Monitoring changes in membrane polarity, membrane integrity, and intracellular ion concentrations in Streptococcus pneumoniae using fluorescent dyes.

Membrane depolarization and ion fluxes are events that have been studied extensively in biological systems due to their ability to profoundly impact cellular functions, including energetics and signal transductions. While both fluorescent and electrophysiological methods, including electrode usage and patch-clamping, have been well developed for measuring these events in eukaryotic cells, methodology for measuring similar events in microorganisms have proven more challenging to develop given their small size in combination with the more complex outer surface of bacteria shielding the membrane.

Green-light transilluminator for the detection without photodamage of proteins and DNA labeled with different fluorescent dyes.

The excitation spectra of Nile red and SYPRO red, two currently used dyes for the fluorescent staining of protein bands in sodium dodecyl sulfate (SDS)-polyacrylamide gels, show an excitation peak in the UV region and another in the visible region (maximum at about 550 nm). Ethidium bromide and other intercalating dyes, e.g. propidium iodide, ethidium dimers, and benzoxazolium-4-quinolinium dimer-3 (YOYO), used for the fluorescent staining of DNA bands in agarose gels also show an excitation peak in the same region of the visible spectrum. We have designed and constructed a green-light transil

Development of Intramolecular Charge Transfer-Type NIR Fluorescent Dyes Bearing a Pyrazino[2,3-b]quinoxaline-Based Electron Acceptor.

Near-infrared (NIR) fluorescent dyes are promising materials for bio-imaging and security applications. However, achieving efficient photoluminescence (PL) over 1000 nm is still challenging due to the energy gap law. Herein, we report new intramolecular charge transfer-type fluorescent dyes 1-H and 1-t-Bu bearing pyrazino[2,3-b]quinoxaline-2,3-dicarbonitrile as an electron acceptor unit. In toluene, these dyes exhibited NIR emission at 947 and 979 nm, with PL quantum yields of 1.04% and 0.12%, respectively. Remarkably, crystallization led to substantial red shifts of their PL spectra to 1076 a

Organotin Schiff bases as halofluorochromic dyes: green synthesis, chemio-photophysical characterization, DFT, and their fluorescent bioimaging in vitro.

Fluorescent bioimaging is an excellent tool in cellular biology, and it will be a powerful technique in modern medicine as a noninvasive imaging technology where tumoral and normal cells must be distinguished. One of the differences between normal and cancer cells is the intracellular pH. Therefore, the design and synthesis of pH-responsive fluorescent materials are required. Organotin Schiff bases showed halofluorochromic behavior in solution. Microwave-assisted synthesis showed better reaction times and chemical yields compared with conventional heating. All compounds were fully characterize

Application of Non-Fluorescent Dyes to Assess the Antischistosomal Effect of Antimalarial Drugs on Schistosoma mansoni Adult Worms.

The possible emergence of praziquantel (PZQ)-tolerant and/or -resistant schistosomes requires the study and development of new antischistosomal drugs as alternatives to PZQ. The present study investigates the capability of 3 dyes-methylene blue (MB), neutral red (NR), and trypan blue (TB)-to assess the in vitro antischistosomal effect of antimalarial drugs on Schistosoma mansoni adult worms. S. mansoni adult worms were incubated in the medium alone as the control or in the medium supplemented with 10 μg/ml primaquine (PQ), artesunate (AR), or amodiaquine (AQ) for 5 days. Lasing in strongly resonant metasurfaces with fluorescent dyes: quasi-dark and bright optical states and the reabsorption effect.

A light-emitting metasurface for highly efficient lasing in the visible is proposed and thoroughly theoretically studied. The metasurface comprises split-square slots etched in a thin TiO2 layer and periodically arrayed in a subdiffractive lattice. The slots are filled with highly fluorescent rhodamine 6G (R6G) molecules in a methanol matrix, providing optically pumped gain under pulsed excitation. Lasing is enabled by a slot-type quasi-bound state in the continuum (qBIC) offering enhanced light-matter interaction. Through a meticulous design process, the lasing threshold is minimized by mitig

Dual-emission fluorescent ZnS nanosheets for sensitive detection of carcinogenic Sudan dyes in chili powder and lipstick samples.

Sudan dyes are carcinogenic colorants illicitly added to foods and cosmetics, posing serious health risks. Herein, a rapid dual-emission fluorescent probe based on two-dimensional (2D) ZnS nanosheets is developed for sensitive detection of Sudan dyes (III/IV). Compared with conventional nanoparticles, the nanosheet structure offers high surface accessibility, excellent dispersion, and reduced aggregation, enabling reliable sensing in complex matrices. The ZnS nanosheets (NS), synthesized via the solvothermal method, exhibit stable fluorescence emissions at 521 and 557 nm with photobleaching re

Dye Sensitized Solar Cells Based on 2-Amino-[(Coumarin-3-yl)-4-(4-(Diphenylamino)Phenyl]nicotinonitrile Fluorescent Dyes.

By introducing novel sensitizer dyes such as ACT, BCT, and SCT, this study seeks to significantly contribute to the field of dye-sensitized solar cells (DSSCs). Modern microwave irradiation has been used to carefully design these dyes. These reaction's outcomes clearly demonstrate how superior this technique is to conventional synthetic approaches. It not only reduces reaction time but also yields better results. A detailed analysis of these novel dyes has demonstrated their superior optical and electrochemical characteristics, which have been validated with the help of Cyclic Voltammetry and

D-π-A type fluorescent dyes: Effect of π-bridge units on optical and G4 DNA binding properties.

Fluorescent solvatochromic dyes that are sensitive to the nature of local microenvironmental, have been explored as probes in applications ranging from the imaging biomolecules to understanding of basic biomolecule functions. To expand the scope of fluorescent solvatochromic dyes for G-quadruplex (G4) DNA structures, and to illustrate the relationship between structure and properties, three newly designed D-π-A type fluorescent dyes were synthesized by introducing diarylimidazole to carbazole skeleton linked to benzene, furan or thiophene π-conjugated bridge and connected with pyridinium accep

Synthesis and Application of Two-Photon Active Fluorescent Rhodol Dyes for Antibody Conjugation and In Vitro Cell Imaging.

A novel family of julolidine-containing fluorescent rhodols equipped with a wide variety of substituents was synthesized in a versatile two-step process. The prepared compounds were fully characterized and exhibited excellent fluorescence properties for microscopy imaging. The best candidate was conjugated to the therapeutic antibody trastuzumab through a copper-free strain-promoted azide-alkyne click reaction. The rhodol-labeled antibody was successfully applied for in vitro confocal and two-photon microscopy imaging of Her2+ cells.

Radical C-H arylation of the BODIPY core with aryldiazonium salts: synthesis of highly fluorescent red-shifted dyes.

We describe herein the first radical C-H arylation of BODIPY dyes. This novel, general, one-step synthetic procedure uses ferrocene to generate aryl radical species from aryldiazonium salts and allows the straightforward synthesis of brightly fluorescent (Φ>0.85) 3,5-diarylated and 3-monoarylated boron dipyrrins in up to 86 % yield for a broad range of aryl substituents. In this way, new and complex dyes with red-shifted spectra can be easily prepared.

Synthesis, spectroscopic and DFT studies of novel fluorescent dyes: 3-aminoimidazo[1,2-a]pyridines possessing 4-pyrone moieties.

A series of novel imidazo[1,2-a]pyridines possessing 4-pyrone ring were synthesized by three-component condensation of 4-pyrone carbaldehydes, 2-aminopyridines and isocyanides. Bismuth (III) chloride was used as a catalyst in these reactions and desired products were synthesized in good yields at a very short period of time under solvent free conditions. UV-Vis absorption and fluorescence emission spectra of these compounds were investigated. It shown that two of these compounds (10f and 10g) exhibit intense fluorescence in dichloromethane. Optimized ground-state molecular geometries and orbit

New and improved methods for measuring lymphocyte proliferation in vitro and in vivo using CFSE-like fluorescent dyes.

The use of carboxyfluorescein diacetate succinimidyl ester (CFSE) to measure lymphocyte proliferation by flow cytometry has become one of the most widely utilised assays for assessing lymphocyte responses. The properties of CFSE make it ideal for such a task, covalently labelling cells with a long-lived fluorescence of high intensity and low variance with minimal cell toxicity. No dye in the last 20 years has been capable of replicating CFSE in these respects. However, currently CFSE is limited to following a maximum of 7 cell divisions and is not compatible for use with ubiquitously available

Analysis of tear protein patterns of dry-eye patients using fluorescent staining dyes and two-dimensional quantification algorithms.

Tear proteins of nonstimulated tears of 29 patients (healthy subjects, n = 8; dry-eye syndrome patients, n = 12; diabetic dry-eye patients, n = 9) were electrophoretically separated and stained by SYPRO Orange, followed by Coomassie blue staining. Both, the fluorescent and the Coomassie stains were subsequently analyzed by an automated two-dimensional algorithm for finding and quantification of peaks and by a discriminant analysis. Using SYPRO Orange, an average number of peaks/sample between three (at 200 ms) and 15 (at 3000 ms) could be found. In comparison, Coomassie staining resulted only

From quantitative modeling of fluorescence experiments on biomolecules to the prediction of spectroscopic dye properties.

Fluorescence spectroscopy and modeling provide powerful means to characterize biomacromolecular structures, dynamics, and interactions. Förster resonance energy transfer serves as a key technique for this because of its nanometer scale distance sensitivity. Quantitative interpretation of fluorescence data relies on models that link molecular structure to observable spectroscopic quantities and vice versa. Integrative modeling frameworks combine fluorescence observables with complementary structural information to infer molecular structures and conformational ensembles. This review outlines con

Intrathecal Application of a Fluorescent Dye for the Identification of Cerebrospinal Fluid Leaks in Cochlear Malformation.

In cases of cerebrospinal fluid (CSF) leaks, reliable detection of their origins is needed to seal the leak sufficiently and prevent complications, such as meningitis. A method is presented here using intrathecal administered fluorescein in a clinical case of bilateral congenital ear malformation. A fluorescent dye is administered intrathecally to achieve intraoperative visualization of CSF leaks. The dye is applied 20 min before surgery, and concentration of 5% is used. Per every 10 kg of body weight, 0.1 mL of the fluid is applied intrathecally. The fluorescein is visualized using a fully di

Biological applications of near infrared fluorescence dye probes in monitoring Alzheimer's disease.

Alzheimer's disease (AD) is characterized by progressive decrease in cognitive abilities, language impairment and irreversible memory loss. Amyloid-β (Aβ) and tau deposits are essential as the major factors involved in the pathogenesis of AD. Unfortunately, anti-Alzheimer's disease agents currently available are not potent enough to reverse the cause of the disease. Interestingly, near infrared fluorescence (NIRF) dyes have been marked as promising tools in analytical researches and are often adopted as molecular probes to monitor and diagnose AD, in vitro or in vivo. Compared with other imagi
