Induction of triploidy and gynogenesis in teleost fish with emphasis on marine species.

The induction of triploidy and gynogenesis by chromosome set manipulation has traditionally been studied more intensively in freshwater than in marine fish. In the last years, however, several studies have applied these manipulations in about a dozen marine species, including mainly sparids, moronids and flatfishes. This paper focuses on the methodologies used to induce, verify, and assess performance of both triploids and gynogenetics of these marine species. Since many of them are batch spawners and have small and fragile eggs and larvae, peculiarities relating to broodstock management, game

The hatching mechanism in Atlantic halibut (Hippoglossus hippoglossus).

In general fish larvae emerge from the protective egg after secreting a hatching enzyme (HE) from diffusely located hatching gland cells (HGCs). This proteolytic enzyme is distributed over the entire inner part of the eggshell (zona radiata). In a marine flatfish halibut, (Hippoglossus hippoglossus), we have found a more specialized hatching process. A strategic location of the HGCs in a narrow belt on the anterior part of the yolk sac leads to restricted degradation of the eggshell resulting in cleavage of the eggshell into two distinct rigid parts. This hatching process--termed "rim-hatching

Expressed sequence tags (ESTs) from immune tissues of turbot (Scophthalmus maximus) challenged with pathogens.

BACKGROUND: The turbot (Scophthalmus maximus; Scophthalmidae; Pleuronectiformes) is a flatfish species of great relevance for marine aquaculture in Europe. In contrast to other cultured flatfish, very few genomic resources are available in this species. Aeromonas salmonicida and Philasterides dicentrarchi are two pathogens that affect turbot culture causing serious economic losses to the turbot industry. Little is known about the molecular mechanisms for disease resistance and host-pathogen interactions in this species. In this work, thousands of ESTs for functional genomic studies and potenti

Construction of a multi-tissue immune atlas for Chinese tongue sole (Cynoglossus semilaevis) reveals granulocyte functional adaptation and underlying regulatory mechanisms.

Innate immunity in teleost fish relies heavily on granulocytes for host defense. However, how these cells functionally adapt to distinct tissue microenvironments remains poorly understood. To address this, we constructed a multi-tissue immune atlas for Cynoglossus semilaevis by integrating single-cell RNA sequencing (scRNA-seq) data from blood, liver, spleen, and head kidney. We identified five transcriptionally distinct granulocyte subsets (Gr1-Gr5), each with divergent roles in inflammation, immunoregulation, and complement activity; moreover, these subsets exhibited tissue-specific distribu

Morphology of the tongue dorsal surface of turbot (Scophthalmus maximus).

This study aimed to analyze the morphology of the tongue, which varies among fish species and has not been studied in turbot (Scophthalmus maximus), using macro-anatomical, light and scanning electron microscopy (SEM). As research materials, the tongues of eight adult turbot, which were used for consumption, were examined. The roughly triangular-shaped tongue of the turbot consisted of three parts: apex, body, and root. The thickness of the tongue continued to increase from the apex to the root. Although papillae were not observed in the macroscopic examination of the tongue, microscopic exami

New insight on vertebral anomalies in cultured Senegalese sole (Solea senegalensis, Kaup) at early stages of development.

Senegalese sole (Solea senegalensis, Kaup) is a promising flatfish species in aquaculture. However, skeletal anomalies are still a great concern in sole farming. Investigation of this issue is crucial to improving larval quality and optimizing production. The aim of this study was to thoroughly assess anomalies in the rachis of reared sole at early developmental stages. Sole (n = 507) were sampled at 31 or 32 days after hatching (dah). The specimens were stained with alcian blue and alizarin red and evaluated for the detection of vertebral deformities. Most fish presented 9:34:3 vertebrae in a

The eye of the tongue sole Cynoglossus bilineatus (Lacepède, 1802) (Teleostei: Pleuronectiformes).

We report the ocular features of the tongue sole, Cynoglossus bilineatus (Lacepède, 1802), a marine, bottom-dwelling flatfish. In this species, both eyes are located juxtaposed on the same side of the flat head. Histology revealed the sclera to be fibrous (collagenous) in nature. The choroid possesses the choriocapillaris, and adjacent to it, 3-4 rows of iridophores with stacks of cytoplasmic platelets. No choroidal gland is present. The retinal pigment epithelium (RPE) contains scanty melanin granules. Its vitread half is modified into a dense tapetum with lipid spheres (about 0.34 μm in diam

Skeletal anomalies in reared Senegalese sole Solea senegalensis juveniles: a radiographic approach.

Reared Senegalese sole Solea senegalensis Kaup show a high incidence of vertebral anomalies; however, little is known about its skeletal anomaly profile in the later farming phases. The purpose of this study was to provide a detailed description and quantification of the most common skeletal anomalies in reared Senegalese sole in the juvenile stage by means of computed radiography. A total of 374 Senegalese sole were classified according to the external morphology of the fish as normal or altered and then radiographed in latero-lateral and in dorso-ventral projections. Gonadogenesis analysis and sex differentiation in cultured turbot (Scophthalmus maximus).

As a flatfish, the turbot (Scophthalmus maximus) is one of the most important farmed fish species with great commercial value, which has a strong sexual dimorphism on growth rate and sexual maturity. In this study, using histology, the basic information on proliferation and migration of germ cells and early gonadal development during sex differentiation in turbot were described in detail. There were six to nine individual primordial germ cells (PGCs) with large nuclei until 15 days post-hatching (dph). The PGCs located under the mesonephric ducts undergo migration along the dorsal mesentery to

Fatal Asphyxiation in Two Long-Finned Pilot Whales (Globicephala melas) Caused by Common Soles (Solea solea).

Long-finned pilot whales (Globicephala melas) are rare visitors to the southern North Sea, but recently two individual strandings occurred on the Dutch coast. Both animals shared the same, unusual cause of death: asphyxiation from a common sole (Solea solea) stuck in their nasal cavity. This is a rare cause of death in cetaceans. Whilst asphyxiation has been reported in smaller odontocetes, there are no recent records of this occurring in Globicephala species. Here we report the stranding, necropsy and diet study results as well as discuss the unusual nature of this phenomenon. Flatfish are no

Micro-anatomical characterization of vertebral curvatures in Senegalese sole Solea senegalensis.

The micro-anatomical changes associated with lordotic and kyphotic vertebral curvatures (VC) in juvenile and adult Senegalese sole Solea senegalensis are described. In addition, it is demonstrated that the tissue and cellular structures of individual vertebrae can be severely affected. Two main conformations were found in deformed juvenile specimens: flattened vertebrae with dorso-ventral compression and trapezoidal vertebrae forming concave and convex sides under compressive and tensile stresses. Histological analyses revealed the occurrence of an ectopic cartilaginous tissue within the acell

Effect of triploidy on turbot haematology.

This study was carried out to compare key haematological features of diploid (2n) and triploid (3n) turbot as a first step towards the assessment of the ability of 3n turbot to withstand sub-optimal culture conditions. Morphometric indices of erythrocytes were determined on blood smears by light microscopy. Triploidy significantly (P<0.001) increased all morphometric indices measured in the erythrocytes, including size, surface, and volume, except for the size of minor nuclear axis. The increase in cell size was larger for the major (31.0%) than for the minor (8.3%) axis, thus rendering erythr

Expanding the Fish-Brain Invitrome With the Senegalese Sole SsB-1 Cell Line-A Versatile Model for Neurotropic Virus Research.

Cell lines are essential tools for studying animal physiology and immunopathology, reducing the use of live animals and supporting the 3Rs principle of reduction, refinement, and replacement. The Senegalese sole (Solea senegalensis), a relevant species for Spanish aquaculture diversification, lacks established in vitro models. We developed SsB-1, the first brain-derived cell line from this species, providing a valuable platform for studying virus-host interactions. SsB-1 exhibits glial features, stable morphology, and high susceptibility to several betanodavirus (NNV) genotypes, enabling detai

Assembly of the poorly differentiated Verasper variegatus W chromosome by different sequencing technologies.

The assembly of W and Y chromosomes poses significant challenges in vertebrate genome sequencing and assembly. Here, we successfully assembled the W chromosome of Verasper variegatus with a length of 20.48 Mb by combining population and PacBio HiFi sequencing data. It was identified as a young sex chromosome and showed signs of expansion in repetitive sequences. The major component of the expansion was Ty3/Gypsy. The ancestral Osteichthyes karyotype consists of 24 protochromosomes. The sex chromosomes in four Pleuronectiformes species derived from a pair of homologous protochromosomes resultin

Phosphate perils in marine aquaculture: Systematic toxicity assessment in turbot juveniles in cultivation.

Recirculating Aquaculture Systems (RAS) aim to minimize water usage while maintaining optimal conditions for aquatic organisms, which is increasingly recognized as a crucial strategy for advancing sustainable aquaculture practices and enhancing global food security. However, many current marine RAS lack efficient treatments for removing dissolved wastes like phosphates (PO43--P), impacting water quality and potentially affecting the health of cultured organisms. Here, juveniles of turbot (Scophthalmus maximus), a promising and growing marine culture species, were exposed to PO43--P at differen

Skeletal development and abnormalities of the vertebral column and of the fins in hatchery-reared turbot Scophthalmus maximus.

To describe the skeletal development and abnormalities in turbot Scophthalmus maximus, samples were collected every day from hatching to 60 days after hatching (DAH). A whole-mount cartilage and bone-staining technique was used. Vertebral ontogeny started with the formation of anterior haemal arches at 5·1 mm standard length (L(S) ) c. 11 DAH, and was completed by the full attainment of parapophyses at 16·9 mm L(S) c. 31 DAH. Vertebral centra started to develop at 6·3 mm L(S) c. 16 DAH and ossification in all centra was visible at 11·0 mm L(S) c. 25 DAH. The caudal fin appeared at 5·1 mm L(S)

Effects of exposure to pile-driving sounds on the lake sturgeon, Nile tilapia and hogchoker.

Pile-driving and other impulsive sound sources have the potential to injure or kill fishes. One mechanism that produces injuries is the rapid motion of the walls of the swim bladder as it repeatedly contacts nearby tissues. To further understand the involvement of the swim bladder in tissue damage, a specially designed wave tube was used to expose three species to pile-driving sounds. Species included lake sturgeon (Acipenser fulvescens)--with an open (physostomous) swim bladder, Nile tilapia (Oreochromis niloticus)--with a closed (physoclistous) swim bladder and the hogchoker (Trinectes macul

Influence of constant light and darkness, light intensity, and light spectrum on plasma melatonin rhythms in senegal sole.

Light is the most important synchronizer of melatonin rhythms in fish. This paper studies the influence of the characteristics of light on plasma melatonin rhythms in sole. The results revealed that under long-term exposure to constant light conditions (LL or DD), the total 24 h melatonin production was significantly higher than under LD, but LL and DD conditions influenced the rhythms differently. Under LL, melatonin remained at around 224 pg/ml throughout the 24 h, while under DD a significant elevation (363.6 pg/ml) was observed around the subjective evening. Exposure to 1 h light pulses at

Difference in Serum Scuticocidal Activity Against Miamiensis avidus (Ciliophora, Scuticociliatida) Between Olive Flounder (Paralichthys olivaceus) and Starry Flounder (Platichthys stellatus).

Miamiensis avidus is an opportunistic parasite responsible for significant mortalities in marine aquaculture. Given the rising economic losses associated with scuticociliatosis in olive flounder (Paralichthys olivaceus) cultured in Korea, alternative species such as starry flounder (Platichthys stellatus), which demonstrates resistance to certain pathogens affecting olive flounder, are being considered for aquaculture. However, the underlying biological mechanisms influencing host susceptibility to M. avidus remain unclear. In this study, we investigated differences in susceptibility to M. avi
