Common immunotoxicity mechanisms of hepatotoxicity induced by raw Polygonum multiflorum and Polygonum multiflorum praeparata: Inhibition of M2 macrophage polarization.

Macrophage polarization has been linked to hepatotoxicity induced by raw Polygonum multiflorum (RPM) and Polygonum multiflorum praeparata (PMP), but the regulatory mechanisms behind this remain unclear. This study aims to investigate the regulatory effects of RPM and PMP on M2 macrophages and the potential mechanisms. Sprague-Dawley rats were exposed to RPM and PMP under lipopolysaccharide (LPS) stimulation. RAW264.7 cells induced with IL-4 were treated with RPM and PMP. Under LPS stimulation, both RPM and PMP increased serum enzyme levels and pro-inflammatory factor levels and induced histopa

Processing-dependent hepatic risk and benefit of Polygonum Multiflorum Radix: Integrated chemical profiling, in vivo validation and candidate quality control markers.

To clarify how steaming and sun-drying cycles affect the hepatic risk-benefit profile of Polygoni Multiflori Radix (PMR), the herb was processed with black bean decoction for 1-9 cycles following the traditional "nine steaming and nine sun-drying" protocol. UPLC-MS analysis showed that anthraquinones and trans-THSG progressively decreased with increasing cycles and stabilized after five cycles, whereas cassialactone-8-O-β-D-glucoside and hyperoside emerged as new components during processing. In a low-dose LPS-sensitized mouse model mimicking immune-idiosyncratic liver injury, PMR processed fo

Evaluating the anticancer potential of Polygonum multiflorum root-derived stilbenes against H2452 malignant pleural mesothelioma cells.

A naturally occurring stilbene, resveratrol, shows promising effects in the treatment of malignant pleural mesothelioma (MPM) both as a single agent and in combination with chemotherapeutic drugs. To discover new anticancer agents targeting MPM, stilbene-targeted isolation was performed on the roots of Polygonum multiflorum Thunb., an herbal medicine rich in stilbene compounds. In this study, seven stilbene glycosides (1-7) were isolated, along with four non-stilbenes (8-11), of which compounds 4 and 9-11 have not previously been isolated from this species. Stiquinoside A (1) is a previously u

Exploratory Quality Control Study for Polygonum multiflorum Thunb. Using Dinuclear Anthraquinones with Potential Hepatotoxicity.

In recent years, the hepatotoxicity of Polygoni Multiflora Radix (PMR) has attracted increased research interest. Some studies suggest that anthraquinone may be the main hepatotoxic component. Most of the relevant studies have focused on the mononuclear anthraquinone component rather than binuclear anthraquinones. The hepatotoxicity of dinuclear anthraquinone (dianthrone) was investigated in a cell-based model. Next, a method for the determination of six free and total dianthonones in PMR and PMR Praeparata (PMRP) was established using ultra-high-performance liquid chromatography triple quadru

Identification of potential biomarkers for Polygonum multiflorum-induced liver injury in a clinical cohort: integrating machine learning of metabolomics with transcriptomic profiling.

ETHNOPHARMACOLOGICAL RELEVANCE: Polygonum multiflorum Thunb. (PM), synonymous with Reynoutria multiflora (Thunb.) Moldenke is known as He Shou Wu in traditional Chinese medicine. Despite its long history of use, PM has been associated with drug-induced liver injury (PM-DILI), a rare but potentially serious adverse event. Therefore, identifying reliable biomarkers is crucial for the early detection of PM-DILI, risk prediction, and the safe and rational clinical use of PM. AIM OF THE STUDY: To identify candidate diagnostic biomarkers for PM-DILI and obtain preliminary mechanistic insights by in

The combination of pro-oxidative acting vanicosides and GLUT1 inhibitor (WZB117) exerts a synergistic cytotoxic effect against melanoma cells.

Vanicosides A and B isolated from Reynoutria sachalinensis rhizomes are disaccharide phenylpropanoid esters with proven antioxidant activity. Our earlier study showed the cytotoxic activity of vanicosides against melanoma cells, but the mechanism of cell death has not been elucidated. Based on the chemical structure of vanicosides, we proposed that they may induce cell death by generating reactive oxygen species (ROS) into melanoma cells. Moreover, the glucose molecule in their structure can affect the glucose transporters (GLUTs), upregulated in cancer cells. The A375 (melanotic) and C32 (ame

Polygonum multiflorum Thunb. Induces hepatotoxicity in SD rats and hepatocyte spheroids by Disrupting the metabolism of bilirubin and bile acid.

ETHNOPHARMACOLOGICAL RELEVANCE: The liver damage associated with Polygonum multiflorum Thunb. (P. multiflorum) and its preparations have aroused widespread concern. Opinions on the toxicity mechanisms and targets of P. multiflorum vary, and the toxic components are even more controversial. However, based on the current research results, we believed that any single component in P. multiflorum could not directly lead to liver injury, but may be the synergistic effect of multiple components. In addition, the toxicity mechanism also involved multiple targets. AIM OF THE STUDY: This study aimed to

Clinical correlation between serum cytokines and the susceptibility to Polygonum multiflorum-induced liver injury and an experimental study.

Polygonum multiflorum (PM), a popular functional food, and a herbal and dietary supplement, is widely used as a tonic in China and East Asia. In recent years, it has attracted great concern for its ability to cause idiosyncratic drug-induced liver injury (IDILI). However, identifying individuals susceptible to IDILI remains challenging. This is a prospective study. For 6 patients whose serum alanine aminotransferase (ALT) levels after consuming PM were abnormally elevated (susceptible group), 15 patients with normal levels of liver injury markers were matched (tolerant group) based on similar

Polygonum cuspidatum Exosome-Like Nanovesicles Alleviate Hypoxic Pulmonary Hypertension by Stabilizing PON1 to Inhibit MAPK-Mediated PASMC Phenotypic Switching.

Hypoxic pulmonary hypertension (HPH) is characterized by excessive proliferation of pulmonary vascular cells, ultimately resulting in right heart failure and death. Although resveratrol, an active component of Polygonum cuspidatum (PC), has shown efficacy against HPH, the mechanism of PC exosome-like nanovesicles (PCELNs) in HPH remains unclear. PC exosome-like nanovesicles (PCELNs) were characterized using transmission electron microscopy, dynamic light scattering, and immunofluorescence (IF). A mouse HPH model was established by chronic hypoxic exposure. Hemodynamic parameters were measured,

Polygonum cuspidatum Extract (Pc-Ex) Containing Emodin Suppresses Lung Cancer-Induced Cachexia by Suppressing TCF4/TWIST1 Complex-Induced PTHrP Expression.

Cachexia, which is characterised by the wasting of fat and skeletal muscles, is the most common risk factor for increased mortality rates among patients with advanced lung cancer. PTHLH (parathyroid hormone-like hormone) is reported to be involved in the pathogenesis of cancer cachexia. However, the molecular mechanisms underlying the regulation of PTHLH expression and the inhibitors of PTHLH have not yet been identified. The PTHLH mRNA levels were measured using quantitative real-time polymerase chain reaction, while the PTHrP (parathyroid hormone-related protein) expression levels were measu
