Fibroblast growth factor 23 (FGF23) is produced by bone cells and regulates renal phosphate and vitamin D metabolism, as well as causing left ventricular hypertrophy. FGF23 deficiency results in rapid aging, whereas high plasma FGF23 levels are found in several disorders, including kidney or cardiovascular diseases. Regulators of FGF23 production include parathyroid hormone (PTH), calcitriol, dietary phosphate, and inflammation. We report that insulin and insulin-like growth factor 1 (IGF1) are negative regulators of FGF23 production. In UMR106 osteoblast-like cells, insulin and IGF1 down-regu

Oxidative damage-triggered ferroptosis and apoptosis drive the progression of Parkinson's disease (PD). Forkhead box O1 (FOXO1) is a critical modulator of oxidative stress, while its role in PD remains largely unexplored. Here, the FOXO1 inhibitor AS1842856 (AS) was employed to treat the MPTP-exposed mice and MPP+-challenged SH-SY5Y cells. MPTP/MPP+ markedly upregulated FOXO1 expression and enhanced its transcriptional activity, effects that were significantly abrogated by AS treatment. Importantly, AS treatment rescued the MPTP/MPP+-induced downregulations of sterol regulatory element-binding

Ovarian endometriosis is characterized by the growth of endometrial tissue within the ovary, causing infertility and chronic pain. However, its pathophysiology remains unclear. Utilizing high-precision single-cell RNA sequencing, we profile the normal, eutopic, and ectopic endometrium from 34 individuals across proliferative and secretory phases. We observe an increased proportion of ciliated cells in both eutopic and ectopic endometrium, characterized by a diminished expression of estrogen sulfotransferase, which likely confers apoptosis resistance. After translocating to ectopic lesions, end

Mechanical stimuli including loading after birth promote bone growth. However, little is known about how mechanical force triggers biochemical signals to regulate bone growth. Here, we identified a periosteal-osteoblast-derived secretory peptide, Osteocrin (OSTN), as a mechanotransducer involved in load-induced long bone growth. OSTN produced by periosteal osteoblasts regulates growth plate growth by enhancing C-type natriuretic peptide (CNP)-dependent proliferation and maturation of chondrocytes, leading to elongation of long bones. Additionally, OSTN cooperates with CNP to regulate bone form

Modern lifestyles have altered diet and metabolic homeostasis, with increased sugar intake, glycemic index, and prediabetes. A strong positive correlation between sugar consumption and diabetic incidence is revealed, but the underlying mechanisms remain obscure. Here we show that oral intake of long-term oscillating glucose (LOsG) (4 times/day) for 38 days, which produces physiological glycemic variability in rats, can lead to β-cells gaining metabolic memory in reactive oxygen species (ROS) stress. This stress leads to suppression of forkhead box O1 (FoxO1) signaling and subsequent upregulati

Periodontitis is a chronic disease that includes the pathologic loss of periodontal tissue and alveolar bone. The inflammatory environment in periodontitis impairs the osteogenic differentiation potential and depresses the regeneration capacity of human periodontal ligament stem cells (hPDLSCs). Since Forkhead box protein O1 (FoxO1) plays an important role in redox balance and bone formation, we investigated the role of FoxO1 in oxidative stress resistance and osteogenic differentiation in an inflammatory environment by overexpressing FoxO1 in hPDLSCs. First, we found that FoxO1 overexpression

Canine osteosarcoma (OS) presents a significant clinical challenge in veterinary oncology. Due to the similarities in aggressive biologic behavior, mutation status, and gene expression profiles, the canine patient also provides a spontaneous animal model for OS in humans. Advancements in the treatment of OS have been slow to progress. The phosphatidylinositol 3-kinase (PI3K), AKT serine/threonine kinase (AKT), and mechanistic target of rapamycin (mTOR) signal transduction pathway is implicated in canine and human OS, and presents a potentially valuable therapeutic target. The present study inv

SCIENTIFIC BACKGROUND: Rational control of rodent populations is crucial for maintaining ecosystem balance and mitigating agricultural economic losses. Follicle development plays a pivotal role in determining animal population abundance, and photoperiod serves as the primary environmental cue affecting this process. Investigating the mechanisms through which photoperiod influences follicle development in the striped hamster (Cricetulus barabensis) offers a promising molecular target for the effective and sustainable management of rodent populations.
METHODOLOGY: This study employed hematoxyli

The expression of the forkhead box O (FoxO) transcription factor, FoxO, in the skeletal muscle is involved in muscle atrophy caused by disuse, fasting, diabetes, and cachexia. Since inhibition of FoxO activity has been shown to be effective in preventing muscle atrophy in genetically engineered animals, inhibition of FoxO activity by dietary components may contribute to the prevention of muscle atrophy. In this study, 4,006 plant extracts were evaluated for FoxO1 and FoxO3a inhibitory activity using a reporter gene assay system, and the extracts from Cephalotaxus harringtonia showed potent inh

The forkhead box O1 (FoxO1), the first discovered member of the FoxO family, is a critical transcription factor predominantly found in insulin-secreting and insulin-sensitive tissues. In the pancreas of adults, FoxO1 expression is restricted to islet β cells. We determined that in human islet microarray datasets, FoxO1 expression is higher than other FoxO transcription factors. Our analyses of three human islet datasets revealed that FoxO1 expression tends to shows a negative correlation with type 2 diabetes and no correlation with body mass index (BMI) between individuals with low levels of H

The liver is a key metabolic organ that maintains whole-body nutrient homeostasis. Aging-induced liver function alterations contribute to systemic susceptibility to aging-related diseases. However, the molecular mechanisms of liver aging remain insufficiently understood. In this study, we performed bulk RNA-Seq and single-cell RNA-Seq analyses to investigate the underlying mechanisms of the aging-induced liver function changes. We found that liver inflammation, glucose intolerance, and liver fat deposition were aggravated in old mice. Aging significantly increased pro-inflammation in hepatic m

This study aimed to analyse the expression of Sirt1/FoxO1 pathway in the placenta of patients with preeclampsia (PE). Clinical data of 111 PE patients were retrospectively analysed and divided into mild group (n = 61) and severe group (n = 50) according to the severity of condition. Another 45 healthy mothers were selected as healthy group. The value of Sirt1/FoxO1 pathway-related proteins in predicting prognosis of PE patients was analysed. The severe group had higher Sirt1 and lower FoxO1 protein expressions than the mild and healthy groups (p < 0.05). Sirt1 protein expression was positively

Sirtuin1 (Sirt1)/forkhead box O1 (FoxO1) axis has been reported as a crucial regulator involved in chondral homeostasis of healthy or osteoarthritis (OA) cartilage. In our study, the aim is to investigate whether dioscin functions as an activator of Sirt1/FoxO1 to protect against mechanical stress-induced chondrocyte dysfunction in vitro and in vivo models. HERB and PubChem databases were implemented to predict dioscin-related gene targets. Cell and mouse models of OA were established to determine the pharmacological value of dioscin, a steroidal saponin. Cartilage loss in the knee joint was d

Oxidative stress (OS) is involved in various reproductive diseases and can induce autophagy and apoptosis, which determine the different fates of cells. However, the sequence and the switch mechanism between autophagy and apoptosis are unclear. Here, we reported that chronic restraint stress (CRS) induced OS (decreased T-AOC, T-SOD, CAT and GSH-Px and increased MDA) and then disturbed the endocrine environment of sows during early pregnancy, including the hypothalamic-pituitary-ovarian (HPO) and the hypothalamic-pituitary-adrenal (HPA) axes. Meanwhile, after CRS, the KEAP1/NRF2 pathway was inh

Basal-like breast cancer (BBC) and glioblastoma multiforme (GBM) are aggressive cancers associated with poor prognosis. BBC and GBM have stem cell-like gene expression signatures, which are in part driven by forkhead box O (FOXO) transcription factors. To gain further insight into the impact of FOXO1 in BBC, we treated BT549 cells with AS1842856 and performed RNA sequencing. AS1842856 binds to unphosphorylated FOXO1 and inhibits its ability to directly bind to DNA. Gene Set Enrichment Analysis indicated that a set of WNT pathway target genes, including lymphoid enhancer-binding factor 1 (LEF1)

BACKGROUND: Exosomes derived from cardiac microvascular endothelial cells (CMECs) under hypoxia can mediate cardiac repair functions and alleviate pyroptosis and oxidative stress during ischemia-reperfusion (I/R) injury. This study is aimed at investigating the effect and mechanism of miR-27b-3p underlying hypoxic CMECs-derived exosomes against I/R injury.
METHODS: CMECs were isolated from the left ventricle of Sprague-Dawley rats, followed by culturing under hypoxic conditions or pretreatment with the miR-27b-3p inhibitor. CMECs-derived exosomes were added into H9C2 cells before hypoxia/reox

Forkhead box class O family member proteins (FoxOs) are evolutionarily conserved transcription factors for their highly conserved DNA-binding domain. In mammalian species, all the four FoxO members, FoxO1, FoxO3, FoxO4, and FoxO6, are expressed in different organs. In bone, the first three members are extensively expressed and more studied. Bone development, remodeling, and homeostasis are all regulated by multiple cell lineages, including osteoprogenitor cells, chondrocytes, osteoblasts, osteocytes, osteoclast progenitors, osteoclasts, and the intercellular signaling among these bone cells. Long-term supplementation with branched-chain amino acids (BCAA) is associated with prolonged survival and decreased frequency of development of hepatocellular carcinoma (HCC) in patients with liver cirrhosis. However, the pharmaceutical mechanism underlying this association is still unclear. We investigated whether continuous BCAA supplementation increases survival rate of rats exposed to a fibrogenic agent and influences the iron accumulation, oxidative stress, fibrosis, and gluconeogenesis in the liver. Further, the effects of BCAA on gluconeogenesis in cultured cells were also investigated

Increased glycolytic metabolism is a key step in the reproduction of Brucella and the induction of brucellosis, however, little is known about how this process is regulated during infection. Forkhead box protein O1 (FOXO1) is a transcription factor that regulates energy metabolism. In this study, we employ the yeast two-hybrid system (Y2H) and immunoprecipitation (Co-IP) to reverse screen for the FOXO1 for the first time and identify interactions between FOXO1 and the Brucella secretory protein VceA. Our findings reveal that the Brucella secretory protein VceA colocalizes with FOXO1 in the cyt

BACKGROUND/OBJECTIVES: The proliferation of appearance-centered values on social media has driven non-obese adolescents towards increasingly extreme diets and exercise programs to achieve weight loss. Despite this, the effects of concurrent diets and exercise on musculoskeletal development during adolescence are unclear. This study examined whether prolonged endurance exercise (EX) with intermittent fasting during adolescence adversely affects musculoskeletal growth.
MATERIALS/METHODS: Eight-week-old male Sprague-Dawley rats were assigned to 1 of 4 groups: sedentary (SED), intermittent fastin
