A 3D Tumor Spheroid Platform to Model Fusobacterium nucleatum-Induced Tumorigenesis.

Cancer cell line spheroids provide a three-dimensional (3D) culture platform that enables the evaluation of cancer cell aggregation, tumor initiation, cell proliferation, and overall tumor growth. Compared with conventional two-dimensional monolayer cultures, spheroid models more faithfully recapitulate the architectural, physiological, and microenvironmental features of solid tumors, making them particularly well suited for studying tumor biology and microbe-host interactions. Previous work has leveraged this 3D system to investigate the role of the oral oncobacterium Fusobacterium nucleatum

Fusobacterium nucleatum exacerbates atherosclerosis progression via ceRNA network-mediated epigenetic reprogramming.

Fusobacterium nucleatum (F. nucleatum), a key periodontal pathogen, is increasingly detected in atherosclerotic plaques, yet its epigenetic regulatory mechanisms in atherosclerosis remain enigmatic. This study investigates how F. nucleatum reshapes the non-coding RNA landscape to drive atherosclerosis progression. Periodontal infection with F. nucleatum significantly increased atherosclerotic lesion area (p < 0.001) and necrotic core ratio, while reducing collagen content (p < 0.05) in ApoE-/- mice. Whole transcriptome sequencing of arterial tissues identified 847 differentially expressed mRNA

Fusobacterium nucleatum confirmed in gastric biopsies of patients without Helicobacter pylori.

OBJECTIVE: Putative associations between certain bacteria and gastric cancer have been described previously; however, the mechanisms underlying such associations are not thoroughly understood. We aimed to confirm the presence of Fusobacterium nucleatum in the biopsy samples of patients with gastroduodenal diseases, including gastric cancer, with concomitant or without Helicobacter pylori infection. RESULTS: Three hundred patients with gastroduodenal disorders, including gastritis, duodenal ulcer, or gastric cancer, were included. One hundred and eighty (60%) patients were negative and 120 (40

Fusobacterium nucleatum-Induced Empyema Masquerading as Lung Cancer: A Diagnostic Challenge.

A 73-year-old man, an ex-smoker with a history of asbestos exposure and hypertension, presented with progressive shortness of breath, weight loss, loss of appetite, and fatigue. He was referred by a general practitioner for evaluation of a round lesion in the right lung. Chest computed tomography (CT) revealed a pleural-based mass and small right-sided pleural effusion that was not amenable to aspiration. Routine blood investigations revealed elevated levels of the inflammatory markers. He was discharged with a course of oral antibiotics, levofloxacin, and scheduled for a two-week follow-up on

The Periodontal Pathogen Fusobacterium nucleatum Exacerbates Alzheimer's Pathogenesis via Specific Pathways.

Alzheimer's Disease (AD) is the most common form of dementia in older adults and has a devastating impact on the patient's quality of life, which creates a significant socio-economic burden for the affected individuals and their families. In recent years, studies have identified a relationship between periodontitis and AD. Periodontitis is an infectious/inflammatory disease that destroys the supporting periodontal structure leading to tooth loss. Dysbiosis of the oral microbiome plays a significant role in the onset and development of periodontitis exhibiting a shift to overgrowth of pathobion

Small Intrahepatic Vein Budd-Chiari Syndrome Complicated by Fusobacterium nucleatum Peritonitis.

Budd-Chiari syndrome is a rare disorder with significant liver-related complications. We present a 28-year-old woman with a 1-month history of weight loss and ascites. Hepatic venogram showed patent hepatic veins and inferior vena cava; however, there was an increased hepatic venous pressure gradient, which is clinically significant for portal hypertension. Hereditary and acquired thrombophilia workup was unrevealing. During admission, she developed peritonitis with Fusobacterium nucleatum and was treated with piperacillin-tazobactam. Liver biopsy showed vascular changes with features of venou

Fusobacterium nucleatum infections: clinical spectrum and bacteriological features of 78 cases.

PURPOSE: Few series describe the clinical spectrum of Fusobacterium spp. infections. Among them, fewer discuss F. nucleatum, even though there are many clinical cases. METHODS: We performed a retrospective study over 8 years (from 2007 to 2014) in Limoges University Hospital, France, to assess clinical and bacteriological aspects of infections due to F. nucleatum. RESULTS: Eighty-one patients with F. nucleatum positive cultures were included in this study, irrespective of sample origin. Abscesses (n = 43), bacteraemia (n = 18) and bone infections (n = 8) were the most common types of infecti

Microbe-driven immune suppression in colorectal cancer: the Fusobacterium nucleatum playbook.

Fusobacterium nucleatum, a key oral-pathobiont in colorectal cancer (CRC), has evolved from a presumed "passenger" to an active "co-conspirator" in tumorigenesis. This review systematically delineates its core role as an "architect of the immune microenvironment," whereby it remodels T-cell immune responses through precise mechanisms to establish an immunosuppressive tumor microenvironment. Specifically, F. nucleatum employs virulence factors such as Fap2 and FadA for specific colonization, and achieves immune evasion by inhibiting NK and T-cell function and recruiting myeloid-derived suppress

2'-Fucosyllactose Attenuates Fusobacterium nucleatum Virulence and Modulates the Oral Microbiota.

Fusobacterium nucleatum (F. nucleatum) is a key periodontal pathobiont associated with oral inflammation. This bacterium forms biofilms and expresses adhesins that facilitate its adhesion to and invasion of gingival epithelial cells. These processes disrupt the epithelial barrier and trigger oral inflammation, and in some cases, systemic inflammation. Conventional antimicrobial strategies predominantly depend on the utilization of antibiotics. Nevertheless, this can result in the proliferation of drug-resistant strains and the disruption of the oral microbiome equilibrium. As the predominant h

Fusobacterium nucleatum infection leading to rare hepatorenal abscess: a case report.

Fusobacterium nucleatum is a gram-negative anaerobic bacterium commonly associated with periodontal disease. However, its role in extraoral infections, particularly in immunocompetent individuals, is increasingly recognized. We report a rare case of hepatorenal abscess caused by F. nucleatum in a previously healthy woman, initially suspected to have a malignant tumor based on PET-CT findings. Next-generation sequencing (NGS) of abscess aspirate confirmed the pathogen. The patient responded well to targeted antibiotic therapy. This case highlights the importance of considering anaerobic pathoge

An Orbital Abscess Caused by Fusobacterium nucleatum Mimicking Tolosa-Hunt Syndrome.

Fusobacterium species, particularly Fusobacterium nucleatum, are anaerobic gram-negative bacteria found in the oral cavity and gastrointestinal tract. Infections caused by these organisms exhibit diverse clinical manifestations. Here we report a rare case of orbital abscess caused by Fusobacterium nucleatum, which was initially misdiagnosed as Tolosa-Hunt syndrome. The patient was controlled conservatively, and surgical intervention was withheld due to poor prognosis, diabetes, and logistical limitations. This case report highlights the need to consider infection even when imaging and laborato

Effect of High Intensity Blue Light on Fusobacterium nucleatum Membrane Integrity.

Oral malodour is considered to be caused mainly by the production of volatile sulfide compounds (VSC) by anaerobic gram-negative oral bacteria. Previous studies showed that these bacteria were susceptible to blue light phototoxicity mediated by the production of reactive oxygen species (ROS). In the present study, we tested the effect of blue light on the integrity Fusobacterium nucleatum's membrane, cellular proteins and DNA. Bacterial samples were exposed to high intensity blue light for 0, 70, 140 and 280 s (i.e. fluences of 0, 96, 192 and 384 J cm-2 , respectively). Following light exposur

Full-mouth antimicrobial photodynamic therapy in Fusobacterium nucleatum-infected periodontitis patients.

BACKGROUND: The purpose of this study is to clinically and microbiologically evaluate the effect of photodynamic therapy (PDT) as a full-mouth procedure in Fusobacterium nucleatum-infected patients with periodontitis. METHODS: In the present study, PDT is administered adjuvantly after scaling and root planing (SRP) to patients with localized chronic periodontitis (LCP). Twenty-four patients, in whom only F. nucleatum was detected by baseline polymerase chain reaction (PCR) after SRP, were randomly assigned to PDT and control groups. PDT was carried out once as a full-mouth disinfection in the

Fusobacterium nucleatum exacerbates the progression of pulpitis by regulating the STING-dependent pathway.

Bacterial infection is the main cause of pulpitis. However, whether a dominant bacteria can promote the progression of pulpitis and its underlying mechanism remains unclear. We provided a comprehensive assessment of the microbiota alteration in pulpitis using 16S rRNA sequencing. Fusobacterium nucleatum was the most enriched in pulpitis and played a pathogenic role accelerating pulpitis progression in rat pulpitis model. After odontoblast-like cells cocultured with F. nucleatum, the stimulator of interferon genes (STING) pathway and autophagy were activation. There was a float of STING express

Apelin Enhances the Effects of Fusobacterium nucleatum on Periodontal Ligament Cells In Vitro.

This study aimed to explore effects of Fusobacterium nucleatum with or without apelin on periodontal ligament (PDL) cells to better understand pathomechanistic links between periodontitis and obesity. First, the actions of F. nucleatum on COX2, CCL2, and MMP1 expressions were assessed. Subsequently, PDL cells were incubated with F. nucleatum in the presence and absence of apelin to study the modulatory effects of this adipokine on molecules related to inflammation and hard and soft tissue turnover. Regulation of apelin and its receptor (APJ) by F. nucleatum was also studied. F. nucleatum resul

Salivary Fusobacterium nucleatum serves as a potential diagnostic biomarker for gastric cancer.

BACKGROUND: As one of the most common tumors, gastric cancer (GC) has a high mortality rate, since current examination approaches cannot achieve early diagnosis. Fusobacterium nucleatum (Fn) primarily colonized in the oral cavity, has been reported to be involved in the development of gastrointestinal tumor. Until now, little is known about the relationship between salivary Fn and GC. AIM: To determine whether salivary Fn could be a biomarker to diagnose GC and explore the influence of Fn on GC cells. METHODS: The abundance of Fn in saliva was quantified by droplet digital polymerase chain r

Fusobacterium nucleatum Pleural Empyema in a Patient with Progressive Rheumatoid Arthritis and Immunosuppression.

Fusobacterium nucleatum is an anaerobic oral commensal organism that is often associated with inflammatory bowel disease, adverse pregnancy outcomes, respiratory tract infections, and Lemierre's syndrome. Rheumatoid arthritis is often associated with pleuropulmonary manifestations including noninfectious pleural effusions and interstitial lung disease. We present a case of a 47-year-old man with progressive rheumatoid arthritis on immunosuppressive therapy who was found to have a left-sided pleural effusion, thought secondary to possible pneumonia, and was treated with levofloxacin and methylp
