Rational Design of a "Turn-on" Supramolecular Fluorescent Probe for Gossypol Sensing Based on the Restriction of Intramolecular Bending Mechanism.

The development of ″turn-on″ supramolecular fluorescent probes for detecting biologically important molecules lacking reactive sites, with a clear mechanistic understanding, remains challenging. To address this challenge, this study establishes and validates a rational design strategy based on the restriction of intramolecular bending (RIB) mechanism for developing supramolecular fluorescent probes. The core of this strategy is to activate fluorescence by physically confining a fluorophore, thereby suppressing its nonradiative decay channels. A BODIPY-based fluorophore IPR featuring sterically

An AIEgen-porphyrin co-assembly sensor array for fingerprinting glycosaminoglycans.

BACKGROUND: Although sensor arrays are powerful for discriminating glycosaminoglycans, the construction of a sensor array is usually cost and labour inefficient. With the purpose to acquire a high dimensional response data using only a limited number of dyes, the rational assembly of fluorescent dye molecules may provide an alternative approach. We envision that co-assembling different dyes within a supramolecular structure can trigger multiple photophysical processes, which should produce high dimensional optical signals. RESULTS: A supramolecular AIEgen-porphyrin co-assembly is synthesized

A Water-Soluble Fluorogenic Probe to Quantify Azide Concentrations in Biologically Relevant Environments.

Fluorogenic probes are highly useful tools for the detection of target molecules in chemical biology and (bio)material science, as they can often be applied in situ and do not depend on purification. Over the past years, a variety of alkyne-based fluorogenic probes have been proposed to detect azides, paving the way for live cell imaging and biomaterial analysis. Unfortunately, undesirable photophysical properties and poor solubility in aqueous solutions hamper the widespread use of these fluorogenic compounds. This work describes a water-soluble azide probe with high fluorescence enhancement

A Dual-Channel Fluorescent Probe for Visualizing Glutathione Dynamics in Plants under Abiotic Stress.

Glutathione (GSH), as an important class of antioxidants, exhibits a close correlation with cellular oxidative stress. Usually, GSH serves as an essential biomarker for evaluating redox homeostasis in plants. In this work, we developed a GSH-specific fluorescent probe by employing the aza-IR780 cyanine scaffold as the near-infrared fluorophore, the naphthalimide block as the visible light fluorophore, and sulfonamide as the GSH-responsive unit. The optical performance showed that this probe could highly selectively detect GSH in both visible light (emission at 498 nm) and near-infrared (emissi

Harnessing aggregation-induced emission-active fluorescent probes to illuminate intracellular metal ions.

Metal ions are indispensable components in numerous physiological processes, and their homeostatic imbalance can lead to serious pathological disorders. Therefore, achieving accurate tracking and real-time monitoring of these ions within living systems is crucial for biological research and clinical diagnostics. Aggregation-induced emission luminogens (AIEgens), which exhibit enhanced fluorescence in the aggregated state due to the restriction of intramolecular motions, have become promising tools for metal ion sensing offering superior photostability, high signal-to-noise ratio, and excellent

Fluorogenic probes for intracellular copper detection and the study of cuproptosis.

Copper is an essential trace element involved in key biological processes, including cellular respiration, antioxidant defense, and enzymatic activity. Intracellular copper homeostasis is regulated through coordinated uptake, trafficking, storage, and excretion mechanisms. Disruption of this balance can result in copper overload, leading to cuproptosis-a recently characterized, copper-dependent form of regulated cell death marked by aggregation of lipoylated mitochondrial proteins, disruption of the tricarboxylic acid cycle, and proteotoxic stress. Cuproptosis has been implicated in the pathog

Synthesis and Fluorescence Profiling of Naphthylamine Analogs.

N-phenyl-1-naphthylamine (NPN) has been widely used to probe the integrity of the outer membrane of gram-negative bacteria. NPN fluoresces in the hydrophobic environment of lipid bilayers, whereas its fluorescence is quenched in the aqueous extracellular milieu. Because NPN does not intrinsically accumulate in intact gram-negative bacteria, it has been broadly applied as a reporter of outer membrane permeabilization. Despite its widespread utility, systematic derivatization of this probe has remained largely unexplored. Here, we investigated the structure-fluorescence relationships of NPN thro

Leveraging fluorescent sensor with prominent-response viscosity for evaluating metal-ion stress in plants.

Effectively imaging the variation of heavy metal induce stress (HMIS) in plant is significantly important for stress resistance research in the fields of environmental and plant biology. However, due to the absence of distinctive parameter to reveal the relationship between HMIS and plant homeostasis, the reported fluorescence sensors fail to assess HMIS. Herein, a new fluorescent sensor (quinoline-based viscosity probe, QVP) with prominent-responsive viscosity was first developed for evaluating HMIS in plants. Spectral experiments indicate that QVP exhibited selectivity, sensitive, photochemi

Dicyanoisophorone-based fluorescent probe for detecting H2S and monitoring nitroprusside metabolism.

Hydrogen sulfide (H2S) is an important multifunctional gasotransmitter that plays a pivotal role in regulating various physiological processes. Recent studies have shown that sodium nitroprusside (SNP), a commonly used drug for treating hypertension and angiectasis, will produce H2S during its metabolism. Thus, real-time monitoring of H2S can track the metabolic processes of SNP in vivo. In this study, we designed and synthesized a novel near-infrared (NIR) fluorescent probe, RM, based on the mechanism of excited-state intramolecular proton transfer (ESIPT). The probe features dicyanoisophoron
