Isolating RNAs from rat facial motor neurons with laser capture microdissection after facial-facial anastomosis.

Both reinnervated and dormant neurons can be observed in facial nucleus following facial-facial anastomosis. Although they may play different roles in the remodeling mechanism of facial nucleus during facial nerve injury and regeneration, comprehensive gene profiling analysis of these two neuron types has never been performed due to the difficulty in isolating specific neuron populations and extracting sufficient amount of RNAs from the heterogeneous facial nucleus. In this study, we developed a method to isolate the Fluoro-Ruby (FR) retrogradely labeled reinnervated facial motor neurons and t

Up-regulation of vasopressin V(1a) receptor mRNA in rat facial motoneurons following axotomy.

We have reported previously that axotomy induced a marked increase of vasopressin receptor binding in the adult rat facial nucleus, suggesting an increased number of vasopressin receptors. These receptors were pharmacologically undistinguishable from peripheral V(1a) vasopressin receptors. In the present study, we show, using in situ hybridization and reverse transcriptase-polymerase chain reaction (RT-PCR), that axotomy regulates the expression of the vasopressin V(1a) receptor mRNA in the facial nucleus. Results were obtained from adult male rats killed 1 week following crush of the right fa

Recovery of motor neuron excitability after facial nerve impairment in rats.

Multiple studies have demonstrated alterations in excitability in the central nervous system after peripheral nerve injury. However, there are few reports on changes in the central nervous system after peripheral facial nerve injury. Our objective was to determine the excitability changes that occur in the facial nucleus after facial nerve impairment. The excitability changes in the facial nucleus were investigated by assessing two types of compound muscle action potentials (M and F waves) in the orbicularis oculi muscles, evoked by electrical stimulation of the zygomatic branch of the facial

Topography and cytoarchitecture of the motor nuclei in the brainstem of salamanders.

The organization of the motor nuclei of cranial nerves V (including mesencephalic nucleus), VI, VII, IX, and X is described from HRP-stained material (whole mounts and sections) for 25 species representing five families of salamanders, and the general topology of the brainstem is considered. Location and organization of the motor nuclei, cytoarchitecture of each nucleus, and target organs for nuclei and subnuclei are described. The trigeminal nucleus is separated distinctly from the facial and abducens nuclei and consists of two subnuclei. The abducens nucleus consists of two distinct subnucle

Distribution of the galectin-1 mRNA in the rat nervous system: its transient upregulation in rat facial motor neurons after facial nerve axotomy.

Galectin-1 is a member of the animal lectin family that displays conserved consensus sequences and similar carbohydrate binding specificities. Recent analyses revealed that galectin-1 plays an important role in the process of nerve regeneration. We analyzed the topological expression of galectin-1 mRNA in adult rat nervous system. Galectin-1 mRNA was predominantly observed in the cell bodies of neurons such as oculomotor nucleus (III), trochlear nucleus (IV), trigeminal motor nucleus (V), abducens nucleus (VI), facial nucleus (VII), hypoglossal nucleus (XII), red nucleus, and locus ceruleus. [Pontine hemorrhage as a cause of peripheral facial paralysis].

Bell's palsy is the most frequent type of peripheral facial nerve paralysis. Nevertheless there are other less frequent causes of peripheral facial palsy, among which, we can find lesions in the brain stem affecting the seventh cranial nerve nucleus or its fibers. These lesions of the pons which paralyze the muscles of the face are often accompanied by lesions to other structures in the vicinity of the nucleus. We present the case of peripheral facial nerve palsy caused by hemorrhage in the pons, seen both on brain CT-scan and MRI, which affected the nucleus of the seventh cranial nerve and wa

Efficacy of Forward and Reverse Suturing Techniques in Enhancing Neural Regeneration and Motor Function Recovery Following Facial Nerve Axotomy.

Background/Objectives: Facial nerve injury from conditions such as Bell's palsy, trauma, surgery, and infection leads to facial asymmetry and motor deficits. Axotomy models reproduce peripheral nerve disruption and consequent motor impairment. To compare the effects of forward versus reverse autologous nerve suturing on neural regeneration and motor recovery within the facial nucleus after axotomy. Methods: In rats subjected to facial nerve axotomy, motor recovery was assessed at 8 weeks using whisker movement and blink reflex tests. Immunohistochemistry quantified choline acetyltransferase (C

Rat facial motoneurons express increased levels of calcitonin gene-related peptide mRNA in response to axotomy.

The expression and localization of the mRNA encoding the calcitonin gene-related peptide (CGRP) were analyzed in the rat facial nucleus after axotomy by Northern blot analysis and by in situ hybridization histochemistry (ISH) using a synthetic 32P-labeled oligonucleotide probe. Northern blot analysis revealed the presence of the 1.2 kb CGRP mRNA in RNA extracted from the facial nucleus. This mRNA species was strongly increased after axotomy of the facial nerve. By ISH increased levels of CGRP mRNA were observed as soon as 16 hr after axotomy compared with the unoperated nucleus. CGRP mRNA coul

Central projection of proprioceptive afferents arising from maxillo-facial regions in some animals studied by HRP-labeling technique.

20 ICR mice (adult females) were used for analyzing the axonal projection of the trigeminal mesencephalic tract neurons and 10 Japanese shrew-moles for analyzing that of the snout proprioceptive neurons. The HRP-labeled axons were found to be ipsilaterally terminated in the trigeminal motor nucleus, supratrigeminal nucleus and trigeminal main and spinal tract nuclei, lateral pontine-medullary reticular formation, vagal dorsal motor and hypoglossal nuclei and the lamina V of the C2 spinal cord segments. No HRP-labeled axons were found in the facial and solitary nuclei and the cerebellum. Also,

Facial paralysis: a critical review of accepted explanation.

Historically, paralysis of facial muscles has been divided into "upper motor neuron injury" and "lower motor neuron injury". Patients who experience a stroke in the cortex or internal capsule have UMN injury and cannot purse their lips or smile on command. They are, however, able to wrinkle their forehead, raise their eyebrows, and completely close their eyes. Patients with LMN injury, in addition to the aforementioned impairments cannot raise their eyebrows. The classical explanations for these clinical findings are that the upper facial muscles receive bilateral innervation from the cerebral

Mechanisms and predictors of chronic facial pain in lateral medullary infarction.

The purpose of this study was to identify clinical predictors and anatomical structures involved in patients with pain after dorsolateral medullary infarction. Eight out of 12 patients (67%) developed poststroke pain within 12 days to 24 months after infarction. The pain occurred in the ipsilateral face (6 patients) and/or the contralateral limbs and trunk (5 patients, 3 of whom also had facial pain). Ipsilateral facial pain was significantly correlated with lower medullary lesions, including those of the spinal trigeminal tract and/or nucleus, as documented by magnetic resonance imaging. The

[Influence of p75 neurotrophin receptor knockout on the regeneration of facial nerves after crush injury in mouse].

OBJECTIVE: To investigate the role of p75 neurotrophin receptor (p75NTR) in the regeneration of facial nerve crush injury. METHODS: In p75NTR knockout mice and wild type mice, the regenerating fibres in the facial nerve were also labelled by an anterograde tracer cholera toxin B (CTB). The next day after injury of facial nerve, CTB was injected into the trunk of the nerve in the proximal side of the crush, and then anterograde tracing and immunohistochemistry were used to examine the regeneration of axons after facial nerve crush injury. In p75NTR knockout mice and wild type mice, the facial

Facial myokymia in syringobulbia.

A patient with a 2-year history of progressive ataxia and tingling in the right hand had prominent facial myokymia. Magnetic resonance imaging revealed syringomyelia and syringobulbia. After successful syringosubarachnoid shunting, the patient died of massive pulmonary embolism. Postmortem examination revealed a syrinx involving the spinal cord and lower half of the medulla; neither the facial nucleus nor facial nerve fibers were directly involved. We hypothesize that interruption of aberrant corticobulbar fibers in the medulla produced disinhibition of a rhythmic neural generator in the facia

Facial myokymia. Pathological features.

Pathologic examination in a case of facial myokymia showed edema and mild astroglial proliferation of the ipsilateral seventh nerve nucleus; gliomatous tumor involvement occurred rostral to this nucleus. These findings give circumstantial support to functional deafferentation as the cause of facial myokymia.

Transient down-regulation and restoration of glycogen synthase levels in axotomized rat facial motoneurons.

In adult rats, transection of the facial nerve causes a functional down-regulation of motoneurons and glial activation/proliferation. It has not been clear how energy-supplying systems are regulated in an axotomized facial nucleus. Here we investigated the regulation of molecules involved in glycogen degradation/synthesis in axotomized facial nuclei in rats. Immunoblotting revealed that the amounts of glycogen phosphorylase in the contralateral and ipsilateral nuclei were unchanged for the first 14 days, whereas the amount of glycogen synthase in the axotomized facial nuclei was significantly

Non-motoneurons in the facial and motor trigeminal nuclei projecting to the cerebellar flocculus in the cat. A fluorescent double-labelling and WGA-HRP study.

The cerebellar projection from the facial and motor trigeminal nuclei was studied in the cat by means of retrograde axonal transport of wheat germ agglutinin-horseradish peroxidase and fluorescent tracers. The feline facial nucleus was cytoarchitectonically subdivided into ventromedial, ventrolateral, lateral, dorsal, intermediate and medial divisions (see Papez 1927), and the motor trigeminal nucleus into medial, ventral, intermediate, lateral and dorsal divisions. The neurons in the facial and motor trigeminal nuclei were classified as small (ovoid to round cells with a maximum diameter of t

Central distribution of afferent and efferent components of the glossopharyngeal nerve: an HRP study in the cat.

Central distribution of efferent and afferent components of the glossopharyngeal (IX) nerve in the cat was studied by applying horseradish peroxidase (HRP) to the lesser petrosal nerve (LPN), carotid sinus nerve (CSN) or cervical trunk (CT) distal to the branching point of the CSN. After applying HRP to the LPN or CT, HRP-labeled neurons were distributed ipsilaterally in the lateral regions of the reticular formation from the caudalmost levels of the facial nerve genu to the rostral levels of the solitary nucleus. HRP-labeled axons of these neurons ran dorsomedially and then medially along the
