[Ethanol and the gastrointestinal tract].

The effects of ethanol upon the gastrointestinal tract (mouth, pharynx, esophagus, stomach, duodenum, Oddi's sphincter, small bowel, colon and rectum) were reviewed. Several studies showed that the incidence of cancer in the mouth and pharynx is increased in alcoholics as a consequence of ethanol effects and probably those of other compounds found in liquors. The gastroesophageal reflux disease may be induced by alcohol since it reduces the pressure in the lower and the upper esophageal sphincter, as well as the extent of primary peristalsis. Several studies showed a strong correlation between

Effect of ethanol on thrombopoiesis.

Chronic ethanol abuse causes thrombocytopenia but the underlying mechanism is unknown. To determine the target cells involved, we examined the effects of the drug in vitro on both megakaryocyte progenitor cells (CFU-Meg) and isolated, maturing megakaryocytes. In the presence of ethanol concentrations of 0.05-2.0 g/dl, megakaryocyte colony formation by mouse CFU-Meg in soft agar was normal. At 5 g/dl ethanol, colony formation was reduced by 50%; with 7 g/dl ethanol, no megakaryocyte colonies were formed. Acetaldehyde did not inhibit colony formation unless very high concentrations (100 mg/dl) w

Ethanol-induced inhibition of liver cell function: I. Effect of ethanol on hormone stimulated hepatocyte DNA synthesis and the role of ethanol metabolism.

We have studied a short term effect (96 hr) of ethanol on hormone-stimulated DNA synthesis in a primary rat hepatocyte culture system. Studies were also performed with respect to total RNA and protein synthesis as well as albumin secretion measured by a solid-phase radioimmunoassay. We found that ethanol, when added to cultured hepatocytes, resulted in a substantial reduction in hormone-stimulated hepatocyte DNA synthesis and this effect was concentration dependent and occurred in serum-free medium. Ethanol also had an inhibitory effect on total RNA synthesis but protein synthesis and albumin

Exploring the effect of ethanol-water structuring on the transport properties of ethanol in porous silicas.

The transport properties of liquid mixtures confined within porous media can change significantly from those observed for bulk mixtures due to changes in the liquid structuring within the pore space. Here, pulsed field gradient NMR was used to measure the diffusion coefficient of ethanol in ethanol-water liquid mixtures confined within silicas with pore diameters of 6 nm and 3 nm as a function of composition. For liquids imbibed within the 6 nm pores, the composition dependence of the ethanol diffusion coefficient closely followed that of the bulk liquid mixture and the absolute diffusion coef

Sugar, a powerful substitute for ethanol in ethanol postdependent rats: Relevance for clinical consideration?

Sugar has been shown to be a powerful substitute for drugs in preclinical studies on addiction. However, the link between sugar intake and alcohol use disorder (AUD) is poorly understood. We assessed the influence of sucrose on ethanol drinking in both nondependent (ND) and dependent (D) Long-Evans rats during acute withdrawal using the postdependent state model. Ethanol (10%-40%) and sucrose (1%-4%) solutions were offered in an operant paradigm either independently or concurrently under ratio schedules of reinforcement. We showed that D rats displayed an enhanced motivation for both 10% ethan

Effects of pregnancy and progesterone on the consumption of ethanol by the high ethanol preferring (HEP) rat.

A significant fraction of women continue to drink heavily during pregnancy, which is associated with the fetal alcohol syndrome, alcohol-related birth defects, alcohol-related neurodevelopmental disorder, and spontaneous abortion. The objective of this study was to determine whether the selectively bred genetic drinking Myers High Ethanol Preferring (HEP) rat would continue to drink through pregnancy. Rats from the F7 generation were screened by a 10-day 3-30% (v/v) ethanol concentration 'step up' procedure in order to determine the concentration which resulted in maximal drinking with an etha

Influence of ethanol consumption on immune competence of adult animals exposed to ethanol in utero.

Ethanol consumption results in significant changes in the immune system of experimental animals and humans. Previous work by ourselves and others has established that in utero exposure to ethanol results in alterations in the immune system of the offspring that persist into adult life. The present study was designed to determine if prenatal exposure to ethanol results in increased vulnerability to the immunosuppressive effects of ethanol consumption in adulthood. Male and female Sprague-Dawley offspring were selected in adulthood from prenatal ethanol (E), pair-fed (PF), and ad libitum-fed con

Effects of ethanol on NMDA receptors in brain: possibilities for Mg(2+)-ethanol interactions.

The major excitatory neurotransmitter in the CNS is L-glutamate, and one of the subtypes of L-glutamate receptors, the N-methyl-D-aspartate (NMDA) subtype, has been found to be quite sensitive to inhibition by low concentrations of ethanol (5-50 mM). The NMDA receptor-ion channels are unique in that they exhibit a voltage-dependent blockade by physiological concentrations of Mg2+, a blockade that is relieved as the cell membrane is depolarized. Several lines of evidence also suggest that the activity of this receptor-channel complex may be regulated through a high-affinity Mg2+ site, which is

Differential response to ethanol, pentobarbital and morphine in mice selectively bred for ethanol sensitivity.

Males of two lines of mice, long sleep (LS) and short sleep (SS), that had been selectively bred for their differential sensitivity to ethanol-induced sleep, were examined for their responses to the hypothermic and analgesic effects of ethanol, pentobarbital and morphine, and to the cataleptic effect of morphine. SS mice were found to be less sensitive than the LS animals to ethanol but not pentobarbital-induced analgesia and hypothermia. The SS animals were also less sensitive to morphine-induced hypothermia, but were, by contrast, more sensitive than their LS counterparts to morphine-induced

Ethanol Reactions over FeMoO Using Low O2/Ethanol Molar Ratio: Reaction Network and Kinetics.

The search for sustainable paths for the valorization of ethanol to produce other substances, such as acetaldehyde, is an object of study in heterogeneous catalysis. This research investigates the conversion of ethanol using an iron-molybdenum oxide catalyst with low O2/EtOH molar ratios (0.0 and 0.05) in a Berty internal recycling reactor to clarify the dominant pathways and associated kinetics at an ethanol partial pressure above the flammability limit and to reveal the significant presence of dehydrogenation, dehydration reactions, and hydrogenation of ethylene to ethane. Adolescent ethanol exposure and differential rearing environment affect taste reactivity to ethanol in rats.

The identification and characterization of variables that influence "liking" and enhance vulnerability to repeated alcohol use are vital to understanding and treating alcohol use disorders. In the current study, we explore the influence of rearing environment and experimenter-administered adolescent ethanol on the hedonic value of ethanol, sucrose, and quinine. Male and female rats were reared for 30 days starting at postnatal day (PND) 21 in either an enriched, isolated, or standard condition and received 1.5 g/kg (intraperitoneally [i.p.]) 20% (w/v) ethanol or saline every other day for 12 d

The role of lymphocytic CD5 in the effects of ethanol on mesolimbic circuitry and ethanol consumption behavior.

Cluster of differentiation 5 (CD5) is a negative regulatory molecule that alters lymphocyte activation pathways and is expressed on the surface of T cells, a subset of B cells (B1 cells), and dendritic cells. CD5 expression is sensitive to ethanol (EtOH) exposure, as the number of CD5+ B cells is reduced during chronic EtOH use. Given the apparent sensitivity of these cells to chronic EtOH consumption and the role of some lymphocytes in regulating the development of inhibitory GABA synapses, we assessed the impact of a murine CD5 genetic knockout (CD5KO) on measures related to EtOH reward, sed

Role of ethanol-derived acetaldehyde in operant oral self-administration of ethanol in rats.

RATIONALE: The role of ethanol-derived acetaldehyde has not been examined yet on performance in a model of operant oral self-administration. However, previous studies reported that an acetaldehyde-sequestering agent, D-penicillamine (DP) and an inhibitor of catalase-mediated acetaldehyde production, 3-amino-1,2,4-triazole (3-AT) reduce voluntary ethanol consumption. OBJECTIVES: The aim of our investigation was to evaluate the effects of DP and 3-AT on acquisition and maintenance of oral operant ethanol self-administration. METHODS: Using operant chambers, rats learned to nose poke in order t

Ethanol withdrawal in mice bred to be genetically prone or resistant to ethanol withdrawal seizures.

We are engaged in a selective breeding program developing lines of mice which differ in severity of withdrawal convulsions after ethanol treatment. Withdrawal seizure prone (WSP) mice show greater handling-induced convulsion scores than withdrawal seizure resistant (WSR) mice after 3 days of ethanol intoxication. In the present experiments, we sought to characterize these mice further as a model of genetic susceptibility to ethanol dependence and withdrawal. During withdrawal after chronic treatment with ethanol, WSP mice displayed more severe handling-induced convulsions and tremor than WSR m

Effect of prenatal exposure to ethanol on adult ethanol preference and response to zimelidine in rats.

The effects of prenatal ethanol (ETOH) exposure (2.8-3.5 g/kg per day) on subsequent adult ETOH preference and on ETOH consumption after treatment with zimelidine, a serotonin uptake inhibitor, were investigated. Pregnant rats were exposed to an ETOH/saccharin solution in the first, second, third, all three, or none, of the trimesters of the gestation period. As adults, the offspring were subjected to a 40-day screening procedure to determine ETOH preference (greater than 50% of total daily fluid consumption) over water (experiment 1). Only animals which preferred ETOH were included in experim

Maternal ethanol consumption and fetal development in the rat: a comparison of ethanol exposure techniques.

Comparison of the polydipsia and liquid diet procedures for ethanol exposure were used in female rats, either before and/or during gestation. Autopsy of the fetuses on day 20 of gestation found a significant weight reduction in the polydipsia-exposed animals that could be attributed to ethanol. No such effect was found in the liquid diet-exposed animals. The data are discussed in relation to the human fetal alcohol syndrome.

On the role of ascending dopamine systems in the control of voluntary ethanol intake and ethanol intoxication.

Selective lesions of the ascending dopamine pathways were made by bilateral injection of the neurotoxin 6-OHDA (8 micrograms/4 microliters). Catecholamine fluorescence histochemistry revealed a marked degeneration of the ascending mesostriatal and mesolimbic dopamine systems, while the hypothalamic dopamine and noradrenaline nerve terminals were unaffected. After recovery of feeding and drinking behaviors the voluntary ethanol intake was not different from that of the controls. The time of ethanol-induced narcosis and the extent of ethanol-induced hypothermia were not affected. In contrast, in
