A cardioviral 2C-ATP complex structure reveals the essential role of a conserved arginine in regulation of cardioviral 2C activity.

UNLABELLED: 2C is a highly conserved picornaviral non-structural protein with ATPase activity and plays a multifunctional role in the viral life cycle as a promising target for anti-picornavirus drug development. While the structure-function of enteroviral 2Cs have been well studied, cardioviral 2Cs remain largely uncharacterized. Here, an endogenous ATP molecule was identified in the crystal structure of 2C from encephalomyocarditis virus (EMCV, Cardiovirus A). The ATP is bound into the ATPase active site with a unique compact conformation. Notably, the γ-phosphate of ATP directly interacts w

Discovery of broad-spectrum antiviral activity in Amaryllidaceae alkaloids.

RNA virus infections pose a serious threat to public health, underscoring the need for broad-spectrum antiviral agents. In this study, we screened 383 plant-derived natural products with distinctive and structurally unique frameworks. We identified three Amaryllidaceae alkaloids, haemanthamine, crinamine, and 6-hydroxycrinamine, that exhibited inhibitory activity against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in Vero E6/TMPRSS2 cells. Notably, these compounds concentration-dependently inhibited the replication of not only SARS-CoV-2 but also encephalomyocarditis virus, Si

[Viral etiology of insulin-dependent diabetes (author's transl)].

Data are collected here that may support the hypothesis that there is an infectious component in the genesis of insulin-dependent diabetes mellitus. The clinical and experimental data on which that hypothesis is based referring to viruses such as: mumps virus, rubella virus, encephalomyocarditis virus, and Coxsackie B viruses, are presented. The authors mention the pathogenic mechanism, which extends over the existence of diabetogenic genes linked to the major complex of the histocompatibility HLA system, as well as the existence of an autoimmune reaction induced by the infecting virus. The interferon-inducible 204 gene, a member of the Ifi 200 family, is not involved in the antiviral state induction by IFN-alpha, but is required by the mouse cytomegalovirus for its replication.

To examine whether Ifi 200 genes are involved in antiviral state induction by IFNs we expressed mutant forms capable of inactivating the endogenous p204 and analyzed replication of both RNA and DNA viruses following IFN-alpha treatment. Inactivation of p204 does not impair replication of vesicular stomatitis virus, encephalomyocarditis virus, ectromelia virus, and herpes simplex virus 1 and does not alter an IFN-alpha induced antiviral state. By contrast, in cells lacking functional p204, mouse cytomegalovirus (MCMV) replication is strongly inhibited and is not further modulated by IFN-alpha.

[Seroepidemiological studies of zoonotic infections in hunters in southeastern Austria--prevalences, risk factors, and preventive methods].

The aim of this study was to investigate the seroprevalences to zoonotic pathogens in hunters, to propose preventive measures and to obtain more information about the occurrence of zoonotic pathogens in local wild animal populations. From 146 male and 3 female hunters originating from the south-eastern Austrian federal states of Styria and Burgenland blood samples were taken and anamnestic data were obtained using a questionnaire. The serological investigations included the following viral, bacterial and parasitic zoonotic agents or zoonoses, respectively (antibody-seroprevalences in brackets)

Induction of 2',5'-oligoadenylate synthetase by retinoic acid in two transformed human cell lines.

2',5'-Oligoadenylate (2-5A) synthetase, which polymerizes adenosine triphosphate into 2-5A, is induced upon treatment of cells with interferon (IFN) and is thought to be involved in its antiviral and anticellular action. We report here that retinoic acid (RA) enhanced the level of this enzyme in two human transformed cell lines, WISH and Namalva. Like IFN, RA induced 2-5A synthetase activity in a time- and dose-dependent manner. Addition of anti IFN-alpha, -IFN-beta, or -IFN-gamma antibodies to the medium concomitantly with RA did not prevent such induction; therefore, the effect of RA is clea

Human cell extract-derived cell-free systems for virus synthesis.

Cell-free synthesis of an infectious virus is an ideal tool for elucidating the mechanism of viral replication and for development of antiviral drugs. In this chapter, the synthesis of Encephalomyocarditis virus (EMCV) from RNA and DNA in a HeLa cell extract-derived in vitro protein expression system is described. When a synthetic EMCV RNA with a hammerhead ribozyme sequence at its 5'-end is incubated with a HeLa cell extract using a dialysis system, EMCV particles are progressively synthesized. For EMCV synthesis from DNA, a plasmid harboring the full-length cDNA of EMCV with the T7 promoter/

Passive transfer of virus induced diabetes mellitus with spleen cells.

The study describes passive transfer of diabetes mellitus by transplantation of spleen cells from donor mice infected with a diabetogenic encephalomyocarditis virus. Inbred normal C57 mice were used as both donors and recipients, and other recipients were C57 athymic, nude mice. Transplants were made in two series after 12 and 22 days duration of infection of the donors, respectively. All recipients became diabetic. The possibility of virus transfer with the spleen cell transplant is discussed, and found to be highly improbable. The fact that passive cellular transfer can be effected points to

[An enzyme hydrolyzing the covalent bond between RNA and VPg of picornaviruses. Substrates and methods of analysis of activity].

Some new substrates--RNA-VPg, RNA-peptide and a small fragment of RNA peptide--labelled at the peptide component with [125I]Bolton-Hunter reagent have been proposed for use in the isolation and characterization of the enzyme hydrolyzing the phosphodiester bond between the VPg protein and encephalomyocarditis virus RNA. A novel procedure for the analysis of the specific enzyme activity is based on thin-layer chromatography of hydrolytic products on silicagel or polyethylenimine cellulose. The molecular mass of the enzyme isolated by a modified procedure involving FPLC is 90-95 kD.

Comparative effects of an inflammatory reaction on the resistance of mice to bacterial and viral infections.

The induction in mice of a sterile subcutaneous granuloma exerted no influence upon the mortality following their infection with herpes type 1, murine hepatitis or encephalomyocarditis viruses. Attempts to reproduce the resistance -- which has been found to occur as a result of the granulomatous reaction, in the case of bacterial, fungal or protozoa infections and tumour invasions -- by varying the route and timing of the virus inoculation or the strain of mice have failed. We conclude that it is not merely through their inflammatory properties that some non-specific immunostimulating substanc

Structural insights into the recruitment of viral type 2 IRES to ribosomal preinitiation complex for protein synthesis.

Picornaviruses employ internal ribosome entry sites (IRESs) in their genomic RNA to hijack the host's translational machinery. The picornavirus, encephalomyocarditis virus, employs a type 2 IRES present in its 5' untranslated region (5'UTR) and requires 43S ribosomal preinitiation complex (PIC), the central domain of eukaryotic initiation factor (eIF) 4G, eIF4A, and an essential ITAF (IRES trans-acting factor)-polypyrimidine tract binding protein 1 (PTB1) to form 48S PIC. In this study, we have used cryo-electron microscopy (cryo-EM) to determine the structure of encephalomyocarditis virus (EM

L-tryptophan-kynurenine pathway metabolites regulate type I IFNs of acute viral myocarditis in mice.

The activity of IDO that catalyzes the degradation of tryptophan (Trp) into kynurenine (Kyn) increases after diseases caused by different infectious agents. Previously, we demonstrated that IDO has an important immunomodulatory function in immune-related diseases. However, the pathophysiological role of IDO following acute viral infection is not fully understood. To investigate the role of IDO in the l-Trp-Kyn pathway during acute viral myocarditis, mice were infected with encephalomyocarditis virus, which induces acute myocarditis. We used IDO-deficient (IDO(-/-)) mice and mice treated with 1

The degree of apoptosis as an immunostimulant for a DNA vaccine against HIV-1 infection.

To regulate the expression of the apoptotic gene, we constructed bicistronic DNA vaccines that encode for HIV env and caspase-3 mutant (casp 3m) that are expressed via the encephalomyocarditis virus internal ribosomal entry site (IRES) or cytomegalovirus (CMV) promoter-dependent translations. While IRES-casp 3m induced weak apoptosis and caused little reduction in antigen expression, CMV-casp 3m elicited strong apoptosis and led to a marked decrease in the antigen expression. Therefore, IRES-casp 3m augmented HIV-specific immune responses, and IRES-casp 3m induced significant protection agains

Interleukin-8 selectively enhances cytopathic effect (CPE) induced by positive-strand RNA viruses in the human WISH cell line.

Interleukin-8 (IL-8), a proinflammatory chemokine, is induced by viruses and appears in circulation during viral infections. We found that IL-8 enhanced cytopathic effect induced by the positive strand RNA virus, encephalomyocarditis virus (EMCV), in the human WISH cell line. The enhancement was dependent on IL-8 dose and virus dose and was reversible by specific monoclonal antibodies to IL-8. The chemokine was also able to increase EMC viral RNA synthesis and infectious virus yield. This IL-8 enhancing action was not observed in the case of the negative strand RNA virus, vesicular stomatitis

Testicular atrophy in Columbian black-tailed deer in California.

During an 18-year period, 4.1% (34/831) of male deer (Odocoileus hemionus columbianus) killed on a field station during the autumn hunting season had velvet-covered, often misshapen antlers, and at least two deer had testicular atrophy (gonads from most deer were not available for examination). Testes from six similarly affected deer and several normal deer were compared histologically. Lesions ranged from hypocellularity of the semeniferous tubules and relative hyperplasia or degeneration of interstitial cells to complete connective tissue replacement of the testicular parencyma. Chronic vasc

IRES-based Venezuelan equine encephalitis vaccine candidate elicits protective immunity in mice.

Venezuelan equine encephalitis virus (VEEV) is an arbovirus that causes periodic outbreaks that impact equine and human populations in the Americas. One of the VEEV subtypes located in Mexico and Central America (IE) has recently been recognized as an important cause of equine disease and death, and human exposure also appears to be widespread. Here, we describe the use of an Internal Ribosome Entry Site (IRES) from encephalomyocarditis virus to stably attenuate VEEV, creating a vaccine candidate independent of unstable point mutations. Mice infected with this virus produced antibodies and wer

Reduced Tyk2 gene expression in β-cells due to natural mutation determines susceptibility to virus-induced diabetes.

Accumulating evidence suggests that viruses play an important role in the development of diabetes. Although the diabetogenic encephalomyocarditis strain D virus induces diabetes in restricted lines of inbred mice, the susceptibility genes to virus-induced diabetes have not been identified. We report here that novel Tyrosine kinase 2 (Tyk2) gene mutations are present in virus-induced diabetes-sensitive SJL and SWR mice. Mice carrying the mutant Tyk2 gene on the virus-resistant C57BL/6 background are highly sensitive to virus-induced diabetes. Tyk2 gene expression is strongly reduced in Tyk2-mut

Overlapping and distinct molecular determinants dictating the antiviral activities of TRIM56 against flaviviruses and coronavirus.

UNLABELLED: The tripartite motif-containing (TRIM) proteins have emerged as a new class of host antiviral restriction factors, with several demonstrating roles in regulating innate antiviral responses. Of >70 known TRIMs, TRIM56 inhibits replication of bovine viral diarrhea virus, a ruminant pestivirus of the family Flaviviridae, but has no appreciable effect on vesicular stomatitis virus (VSV), a rhabdovirus. Yet the antiviral spectrum of TRIM56 remains undefined. In particular, how TRIM56 impacts human-pathogenic viruses is unknown. Also unclear are the molecular determinants governing the a
