A mechanistic modeling framework to interpret ACTH stimulation tests across HPA axis adaptation states and glucocorticoid feedback dynamics.

The hypothalamic pituitary adrenal (HPA) axis is a key regulatory system coordinating endocrine responses to physiological and psychological stress. While the ACTH stimulation test remains a cornerstone of adrenal function assessment, its interpretation is complicated by the dynamic and adaptive nature of the HPA axis under chronic stress exposure. In particular, prolonged stress induces glandular remodeling, glucocorticoid receptor (GR) resistance and delayed feedback recovery, all of which may alter test outcomes without indicating primary adrenal failure. In this study, we present a mechani

Negative feedback regulation of thyrotropin subunits and pituitary deiodinases in red drum, Sciaenops ocellatus.

Thyroxine (T4) undergoes dynamic daily cycles in the perciform fish the red drum, Sciaenops ocellatus, that are inversely timed to cycles of thyrotropin (TSH) subunit mRNA expression in the pituitary gland. We have proposed that these daily cycles are regulated by negative feedback of circulating T4 on expression of pituitary thyroid hormone deiodinase type 3 (Dio3), such that elevated circulating T4 results in diminished pituitary thyroid hormone catabolism and consequent increased negative feedback on expression of TSH subunits during the day. To determine whether thyroid hormones function t

Lifetime achievement from a brain-adrenal perspective: on the CRF-urocortin-glucocorticoid balance.

This contribution dedicated to Wylie Vale is focused on the action of the glucocorticoid hormone aimed to counterbalance the stress response orchestrated by the corticotrophin releasing factor (CRF) and urocortin (Ucn) family of peptides. It appears that the release and action of these stress hormones themselves are subjected to intrinsic self-regulatory feedback loops that operate as checks and balances in stress adaptation. One of these feedback loops is operated by the mineralocorticoid (MR) and glucocorticoid receptors (GR) that mediate in complementary fashion the action of endogenous cor

Predicting neuronal response properties from hemodynamic responses in the auditory cortex.

Recent functional MRI (fMRI) studies have highlighted differences in responses to natural sounds along the rostral-caudal axis of the human superior temporal gyrus. However, due to the indirect nature of the fMRI signal, it has been challenging to relate these fMRI observations to actual neuronal response properties. To bridge this gap, we present a forward model of the fMRI responses to natural sounds combining a neuronal model of the auditory cortex with physiological modeling of the hemodynamic BOLD response. Neuronal responses are modeled with a dynamic recurrent firing rate model, reflect

De novo motor learning as controller synthesis: a conceptual framework.

Most studies of motor learning focus on adaptation, which can be described as recalibration within an existing controller following changes in dynamics, kinematics, or sensory feedback. In these paradigms, control variables, coordinate representations, and feedback organization are treated as available a priori, and learning as parameter tuning within a specified controller architecture. However, de novo motor-learning tasks require learners to establish novel relationships between intention, sensory feedback, and action, rather than merely recalibrating a familiar control organization. These

Reward prediction errors shape sensory-error-driven single-trial motor learning.

Short-term motor adaptation is a form of motor learning that optimizes how sensory information about a target's location is translated into a target-directed movement, using sensory feedback on performance errors. When actual and predicted sensory feedback differ, sensory prediction errors (SPEs) are generated, enabling the cerebellum to adjust this mapping. In contrast, reinforcement learning uses reward prediction error (RPEs) based on fulfilled or unfulfilled reward expectations to shape future actions. As RPE-related information also reaches the cerebellum, we wondered whether it interacts

Understanding negative feedback: Changes in high-molecular-weight adrenocorticotropic hormone in adrenocorticotropic hormone-independent Cushing's syndrome.

Cushing's syndrome is characterized by chronic glucocorticoid oversecretion and diverse clinical manifestations. Distinguishing between adrenocorticotropic hormone (ACTH)-independent and ACTH-dependent forms is crucial for determining treatment options. Plasma ACTH levels aid in the differential diagnosis, with undetectable or low levels suggesting ACTH-independent hypercortisolemia. ACTH is derived from pro-opiomelanocortin, and its processing involves prohormone convertase 1/3. High-molecular-weight ACTH is generally found in ACTH-producing pituitary tumors and ectopic ACTH syndrome. The mec

The Role of the Neutrophilic Network in the Pathogenesis of Psoriasis.

One role of neutrophils, the most abundant innate immune sentinels, is neutrophil extracellular trap (NET) formation, which plays a significant role in immune surveillance. However, NET operation is bidirectional. Recent studies report that NETs may contribute to the development of autoimmune diseases such as psoriasis. The participation of neutrophils in the pathogenesis of that disease is dependent on an autoinflammatory feedback loop between neutrophils, lymphocytes, dendritic cells and keratinocytes. Our aim was to clarify the field of NET research in psoriasis and highlight the main facto

A wearable skin stretch haptic feedback device: Towards improving balance control in lower limb amputees.

Haptic feedback to lower limb amputees is essential to maximize the functionality of a prosthetic device by providing information to the user about the interaction with the environment and the position of the prostheses in space. Severed sensory pathway and the absence of connection between the prosthesis and the Central Nervous System (CNS) after lower limb amputation reduces balance control, increases visual dependency and increases risk of falls among amputees. This work describes the design of a wearable haptic feedback device for lower limb amputees using lateral skin-stretch modality int

In-vitro evaluation of physiological controller response of rotary blood pumps to changes in patient state.

Rotary blood pumps (RBPs) have a low sensitivity to preload changes when run at constant speed, which can lead to harmful ventricular suction events. Therefore a control mechanism is needed to adjust RBP speed in response to patient demand, but an appropriate response time for physiological control strategies to these changes in patient demand has not been determined. This paper aims to evaluate the response of a simulated healthy heart with those of different RBP control techniques during exercise simulations and a Valsalva manoeuver. A mock circulation loop was used to simulate the response

Regulation of copper transport in Arabidopsis thaliana: a biochemical oscillator?

Plants are among the most versatile higher eukaryotes in accommodating environmental copper availability to largely variable demands. In particular, copper deficiency in soils is a threat for plant survival since it mostly affects reproductive structures. One of the strategies that plant cells use to overcome this situation is to increase copper levels by expressing high-affinity copper transporters delivering the metal to the cytosol. In this minireview, we discuss recent advances in the structure, function, and regulation of the CTR/COPT family of copper transporters, and pay special attenti

Effect of sensory-motor latencies and active muscular stiffness on stability for an ankle-hip model of balance on a balance board.

To achieve human upright posture (UP) and avoid falls, the central nervous system processes visual, vestibular, and proprioceptive information to activate the appropriate muscles to accelerate or decelerate the body's center of mass. In this process, sensory-motor (SM) latencies and muscular deficits, even in healthy older adults, may cause falls. This condition is worse for people with chronic neuromuscular deficits (stroke survivors, patients with multiple sclerosis or Parkinson's disease). One therapeutic approach is to recover or improve quiet UP by utilizing a balance board (BB) (a rotati

Feedback control indirect response models.

A general framework is introduced for modeling pharmacodynamic processes that are subject to autoregulation, which combines the indirect response (IDR) model approach with methods from classical feedback control of engineered systems. The canonical IDR models are modified to incorporate linear combinations of feedback control terms related to the time course of the difference (the error signal) between the pharmacodynamic response and its basal value. Following the well-established approach of traditional engineering control theory, the proposed feedback control indirect response models incorp

Spinal circuits for motor learning.

Studies of motor learning have largely focussed on the cerebellum, and have provided key concepts about neural circuits required. However, other parts of the nervous system are involved in learning, as demonstrated by the capacity to 'train' spinal circuits to produce locomotion following spinal cord injury. While somatosensory feedback is necessary for spinal motor learning, feed forward circuits within the spinal cord must also contribute. In fact, motoneurons themselves could act as comparators that integrate feed forward and feedback inputs, and thus contribute to motor learning. Applicati

The role of luminance and chromatic cues in emmetropisation.

PURPOSE: At birth most, but not all eyes, are hyperopic. Over the course of the first few years of life the refraction gradually becomes close to zero through a process called emmetropisation. This process is not thought to require accommodation, though a lag of accommodation has been implicated in myopia development, suggesting that the accuracy of accommodation is an important factor. This review will cover research on accommodation and emmetropisation that relates to the ability of the eye to use colour and luminance cues to guide the responses. RECENT FINDINGS: There are three ways in whi

The effect of non-contingent and accurate performance feedback on pacing and time trial performance in 4-km track cycling.

OBJECTIVE: To determine whether the provision of comparative performance feedback during 4 km track cycling affects completion time. DESIGN: Five highly trained male cyclists first performed a baseline (BL) 4-km time trial (TT) on a velodrome track, followed by two further randomised 4-km TT, during which riders received either correct (COR) or non-contingent (FAL) feedback based on their BL performance. RESULTS: Participants completed the COR TT in a significantly faster time (t(4)=-3.10, p<0.05) than the FAL TT (341 (8) s vs 350 (12) s). Over the TT, a significant difference in mean speed

Extracellular calcium-sensing receptors in the parathyroid gland, kidney, and other tissues.

PURPOSE OF REVIEW: The discovery of the extracellular calcium-sensing receptor, CasR has broadened our understanding of calcium homeostasis and led to the development of new pharmacological agents, calcimimetics, for treating hyperparathyroidism. In the present review, I discuss the function of CasR as well as provide evidence for the presence of additional calcium-sensing mechanisms in the skeleton and possibly other tissues. RECENT FINDINGS: Inactivating and activating mutations of the CasR respectively cause hereditary hyperparathyroidism, and demonstrate the predominant role of the CasR i

Cushing's syndrome: from physiological principles to diagnosis and clinical care.

The physiological control of cortisol synthesis in the adrenal cortex involves stimulation of adrenocorticotrophic hormone (ACTH) by hypothalamic corticotrophin-releasing hormone (CRH) and then stimulation of the adrenal by ACTH. The control loop of the hypothalamic-pituitary-adrenal (HPA) axis is closed by negative feedback of cortisol on the hypothalamus and pituitary. Understanding this system is required to master the diagnosis, differential diagnosis and treatment of endogenous hypercortisolism--Cushing's syndrome. Endogenous Cushing's syndrome is caused either by excess ACTH secretion or

The effects of video and nonnegative social feedback on distorted appraisals of bodily sensations and social anxiety.

The effects of video feedback and nonnegative feedback from other people were examined as possibly ameliorating distorted appraisals of bodily sensations, as well as subjective and physiological anxiety in socially anxious individuals. Nonnegative feedback from a confederate emphasized the absence of negative outcomes (e.g., did not seem to tremble) rather than the presence of positive outcomes (e.g., looked calm). Socially anxious students were randomly assigned to either the experimental group, which received video and social feedback (n=12), or the control group (n 13). Participants were as
