Pharmacokinetic interactions of fruit juices with antihypertensive drugs in humans: A systematic review and meta-analysis.

INTRODUCTION: Fruit juice-antihypertensive drug interactions can lead to subtherapeutic or supratherapeutic outcomes. This systematic review and meta-analysis assesses such interactions and their potential clinical relevance. METHODS: PubMed, Scopus, and Science Direct databases were searched from their inception through June 2024. Eligible studies were those that investigated the effects of fruit juice on the pharmacokinetics of antihypertensive drugs. I2 was used to determine heterogeneity among studies, and a random effect model was employed for meta-analysis. This review adhered to PRISMA

A clinician's guide to drug-drug interactions with direct-acting antiviral agents for the treatment of hepatitis C viral infection.

UNLABELLED: The US Food and Drug Administration has recently approved a number of new direct-acting antiviral agents for the treatment of chronic hepatitis C virus that have significantly increased the likelihood of a virological cure. These agents are highly effective but present a substantial risk for a host of clinically relevant drug-drug interactions. These interactions must be considered both when starting and stopping any medication, including over-the-counter medications and herbal supplements. These drug-drug interactions can increase the risk of toxicity or decrease the likelihood of

[Notable Adverse Events Associated with Concomitant Use of Health Foods and Drugs Derived from the Analysis of HFNet Data on the Safety and Effectiveness of Health Foods].

Health foods are commonly consumed at their own discretion by patients with various diseases who are also being treated with conventional drugs. Both health foods and drugs are diverse, and enormous numbers of possible combinations exist, so that it is very difficult to identify adverse events that may occur due to their interactions. Here, we analyzed the characteristics of adverse events related to the concomitant use of health foods and drugs using data from the "Information system on safety and effectiveness for health foods (HFNet)" website (https://hfnet.nibiohn.go.jp/) compiled by the F

An update on drug-drug interactions with biologics for the treatment of moderate-to-severe psoriasis.

OBJECTIVE: Moderate-to-severe psoriasis is a chronic skin condition that often requires systemic therapy and biologics are the newest systemic treatment available. A favorable aspect of biologics is that they are thought to have minimal risks for drug-drug interactions compared to oral systemic medications such as cyclosporine and methotrexate. However, this assumption has not been recently or adequately reviewed. We reviewed the literature to identify possible drug-drug interactions with biologics and other medications. METHODS: We searched PubMed for published case reports, clinical studies

Drug-Drug Interactions in the Management of Patients With Pulmonary Arterial Hypertension.

The management of pulmonary arterial hypertension (PAH) has become more complex in recent years because of increased pharmacotherapy options and longer patient survival with increasing numbers of comorbidities. As such, more opportunities exist for drug-drug interactions between PAH-targeted medications and medications potentially used to treat comorbid conditions. In this review, we provide an overview of pharmaceutical metabolism by cytochrome P450 and discuss important drug-drug interactions for the 14 Food and Drug Administration-approved medications for PAH in the nitric oxide (NO), endot

[Drug interactions as a cause of drug side effects].

Adverse drug interactions are a frequent clinical problem. Mechanistically, they can be classified as pharmaceutic, pharmacokinetic and pharmacodynamic interactions. Mostly, however, several different mechanisms are involved in the pathogenesis of adverse drug interactions, which makes their predictability as well as their prevention strongly dependent on the early and adequate recognition of high-risk patients. Such high-risk situations include drug-related (e.g. galenic form, stability, enantiomers), patient-related (e.g. pharmacokinetics, age, genetic disposition) and various exogenous (e.g

Food Bioactive Compounds and Their Interference in Drug Pharmacokinetic/Pharmacodynamic Profiles.

Preclinical and clinical studies suggest that many food molecules could interact with drug transporters and metabolizing enzymes through different mechanisms, which are predictive of what would be observed clinically. Given the recent incorporation of dietary modifications or supplements in traditional medicine, an increase in potential food-drug interactions has also appeared. The objective of this article is to review data regarding the influence of food on drug efficacy. Data from Google Scholar, PubMed, and Scopus databases was reviewed for publications on pharmaceutical, pharmacokinetic,

Concomitant therapy in people with epilepsy: potential drug-drug interactions and patient awareness.

People with epilepsy (PWE) may use prescription and over-the-counter (OTC) drugs for the treatment of concomitant diseases. Combinations of these drugs, as well as dietary supplements, with antiepileptic drugs (AEDs) may lead to reduced control of seizures and of coexisting medical conditions and increased risk of adverse drug reactions (ADRs). The aims of this study were to obtain comprehensive lists of medications, dietary supplements, botanicals, and specific food components used by adult PWE and to evaluate the potential for interactions involving AEDs and patients' awareness of such poten

[Valproic acid and protein supplements: possible clinically relevant drug interaction].

Introduction: valproic acid (VPA) is a drug with narrow therapeutic index (50-100 µg/mL) with a high pharmacokinetic variability due to plasma protein binding saturation (PPBU). Case report: we present a clinical case of a 60-year-old patient under treatment with VPA, whose plasma VPA concentrations were analyzed and a decrease in these concentrations was detected when administrated with protein supplement. When the intake was spaced out, VPA serum concentrations returned to the therapeutic range. Discussion: administration with food does not appear to be important according to the majority of

Statin drug interactions and related adverse reactions.

INTRODUCTION: Statin monotherapy is generally well tolerated, with a low frequency of adverse events. The most important adverse effects associated with statins are myopathy and an asymptomatic increase in hepatic transaminases, both of which occur infrequently. Because statins are prescribed on a long-term basis, their possible interactions with other drugs deserve particular attention, as many patients will typically receive pharmacological therapy for concomitant conditions during the course of statin treatment. AREAS COVERED: This review summarizes the pharmacokinetic properties of statin

Transdermal selegiline.

Although older monoamine oxidase inhibitors (MAOIs) are effective in the treatment of depressive disorders, they are underutilized in clinical practice due to main concerns about interaction with tyramine-containing food, and side effects. Efforts to address these safety issues led to the development of a transdermal formulation of selegiline, called selegiline transdermal system (STS). STS has been approved by the U.S. Food and Drug Administration (FDA) for the treatment of major depression. Transdermal administration of selegiline bypasses gastrointestinal absorption and first-pass metabolis

Drug interactions involving antiepileptic drugs: assessment of the consistency among three drug compendia and FDA-approved labels.

Interactions of antiepileptic drugs (AEDs) with other substances may lead to adverse effects and treatment failure. To avoid such interactions, clinicians often rely on drug interaction compendia. Our objective was to compare the concordance for twenty-two AEDs among three drug interaction compendia (Micromedex, Lexi-Interact, and Clinical Pharmacology) and the US Food and Drug Administration-approved product labels. For each AED, the overall concordance among data sources regarding existence of interactions and their classification was poor, with less than twenty percent of interactions liste

Diet and drug interactions in the treatment of mental illness--a review.

Drug-diet interactions in the treatment of mental illness are reviewed from two perspectives: the potential effects of drugs on a person's nutritional status, and the effect of diet on drug action through modification of its absorption or various stages of its metabolism. As mental illness is often chronic and recurrent, requiring long-term drug therapy, it is important to become aware of the effects of drugs and their possible interactions with other important factors such as food.

Factors contributing to variability in drug pharmacokinetics. I. Absorption.

Apart from the physical and chemical properties of a drug, and also the dosage form in which it is presented, many other factors may affect the absorption of orally administered compounds and give rise to variable systemic availability. Three such factors, gastrointestinal (GI) disease, drug-drug interactions, and drug-food interactions are considered here. Although information regarding these factors, particularly GI disease, is scarce and sometimes conflicting, it is clear that they may give rise to variable drug absorption. The wide spectrum of effects on different drugs precludes the appli
