Vitamin B12 Availability and Interaction with Bacterium Mesorhizobium loti Affect Growth and Biochemical Composition of Mixotrophic Alga Euglena Gracilis.

Over 90% of the mixotrophic algae surveyed require vitamin B12 (B12), a micronutrient synthesized exclusively by certain prokaryotes. Bacteria-mediated B12 supply presents an important but poorly understood control on physiology and nutritional quality of freshwater algae. Here, we examined the effects of B12 availability and the B12-synthesizing bacterium, Mesorhizobium loti, on the growth and biochemical composition of the freshwater bloom-forming mixotrophic alga, Euglena gracilis. We also investigated bacteria-alga interactions under highly labile dissolved organic carbon (DOC) supplementa

Engineering Anaerobic Metabolic Branch Points to Control Carbon Allocation Between Wax Ester Synthesis and Succinate Accumulation in Euglena gracilis.

Under anaerobic conditions, Euglena gracilis degrades paramylon and synthesizes wax esters (WEs) while accumulating succinate in the culture supernatant. Succinate can either be converted to succinyl-CoA by succinyl-CoA synthetase (SCS) and enter the propionyl-CoA biosynthetic pathway or accumulate extracellularly. Propionyl-CoA serves as a primer for odd-chain WE synthesis. Prior research revealed that knockout (KO) of the SCS α-subunit (SCSα) nearly eliminated odd-chain WE synthesis while preserving overall WE accumulation. In the present study, the effects of targeted modifications at anaer

Oral Administration of Euglena Gracilis Z Alleviates Constipation and Cardiac Dysfunction in a Mouse Model of Isoproterenol-Induced Heart Failure.

Background: Patients with heart failure (HF) often experience gastrointestinal problems such as constipation, diarrhea, and disturbances to drug absorption. In HF, hypoperfusion and congestion cause structural and functional changes in the gut, which, in turn, lead to impaired cardiac function. Euglena gracilis Z (hereafter "Euglena"), called Midorimushi in Japanese, is a microalga that is used as a food or nutritional supplement. It is unclear whether Euglena is beneficial for bowel habitus and cardiac function in subjects with HF. Methods and Results: We injected C57BL/6 male mice subcutaneo

The coupling effects of some thiol and other sulfur-containing compounds on the circadian rhythm of cell division in photosynthetic mutants of Euglena.

Previous work has demonstrated a persisting, free-running, circadian rhythm of cell division in the P4ZUL photosynthetic mutant of the alga Euglena gracilis Klebs (Strain Z) Pringsheim grown organotrophically in continuous light or darkness at 19 degrees C following prior synchronization by a repetitive LD:10,14 light cycle. A similar circadian rhythmicity has been recently discovered in the W6ZHL heat-bleached and the Y9ZNalL naladixic acid-induced mutants of Euglena grown under comparable conditions. Over extended timespans, however, these mutants appear to gradually lose first their ability

Metabolic division of labor drives synergistic production of biomass, lipid and α-tocopherol in Euglena gracilis-Chlorella pyrenoidosa co-cultures.

Microalgal co-culture is a promising strategy to enhance biomass and high-value metabolite production, yet the underlying metabolic coordination remains to be fully elucidated. This study developed a synergistic co-culture system using Euglena gracilis (EG) and Chlorella pyrenoidosa (CP) to maximize biomass, lipid, and α-tocopherol yields. Under an optimized inoculation ratio of EG:CP = 3:7 in TAP medium, the co-culture achieved a biomass concentration of 1.23 g/L, lipid productivity of 163.7 mg/L/d, and α-tocopherol yield of 1976.28 μg/L, corresponding to 1.18-1.52-fold, 1.58-2.81-fold, and 2

Regulation of cell shape in Euglena gracilis. II. The effects of altered extra- and intracellular Ca2+ concentrations and the effect of calmodulin antagonists.

When cultures of Euglena gracilis Z., normally grown in medium containing 180 microM-Ca2+, are resuspended in Ca2+-free medium cells assume round shapes within 10 min, from which they recover slowly when Ca2+ is returned to the cultures. Cultures grown in 10 microM-Ca2+ do not display the typical circadian rhythm in cell shape even though the photosynthesis and cell division circadian rhythms are unaffected. Elevating intracellular Ca2+ levels by the addition of the Ca2+ ionophore A23187 prevents cells from undergoing the two daily shape changes characteristic of growth-synchronized cultures,

Lipid metabolism of manganese-deficient algae. I. Effect of manganese deficiency on the greening and the lipid composition of Euglena gracilis Z.

The growth of photoautotrophic Euglena gracilis Z is strongly inhibited by manganese deficiency, whereas chlorophyll formation is not appreciably affected. The galactosyldiglyceride content of the manganese-deficient photo-autotrophic Euglena was about 40% lower on the basis of either chlorophyll content or dry weight. When dark-grown cultures of Euglena were grown photoheterotrophically in light sufficient for the greening of the cells, or photosynthesis, manganese deficiency resulted in a reduction of the cellular content of chlorophyll and galactosyldiglycerides to 40% of control values, in

Incorporating Euglena gracilis into the habitual diet modulates immune function and reduces common cold-like symptoms: a randomized controlled trial.

BACKGROUND: Dietary approaches to support immune function have emphasized the importance of dietary fiber, including non-digestible polysaccharides, although intake remains below recommended levels in many populations. In this context, microalgae have attracted attention as sustainable sources of these components. Euglena gracilis, a microalga used in foods, contains paramylon, a β-1,3-glucan with reported immunomodulatory properties; however, its effects on plasmacytoid dendritic cell (pDC) activation in humans within a dietary context remain incompletely understood. METHODS: This randomized

Effects of Paramylon Extracted from Euglena gracilis EOD-1 on Parameters Related to Metabolic Syndrome in Diet-Induced Obese Mice.

Paramylon (PM), a type of β-glucan, functions like dietary fiber, which has been suggested to exert a protective effect against obesity. We evaluated the potential beneficial effects of PM powder on obesity in mice. Male C57BL/6J mice were fed a high-fat diet supplemented with either 2.5 or 5% PM powder, extracted from Euglena gracilis, for 74 days. Growth parameters, abdominal fat content, serum biochemical markers, hepatic lipid accumulation and hepatic mRNA expression were measured. Dietary supplementation with PM resulted in decreased food efficiency ratios and abdominal fat accumulation.

On the Role of Manganese in Photosynthesis: Kinetics of Photoinhibition in Manganese-deficent and 3-(4-Chlorophenyl)-1, 1-dimethylurea-inhibited Euglena gracilis.

Euglena gracilis (Klebs) cultures were grown under conditions where limitation in supply of manganese limited chlorophyll content much more than growth. Although the initial rates of photosynthetic oxygen evolution were not affected by the level of manganese, photoinhibition in high intensity light was markedly influenced. All cultures showed first order kinetics for photoinhibition, with the half-time exponentially related to the Mn concentration in the medium. Treatment with 3-(4-chlorophenyl)-1, 1-dimethylurea (CMU) also increased the rate of photoinhibition. Manganese-deficient cells were

β-1,3-glucans from Euglena Gracilis protects against ulcerative colitis via modulating the gut barrier, T-cell immunity and gut microbiota.

Ulcerative colitis (UC) is characterized by impaired gut barrier, dysregulated immune responses and pronounced gut dysbiosis. Euglena gracilis (EG), rich in β-1,3-glucan (EGP), exhibits immunomodulatory properties, yet its effects on colitis and EGP's role as a core bioactive component are unclear. The aim of this study was to investigate the protective effects of EGP against UC by targeting gut barrier, T-cell immunity and gut microbiota. Results indicated that EG and EGP effectively improved the body weight, colon growth and reduced disease activity index of the DSS-induced mice. Both treatm

Euglena gracilis-derived β-1,3-glucan as a growth substrate for Lactococcus lactis-a snapshot of accompanying changes in transcriptomic and metabolomic profiles.

β-1,3-glucan from Euglena gracilis is an emerging microalgal prebiotic. However, its potential to reprogram the metabolic profile of lactic acid bacteria has not been reported previously. In the present study, the effects of insoluble β-1,3-glucan on growth dynamics, gene expression, and the extracellular metabolite profile of Lactococcus lactis DSM20175 were investigated using transcriptomic and untargeted metabolomic approaches. Real-time growth monitoring combined with flow cytometric cell enumeration showed that β-1,3-glucan supplementation significantly increased both growth rate and fina

Gamma radiation-induced single strand breaks in DNA and their repair in spheroplasts and nuclei of light-grown and dark-grown Euglena cells.

Exposure of light-grown and dark-grown Euglena cells to gamma radiation causes single strand breaks in nuclear DNA as assessed by sedimentation analysis in alkaline sucrose density gradients. The number of radiation-induced single strand breaks in nuclear DNA of light-grown cells is found to be less than that in dark-grown cells. Post-irradiation incubation of both types of cells in 0 . 1 M phosphate buffer, pH 7 . 0 at 25 degrees C for 1 hour results in restitution of the strand breaks in DNA. Light-grown cells (cells with chloroplasts) are able to rejoin all the single strand breaks in DNA p

Prospective Uncontrolled Interventional Study of Itraconazole and β-Glucans (Euglena gracilis) to Assess Safeness and Clinical Effectiveness in Cats with Cutaneous and Mucosal Sporotrichosis.

Brazil is an endemic region for both human and feline sporotrichosis, with Sporothrix brasiliensis being the main etiological agent. Currently, few effective antifungal agents are available for treating this mycosis in cats, and therapeutic studies remain limited. Itraconazole (ITZ) is the first-line drug; however, its effectiveness is variable. To evaluate the use of ITZ combined with β-glucans (Euglena gracilis) in feline sporotrichosis, a prospective, uncontrolled interventional study was conducted in 29 cats. Clinical cure was achieved in 21 animals (72.4%) with a median treatment duration

Neuroprotective Effect of β-1,3-Glucans-Rich Euglena gracilis Against Ischemic Stroke in Middle-Aged Mice Fed With a High-Fat-High-Fructose Diet.

Ischemic stroke leads to significant neurological deficits and remains a leading cause of disability and mortality worldwide. β-1,3-glucans-rich Euglena gracilis (EG), a type of dietary supplement, was reported to regulate metabolic perturbation and the gut-brain axis that have been implicated in aggravation of the disease. The present study aimed to investigate the neuroprotective effects of EG supplementation in an ischemic stroke model in middle-aged mice fed with a high-fat high-fructose (HFHF) diet. The mice were divided into three groups: HFHF, HFHF+EG (HFHF_EG), and HFHF+Sham (HFHF_Sham

Microarray Analysis of Paramylon, Isolated from Euglena Gracilis EOD-1, and Its Effects on Lipid Metabolism in the Ileum and Liver in Diet-Induced Obese Mice.

We previously showed that supplementation of a high fat diet with paramylon (PM) reduces the postprandial glucose rise, serum total and LDL cholesterol levels, and abdominal fat accumulation in mice. The purpose of this study was to explore the underlying mechanism of PM using microarray analysis. Male mice (C57BL/BL strain) were fed an experimental diet (50% fat energy) containing 5% PM isolated from Euglena gracilis EOD-1 for 12 weeks. After confirming that PM had an improving effect on lipid metabolism, we assessed ileal and hepatic mRNA expression using DNA microarray and subsequent analys

Participation of the inositol phospholipid signaling pathway in the increase in cytosolic calcium induced by tributyltin chloride intoxication of chlorophyllous protozoa Euglena gracilis Z and its achlorophyllous mutant SM-ZK.

Exposure of tri-n-butyl tin chloride (TBTCl) as a stressor to Euglena gracilis Z causes rapid alteration of cell morphology followed by deflagellation. The present study was undertaken to reveal the mechanism of the cell response at a molecular level. Chlamydomonas reinhardtii, in this study E. gracilis Z and its achlorophyllous mutant SM-ZK, gave similar results when subject to the same stressor. Indeed, similar results were obtained with both strains. Next, assuming that the morphological alteration caused by TBTCl is mediated by the inositide phosphate-lipid signaling pathway, the effects o
