The first is an in vitro technique for lineage analysis, where a chosen population of cells within the mouse embryo is labeled with a fluorescent cell tracker dye. The embryos are then transferred to whole embryo culture for a defined period, after which time the location of the labeled cells is determined with respect to the developing thymus. In the second method, an in vivo assay is used to determine the ability of a specific tissue type to form a structurally and functionally normal thymus. This

A successful pregnancy requires a synchronous dialogue between endometrium and embryo within the endometrial milieu. The aim of this study was to assess the role in the implantation of mediators in the endometrial milieu. Total secretome (TS), soluble secretome (SS) and small extracellular vesicles (containing exosomes) were generated from hormonally primed human endometrial epithelial cell culture medium. Human trophectoderm stem cell-derived spheroids were cultured with TS, SS or exosomes (30 µg/ml) on hormonally primed epithelial cells, with exosomes significantly increasing cell adhesion a

STUDY QUESTION: Can a generally applicable morphokinetic algorithm suitable for Day 3 transfers of time-lapse monitored embryos originating from different culture conditions and fertilization methods be developed for the purpose of supporting the embryologist's decision on which embryo to transfer back to the patient in assisted reproduction?
SUMMARY ANSWER: The algorithm presented here can be used independently of culture conditions and fertilization method and provides predictive power not surpassed by other published algorithms for ranking embryos according to their blastocyst formation po

Co-culture techniques using fetal bovine uterine fibroblasts or bovine oviductal epithelial cells have improved embryonic development prior to replacement in humans. In initial co-culture trials, embryo development and implantation rates increased after just 1 day in culture. The most overt characteristics noted following co-culture were improved blastomere development and characteristics, reduced fragmentation, and the appearance of swollen blastomeres. In addition, an increase in the incidence of zona thickness variation was detected. Improved development of polyspermic and supernumerary emb
