To clarify the characteristics of late-arising (delayed) chromosome aberrations after irradiation in human lymphocytes, 30 B-cell lines were established from the peripheral blood from a healthy adult donor, the lymphocytes of which were exposed to alpha-rays or gamma-rays and then used for experiments. Chromosome aberrations were serially observed at several passages by both conventional cytogenetics and fluorescence in situ hybridization analysis using subtelomere probes. These B-cell lines derived from lymphocytes with a history of radiation exposure had higher percentages of delayed chromos

Nuclear morphology and DNA synthesis were analyzed to determine the mechanism through which hydatidiform moles proliferate. Hydatidiform moles are characterized by a great variation in nuclear morphology and size. There are cells with small nuclei of variable size that have chromocenters and Barr bodies which do not undergo DNA synthesis or mitosis, as well as cells in the diploid range that have evenly stained nuclei that display numerous mitoses and a high proportion of interphase nuclei in the process of DNA synthesis. Nuclei in the medium range show classical endomitotic stages. Endomitoti

Anthropogenic pollution of agricultural soils with cadmium (Cd) should receive adequate attention as Cd accumulation in crops endangers human health. When Cd is present in the soil, plants are exposed to it throughout their entire life cycle. As it is a non-essential element, no specific Cd uptake mechanisms are present. Therefore, Cd enters the plant through transporters for essential elements and consequently disturbs plant growth and development. In this review, we will focus on the effects of Cd on the most important events of a plant's life cycle covering seed germination, the vegetative

Polyploidy represents a major driver of eukaryotic genome evolution. In vertebrates, premeiotic endoreplication is considered as an important route to polyploidization, yet the molecular basis governing this process remains elusive. Here, we reported that heterozygous mutation of cdk1 in the loach, Misgurnus anguillicaudatus (Teleostei, Cobitidae), triggered premeiotic endoreplication in spermatogonia, resulting in the production of unreduced diploid sperms. Through integrated proteomic and phosphoproteomic profiling, we uncovered that Cdk1 heterozygosity led to widespread dysregulation of the

BACKGROUND & AIMS: Large liver cell changes (LLCC) are characterized by pleomorphic large nuclei frequently found in liver diseases as chronic viral hepatitis and liver cirrhosis. The origin of this lesion remains cryptic, but the presence of LLCC is correlated with an increased risk of hepatocellular carcinoma. Telomeric repeat binding factor 1 (TRF1) is part of the shelterin complex and is essential for telomere protection. Ablation of TRF1 induces telomere fragility and fusions and chromosomal instability.
METHODS: In this study, we addressed the role of TRF1 in liver regeneration generati

Common wheat is an allohexaploid species, derived through endoreduplication of an interspecific triploid hybrid produced from a cross between cultivated tetraploid wheat and the wild diploid relative Aegilops tauschii. Hybrid incompatibilities, including hybrid necrosis, have been observed in triploid wheat hybrids. A limited number of A. tauschii accessions show hybrid lethality in triploid hybrids crossed with tetraploid wheat as a result of developmental arrest at the early seedling stage, which is termed severe growth abortion (SGA). Despite the potential severity of this condition, the ge

Each neuron has 100-10000 connections (synapses) with other neural cells, therefore genome pathologies affecting a small proportion of brain cells are capable of causing dysfunction of the entire central nervous system (CNS). Recently, genome and chromosome instability has been uncovered in neurodegeneration (Alzheimer's disease, ataxia telangiectasia). Somatic tissue-specific mosaicism was observed in the brain of individuals with neuropsychiatric diseases including schizophrenia, autism, intellectual disability, and epilepsy. The study of genetic processes in neurons allows determination of

The Japanese quail was used as an experimental system to detect the effects of genes that affect chromosome behavior and distribution. From a random-bred population, three inbred generations were produced by full-sib matings in 36 families. The expectation from such a breeding scheme was that embryos bearing aberrations induced by recessive mutant genes would cluster within families and recur in particular lineages. Chromosomal aberrations caused by errors during fertilization, cleavage mitosis, and gametogenesis were scored in 2,037 16- to 18-h embryos from 107 families. Comparisons of the ob

Peripheral T-cell lymphomas (PTCL) are a diverse group of rare non-Hodgkin lymphomas (NHL) that carry a poor prognosis and are in need of effective therapies. Alisertib (MLN8237) an investigational agent that inhibits Aurora A Ser/Thr kinase has shown activity in PTCL patients. Here we demonstrate that aurora A and B are highly expressed in T-cell lymphoma cell lines. In PTCL patient samples aurora A was positive in 3 of 24 samples and co-expressed with aurora B. Aurora B was positive in tumor cells in 22 of 32 samples. Of the subtypes of PTCL, aurora B was over-expressed in PTCL (NOS) [73%],

Regioisomers of pyrene and benzo[a]pyrene quinones were tested for their ability to induce structural and numerical aberrations and spindle disturbance in Chinese hamster epithelial liver (CHEL) cells in culture. All quinones tested were clastogenic. Pyrene-1,8-quinone (P-1,8-Q) and benzo[a]pyrene-3,6-quinone (BP-3,6-Q) induced strikingly high levels of triradials. In addition, dicentrics and ring chromosomes were very common in BP-3,6-Q-treated cultures. Isomers of these compounds, pyrene-1,6-quinone (P-1,6-Q) and benzo[a]pyrene-1,6-quinone (BP-1,6-Q), induced unobtrusive patterns of chromoso

Multiple aspects of organismal physiology influence the number and activity of stem cells and their progeny, including nutritional status. Previous studies demonstrated that Drosophila germline stem cells (GSCs), follicle stem cells (FSCs), and their progeny sense and respond to diet via complex mechanisms involving many systemic and local signals. AMP-activated protein kinase, or AMPK, is a highly conserved regulator of energy homeostasis known to be activated under low cellular energy conditions; however, its role in the ovarian response to diet has not been investigated. Here, we describe n

The E2F family plays a pivotal role in cell cycle control and is conserved among plants and animals, but not in fungi. This provides for the possibility that the E2F family was integrated during the development of higher organisms, but little is known about this. We examined the effect of E2F ectopically expressed in transgenic tobacco (Nicotiana tabacum) plants on growth and development using E2Fa (AtE2F3) and DPa from Arabidopsis. E2Fa-DPa double transgenic lines exhibited altered phenotypes with curled leaves, round shaped petals, and shortened pistils. In mature but not immature leaves of

Endopolyploidy (within-organism polyploidy) is an often-beneficial aspect of development. But polyploids, which have a higher baseline genome content, have been reported to have less endoreduplication than diploid relatives. Why has remained unclear; we explored this in Arabidopsis arenosa. We measured endopolyploidy levels and phenotyped cellular traits, comparing diploids, neo-autotetraploids, and established autotetraploids of A. arenosa. We used a genetic approach to ask whether CYCD3;2, a polyploidy regulator with evidence of selection in tetraploids, might be responsible for endopolyploi

Intracellular plant-microbe interactions rely on host-derived interface membranes: as sites for reciprocal nutrient and signal exchange during symbiosis, or as conduits for asymmetrical nutrient acquisition and effector delivery by pathogens. Sustaining these dynamic structures places substantial metabolic and vesicular trafficking demands on host cells. This review examines how cell cycle reprogramming may help plants meet these demands. During plant-pathogen interactions, biotrophic pathogens can reprogram host cell cycle pathways to establish metabolically favorable niches, whereas plant im

Non-functional parathyroid carcinoma (NFPC) is an exceptionally rare malignancy, with approximately 50 cases reported since the parathyroid glands were first described in the late nineteenth century. Unlike functional parathyroid carcinomas (PC), which typically present with severe hyperparathyroidism (HPT) and are monitored via serum calcium and parathyroid hormone (PTH) levels, NFPC lacks biochemical markers and is monitored solely through imaging. In this study, we present two new cases of primary NFPC and provide a comprehensive review of the literature, including a detailed analysis of im

Nutrition is one of the most important influences on growth and the timing of maturational transitions including mammalian puberty and insect metamorphosis. Childhood obesity is associated with precocious puberty, but the assessment mechanism that links body fat to early maturation is unknown. During development, the intake of nutrients promotes signaling through insulin-like systems that govern the growth of cells and tissues and also regulates the timely production of the steroid hormones that initiate the juvenile-adult transition. We show here that the dietary lipid cholesterol, which is r

Near-triploid human tumors are frequently resistant to radio/chemotherapy through mechanisms that are unclear. We recently reported a tight association of male tumor triploidy with XXY karyotypes based on a meta-analysis of 15 tumor cohorts extracted from the Mitelman database. Here we provide a conceptual framework of the digyny-like origin of this karyotype based on the germline features of malignant tumors and adaptive capacity of digyny, which supports survival in adverse conditions. Studying how the recombinatorial reproduction via diploidy can be executed in primary cancer samples and He

Exposure to particulate matter (PM) is a major world health concern, which may damage various cellular components, including the nuclear genetic material. To assess the impact of PM on nuclear genetic integrity, structural chromosomal aberrations are scored in the metaphase spreads of mouse RAW264.7 macrophage cells. PM is collected from ambient air with a high volume total suspended particles sampler. The collected material is solubilized and filtered to retain the water-soluble, fine portion. The particles are characterized for chemical composition by nuclear magnetic resonance (NMR) spectro

The DNA damage response (DDR) is a critical mechanism to maintain the genome stability of an organism upon exposure to endogenous and exogenous DNA-damaging factors. The DDR system is particularly important for plants as these organisms, owing to their intrinsic immobility, are inevitably exposed to environmental stress factors, some of which induce DNA damage. Arabidopsis thaliana has orthologs of several DDR factors that are present in animals; however, some of the important animal regulators, such as the tumor suppressor p53 and the DDR kinases CHK1 and CHK2, have not been found in plants.
