Exploring the vector potential of Lipoptena cervi (Diptera: Hippoboscidae): first record of Setaria cervi (Filarioidea: Onchocercidae) DNA in deer keds.

Setaria cervi is a filarial nematode that infects both wild and domestic ungulates. It is primarily transmitted by mosquitoes, although the possible role of other hematophagous insects is still uncertain. We investigated 83 wingless deer keds (Lipoptena spp.) from red and fallow deer in northern and eastern Slovakia, as well as 43 red deer liver samples for the presence of filarial DNA. Deer keds were identified as Lipoptena cervi (n = 80) and Lipoptena fortisetosa (n = 3). Genomic DNA from individual ectoparasites was screened by PCR targeting a mitochondrial cox1 gene fragment of filaroid ne

Setaria cervi (Filarioidea, Onchocercidae) undressing in ungulates: altered morphology of developmental stages, their molecular detection and complete sequence cox1 gene.

This work introduces new morphological and molecular information on the filaroid nematode Setaria cervi (Rudolphi, 1819) obtained from 13 infected game ungulates out of 96 dissected. The hosts comprised the following: a single moose (Alces alces), ten red deer (Cervus elaphus) and two sika deer (Cervus nippon) originating from the western and northern regions of the Czech Republic. Based on the complete sequences of the gene encoding mitochondrial cytochrome c oxidase subunit 1 (cox1), all 20 females and four males belonged to the species S. cervi. We detected three developmental female stages

PREVALENCE AND MOLECULAR CHARACTERIZATION OF DIROFILARIA IMMITIS (FILARIOIDEA: ONCHOCERCIDAE) IN DOGS FROM ENDEMIC AREAS OF RIO DE JANEIRO STATE, BRAZIL.

Canine dirofilariasis is common in Brazil, but molecular diagnosis is rare even though molecular studies increase our knowledge about molecular epidemiology and circulating genotypes from helminths worldwide. This study aims to estimate the prevalence of infection with a modified Knott's test and to perform molecular characterization of Dirofilaria immitis (Leidy, 1856) Railliet and Henry, 1911, in dogs from endemic areas of Maricá and Niterói municipalities, Rio de Janeiro State, Brazil. Molecular characterization was performed in 33 blood samples from dogs positive for microfilariae and 4 ad

In cold blood: screening wild Alaskan moose (Alces alces gigas) for filarial nematode (Filarioidea: Onchocercidae) infections using a deep-amplicon sequencing approach.

Moose are significant ecological, economical and cultural animals for the stakeholders of Alaska, USA. Thus, the impact of pathogens, like filarial nematodes, is a critical area of moose research. These vector-borne parasites, including Setaria yehi, and Rumenfilaria andersoni, can lead to severe health consequences (e.g., peritonitis). However, little is known about filarial nematode distribution, diversity and its associated life cycle with Alaskan moose hosts. Newly developed next-generation sequencing techniques offer the ability to efficiently screen multiple species of co-infecting filar

Validation of a species-specific probe-based qPCR for detection of Setaria yehi (Filarioidea: Onchocercidae) in Alaskan moose (Alces alces gigas).

Northern ungulates contend with Setaria yehi and Rumenfilaria andersoni, filarioid nematodes that are transmitted by ectoparasitic blood-feeding arthropods, which can result in animal and population level impacts. Setaria yehi microfilariae can be detected in fresh blood samples using a modified Knott's test, or by postmortem detection by genetic sampling or through the retrieval of adult specimens in the peritoneal cavity. In this study we validated a novel qPCR for detection of S. yehi DNA in blood samples of moose (Alces alces). Additionally, we compared quantitative values from modified Kn

Complement-mediated leukocyte adherence to infective larvae of Dipetalonema viteae (Filarioidea): requirement for eosinophils or eosinophil products in effecting macrophage adherence.

The present study reports the existence of C-mediated adherence of eosinophils and/or macrophages to filarial infective larvae of Dipetalonema viteae. C3 molecules are present on the surface of the parasite, as shown by immunofluorescence studies. Samples of fNRS depleted of AP of complement by treatment with Zymosan A or of factor B by heating at 50 degrees C for 20 min fail to mediate cell adherence to the parasite. In contrast, fNRS inactivated for CP of complement by the chelating agent EGTA retains its activity in mediating cell adherence to the parasite. There is a significant consumptio

Setaria labiatopapillosa (Filarioidea, Nematoda) in Moroccan cattle: atypical localization and morphological characterization of females and microfilariae by light and scanning electron microscopy.

Filarioid nematodes are parasites of the tissues and tissue spaces of all vertebrates except fish. Females produce microfilariae that enter the host's blood circulation or skin and may cause ocular and neurological pathology, leading to important implications in veterinary and public health. The present work is the first investigation on Setaria labiatopapillosa conducted in Morocco to characterize the morphological features of both adult and microfilaria forms. Two adult female nematodes were found free in the thoracic cavity of a slaughtered 3.5-year-old (6 teeth) Moroccan enhanced cross-bre

Comparative susceptibility of species A, B and C of Anopheles quadrimaculatus complex to infection with subperiodic Brugia malayi and Brugia pahangi (Nematoda: Filarioidea).

Susceptibilities of natural populations of sibling species A, B and C of the Anopheles quadrimaculatus complex and the colonized strain A to subperiodic Brugia malayi and Brugia pahangi were compared. All 3 sibling species showed varying degrees of susceptibility to both B. pahangi and B. malayi, and they were considerably more susceptible to B. pahangi than to B. malayi. The rate and intensity of infection to B. pahangi were highest for species A (66.2% and 7.4 L3/female, respectively) and lowest for species B (21.3% and 1.7 L3/female). For B. malayi these values were higher for species A (29

A preliminary analysis of the population genetics and molecular phylogenetics of Onchocerca volvulus (Nematoda: Filarioidea) using nuclear ribosomal second internal transcribed spacer sequences.

Nuclear internal transcribed spacer 2 (ITS2) rDNA sequences were used for a molecular phylogenetics analysis of five Onchocerca species. The sister species of the human parasite O. volvulus was found to be the cattle parasite O. ochengi and not O. gibsoni, contrary to chromosomal evidence. The genetic differentiation of two African populations (representing the two African strains) and a Brazilian population of O. volvulus was also studied. Phylogenetic and network reconstruction did not show any clustering of ITS2 alleles on geographic or strain grounds. Furthermore, population genetics tests

Course of antibody production by the DIG-ELISA method in neonatally infected and juvenile infected rats after primary infection with Breinlia booliati (Filarioidea: Onchocercidae).

The course of antibody production in Wistar neonatal and juvenile rats after primary infection with Breinlia booliati was studied by the DIG-ELISA technique using filter papers impregnated with capillary blood drawn from the infected rat tails at 7, 14, 28, 60 and 90 days post infection. Sera of neonatally infected rats did not react with adult worm antigen until day 7 and the titers of antibody remained at very low levels for the next 7 days. There was little tendency to eliminate the filarial larvae during this time. The antibody levels then rose rapidly throughout the next fortnight and inc

Detection of Dirofilaria immitis (Nematoda: Filarioidea) by polymerase chain reaction in Aedes albopictus, Anopheles punctipennis, and Anopheles crucians (Diptera: Culicidae) from Georgia, USA.

Potential mosquito vectors of Dirofilaria immitis (Leidy) (Nematoda: Filarioidea), the causative agent of dog heartworm in the southeastern region of the United States, were collected with CDC light traps and gravid traps in seven counties in the state of Georgia, USA. The presence of D. immitis in these mosquitoes was detected by polymerase chain reaction using species-specific primers for the D. immitis surface or cuticular antigen. Overall, 1,574 mosquitoes of 13 species in seven genera were collected; 92% of the specimens were Aedes albopictus (Skuse), Aedes vexans (Meigen), or Anopheles p

Onchocerca eberhardi n. sp. (Nematoda: Filarioidea) from sika deer in Japan; relationships between species parasitic in cervids and bovids in the Holarctic region.

Onchocerca eberhardi n. sp. from the sika deer, Cervus nippon, in Japan is described. Adult worms lived in the carpal ligament; infection reached high levels (up to 25 female and 16 male worms in a single carpal limb). Skin dwelling microfilariae were mainly found in the ears. Prevalence of infection was 81% at the type locality, Mt. Sobo, in Kyushu. The new material was compared to the 31 species of Onchocerca presently known. Onchocerca eberhardi n. sp. females were characterized by a long slender anterior end and a thin esophagus < or =1 mm long with no or only a slight glandular region. Th

Affinities between Cutifilaria (Nematoda: Filarioidea), parasites of deer, and Mansonella as seen in a new onchocercid, M. (C.) perforata n. sp., from Japan.

A new dermal filarioid nematode, collected from Cervus nippon nippon (sika deer) on Kyushu Island, Japan, showed close affinities between the genera Cutifilaria and Mansonella (Onchocercidae: Onchocercinae): no buccal capsule, esophagus reduced to a thin fibrous tube, and female tail with four lappets. We propose Cutifilaria as a subgenus of Mansonella. Cutifilaria was distinguished from the five other subgenera, Mansonella, Tetrapetalonema, Esslingeria, Sandnema, and Tupainema, in having an area rugosa composed of transverse bands with tiny points, 14-16 papillae around the cloacal aperture,

Demonstration of anti-cuticular antibodies by immuno-electron microscopy in sera of mice immunized with cuticular extracts and isolated cuticles of adult Dipetalonema viteae (Filarioidea).

The immunogold technique was used for the ultrastructural localization of antibody-binding sites on thin sections of Lowicryl K4M embedded adult females, infective larvae and pieces of adult cuticles of Dipetalonema viteae insoluble in SDS-2-ME. The antisera used were either produced against SDS-2-ME extracts of cuticles or the insoluble pellet after SDS-2-ME extraction. With both types of antisera a labelling of epitopes on fibers was achieved in intact cuticles. In isolated cuticles the corresponding structures were absent. The same sera crossreacted with the larval and microfilarial cuticle

Multiplex polymerase chain reaction for detection of Dirofilaria immitis (Filariidea: Onchocercidae) and Wuchereria bancrofti (Filarioidea: Dipetalonematidae) in their common vector Aedes polynesiensis (Diptera: Culicidae).

In French Polynesia, Aedes polynesiensis (Marks) is the vector of the human filarial parasite Wuchereria bancrofti (Cobbold) and dog heartworm, Dirofilaria immitis (Leidy). A multiplex polymerase chain reaction (PCR) assay was designed to screen pools of field-collected Ae. polynesiensis for the presence of both parasites simultaneously using primers specific for each parasite. The sensitivity of detection on purified DNA was 1 and 10 pg, equivalent to 0.1 and 1 L3 larva per pool for W. bancrofti and D. immitis, respectively. Codetection was performed at an hybridization temperature of 58 degr

Discovery of potentially novel species of the Onchocercidae (Nematoda: Filarioidea) in Burmese fighting chickens (Gallus gallus): Genetic insights into avian filariasis and co-infection with Plasmodium juxtanucleare.

Burmese fighting chickens (Gallus gallus) raised in Nakhon Ratchasima Province, eastern Thailand, were investigated for filarioids and co-infection with other haemoparasites. Microscopy and molecular diagnostic targeting the cox1 gene revealed that 2 out of 12 chickens tested positive for onchocercid filarioids. One chicken carried a single infection (Onchocercidae sp. CH09), characterized by unsheathed microfilariae with a cross-striated cuticle, a blunt anterior end, a short cephalic space, and a hook-like tail tip. Another chicken exhibited a mixed filarioid infection, necessitating the sub
