Cellular self-organization orchestrates ferroptosis propagation.

Self-organization generates cellular patterns that guide morphogenesis, but how it regulates collective cell fate remains underexplored. We show that cellular self-organization constrains the propagation of ferroptosis, an iron-dependent form of oxidative cell death that can otherwise spread as unbounded trigger waves. The spatial patterning of cell density and alignment encodes transcriptional programs, priming cells toward ferroptosis-sensitive or -resistant fates. Low-density, misaligned regions with elevated YAP/TEAD1 activity are sensitive, whereas high-density, aligned regions with incre

PLA2G2F suppresses ferroptosis through phospholipid remodeling.

Ferroptosis is an iron-dependent form of regulated cell death driven by phospholipid peroxidation, and it has emerged as a potential therapeutic vulnerability of cancer. Here we identify the secretory phospholipase PLA2G2F (phospholipase A2 group IIF) as a ferroptosis suppressor in bladder cancer and elucidate its regulation and mechanism of action. PLA2G2F functions through an intracellular mechanism by localizing to the endoplasmic reticulum to inhibit ferroptosis. Our genetic and pharmacological analyses reveal that peroxisome proliferator-activated receptor γ (PPARG), a nuclear hormone rec

Pro-inflammatory Cytokines, Ferroptosis, and Cancer.

Ferroptosis, iron-dependent regulated cell death, is induced by the polyunsaturated fatty acid peroxidation of membrane phospholipids and is controlled by glutathione peroxidase 4. In recent years, convincing evidence has emerged, demonstrating a close relationship between chemo-, radio-, immuno-, and targeted therapy resistance and ferroptosis resistance. In this review, we discuss the basic principles of ferroptosis in cancer. Considerable attention is paid to the formation of an immunosuppressive tumor microenvironment. The main focus is centered on the involvement of the excessive, chronic

Ferroptosis: when metabolism meets cell death.

We present here a comprehensive update on recent advancements in the field of ferroptosis, with a particular emphasis on its metabolic underpinnings and physiological impacts. After briefly introducing landmark studies that have helped to shape the concept of ferroptosis as a distinct form of cell death, we critically evaluate the key metabolic determinants involved in its regulation. These include the metabolism of essential trace elements such as selenium and iron; amino acids such as cyst(e)ine, methionine, glutamine/glutamate, and tryptophan; and carbohydrates, covering glycolysis, the cit

Targeting ferroptosis for treating kidney disease.

Ferroptosis is a type of regulated cell death hallmarked by iron-mediated excessive lipid oxidation. Over the past decade since the coining of the term ferroptosis, advances in research have led to the identification of intracellular processes that regulate ferroptosis such as GSH-GPX4 pathway and FSP1-coenzyme Q10/vitamin K pathway. From a disease perspective, the involvement of ferroptosis in pathological conditions including kidney disease has attracted attention. In terms of renal pathophysiology, ferroptosis has been widely investigated for its involvement in ischemia-reperfusion injury,

Berberine protects cardiac cells against ferroptosis.

OBJECTIVES: Cardiovascular diseases are one of the primary causes of death. Cardiomyocyte loss is a significant feature of cardiac injury. Ferroptosis is iron-dependent cell death, which occurs due to excess iron and reactive oxygen species (ROS) accumulation causing lipid peroxidation, and subsequent cell death. Ferroptosis has been confirmed to mediate ischemia/reperfusion-induced cardiomyopathy and chemotherapy-induced cardiotoxicity. Berberine (BBR) has been proven to protect the heart from cardiomyopathies, including cardiac hypertrophy, heart failure, myocardial infarction, and arrhythmi

Embryonal erythropoiesis and aging exploit ferroptosis.

Ferroptosis is a form of regulated cell necrosis, as a consequence of Fe(II)-dependent lipid peroxidation. Although ferroptosis has been linked to cancer cell death, neurodegeneration and reperfusion injury, physiological roles of ferroptosis have not been elucidated to date mostly due to the lack of appropriate methodologies. Here, we show that 4-hydroxy-2-nonenal (HNE)-modified proteins detected by a HNEJ-1 mouse monoclonal antibody is a robust immunohistochemical technology to locate ferroptosis in tissues in combination with morphological nuclear information, based on various models of fer

Dysregulation of ferroptosis-related genes and the ACSL4-mediated ferroptosis-PD-L1 axis in prostate cancer pathogenesis.

Prostate cancer (PCa) is a major cause of morbidity and mortality worldwide. Ferroptosis, an iron-dependent form of regulated cell death, has emerged as a crucial mechanism in various cancers, including PCa. This study focused on two interrelated aspects of ferroptosis in PCa. Firstly, it sought to determine whether ferroptosis-related genes are dysregulated in PCa and correlate with disease subtypes and prognosis. Secondly, it aimed to investigate whether the ACSL4/PD-L1 axis mediates crosstalk between ferroptosis and the immune microenvironment. Using TCGA data, we identified 1019 differenti

Ethanolamine phosphate phospholyase drives non-alcoholic fatty liver disease via hepatocyte ferroptosis and ferroptosis-mediated macrophage reprogramming.

Obesity is closely linked to the development of non-alcoholic fatty liver disease (NAFLD), yet its mechanisms are not fully understood. Ethanolamine phosphate phospholyase (ETNPPL), an enzyme highly expressed in the liver, catalyzes the conversion of phosphoethanolamine to ammonia, inorganic phosphate, and acetaldehyde. This study shows that ETNPPL plays a crucial role in obesity-associated NAFLD by regulating lipid accumulation, liver dysfunction, and lipid homeostasis. ETNPPL expression was increased in high-fat diet (HFD)-induced NAFLD mice, while its deficiency alleviated NAFLD. ETNPPL ove

ALDH3A2 negatively orchestrates gastric cancer progression through a synergistic induction of ferroptosis and ferroptosis-driven macrophage reprogramming.

Gastric cancer (GC) is a prevalent gastrointestinal malignancy in which ferroptosis, mitochondrial dysfunction, and macrophage reprogramming remarkably contribute to disease progression. However, the molecular interplay among these processes in contributing to GC remains poorly understood. In this study, we identified ferroptosis- and mitochondrial dysfunction-related genes (FMDRGs) implicated in GC through bioinformatics analyses. Among them, aldehyde dehydrogenase 3 family member A2 (ALDH3A2) was identified as a key FMDRG significantly downregulated in GC tissues and cell lines. Functional a

WTAP targets SOX2 to inhibit trophoblast ferroptosis in the pathogenesis of preeclampsia via m6A-dependent ferroptosis regulation.

BACKGROUND: Preeclampsia (PE) is associated with trophoblast damage, which may be caused by aberrant ferroptosis, an important process in placental development and function. The key component of N6-methyladenosine (m6A)-wilms tumor 1 associated protein (WTAP) has been reported to be involved in the migration and invasion of trophoblasts in the pathogenesis of PE. However, the connection between WTAP and ferroptosis in PE remained poorly understood. Accordingly, in order to explore potential treatments for PE, and understand how WTAP influences the trophoblasts' ferroptosis in PE, researchers c

Applying Multimodal Mass Spectrometry to Image Tumors Undergoing Ferroptosis Following In Vivo Treatment with a Ferroptosis Inducer.

Epithelial ovarian cancer (EOC) is the most common form of ovarian cancer. The poor prognosis generally associated with this disease has led to the search for improved therapies such as ferroptosis-inducing agents. Ferroptosis is a form of regulated cell death that is dependent on iron and is characterized by lipid peroxidation. Precise mapping of lipids and iron within tumors exposed to ferroptosis-inducing agents may provide insight into processes of ferroptosis in vivo and ultimately assist in the optimal deployment of ferroptosis inducers in cancer therapy. In this work, we present a metho

FerroEnrich: an interactive web platform for liver-focused ferroptosis index scoring, enrichment, and ferroptosis-senescence cross-talk analysis.

MOTIVATION: Ferroptosis is an iron-dependent regulated cell death process involving lipid peroxidation, redox imbalance, and impaired antioxidant defense. It is increasingly relevant to metabolic liver disease and non-alcoholic steatohepatitis. Existing ferroptosis resources mainly provide gene annotations and disease associations, but they do not offer an integrated workflow for user-uploaded transcriptome data, liver-focused enrichment, dataset-level scoring, and ferroptosis-senescence cross-talk analysis. RESULTS: We developed FerroEnrich, an open-source web platform for analyzing ferropto

Combine immune checkpoint indoleamine 2,3-dioxygenase 1 knockdown with ferroptosis inducer Erastin/RSL3 accelerates colorectal cancer cell ferroptosis.

OBJECTIVE: Colorectal cancer (CRC) is a malignancy known for its aggressive behavior and notable mortality burden. Indoleamine 2,3-dioxygenase 1 (IDO1), an immune checkpoint molecule, is markedly upregulated in CRC, and it has been implicated in the regulation of key components of the ferroptosis pathway. This study aimed to elucidate the regulatory interaction between IDO1 and ferroptosis and assess the potential therapeutic effect of combined IDO1 inhibition with ferroptosis inducers Erastin and RSL3 in CRC. MATERIAL AND METHODS: Three CRC cell lines (CT26, MC38, and HT-29) and one normal c

Ferroptosis in skin diseases and cutaneous immunology.

Ferroptosis is a regulated form of cell death driven by iron-dependent phospholipid peroxidation, but its immunological identity is not fixed. This context dependence complicates therapeutic development: ferroptosis may eliminate malignant cells but can also amplify inflammation or impair protective immune responses. Moreover, sublethal ferroptosis-associated stress can alter cellular function without causing cell death. The skin provides an informative model for investigating these divergent effects because epidermal differentiation and cutaneous compartmentalization create spatial heterogene

Fueling Death: The Metabolic Control of Ferroptosis.

Ferroptosis is a metabolically regulated, iron-dependent form of cell death driven by lipid peroxidation. This chapter dissects the core metabolic circuits that converge to determine ferroptotic susceptibility and explores the interplay between ferroptosis and other cell death programs, highlighting its physiological roles in development, immunity, and aging, and examines its dual function in disease. Emerging pharmacological strategies to modulate ferroptosis in cancer and degenerative conditions are discussed. By integrating metabolic checkpoints with redox biology, ferroptosis emerges as a

Role of ferroptosis on immunotherapy in breast cancer.

Breast cancer is the most prevalent cancer among women globally, and its rising incidence and mortality rates underscore the urgent need for new treatment strategies. Current treatment methods including surgery, radiotherapy, chemotherapy have limited efficacy. In recent years, immunotherapy has attracted considerable attention as an innovative cancer treatment method due to its ability to activate the immune system against tumor cells, particularly in breast cancer. Ferroptosis, a newly identified form of programmed cell death, has been demonstrated to be closely associated with the initiatio

Targeting ferroptosis to halt MASLD and MASH.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global epidemic, with its inflammatory form, metabolic dysfunction-associated steatohepatitis (MASH), driving cirrhosis and hepatocellular carcinoma. Ferroptosis, an iron-dependent form of programmed cell death, is a central pathogenic mechanism in MASLD and MASH. The core drivers of ferroptosis include dysregulated iron metabolism, an increase in peroxidation-susceptible phospholipids, and impaired antioxidant defenses. These changes promote hepatocyte death via a so-called 'ferroptotic explosion' triggered by an increase i

Ferroptosis: a promising therapeutic target for periodontitis.

Periodontitis is a chronic inflammatory disease driven by dysbiotic biofilm and abnormal host adaptive immune responses, characterized by irreversible destruction of periodontal tissues. Recent studies indicate that ferroptosis-a form of iron-dependent lipid peroxidation-mediated cell death-is not a passive outcome of the disease process but rather an important active inducer and pathological amplifier in the development of periodontitis. This review synthesizes existing evidence to propose that ferroptosis acts as an important regulatory node within the pathological network of periodontitis,
