The use of neurophysiological endophenotypes to understand the genetic basis of schizophrenia.

Specifying the complex genetic architecture of the "fuzzy" clinical phenotype of schizophrenia is an imposing problem. Utilizing metabolic, neurocognitive, and neurophysiological "intermediate" endophenotypic measures offers significant advantages from a statistical genetics standpoint. Endophenotypic measures are amenable to quantitative genetic analyses, conferring upon them a major methodological advantage compared with largely qualitative diagnoses using the Diagnostic and Statistical Manual of Mental Health, 4th Edition (DSM-IV). Endophenotypic deficits occur across the schizophrenia spec

[Endophenotype--a new concept for biological characterization of psychiatric disorders].

In the field of psychiatric genetics, the concept of endophenotypes is presently receiving high popularity. Current difficulties in the search for susceptibility genes of complex diseases have been attributed to the etiological heterogeneity of the clinical, psychopathologically defined phenotype of the disease. Neurobiological correlates of the disease which are genetically influenced and stable over time are considered to be more promising targets of phenotypes. They are more directly influenced by gene effects and presumably defined by a genetic determination which is less complex than the

Idiopathic male infertility revisited: can redox endophenotypes reframe the 'idiopathic' label?

Male infertility accounts for nearly half of all infertility cases worldwide, yet up to 30-40% of affected men remain categorized as 'idiopathic', reflecting limitations of conventional diagnostics such as semen analysis, hormonal profiling, karyotyping, and Y-chromosome microdeletion testing. These methods describe sperm quantity and morphology but fail to uncover underlying molecular dysfunctions. Increasing evidence suggests that oxidative stress plays a central role in driving sperm damage, encompassing lipid peroxidation, protein oxidation, mitochondrial dysfunction, DNA fragmentation, an

Immune and metabolic signatures characterise constipation-driven endophenotypes in Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder defined by motor impairments. However, people with PD (PwPD) experience a defined spectrum of non-motor symptoms, with gastrointestinal dysfunction the most common and earliest-presenting. Evidence suggests that PD pathology may originate in the gut, where microbial dysbiosis and immune dysregulation contribute to neuroinflammation, although mechanisms underlying this are unclear. PwPD (n = 31) and healthy controls (n = 28) were evaluated for clinical and gastrointestinal symptoms, faecal and plasma sample metabolomics, and c

Novel ethological endophenotypes in a transgenic mouse model of Huntington's disease.

Huntington's disease (HD) is an autosomal dominant, neurodegenerative disorder with a characteristic triad of cognitive, affective and motor symptoms. Transgenic HD mice show excellent construct and face validity for many of these symptoms, however the decline in some facets of every day activity in humans is difficult to model. One approach is the assessment of species-relevant behaviors. Here we described three ethologically appropriate tests in the mouse-olfactory sensitivity, nest-building and sexually-motivated vocalizations. In R6/1 HD mice, olfactory and nest-building tests were sensiti

Association between AVPR1A, DRD2, and ASPM and endophenotypes of communication disorders.

OBJECTIVES: Speech sound disorder (SSD) is one of the most common communication disorders, with a prevalence rate of 16% at 3 years of age, and an estimated 3.8% of children still presenting speech difficulties at 6 years of age. Several studies have identified promising associations between communication disorders and genes in brain and neuronal pathways; however, there have been few studies focusing on SSD and its associated endophenotypes. On the basis of the hypothesis that neuronal genes may influence endophenotypes common to communication disorders, we focused on three genes related to b

The many-to-many problem of endophenotypes in psychiatry - a biological perspective.

While modern diagnostic classification systems aim to nosologically structure psychiatric disorders, they poorly align with the genetic, neurobiological, and environmental heterogeneity observed in these disorders. This limitation has complicated the search for clinically useful biomarkers for diagnosis and treatment. Recent work on genetic and environmental contributions to mental health indicates that this heterogeneity stems from differential involvement of diverse biological pathways within and across diagnostic clusters. This complex interplay presents a many-to-many mapping problem in ps

Thalamic Reticular Dysfunction as a Circuit Endophenotype in Neurodevelopmental Disorders.

Diagnoses of behavioral disorders such as autism spectrum disorder and schizophrenia are based on symptomatic descriptions that have been difficult to connect to mechanism. Although psychiatric genetics provide insight into the genetic underpinning of such disorders, with a majority of cases explained by polygenic factors, it remains difficult to design rational treatments. In this review, we highlight the value of understanding neural circuit function both as an intermediate level of explanatory description that links gene to behavior and as a pathway for developing rational diagnostics and t

Whole-brain functional hypoconnectivity as an endophenotype of autism in adolescents.

Endophenotypes are heritable and quantifiable markers that may assist in the identification of the complex genetic underpinnings of psychiatric conditions. Here we examined global hypoconnectivity as an endophenotype of autism spectrum conditions (ASCs). We studied well-matched groups of adolescent males with autism, genetically-related siblings of individuals with autism, and typically-developing control participants. We parcellated the brain into 258 regions and used complex-network analysis to detect a robust hypoconnectivity endophenotype in our participant group. We observed that whole-br

Alzheimer's disease: genetic basis and amyloid imaging as endophenotype.

To date, all known Alzheimer's disease genes influence amyloid β (Aβ). Imaging of Aβ deposition in the human brain using positron emission tomography (PET) tracers as [11C]Pittsburgh Compound B ([(11)C]PiB) or [18F]FDDNP offers the possibility of using cortical tracer binding as a quantitative endophenotype for genetic studies of late-onset Alzheimer's disease (AD). In this review we investigate the association between cerebral Aβ burden, as measured by amyloid PET imaging, and different genetic risk factors involved in AD. Through a look at the major genetic risk factors for both early-onset f

EndoPRS-SS: Summary-statistic based incorporation of endophenotypes to improve risk prediction.

Endophenotypes are critical biological markers with genetic links to disease progression. We previously developed endoPRS, a multi-trait polygenic risk score (PRS) method that leverages endophenotype data to improve disease risk prediction. However, endoPRS requires individual-level genotype-phenotype data for model fitting, restricting its broader use. To address this, we introduce endoPRS-SS, an extension which enables PRS inference using genome-wide association study (GWAS) summary statistics and LD reference panels. Through simulation and real data analysis, we demonstrate that endoPRS-SS

Associations of dementia polyexposure scores to Alzheimer's disease endophenotypes in diverse populations.

INTRODUCTION: Dementia clinical risk scores (CRS) provide accessible tools for identifying individuals at risk for Alzheimer's disease (AD), yet their performance across diverse populations and relationships to AD endophenotypes remains unclear. METHODS: We evaluated four CRS, mCAIDE, WHICAP, LIBRA, and CogDRisk, in relation to cognitive impairment diagnoses and endophenotypes for AD, including plasma biomarkers, neuroimaging measures, and cognitive composite scores. Logistic and linear regression models stratified by self-reported race/ethnicity were used to assess the associations of CRS wi

Precision Pharmacotherapy for Obstructive Sleep Apnea: Matching Targeted Drugs to Patient Endophenotypes.

Background: Obstructive sleep apnea (OSA) remains under-treated because continuous positive airway pressure, though efficacious, is frequently limited by poor tolerance and adherence. Long considered a purely anatomical disease refractory to drugs, OSA is now understood as the shared endpoint of four measurable endotypes-a collapsible upper airway, poor pharyngeal dilator-muscle responsiveness, high loop gain, and a low arousal threshold-each a distinct pharmacological target. This review synthesizes the mechanistic rationale and clinical evidence for targeting them. Methods: In this narrative

Unraveling sex differences in Alzheimer's disease and related endophenotypes with brain proteomes.

INTRODUCTION: Sex differences exist in Alzheimer's disease (AD), but the underlying mechanisms remain unclear. METHODS: We examined brain proteomes profiled from the dorsolateral prefrontal cortex of 770 donors (66.2% female). RESULTS: Proteome-wide differential expression analysis in males and females jointly identified many significant proteins for AD dementia (n = 1228), amyloid beta (n = 1183), tangles (n = 1309), and global cognitive trajectory (n = 2325) at a false discovery rate of <0.05. Sex-stratified analyses also identified many proteins associated with AD or its endophenotypes. F

Inflammatory and oxidative endophenotypes in borderline personality disorder: A biomarker cluster analysis.

OBJECTIVES: This study is designed to search for aggrupation of inflammatory/oxidative biomarker alterations in borderline personality disorder (BPD) and their association with phenotypic features. METHODOLOGY: Inflammatory/nitrosative proteins were measures in plasma and peripheral blood mononuclear cells obtained from BPD patients. Patients were assessed on different clinical dimensions of BPD. Oxidative damage was tested by measuring TBARS, nitrites, catalase, GPx and SOD. Protein expression of IκBα, NFκB, iNOS, COX-2, PPARγ, Keap1, NQO1, Nrf2 and α7nAChR was also determined. Western blot

Current directions in biomarkers and endophenotypes for anorexia nervosa: A scoping review.

There are currently no validated biomarkers for anorexia nervosa (AN), though recent literature suggests an increased research interest in this area. Biomarkers are objective, measurable indicators of illness that can be used to assist with diagnosis, risk assessment, and tracking of illness state. Related to biomarkers are endophenotypes, which are quantifiable phenomena that are distinct from symptoms and which link genes to manifest illness. In this scoping review, we sought to provide a summary of recent research conducted in the pursuit of biomarkers and endophenotypes for AN. The finding

Heritability of glaucoma and glaucoma-related endophenotypes: Systematic review and meta-analysis.

We have systematically extracted all available heritability (h2) estimates of glaucoma and related endophenotypes from the literature and summarized the evidence by meta-analysis. Glaucoma endophenotypes were classified into 10 clusters: intraocular pressure, anterior chamber size, central corneal thickness, cup-to-disc ratio, disc size, cup size, corneal hysteresis, retinal nerve fiber layer thickness, cup shape, and peripapillary atrophy. Random-effects meta-analyses were performed for each cluster. For clusters with n ≥ 10 h2 estimates, we also performed subgroup and meta-regression analyse

[Searching for psychosocial endophenotypes in schizophrenia: the innovative role of brain imaging].

Schizophrenia is a disease with heterogeneous features and often a disabling longitudinal outcome. In order to achieve a better understanding of the disease, a detailed characterization and definition of symptomatology, social functioning and cognitive performance of patients is required. Imaging techniques may allow to identify measurable markers of different subgroups of patients, who share common clinical course and, probably, a similar hereditary pathway. The review offers a description of cross-sectional, predictive and longitudinal studies on the relationship between biological, clinical

Modelling facets of mania--new directions related to the notion of endophenotypes.

The lack of appropriate animal models is a major limitation in research of bipolar disorder (BPD): at this time there are very few models for this devastating disease. Whereas limited attempts have been made to develop comprehensive models for BPD, the new notion of endophenotypes encourages us to explore the possibility of developing separate models for separate facets of the disorder. Since more models are available for depression, there is a dire need for models for mania that will be relatively easy and simple to induce and test and will therefore be practical for purposes of screening pos
