The small bovine amelogenin LRAP fails to rescue the amelogenin null phenotype.

Amelogenins are the most abundant secreted proteins in developing dental enamel. These evolutionarily-conserved proteins have important roles in enamel mineral formation, as mutations within the amelogenin gene coding region lead to defects in enamel thickness or mineral structure. Because of extensive alternative splicing of the primary RNA transcript and proteolytic processing of the secreted proteins, it has been difficult to assign functions to individual amelogenins. To address the function of one of the amelogenins, we have created a transgenic mouse that expresses bovine leucine-rich am

Tooth Enamel and its Dynamic Protein Matrix.

Tooth enamel is the outer covering of tooth crowns, the hardest material in the mammalian body, yet fracture resistant. The extremely high content of 95 wt% calcium phosphate in healthy adult teeth is achieved through mineralization of a proteinaceous matrix that changes in abundance and composition. Enamel-specific proteins and proteases are known to be critical for proper enamel formation. Recent proteomics analyses revealed many other proteins with their roles in enamel formation yet to be unraveled. Although the exact protein composition of healthy tooth enamel is still unknown, it is appa

Biological synthesis of tooth enamel instructed by an artificial matrix.

The regenerative capability of enamel, the hardest tissue in the vertebrate body, is fundamentally limited due to cell apoptosis following maturation of the tissue. Synthetic strategies to promote enamel formation have the potential to repair damage, increase the longevity of teeth and improve the understanding of the events leading to tissue formation. Using a self-assembling bioactive matrix, we demonstrate the ability to induce ectopic formation of enamel at chosen sites adjacent to a mouse incisor cultured in vivo under the kidney capsule. The resulting material reveals the highly organize

Aberrant root formation: review of root genesis and three case reports.

The mechanism of root formation and tooth eruption is a complex process which is not fully understood. Prior to a tooth emerging into the oral cavity, root genesis is initiated by derivatives of the enamel organ. The dental follicle mediates an eruption pathway allowing for movement of the developing tooth in a coronal direction. As the tooth moves towards the oral cavity, root formation occurs passively in the resulting space. Failure of the enamel organ and dental follicle to properly coordinate may result in complications in the eruption process. This clinical report presents 3 cases of iso

Effects of fluoride and lead on enamel composition during the maturation stage of amelogenesis in rat mandibular third molars.

Dental enamel formation depends on tightly regulated mineral deposition and organic matrix removal during the maturation stage. Fluoride (F) adversely affects the maturation stage of amelogenesis, causing hypomineralized enamel defects. However, hypomineralized enamel defects have increased worldwide in recent years, making it urgent to understand how co-exposure to other environmental contaminants affects fluorosis and maturation-stage amelogenesis at fluoride levels sufficient to induce defects during early-life exposure conditions in experimental models. This study aimed to determine whethe

Changes in the tartrate-resistant acid phosphatase cell population in dental follicles and bony crypts of rat molars during tooth eruption.

It was the aim of this study to determine the cellular changes that occur in the enamel organ, dental follicle, and surrounding bony crypt of the rat molar prior to and during tooth eruption. By use of light microscope histochemistry to detect cells containing tartrate-resistant acid phosphatase (TRAP), it was seen that TRAP-positive mononuclear cells were present in the dental follicle prior to the onset of eruption (e.g., three days postnatal age) and then declined in number during eruption. Concurrently, TRAP-positive osteoclasts were initially present in large numbers on the surface of the

A scanning electron microscopic study on the differences between newly erupted teeth and old teeth with reference to fissure enamel surfaces and contents.

Scanning electron microscopy of the fissure enamel surfaces of newly erupted permanent teeth and old sound teeth long-standing against caries demonstrated two distinct features; prismatic enamel and prismless enamel. Prismatic enamel was commonly found on the walls of fissures of newly erupted teeth and rarely found on both sides of fissures of old teeth. Prismless enamel was usually found on both sides of fissures of old teeth. Various types of calcified contents were almost confined on the prismless enamel surfaces of the occlusal orifice of fissures of old teeth, while organic materials and

Light and electron microscopical immunohistochemical localization of large proteoglycans in human tooth germs at the bell stage.

The immunohistochemical localization of large hyaluronate-binding proteoglycans has been studied in human tooth germs at the bell stage using a monoclonal antibody, 5D5, which is derived from bovine sclera and specifically recognizes the core protein of large proteoglycans, such as versican, neurocan and brevican, but not that of aggrecan. In the early bell stage before predentine secretion, when the enamel organs consisted of the inner and outer enamel epithelia, stratum intermedium and stellate reticulum, the enamel organs were not stained by 5D5, but the dental papillae and follicles staine

Role of reduced enamel epithelium in root resorption.

BACKGROUND: The precise mechanism of root resorption in human primary teeth is unclear. However, research has suggested that the enamel organ and dental follicle of the successive (permanent) teeth are important for this process. This review focuses on the mechanism of root resorption. HIGHLIGHT: Impaction of the maxillary permanent canines is occasionally seen. Computer tomography studies have shown that these canines cause root resorption in 12.5% of the neighboring central and lateral incisors. In such cases, enlargement of the radiolucent areas around the canine crowns is frequently seen.

Morphological, Histochemical, and Proteomic Analysis of the Effects of Fluoride and Amoxicillin, with Calcium and Vitamin D Supplementation, on Dental Enamel Formation.

Ameloblasts are specialized epithelial cells responsible for enamel formation, a process that requires tightly coordinated matrix secretion, proteolytic turnover, and hydroxyapatite crystal growth. Although genetically regulated, amelogenesis is susceptible to environmental modulation by agents such as fluoride and amoxicillin. Calcium and vitamin D supplementation have been proposed as potential protective factors against fluoride-induced enamel alterations. This study investigated whether combined exposure to fluoride and amoxicillin impairs ameloblast viability and enamel formation, and whe

Alteration of enamel proteins in hypomaturation amelogenesis imperfecta.

Amelogenesis imperfecta (AI) is a diverse group of disorders that affects primarily the enamel of teeth through a number of developmental processes. The purpose of this study was to characterize the enamel proteins in normal enamel and in hypomaturation AI enamel. Impacted teeth, which were at similar stages of development, were obtained for analysis from an individual with AI and from normal healthy controls. Evaluation of the amino acid profile and quantity of organic material collected showed that there was an excess of enamel protein material that had an amelogenin-like amino acid profile

Laminin γ2 knockout mice rescued with the human protein exhibit enamel maturation defects.

The epithelial ameloblasts are separated from the maturing enamel by an atypical basement membrane (BM) that is enriched in laminin 332 (LM-332). This heterotrimeric protein (α3, ß3 and γ2 chains) provides structural integrity to BMs and influences various epithelial cell processes including cell adhesion and differentiation. Mouse models that lack expression of individual LM-332 chains die shortly after birth. The lethal phenotype of laminin γ2 knockout mice can be rescued by human laminin γ2 (LAMC2) expressed using a doxycycline-inducible (Tet-on) cytokeratin 14 promoter-rtTA. These otherwis

Distinctive Amelogenesis Imperfecta in Loeys-Dietz Syndrome Type II.

Loeys-Dietz syndrome (LDS1-6) is caused by mutations along the transforming growth factor-β (TGF-β) signaling pathway and features aortic aneurysms and craniofacial dysmorphology. Mutations that cause LDS can be found in the genes encoding transforming growth factor-β (TGF-β) ligands (TGFB2 and TGFB3), receptors (TGFBR1 and TGFBR2), and signal transducers (SMAD2 and SMAD3). Variable enamel anomalies are seen in patients with LDS, but the most severe enamel defects with advanced attrition have been observed specifically in some patients with mutation in the TGFBR2 gene (LDS2). We used human spe

Immunodetection of enamel- and cementum-related (bone) proteins at the enamel-free area and cervical portion of the tooth in rat molars.

Enamel and dentin at the cervical portion of the tooth are frequently covered by a collagen-free matrix referred to as acellular afibrillar cementum (AAC). It is believed that AAC deposition occurs when the enamel organ is displaced or disrupted, and mesenchymal cells from the dental follicle gain access to the tooth surface, differentiate into cementoblasts, and secrete noncollagenous proteins typically found in collagen-based mineralized tissues. A similar thin layer of mineralized matrix is found at the enamel-free area (EFA) of rodent molars, but in this case the matrix is covered by inner

The glycogen metabolism via Akt signaling is important for the secretion of enamel matrix in tooth development.

Cells alter their energy metabolism depending on the stage of differentiation or various environments. In the ameloblast differentiation of continuous growing mouse incisors, we found temporary glycogen storage in preameloblasts before the start of enamel matrix secretion and investigated the relationship between enamel matrix secretion and glycogen metabolism. Immunohistochemistry showed that in the transitional stage from preameloblasts to secretory ameloblasts, the glycogen synthase changed from the inactive form to the active form, the expression of glycogen phosphorylase increased, and fu

Investigation of the effects of 30% hydrogen peroxide on human tooth enamel by Raman scattering and laser-induced fluorescence.

The safety of tooth bleaching, which is based upon hydrogen peroxide (HP) as the active agent, has been questioned. Our aim was to investigate the effects of 30% HP on human tooth enamel. The specimens were divided randomly into three groups and treated with distilled water, HCl, and HP, respectively. Raman scattering and laser-induced fluorescence of enamel were determined before and after treatment. Microhardness testing and scanning electron microscopy were also used. The results of Raman scattering showed that the Raman relative intensity of enamel changed significantly after HP and HCl tr

The secretory stage of amelogenesis in rat mandibular incisor teeth observed by scanning electron microscopy.

In the scanning electron microscope (SEM), the structural organization of rat incisor enamel was divided into five regions: the initial prismless layer at the dentin-enamel junction; the decussating rods of inner enamel; intermediate enamel, where the structure changes from inner to outer enamel; outer enamel with parallel rods; and the surface prismless layer. There were alterations in the orientation of the ameloblasts and characteristic changes in the morphology of the Tomes' processes during the secretion of each of these regions. Initially, the developing Tomes' processes were seen within

Immunohistochemical similarities and differences between amelogenin and tuftelin gene products during tooth development.

Amelogenins and tuftelins are highly specialized proteins secreted into the developing enamel matrix during mammalian enamel formation. Both tuftelins and amelogenins have been associated with various functions during nucleation and maturation of the developing enamel matrix. In this study we conducted experiments to investigate whether tuftelins and portions of the amelogenin molecule were deposited and processed in spatially distinguished portions of the developing enamel matrix, using antibodies specific against tuftelin or amelogenins. The amelogenin antibodies were raised against recombin

Enamel regeneration - current progress and challenges.

Dental Enamel is the outermost covering of teeth. It is hardest mineralized tissue present in the human body. Enamel faces the challenge of maintaining its integrity in a constant demineralization and remineralization within the oral environment and it is vulnerable to wear, damage, and decay. It cannot regenerate itself, because it is formed by a layer of cells that are lost after the tooth eruption. Conventional treatment relies on synthetic materials to restore lost enamel that cannot mimic natural enamel. With advances in material science and understanding of basic principles of organic ma
