Protective effects of Demethylfuropinnarin on porcine pre-implantation embryos under Tunicamycin-induced oxidative stress and endoplasmic reticulum stress during in vitro culture.

Assisted Reproductive Technologies (ART) are essential for addressing infertility and improving reproductive outcomes in both humans and animals. In vitro culture (IVC) supports embryo development outside the body but is often compromised by oxidative and endoplasmic reticulum (ER) stress, leading to impaired development and apoptosis. Demethylfuropinnarin (DMFP), a furocoumarin from Notopterygium incisum, has not been previously studied for its biological activity. Given the antioxidant properties of furocoumarins, this study aimed to determine whether DMFP could alleviate tunicamycin (TM)-in

Phototoxic dermatitis due to Chenopodium album in a mother and son.

Chenopodium album L. subs. album (Chenopodiaceae) is an annual herb with fibrous roots. The plant grows worldwide and frequently in moist areas. Sometimes, the young parts of this plant can be cooked and eaten as a vegetable. In this article, we report a mother and her adult son, in whom phototoxic reaction developed on the sun-exposed body areas after eating this plant of Chenopodiaceae family because of rare presentation. We thought that this reaction was probably due to furocoumarins constituent within the plant.

Recurrent Bilateral Lime Disease in a Young Female- Case Report.

INTRODUCTION: Lime phytodermatitis, also known as margarita dermatitis, is a condition that results in a skin rash after sunlight exposure when handling certain plants. Misdiagnosis is common due to its resemblance to skin burns or allergic contact dermatitis. Detailed history and disease recognition is important to provide accurate treatment recommendations. CASE REPORT: A 32-year-old woman presented with a recurrent rash on her hands that would only occur in the summer months. She was previously misdiagnosed as allergic contact dermatitis. History revealed yearly vacations involving margari

Ruta angustifolia Pers. (Narrow-Leaved Fringed Rue): Pharmacological Properties and Phytochemical Profile.

The genus Ruta in the family Rutaceae includes about 40 species, such as the well-known plants R. graveolens L. (common rue) or R. chalepensis L. (fringed rue), but also much lesser-known species such as R. angustifolia Pers. (narrow-leaved fringed rue). This rue specie, originating from the Mediterranean region, is well-distributed in Southeast Asia, notably in the Indo-Chinese peninsula and other territories. In some countries, such as Malaysia, the plant is used to treat liver diseases and cancer. Extracts of R. angustifolia display antifungal, antiviral and antiparasitic effects. Diverse b

Phytophotodermatitis as a clinical problem and as a therapeutic option: Case report and review of the literature.

BACKGROUND: Phytophotodermatitis is a contact photodermatitis to furocoumarins, which act as sensitizing psoralens, from certain plants, especially citrus and fig trees. This photosensitizing effect has traditionally been used for the treatment of cutaneous viral warts, a reflection of traditional medicine. However, there are hardly any studies that support this fact. Otherwise, on certain occasions, especially in extensive exposures, they can cause a generalized severe condition that can even put the patient's life at risk. CASE PRESENTATION: We report the case of a 28-year-old man with seve

Severe Photo-oxidative Injury from Over-the-Counter Skin Moisturizer: A Child Abuse Mimic.

BACKGROUND: The cutaneous manifestations of pathological conditions have been described to mirror findings commonly associated with child abuse. Although it is important for clinicians to report suspected abuse, vigilance is required to detect conditions that mimic abuse. Phytophotodermatitis, a phototoxic reaction to furocoumarin-containing plants, is a well-described mimicker of nonaccidental trauma. However, non-furocoumarin-containing chemicals may cause similar presentations through a process called auto-oxidation. Typically, these chemical reactions occur as a result of aero-oxidation or

The induction of prophage expression in different Salmonella typhimurium strains by DNA cross-linking and monoadduct forming psoralens and longwave ultraviolet radiation.

The cytotoxicity, and ability to induce the expression of prophage in Salmonella typhimurium LT2 strains has been examined for 4 furocoumarins, 8-methoxypsoralen (8-MOP), 5-methoxypsoralen (5-MOP), 3-carbethoxypsoralen (3-CP), and 5-methylisopsoralen (5-MI) given with ultraviolet-A radiation (u.v.A). 8-MOP and 5-MOP have a linear tricyclic structure and possess two photoreactive sites, the 3,4 and 4',5' double bonds, enabling them to form both monoadducts and interstrand cross-links with DNA. The angular structure of 5-MI imposes steric constraints preventing it from forming interstrand cross-

Phytophotodermatitis induced by wild parsnip.

Phytophotodermatitis results when skin is exposed to ultraviolet light after previous contact with a phototoxic compound. Wild parsnip (Pastinia sativa), a member of the Umbelliferae family, is an invasive plant species introduced to North America as a root vegetable. Although cultivated less commonly today, the plant is increasingly found growing wild in prairies and roadsides. The stems and leaves contain furocoumarins, which upon activation by UV light interact with oxygen. Resultant reactive oxygen species induce tissue damage manifesting initially as blistering and later as hyperpigmentat

Photocontact dermatitis.

Photocontact dermatitis is not a common condition, but neither is it rare. Both photo-irritant contact dermatitis (PICD) and photoallergic contact dermatitis (PACD) are seen by most dermatologists in general practice. PICD is diagnosed on clinical grounds and is usually caused by furocoumarins in plants like limes and celery. PACD is caused primarily by sunscreens but can also be the result of fragrances and antibacterial agents. PACD can only be diagnosed by photo-patch testing that most dermatologists, even those who patch test and give phototherapy in their office, do not perform. The proce

Photoallergic contact dermatitis to Heracleum giganteum.

Heracleum plants occur in numerous species worldwide and may cause phototoxic reactions due to its content of various furocoumarins. In this case report, a widespread photoallergic contact dermatitis after exposure to Heracleum giganteum (giant bear claw) is described. A photopatch test with extracts from the stem, leaves and seeds of the giant bear claw revealed a positive papulovesicular reaction that already appeared at 24 h and peaked at 72 h after irradiation with 5 J/cm(2) UVA. The unirradiated controls remained negative. We conclude that in rare cases Heracleum plants may cause severe p

[Dermatitis bullosa striata pratensis caused by Dictamnus albus L. (burning bush)].

We report two patients with bullous phototoxic contact dermatitis caused by Dictamnus albus L. (gas plant) which belongs to the Rutaceae family. In both cases long lasting postinflammatory hyper-pigmentations were observed as a result of the phototoxic contact dermatitis. The phototoxic components of Dictamnus albus L. are the furocoumarins 5-methoxypsoralen (bergapten) and 8-methoxypsoralen, and the alkaloid dictamnin whose phototoxic properties were recently discovered. Even though reactions to the gas plant have been infrequently reported, it is becoming more popular with gardenus and shoul

Free radical induced biophysical modification of membrane lipids: a novel mechanism proposed for a haemorheological alteration induced by 4,5' 8-trimethyl psoralen.

Haemostasis is the spontaneous arrest of bleeding from damaged blood vessel. Platelet aggregation is the major event in this process. A number of drugs are known to inhibit this process of platelet aggregation. 4,5' 8-trimethyl psoralen a naturally occurring furocoumarin is used as a photochemotherapeutic agent in PUVA therapy. It shows antithrombotic action by preventing platelet aggregation induced by adenosine diphosphate in vitro. It is proposed that a free radical induces a lipid phase transition of the membrane lipids, the platelet membrane undergoes certain biophysical alteration leadin

Fig leaf tanning lotion and sun-related burns: case reports.

A sun tan is considered a symbol of well-being in our society, but incorrect methods of sun exposure can create serious problems. We present two cases of severe sun-related burns caused by fig leaf decoction used as home-made tanning lotion. Twenty four-thirty six hours after application and sun exposure, patients developed a phytophotodermatitis characterised by erythema, and blister formation involving all the photoexposed areas (45-70% BSA). Their general conditions became rapidly critical and they were admitted to our Burn Centre. The patients were discharged after 11 and 26 days, respecti

Hidden threat lurking in extensive hand hygiene during the Covid-19 pandemic: investigation of sensitizing molecules in gel products by hyphenated chromatography techniques.

During the Covid-19 pandemic, health agencies worldwide have recommended frequent handwashing and sanitizing. A variety of hand gel products were made available on the market, often with fragrances added to curtail the strong smell of alcohol. Commonly used Citrus fragrances contain volatile aroma constituents and non-volatile oxygen heterocyclic compounds (OHCs), consisting mostly of polymethoxyflavones, coumarins, and furocoumarins. The latter have long been investigated for their phototoxic properties, and their safety as cosmetic product ingredients has been debated recurrently. To this co

Scoring function for DNA-drug docking of anticancer and antiparasitic compounds based on spectral moments of 2D lattice graphs for molecular dynamics trajectories.

We introduce here a new class of invariants for MD trajectories based on the spectral moments pi(k)(L) of the Markov matrix associated to lattice network-like (LN) graph representations of Molecular Dynamics (MD) trajectories. The procedure embeds the MD energy profiles on a 2D Cartesian coordinates system using simple heuristic rules. At the same time, we associate the LN with a Markov matrix that describes the probabilities of passing from one state to other in the new 2D space. We construct this type of LNs for 422 MD trajectories obtained in DNA-drug docking experiments of 57 furocoumarins

Effect of addition of dried healthy or diseased parsnip root tissue to a modified AIN-76A diet on cell proliferation and histopathology in the liver, oesophagus and forestomach of male Swiss Webster mice.

Umbelliferous crop plants, including the parsnip (Pastinaca sativa L.), elaborate enhanced levels of furocoumarins, including psoralens, when subjected to biotic or abiotic stress. These furocoumarins are recognized to lead to phototoxicity. In this study, the effect of these agents, which are present in diseased parsnip root tissue, on the liver and two tissues on the route of entry to the body (the oesophagus and forestomach) were investigated. Young male Swiss Webster mice were fed for approximately 30 days with modified AIN-76A diets containing 32.5% dried healthy, 32.5% apparently healthy

Nine case series with phototoxic dermatitis related to Chenopodium album.

Chenopodium album L. ssp. album is a common plant species worldwide, especially in humid areas. Sun exposure after oral intake of the plant can lead to sunburn-like rashes owing to its furocoumarin content. In this article, we reported nine patients who developed dermatitis with edema, erythema and necrosis on the face and dorsum of the hands. Each was exposed to sunlight after eating selemez (the vernacular name of C. album L. ssp. album). The plant samples analyzed by an expert botanist were described as C. album L. ssp. album belonging to Chenopodiacea family.

Plasma and buffy coat concentration of 8-methoxypsoralen in patients treated with extracorporeal photopheresis.

Recently, a new therapy involving an extracorporal activation of orally administered 8-methoxypsoralen (8-MOP), photosensitizing furocoumarin, is established for the treatment of different skin diseases, extracorporeal photopheresis (ECP). The pharmacokinetic profile of 8-MOP has been pursued as part of a clinical study which should assess the efficacy of ECP in patients with progressive systemic sclerosis and cutaneous T-cell lymphoma. The enormous intra-individual variations proofed for plasma as well as buffy coat concentration are unfavourable for oral 8-MOP therapy. Therefore, the introdu

Characterization by 32P-postlabelling of 8-methoxypsoralen adducts.

8-Methoxypsoralen is a bifunctional furocoumarin used in human photochemotherapy. It can form two kinds of DNA adduct, monoadducts and interstrand cross-links. These adducts have been detected by 32P-postlabelling using hydrolysis with DNase1, alkaline phosphatase and snake venom phosphodiesterase, and longer labelling than usual, with more T4 polynucleotide kinase. Using preliminary two-dimensional chromatography (D1, D2) followed by transfer of adducts to separate PEI cellulose sheets for further development (D3, D4), we observed three spots corresponding to the adducts sought. Two experimen
