Multimodal characterization of temperature-induced structural remodeling and aggregation of regenerated silk fibroin.

Regenerated silk fibroin (RSF) is a protein biomaterial whose molecular conformation, aggregation behavior, and functional properties are strongly influenced by processing conditions. The current study investigated the structural response of RSF to thermal treatment following incubation at 50-100 °C for 3 h. Intrinsic fluorescence increased progressively with treatment temperature without an appreciable displacement of the emission maximum, while fluorescence lifetime measurements at selected conditions indicated altered excited-state behavior of the intrinsic fluorophore population. Increased

Manufacturing Silk Fibroin Hollow Nanoyarns as Fundamental Units for Advanced Medical Textiles.

Nature-inspired designs aim to replicate the hierarchical structure observed in native tissues. Fibers serve as fundamental modular units, enabling the fabrication of complex architectures for the engineering of medical textiles and tissue equivalents. However, synthetic yarns lack inherent biological cues to support tissue integration, leading to a growing interest in yarns derived from natural materials. Here, we describe for the first time the fabrication of hollow nanoyarns from pure silk fibroin (SF) using an advanced funnel electrospinning process. This yielded long SF nanoyarns, spannin

Silk Fibroin Gels Fabricated from Ajisawa Reagent with Mechanochromic Sensing Capability.

Silk fibroin (SF) derived from Bombyx mori cocoons was employed to fabricate gels using Ajisawa reagent (AJ) as the extraction medium. The resulting regenerated SF solution was subsequently directly dialyzed against an ethanol-water mixture, inducing a gradual and controllable solution-gel transition. Obtained SF gels exhibit a pronounced increase in compressive strength aligned with rising initial fibroin concentration, along with excellent water stability. FESEM, FTIR, and synchrotron X-ray diffraction analyses reveal a homogeneous network structure, likely governed by uniformly distributed

Silk Fibroin-Based Matrices for the Guidance of Cell Interaction, Tissue Regeneration, and Crosstalk.

The interactions between cells and the extracellular matrix are essential regulators of cell behaviors such as adhesion, proliferation, migration, differentiation, and function. From the perspective of tissue regeneration, some physicochemical characteristics of the material, including hydrophilicity, topology, and charge of the material surface, can significantly affect cell adhesion, proliferation, and differentiation. Many biomaterials are introduced for tissue engineering scaffolds, biomimicking natural tissues. Among the biomaterials, silk proteins (fibroin and sericin) have many excellen

Kinetically Tuning the Structural Assembly of Silk Fibroin for Highly Piezoelectric Wearable Sensors.

To overcome the challenges of extended processing and filler dependency for performance enhancement in pure silk fibroin (SF) piezoelectric devices, this study introduces a regeneration-free, integrated dissolution-spinning-crystallization strategy. This approach effectively enhances the intrinsic piezoelectric potential of SF by controlling the conformational rearrangement of its molecular chains and promoting the formation of ordered β-sheet crystal structures. Uniform nanofibers were fabricated under optimized conditions (6 wt % CaCl2, 18 wt % SF, and 19 kV). A subsequent treatment with 90%

Novel Silk Fibroin Based Bilayer Scaffolds for Bioabsorbable Internal Biliary Stenting.

Biliary duct reconstruction is one of the most challenging parts of liver transplantation and accounts for 40%-60% of complications. While current stent-based devices on the market show promising results in reducing complications, they are manufactured from permanent synthetic materials and require a second reintervention for their removal. This exposes the patients to other potential complications and increases healthcare costs. This study develops a fabrication technique to produce a bioabsorbable biliary stent based on silk fibroin. The process used a dip-coating procedure for silk fibroin

Silk Fibroin Formed Bioadhesive Ophthalmic Gel for Dry Eye Syndrome Treatment.

PURPOSE: Dry eye syndrome (DES), arising from various etiologic factors, leads to tear film instability and ocular surface damage. Given its anti-inflammatory effects, cyclosporine A (CsA) has been widely used as a short-term treatment option for DES. However, poor bioavailability and solubility of CsA in aqueous phase make the development of a cyclosporine A-based eye drop for ocular topical application a huge challenge. METHODS: In this study, a novel strategy for preparing cyclosporine A-loaded silk fibroin nanoemulsion gel (CsA NBGs) was proposed to address these barriers. Additionally, t

Silk fibroin hydrogel adhesive enables sealed-tight reconstruction of meniscus tears.

Despite orientationally variant tears of the meniscus, suture repair is the current clinical gold treatment. However, inaccessible tears in company with re-tears susceptibility remain unresolved. To extend meniscal repair tools from the perspective of adhesion and regeneration, we design a dual functional biologic-released bioadhesive (S-PIL10) comprised of methacrylated silk fibroin crosslinked with phenylboronic acid-ionic liquid loading with growth factor TGF-β1, which integrates chemo-mechanical restoration with inner meniscal regeneration. Hyaluronic acid/silk fibroin nanoparticles loaded with methotrexate for topical treatment of psoriasis.

Systemic administration of methotrexate (MTX), widely regarded as one of the most effective treatments for psoriasis, poses significant challenges due to its high toxicity, limited solubility, and potential for adverse effects. Consequently, developing a topical form of MTX may offer a safer and more effective strategy for psoriasis management. Silk fibroin (SF), a protein-based biomacromolecule, has shown considerable promise as a nanocarrier for sustained and targeted drug delivery, owing to its exceptional physicochemical and biological properties. This study aimed to develop and characteri

Silk Fibroin Encapsulated Icariin Nanoparticles Mitigate Bisphenol A-Induced Spermatogenesis Dysfunction.

Bisphenol A (BPA) is a prevalent endocrine disruptor found in natural environments. Exposure to BPA has been associated with male infertility. The natural phytochemical icariin (ICA) has demonstrated significant promise for the treatment of male infertility. However, its effectiveness is limited due to its low bioavailability, poor water solubility, and insufficient targeting abilities. Herein, novel nanoparticles are generated from the natural silk fibroin, which are used to load ICA. The efficient drug delivery system (ICA-SNPs) result in significantly focused drug distribution to spermatogo

Development of a More Environmentally Friendly Silk Fibroin Scaffold for Soft Tissue Applications.

A push for environmentally friendly approaches to biomaterials fabrication has emerged from growing conservational concerns in recent years. Different stages in silk fibroin scaffold production, including sodium carbonate (Na2CO3)-based degumming and 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP)-based fabrication, have drawn attention for their associated environmental concerns. Environmentally friendly alternatives have been proposed for each processing stage; however, an integrated green fibroin scaffold approach has not been characterized or used for soft tissue applications. Here, we show that

Application of silk fibroin coatings for biomaterial surface modification: a silk road for biomedicine.

Silk fibroin (SF) as a natural biopolymer has become a popular material for biomedical applications due to its minimal immunogenicity, tunable biodegradability, and high biocompatibility. Nowadays, various techniques have been developed for the applications of SF in bioengineering. Most of the literature reviews focus on the SF-based biomaterials and their different forms of applications such as films, hydrogels, and scaffolds. SF is also valuable as a coating on other substrate materials for biomedicine; however, there are few reviews related to SF-coated biomaterials. Thus, in this review, w

A medical adhesive used in a wet environment by blending tannic acid and silk fibroin.

A multifunctional and effective medical adhesive with a combination of high toughness and superior adhesion is highly desired in biomedical fields. However, clinical application of medical adhesives is still limited due to their weak adhesion to wet tissue. In this study, a novel medical adhesive called TASK composed of tannic acid (TA) and silk fibroin (SF) based on polyphenol-gel systems was developed. TASK powder was prepared by a simple physical mixture of pyrogallol-rich tannic acid and silk fibroin in aqueous solution and further freeze drying, which was stable and convenient for sterili

Meniscal Replacement With a Silk Fibroin Scaffold Reduces Contact Stresses in the Human Knee.

The aim of the current study was to verify if a previously developed silk fibroin scaffold for meniscal replacement is able to restore the physiological distribution of contact pressure (CP) over the articulating surfaces in the human knee joint, thereby reducing peak loads occurring after partial meniscectomy. The pressure distribution on the medial tibial articular surface of seven human cadaveric knee joints was analysed under continuous flexion-extension movements and under physiological loads up to 2,500 N at different flexion angles. Contact area (CA), maximum tibiofemoral CP, maximum pr
