Preparation, characterization, antioxidant activity evaluation and application of high-thickening instant Tremella fuciformis polysaccharide products under the IDDSI framework.

Tremella fuciformis polysaccharides (TP), a natural polysaccharide with dual medicinal and edible values, possesses excellent gelling properties, water-holding capacity, and biological activities, rendering it a promising thickening agent for dysphagia-friendly foods. However, its poor solubility and tendency to agglomerate severely limit its practical applications. To overcome this limitation, the present study developed two optimized processing techniques: fluffy tablets prepared by heating at 95 °C (named TPS1, where S stands for fluffy tablets) and tablets treated with a single cycle of hi

The use of β-glucans and other polysaccharides from edible fungi for improving obesity: A review.

Obesity, primarily driven by long-term high-calorie diets, has become a major global public health issue and a key contributor to metabolic diseases. Current obesity treatments often have significant side effects and socioeconomic burdens. In contrast, bioactive β-glucans and other polysaccharides from edible fungi show obesity prevention effects with few adverse effects, making them potential candidates for obesity prevention and treatment. Although many studies have explained obesity development mechanisms, primarily through gut microbiota and intestinal barrier modulation. Moreover, several

The Gut Microbiota as a Mediator of the Metabolic Benefits of Fungi and Their Polysaccharides: A Review of Evidence Addressing Causality.

Prebiotic supplementation has emerged as a promising strategy to ameliorate metabolic disorders through beneficial reshaping of the gut microbiota. Accumulating evidence suggests that fungi and their polysaccharides (e.g., those from Auricularia auricula-judae, Boletus edulis, Hirsutella sinensis, Isaria cicadae, Ramaria botrytoides, and Wolfiporia cocos) possess considerable potential in this context. The present review summarizes studies providing data relevant to the potential involvement of the gut microbiota in mediating the metabolic benefits of fungal interventions. Observed outcomes, p

Structural characterization and antioxidant activity evaluation of a polysaccharide from pink Auricularia cornea.

An acidic polysaccharose (YL-D2N2) was isolated from crude polysaccharides of pink Auricularia cornea and characterized for its structural and antioxidant properties. YL-D2N2 consists of fucose, galactose, glucose, xylose, mannose and glucuronic acid in a molar ratio of 0.85: 1.50: 4.44: 27.52: 46.56: 19.13. It has a number-average molecular weight of about 52.811 kDa and a weight-average molecular weight of about 135.457 kDa. Structural characterization showed that YL-D2N2 consists of nine residues (Xylp-(1→, GlcpA-(1→, →2)-Xylp-(1→, →3)-Galp-(1→, →3)-Manp-(1→, →4)-GlcpA-(1→, →2,3)-Manp-(1→,

The Cell Wall-Associated Proteins in the Dimorphic Pathogenic Species of Paracoccidioides.

Paracoccidioides brasiliensis and P. lutzii cause human paracoccidioidomycosis (PCM). They are dimorphic ascomycetes that grow as filaments at mild temperatures up to 28oC and as multibudding pathogenic yeast cells at 37oC. Components of the fungal cell wall have an important role in the interaction with the host because they compose the cell outermost layer. The Paracoccidioides cell wall is composed mainly of polysaccharides, but it also contains proportionally smaller rates of proteins, lipids, and melanin. The polysaccharide cell wall composition and structure of Paracoccidioides yeast cel

The environmental yeast Cryptococcus liquefaciens produces capsular and secreted polysaccharides with similar pathogenic properties to those of C. neoformans.

Invasive fungal infections, including cryptococcosis, are a growing threat to immunocompromised patients. Although Cryptococcus neoformans and Cryptococcus gattii are the main agents of human cryptococcosis, opportunistic infections by environmental species, such as C. liquefaciens, have been observed recently. The main Cryptococcus virulence factor is the production and secretion of polysaccharides (PS). Previously, we showed that both species produce PS of similar composition. Here, we examined the ultrastructure and biological activity of capsular and secreted PS from C. liquefaciens, and y

Polysaccharides from bamboo-associated edible and medicinal fungi: Structural features, bioactivities, and evidence for structure-activity relationships.

Polysaccharides from edible and medicinal fungi have been widely investigated, but the relationships among fungal ecological context, polysaccharide characteristics, and structure-activity associations (SARs) remain insufficiently resolved. This review examines bamboo-associated edible and medicinal fungi (BEMF), defined as fungi associated with bamboo, bamboo-derived substrates, or bamboo-forest habitats. We summarize reported polysaccharide characteristics, including molecular weight, monosaccharide composition, chemical modifications, glycosidic linkages, branching, conformation, and biolog

Lonicera caerulea polysaccharide alleviates alcoholic liver disease in mice associated with modulation of the gut bacteriome and mycobiome.

Lonicera caerulea is a commercial fruit crop. A polysaccharide (LCP) was extracted from its fruits and characterized as a pyranose polysaccharide with a molecular weight of 4.096 × 105 Da. Monosaccharide composition analysis revealed that LCP was a heteropolysaccharide composed of glucose, mannose, xylose, glucuronic acid, galacturonic acid, rhamnose, and galactose. In a mouse model of acute alcoholic liver injury, oral administration of LCP for 14 days was associated with dose-dependent improvements in serum transaminases, lipid accumulation, oxidative stress markers, and pro-inflammatory cyt

A Review on Antifungal Efficiency of Plant Extracts Entrenched Polysaccharide-Based Nanohydrogels.

Human skin acts as a physical barrier; however, sometimes the skin gets infected by fungi, which becomes more severe if the infection occurs on the third layer of the skin. Azole derivative-based antifungal creams, liquids, or sprays are available to treat fungal infections; however, these formulations show various side effects on the application site. Over the past few years, herbal extracts and various essential oils have shown effective antifungal activity. Additionally, autoxidation and epimerization are significant problems with the direct use of herbal extracts. Hence, to overcome these

Immunological down-regulation of host defenses in fungal infections.

Fungal pathogens use multiple virulence factors to cause progressive disease. A mechanism that could be regarded as a virulence factor is the fungal pathogen's ability to evade or down-regulate host protective mechanisms. Cryptococcus neoformans is an excellent example of a fungal pathogen that can down-regulate both innate and immune host protective mechanisms. Cr. neoformans is a basidiomycetous yeast-like organism that causes cryptococcosis, a frequently fatal disease in man. This organism produces a capsule that inhibits phagocytosis, and the excess capsular material sloughs off and gets i

Functional variation among LPMOs revealed by the inhibitory effects of cyanide and buffer ions.

Enzymes known as lytic polysaccharide monooxygenases (LPMOs) are mono-copper polysaccharide-degrading peroxygenases that engage in several on- and off-pathway redox reactions involving O2 and H2O2. Herein, we show that the known metalloenzyme inhibitor cyanide inhibits reductive activation of LPMOs by binding to the LPMO-Cu(II) state and that the degree of inhibition depends on the concentrations of the polysaccharide substrate, the reductant and H2O2. Importantly, this analysis revealed differences between fungal NcAA9C and bacterial SmAA10A, which have different secondary copper coordination

Medicinal Mushrooms: Past, Present and Future.

The survival of Homo sapiens is continually under threat from agencies capable of inflicting calamitous damage to the overall health and well-being of humankind. One strategy aimed at combatting this threat is focused on medicinal mushrooms and derivatives thereof. Mushrooms themselves have been consumed as part of the human diet for centuries, whereas 'mushroom nutriceuticals' is a more recently adopted term describing mushroom-derived products taken as dietary supplements to enhance general health and fitness. Among the most extensively studied pharmacologically active components of mushroom

Characterization of Carbohydrate Specificity of Monoclonal Antibodies to Fungal Antigenic Markers Using Biotinylated Oligosaccharides as Coating Antigens.

Mannan and β-(1→3)-glucan are two polysaccharide markers that are characteristic for a number of fungal pathogens, including Candida albicans, which is the most common cause of invasive mycoses in humans. In this study, we examined epitope specificity of two monoclonal antibodies, CM532 and FG70, which recognize certain oligosaccharide fragments of these fungal polysaccharides. Using a panel of biotinylated oligosaccharides as coating antigens, we found that the CM532 antibody obtained by immunization with the pentamannoside β-Man-(1→2)-β-Man-(1→2)-α-Man-(1→2)-α-Man-(1→2)-α-Man KLH conjugate,

Amphotericin B-loaded polyphenolic nanoparticles for synchronous fungal eradication and immunomodulation in treating superficial fungal infections.

Fungal infection represents an escalating global health threat, yet effective treatment remains clinically challenging due to the poor solubility, limited bioavailability, and pronounced cytotoxicity of conventional antifungal agents. To overcome these limitations, a self-assembled nanoparticle (ATE NPs) is developed by integrating amphotericin B (AmB), epigallocatechin gallate (EGCG), and tri(phenylboronic acid)-derived crosslinker pentamethyldiethylenetriamine (T-PBA) for efficient fungal eradication and accelerated tissue repair. Benefiting from the strong affinity of T-PBA towards fungal c

Micro- to nano-scale chemical and mechanical mapping of antimicrobial-resistant fungal biofilms.

A fungal biofilm refers to the agglomeration of fungal cells surrounded by a polymeric extracellular matrix (ECM). The ECM is composed primarily of polysaccharides that facilitate strong surface adhesion, proliferation, and cellular protection from the surrounding environment. Biofilms represent the majority of known microbial communities, are ubiquitous, and are found on a multitude of natural and synthetic surfaces. The compositions, and in-turn nanomechanical properties, of fungal biofilms remain poorly understood, because these systems are complex, composed of anisotropic cellular and extr

Natural Polysaccharides as Preventive and Therapeutic Horizon for Neurodegenerative Diseases.

Neurodegenerative diseases are a serious and widespread global public health burden amongst aging populations. The total estimated worldwide global cost of dementia was US$818 billion in 2015 and has been projected to rise to 2 trillion US$ by 2030. While advances have been made to understand different neurodegenerative disease mechanisms, effective therapeutic strategies do not generally exist. Several drugs have been proposed in the last two decades for the treatment of different types of neurodegenerative diseases, with little therapeutic benefit, and often with severe adverse and side effe

Preliminary Structural Characterization and Bioactivity of Exopolysaccharides Produced by Sanghuangporus vaninii (Agaricomycetes).

Sanghuangporus vaninii produces bioactive polysaccharides with therapeutic potential, but the structural features and anti-obesity mechanisms of its exopolysaccharides (EPS) remain unclear. Two polysaccharide fractions (Fr-I and Fr-II) have been purified from the crude EPS of S. vaninii via gel filtration chromatography. The structural characterization of the two fractions was comprehensively characterized at multiple levels, employing SEC/MALLS, GC and FT-IR. SEC/MALLS indicated their average molecular weights (Mw) have been 3.162 ȕ 106 (Fr-I) and 2.613 ȕ 106 (Fr-II). Both fractions exhibited
