Protective Effect of [Cu(NN1)2](ClO4) Complex in Rainbow Trout Challenged against Flavobacterium psychrophilum.

Previously, we reported an in vitro evaluation regarding antibacterial effects against F. psychrophilum by a new Cu (I) complex, [Cu(NN1)2](ClO4). This study presents the results of an in vivo evaluation of [Cu(NN1)2](ClO4) added as a dietary supplement against F. psychrophilum in rainbow trout. The results showed that the administration of [Cu(NN1)2](ClO4) at 29 and 58 µg/g of fish for 15 days does not affect the growth of rainbow trout. On the other hand, the amount of copper present in the liver, intestine, and muscle of rainbow trout was determined. The results showed that the amount of co

Erythromycin and florfenicol treatment of rainbow trout Oncorhynchus mykiss (Walbaum) experimentally infected with Flavobacterium psychrophilum.

Flavobacterium psychrophilum is responsible for significant economic losses in rainbow trout aquaculture. Antimicrobial treatment remains the primary means of control; however, there are limited choices available for use. The objectives of the study were therefore to determine the minimum inhibitory concentrations for erythromycin and florfenicol in selected F. psychrophilum isolates and to evaluate their clinical treatment efficacy in experimentally infected rainbow trout. All isolates tested had moderate susceptibility to florfenicol and erythromycin except one isolate, which had low suscept

More than just antibodies: Protective mechanisms of a mucosal vaccine against fish pathogen Flavobacterium columnare.

A recently developed attenuated vaccine for Flavobacterium columnare has been demonstrated to provide superior protection for channel catfish, Ictalurus punctatus, against genetically diverse columnaris isolates. We were interested in examining the mechanisms of this protection by comparing transcriptional responses to F. columnare challenge in vaccinated and unvaccinated juvenile catfish. Accordingly, 58 day old fingerling catfish (28 days post-vaccination or unvaccinated control) were challenged with a highly virulent F. columnare isolate (BGSF-27) and gill tissues collected pre-challenge (0

Biochemical reference intervals and pathophysiological changes in Flavobacterium psychrophilum-resistant and -susceptible rainbow trout lines.

Host genetic resistance against disease-causing pathogens can be enhanced through family-based selective breeding. At present, there is an incomplete understanding of how artificial selection of fish alters host physiology and response following pathogen exposure. We previously reported the generation of selectively-bred rainbow trout Oncorhynchus mykiss lines with either increased resistance (ARS-Fp-R) or susceptibility (ARS-Fp-S) to bacterial cold water disease (BCWD). This study (1) determined baseline reference-range intervals for packed cell volume (PCV) and 18 plasma biochemistry analyte

Identification and Characterization of Flavobacterium psychrophilum as a Causative Pathogen in Farmed Grass Carp Ctenopharyngodon idella in China.

In March 2024, a severe mass mortality event occurred at a grass carp (Ctenopharyngodon idella) farm in Zhejiang, China, resulting in a cumulative mortality rate of 70%. To identify the causative agent, 12 bacterial isolates that exhibited the same phenotypic characteristics were isolated from diseased fish. A further phylogenetic (16S rDNA) characterization identified the bacterium as Flavobacterium psychrophilum. Genetic characterization revealed that all 12 bacterial isolates belonged to serotype Type-1 and sequence type ST418, a newly documented ST closely associated with cyprinid fish hos

Biochemical properties of a Flavobacterium johnsoniae dextranase and its biotechnological potential for Streptococcus mutans biofilm degradation.

Cariogenic biofilms have a matrix rich in exopolysaccharides (EPS), mutans and dextrans, that contribute to caries development. Although several physical and chemical treatments can be employed to remove oral biofilms, those are only partly efficient and use of biofilm-degrading enzymes represents an exciting opportunity to improve the performance of oral hygiene products. In the present study, a member of a glycosyl hydrolase family 66 from Flavobacterium johnsoniae (FjGH66) was heterologously expressed and biochemically characterized. The recombinant FjGH66 showed a hydrolytic activity again

Immune Suppression and Rapid Invasion of Nile Tilapia Gills Following an Acute Challenge by Flavobacterium davisii.

Flavobacterium davisii is one of the causative agents of columnaris disease, significantly impacting Nile tilapia aquaculture. This study examines the invasion and immune evasion mechanisms of a highly virulent F. davisii strain through transcriptomic profiling of tilapia gills following acute immersion. We identified 8192 differentially expressed genes (DEGs) at 2 h, 6 h, and 12 h post-infection. They are enriched in pathways related to oxidative stress, immune suppression, tissue necrosis, and bacterial infection. Notably, early overexpression of rhamnose-binding lectin and mucin genes facil

Palladium Phthalocyanines Varying in Substituents Position for Photodynamic Inactivation of Flavobacterium hydatis as Sensitive and Resistant Species.

Antimicrobial photodynamic therapy (aPDT) has been considered as a promising methodology to fight the multidrug resistance of pathogenic bacteria. The procedure involves a photoactive compound (photosensitizer), the red or near infrared spectrum for its activation, and an oxygen environment. In general, reactive oxygen species are toxic to biomolecules which feature a mechanism of photodynamic action. The present study evaluates two clinical isolates of Gram-negative Flavobacteriumhydatis (F. hydatis): a multidrug resistant (R) and a sensitive (S) strain. Both occur in farmed fish, leading to

A salicylic acid-associated plant-microbe interaction attracts beneficial Flavobacterium sp. to the Arabidopsis thaliana phyllosphere.

Both above- and below-ground parts of plants are constantly challenged with microbes and interact closely with them. Many plant-growth-promoting rhizobacteria, mostly interacting with the plant's root system, enhance the immunity of plants in a process described as induced systemic resistance (ISR). Here, we characterized local induced resistance (IR) triggered by the model PGPR Pseudomonas simiae WCS417r (WCS417) in Arabidopsis thaliana. Hydroponic application of WCS417 to Arabidopsis roots resulted in propagation of WCS417 in/on leaves and the establishment of local IR. WCS417-triggered loca

First detection of Flavobacterium psychrophilum in juvenile Siberian sturgeons (Acipenser baerii) and description of the pathological findings.

Flavobacterium psychrophilum affects many cultured fish species and is considered one of the most important bacterial pathogens causing substantial economic losses in salmonid aquaculture worldwide. Here, F. psychrophilum was identified by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) and nested PCR as the aetiological agent causing mortality in diseased juvenile Siberian sturgeons (Acipenser baerii) reared on a freshwater fish farm. Diseased sturgeons were lethargic and displayed dark skin pigmentation, increased mucus production and the presence

Unravelling mass mortality in transported rohu (Labeo rohita) fingerlings: evidence of Flavobacterium columnare from Western India.

Columnaris disease caused by Flavobacterium columnare in freshwater farmed fishes is a severe problem, leading to considerable mortality. The present study investigated the mass mortality happened in Labeo rohita fingerlings within 48 h post-transportation from an organized fish farm in Maharashtra, India, to the laboratory. The moribund fish showed clinical signs analogous to columnaris and the bacterial isolation showed pale yellow rhizoid edged tightly adhered colonies which are found as slender rod-shaped bacteria by TEM. Molecular analysis of the isolate using 16S rRNA nucleotide homology

Abscisic acid metabolism to dehydrovomifoliol by Candidatus Liberibacter asiaticus and CLas-associated Flavobacterium strains CGRL1 and CGRL2.

INTRODUCTION: Citrus huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas), is a globally devastating plant disease characterized by severe disruption of host physiology. Although alterations in plant hormone homeostasis, including salicylic acid and abscisic acid (ABA), are hallmarks of HLB, the contribution of plant-associated microbiota to these hormonal changes remains poorly understood. Here, we investigated whether ABA depletion in HLB-affected citrus is associated with microbial metabolism. METHODS: ABA and its metabolites were quantified in HLB-affected citrus tissue

Partial purification and characterization of an extracellular cold-active protease from Flavobacterium azizsancarii, a novel Antarctic isolate.

Cold-active enzymes exhibit high catalytic efficiency at low temperatures, making them valuable biocatalysts for energy-efficient and environmentally friendly industrial processes. The enzymatic activity of bacteria that thrive in low-temperature environments has attracted the attention of researchers. In this study, an extracellular cold-active protease produced by Flavobacterium azizsancarii (F. azizsancarii), a novel Antarctic isolate, was partially purified by ammonium sulfate precipitation and dialysis. The enzyme showed maximal activity toward casein at pH 8.0 and 20 °C. The effects of v

First report on Flavobacterium columnare causing disease outbreaks in cultured black carp, Mylopharyngodon piceus, in Northern Vietnam.

Columnaris disease has recently emerged as a concern for black carp (Mylopharyngodon piceus), an economically important aquaculture species in northern Vietnam. This study investigated the disease outbreaks from five affected farms. A total of 47 naturally diseased fish were sampled for bacterial isolation and identification. Five representative isolates resembling Flavobacterium were selected for phenotypic and genetic characterization. Through a combination of biochemical tests, PCR assays, and 16 S rRNA sequencing, these bacterial isolates were identified as Flavobacterium columnare. Experi

Purification and characterisation of 3-hydroxyphenylacetate 6-hydroxylase: a novel FAD-dependent monooxygenase from a Flavobacterium species.

3-Hydroxyphenylacetate 6-hydroxylase was purified 70-fold from a Flavobacterium sp. grown upon phenylacetic acid as its sole carbon and energy source. The presence of FAD and dithiothreitol during purification is essential for high recovery of active enzyme. SDS/PAGE of purified enzyme reveals a single band with a minimum molecular mass of 63 kDa. Analytical gel-filtration, sedimentation-equilibrium and sedimentation-velocity experiments indicate that the purified enzyme exists in solution mainly as a dimer, containing 1 molecule non-covalently bound FAD/subunit. 3-Hydroxyphenylacetate 6-hydro

endo-beta-N-acetylglucosaminidase F: endoglycosidase from Flavobacterium meningosepticum that cleaves both high-mannose and complex glycoproteins.

We have detected an endoglycosidase activity produced by Flavobacterium meningosepticum. This enzyme, named endo F, cleaves glycans of both the high-mannose and the complex type linked through asparagine to the protein backbone. The data indicate that cleavage occurs via hydrolysis of the glycosidic bond of the N,N'-diacetylchitobiose core structure adjacent to asparagine, similar to that due to endo H and endo D. Extreme variability was noted in the availability of this cleavage site among N-linked glycoproteins. Glycoproteins of retrovirus, lymphocytic choriomeningitis virus, Pichinde virus,

The first report of submandibular lymph node infection with Flavobacterium ceti in a horse and its complete genome sequence.

This is the first report describing the isolation and genome sequence of Flavobacterium ceti (F. ceti IJFC025) from a draining submandibular lymph node abscess in a 6-year-old mixed-breed mare in Korea. Since F. ceti is not a usual infectious pathogen that causes equine health problems, this study presents the complete genome sequence of F. ceti isolated from a submandibular lymph node abscess sample in Jeju, South Korea, in 2022. The entire genome sequence of F. ceti IJFC025 consisted of a 3,144,426-bp chromosome with 35.6 % G + C content. Gene prediction revealed that this strain possesses 2

Isolation and characterization of a new plasmid from a Flavobacterium sp. which carries the genes for degradation of 2,4-dichlorophenoxyacetate.

A Flavobacterium sp. (strain 50001), capable of degrading 2,4-dichlorophenoxyacetate (2,4-D), 2-methyl-4-chlorophenoxyacetate, and 2-chlorobenzoate and imparting resistance to mercury, harbored a degradative plasmid, pRC10. Cured strains of the Flavobacterium sp. lost the plasmid as well as the ability to degrade these chlorinated compounds. Comparison of this plasmid with the well-characterized 2,4-D-degradative plasmid pJP4 from Alcaligenes eutrophus showed regions of homology between the two plasmids.
