Famotidine-induced reversal of meperidine-related serotonin syndrome: a case report.

Serotonin syndrome is an unexpected fatal adverse event related to serotonergic medication. This case report is the first report describing the possible treatment effect of famotidine on serotonin syndrome. Furthermore, this is the first case report of serotonin syndrome induced by meperidine alone in a patient with no previous history suggesting a susceptibility to serotonin syndrome. A 70-year-old male with no recent history of serotonergic drug use presented with severe serotonin syndrome following ureteroscopy, possibly due to postoperative meperidine administration. The patient's symptoms

Outpatient intravenous interleukin-2 with famotidine has activity in metastatic melanoma.

UNLABELLED: Daily short intravenous interleukin-2 (IL-2) infusions have been developed to decrease toxicity while maintaining the anticancer activity of this agent against melanoma. Such IL-2 schedules have previously been shown to promote lymphokine-activated killer cell (LAK) activity. Famotidine may increase LAK activity by increasing IL-2 internalization by the IL-2 receptor on lymphocytes. Twenty-one patients with metastatic melanoma were treated with IL-2 18 million IU/m² intravenously (i.v.) over 15-30 minutes and famotidine 20 mg i.v. daily for 3 days for 6 consecutive weeks on an outp

Lansoprazole versus famotidine in symptomatic reflux esophagitis: a randomized, multicenter study.

We compared the usefulness of lansoprazole and famotidine for treatment of reflux esophagitis. Fifty-four subjects with moderate to severe reflux esophagitis were divided into two groups. The lansoprazole group (LPZ) received lansoprazole 30 mg once daily for 4 weeks, followed by famotidine 20 mg twice daily for 4 weeks. The famotidine group (FAM) received the same medications in the reverse order. Patient evaluation was based on six factors found on endoscopy, and on patient reports of heartburn. Evaluations were done before the start of the study and at 4 and 8 weeks. The improvement rates i

High dose famotidine in ranitidine resistant severe oesophagitis: a pilot study.

Fifty-three asymptomatic patients who, in spite of continuous treatment with ranitidine 300 mg bid for six months, had active severe oesophagitis, Savary and Miller grade III-IV or Barretts ulceration at endoscopy were treated with high dose famotidine. In the severe oesophagitis group famotidine 40 mg bid, 13/29 (44%) healed at eight weeks and 17/29 (58%) at sixteen weeks. In the Barrett's ulcer group famotidine 40 mg tid, 10/24 (41%) healed at eight weeks and 18/24 (75%) at sixteen weeks. Famotidine in high doses was well tolerated in this age group (mean 66.75 years), with 5/53 (9%) minor a

Comparison of gastric pH with omeprazole magnesium 20.6 mg (Prilosec OTC) o.m. famotidine 10 mg (Pepcid AC) b.d. and famotidine 20 mg b.d. over 14 days of treatment.

BACKGROUND: The onset of acid inhibition for proton pump inhibitors is slower than with H2RAs and generally considered to be at a steady-state after 5 days. Thus, little direct comparison data exists between H2RAs and proton pump inhibitors for gastric acid suppression on day 1 of therapy. Furthermore, the durability of their acid suppression has not been systematically compared. AIM: To compare the effects of 20.6 mg omeprazole magnesium o.m. (Ome-Mg 20), famotidine 10 mg b.d. (Fam 10) and famotidine 20 mg b.d. (Fam 20) on intragastric pH on day 1 and throughout 14 days of dosing. METHODS:

Valproic Acid and Famotidine Drug-Drug Interaction: Report of a Pediatric Case.

Valproic acid is a broad-spectrum anticonvulsant medication that is used to treat multiple neurologic and psychiatric disorders such as epilepsy, bipolar disorder, schizophrenia, and migraine prophylaxis. Famotidine is an antacid used for various gastrointestinal conditions, including gastric and duodenal ulcers and gastroesophageal acid reflux disease (GERD). Acute ingestion of famotidine was found to inhibit anticonvulsant action and increase brain concentrations relative to free plasma levels in a mouse model. While they are not known to interact with one another, one of their proposed meta

Omeprazole is more effective than famotidine for preventing acute gastritis in rats.

PURPOSE: Acute gastric mucosal lesions, which can develop within a few hours after polytrauma, shock, major operations, central nervous system lesions, or severe infection, cause about 33% of cases of gastrointestinal bleeding. We analyzed and compared the effectiveness of famotidine and omeprazole on acute gastric mucosal lesions. METHODS: Thirty male albino Wistar rats were given ketalar anesthesia after 12 h fasting, then immobilized and exposed to stress according to Brodie's protocol, without restricting their respiration. We divided the rats into three groups of ten according to whether

Famotidine for the prevention of gastric and duodenal ulcers caused by nonsteroidal antiinflammatory drugs.

BACKGROUND: Acid suppression with famotidine, a histamine H2-receptor antagonist, provides protection against gastric injury in normal subjects receiving short courses of aspirin or naproxen. The efficacy of famotidine in preventing peptic ulcers in patients receiving long-term therapy with nonsteroidal antiinflammatory drugs (NSAIDs) is not known. METHODS: We studied the efficacy of two doses of famotidine (20 mg and 40 mg, each given orally twice daily), as compared with placebo, in preventing peptic ulcers in 285 patients without peptic ulcers who were receiving long-term NSAID therapy for

Changes in the effects of nizatidine and famotidine on cardiac performance after pretreatment with ranitidine.

This was an open, randomized study of the cardiovascular effects of the histamine H2 receptor antagonists ranitidine, famotidine, and nizatidine after single oral doses alone or in combination in healthy volunteers. When compared with placebo ranitidine (450 mg) did not have any haemodynamic effects. Nizatidine (300 mg) caused significant falls in heart rate and cardiac output. Famotidine (40 mg) caused significant falls in stroke volume and cardiac output and an increase in pre-ejection period. Pretreatment with ranitidine abolished the haemodynamic effects of nizatidine and caused a time-shi

Effect of H2-blockers, cimetidine and famotidine, on histamine nasal provocative test.

The H2-receptor is considered to mediate the stabilization of the membrane of mast cells by enhancing cAMP synthesis. In the present study, we investigated the effect of H2-receptor antagonists, i.e. H2-blockers, on nasal hypersensitivity using a histamine nasal provocative test. A 7-day administration of cimetidine tended to lower the histamine threshold. Famotidine administered in the same way caused no significant decrease in the histamine threshold. Based on the above results, we postulate that famotidine should be preferred to cimetidine as an H2-blocker in patients with gastric and/or du

Effects of famotidine on upper gastrointestinal motility in patients with progressive systemic sclerosis.

The effects of famotidine on human upper gastrointestinal motility were investigated, together with the relationship of gastric alkalinization and serum gastrin levels to changes produced by famotidine. Intravenous famotidine (20 mg), at a dose level in which an inhibitory effect on acetylcholinesterase activity is not recognized, was given to 13 patients with progressive systemic sclerosis but no other disorders. Gastric phasic motor activity was not changed significantly, but the lower esophageal sphincter pressure was elevated significantly in comparison with 15 controls given physiological
