A Novel Sialoglycopeptide from Gadus morhua Eggs Prevents Liver Fibrosis Induced by CCl4 via Downregulating FXR/FGF15 and TLR4/TGF-β/Smad Pathways.

Liver fibrosis plays a critical role in liver disease progression. A sialoglycopeptide from the Gadus morhua eggs (Gm-SGPP) was identified having a 7000 Da molecular weight with a core pentasaccharide structure and osteogenesis activity. However, whether Gm-SGPP is beneficial to liver fibrosis remains unknown. In this study, mice with liver fibrosis were intraperitoneally injected with 2.5% CCl4 (10 mL/kg) and orally administered with Gm-SGPP (500 mg/kg) for 30 days. Results showed that Gm-SGPP alleviated oxidative liver damage and lipid metabolism disorder and reduced hepatocyte necrosis and

Susceptibility and Pathology in Juvenile Atlantic Cod Gadus morhua to a Marine Viral Haemorrhagic Septicaemia Virus Isolated from Diseased Rainbow Trout Oncorhynchus mykiss.

The first known outbreak caused by a viral haemorrhagic septicaemia virus (VHSV) strain of genotype III in rainbow trout occurred in 2007 at a marine farm in Storfjorden, Norway. The source of the virus is unknown, and cod and other marine fish around the farms are suspected as a possible reservoir. The main objective of this study was to test the susceptibility of juvenile Atlantic cod to the VHSV isolate from Storfjorden. As the pathology of VHS in cod is sparsely described, an additional aim of the study was to give a histopathological description of the disease. Two separate challenge expe

Characterization and expression analyses of anti-apoptotic Bcl-2-like genes NR-13, Mcl-1, Bcl-X1, and Bcl-X2 in Atlantic cod (Gadus morhua).

NR-13, Mcl-1, and BCL-X(L), are conserved anti-apoptotic proteins that belong to the anti-apoptotic Bcl-2 sub-family, which inhibits cell death by preventing mitochondrial membrane permeabilization (MMP). Given the anti-apoptotic functions of these proteins in vertebrates (e.g. human, mouse, and zebrafish) and the involvement of apoptotic regulation in immune responses, we studied the sequences of these genes and their transcript expression in Atlantic cod (Gadus morhua) during innate immune responses to viral and bacterial stimuli. Based on previously generated Atlantic cod expressed sequence

Effects of long-term temperature acclimation on thyroid hormone deiodinase function, plasma thyroid hormone levels, growth, and reproductive status of male Atlantic cod, Gadus morhua.

The recent collapse of the Northwestern Atlantic cod fisheries has coincided with a cooling of water temperatures. During this time the condition factor of cod has been poor. The objective of the present study was to determine the effects of long-term temperature acclimation on growth reproduction and thyroid function in laboratory held Atlantic cod (Gadus morhua). One of the key parameters used to assess thyroid function is the peripheral metabolism of L-thyroxine (T4) by microsomal deiodinase enzymes. Deiodinase function has not been described for gadid fish. T4 outer-ring deiodinating activ

Deformation of the notochord by pressure from the swim bladder may cause malformation of the vertebral column in cultured Atlantic cod Gadus morhua larvae: a case study.

This study describes a malformation that frequently occurs in Atlantic cod Gadus morhua in intensive culture systems. The malformation is characterised by a slight upward tilt of the head and an indented dorsal body contour at the transition between the head and the trunk, and is first evident to the fish farmer when the cod reach the juvenile stage. These abnormalities are associated with malformations of the neurocranium, the cranial region of the vertebral column and the cranial part of the epaxial lateral muscles. The pathogenesis involves deformation of the notochord, which can be observe

Effects of phosphates and salt in ground raw and cooked farmed cod (Gadus morhua) muscle studied by the water holding capacity (WHC), and supported by P-NMR measurements.

A model system consisting of ground farmed cod muscle (80%, w/w) and added brine (20%, w/w) with different content and combinations of salt (0% and 3% in brine) and phosphorus compounds (mono-, di-, tri- and hexametaphosphates; 0% and 3% in brine) was used to simulate industrial brining of muscle foods. Individual phosphorus component concentrations and breakdown as function of time (0, 23 h) were analyzed using (31)P-NMR spectroscopy. The effects of salt and phosphate on water holding capacity (WHC) were measured at similar sampling times, and interrelations between phosphorous components det

Infectious Pancreatic Necrosis Virus Can Be Transmitted Through Cohabitation From Atlantic Cod (Gadus morhua) to Atlantic Cod, Atlantic Salmon (Salmo salar), Atlantic Halibut (Hippoglossus hippiglossus) and Lumpfish (Cyclopterus lumpus).

Infectious pancreatic necrosis virus (IPNV) can cause devastating disease in Atlantic salmon (Salmon salar). IPNV has a broad host range and may threaten other aquaculture species. Understanding interspecies transmission of IPNV is crucial for protecting the aquaculture industry. With the expansion of fish farming (in Norway), it is important to assess whether a pathogen can transmit from one fish species to another and cause disease. We investigated whether IPNV-infected Atlantic cod can shed IPNV, leading to infection in other fish important to Norwegian aquaculture: halibut, salmon and lump

A comparative study of quality and safety of Atlantic cod (Gadus morhua) fillets during cold storage, as affected by different thawing methods of pre-rigor frozen headed and gutted fish.

BACKGROUND: The catch of marine whitefish is typically seasonal, whereas the land-based processing industry has a need for all-year stable supply of raw materials. This challenge can be met by applying fish frozen at sea. When using frozen fish, the methods employed for thawing may influence the safety and quality of the final product. This study aimed to investigate the applicability of novel thawing strategies in order to provide an all-year supply of high-quality and safe cod products. RESULTS: Comparative investigations of quality and safety factors after thawing in water, with and withou

Distribution of whaleworm (Anisakis simplex, Nematoda, Ascaridoidea) L3 larvae in three species of marine fish; saithe (Pollachius virens (L.)), cod (Gadus morhua L.) and redfish (Sebastes marinus (L.)) from Norwegian waters.

The frequency distribution of Anisakis simplex L3 larvae between host tissues was investigated in three host species: saithe, cod and redfish. Fish were sampled from Norwegian coastal waters and examined for the presence of A. simplex in muscle and viscera. In all three of the host species, A. simplex larvae were most frequently detected in the viscera; the percentages of total infection for saithe, cod and redfish were 99.6%, 97.8% and 88.0%, respectively. In general, the distribution patterns of A. simplex L3 between muscle and viscera were not significantly affected by host size. The observ

A Novel Method for Predicting Anisakid Nematode Infection of Atlantic Cod Using Rough Set Theory.

Atlantic cod ( Gadus morhua L.) is one of the most important fish species in the fisheries industries of many countries; however, these fish are often infected with parasites. The detection of pathogenic larval nematodes is usually performed in fish processing facilities by visual examination using candling or by digesting muscles in artificial digestive juices, but these methods are both time and labor intensive. This article presents an innovative approach to the analysis of cod parasites from both the Atlantic and Baltic Sea areas through the application of rough set theory, one of the meth

More often, less each: Averaging can hide opposing trends in prey occurrence and consumer stomach content.

Averages of prey mass in fish stomachs are widely used to track changes in feeding. Yet, a mean over all stomachs is the exact product of two quantities-the frequency of occurrence of a prey and its mean content in the stomachs that contained it-which can move in opposite directions. In Atlantic cod (Gadus morhua L.) in the Baltic Sea, a stable mean benthos content concealed a falling content of consumers and a rising frequency of occurrence, opposing trends that can signal density dependence or responses to hypoxia.

Abolition of reflex bradycardia by cardiac vagotomy has no effect on the regulation of oxygen uptake by Atlantic cod in progressive hypoxia.

The functional significance of chemoreflexive hypoxic bradycardia was explored in Atlantic cod Gadus morhua L. (mean mass approximately 800 g, acclimated to a seawater temperature of 11 degrees C) by investigating responses to progressive hypoxia following section of the cardiac branches of cranial nerve X. Cardiac denervation had no effect on oxygen uptake rate (MO(2)), gill ventilation rate (f(G)) or opercular pressure amplitude (P(OP)) under normoxic conditions, but caused a significant increase in heart rate (f(H)), to 50+/-1 beats min(-1) by comparison to 40+/-2 beats min(-1) in sham-oper

Mitochondrial DNA of Anisakis simplex s.s. as a potential tool for differentiating populations.

Anisakis simplex s.s. specimens from Gadus morhua, Gadus ogac, Reinhardtius hippoglossoides, Clupea harengus, Micromesistius poutassou, Pleurogrammus monopterygius, and Oncorhynchus nerka, caught in the north Atlantic and Pacific, were examined. Identity of the parasite species was confirmed using a molecular key. All specimens were used for amplifying a part of the non-coding sequence of the AT-rich region, flanked by the nad6-nad4L-trnW-trnE-rrnS-trnS2, and trnN-trnY-nad1 genes. This part of the mtDNA was chosen as a promising fragment, which could yield information regarding population diff

Comparative histopathology of livers from Baltic cod infected with the parasitic nematode Contracaecum osculatum.

Infection levels with the parasitic nematode Contracaecum osculatum in Eastern Baltic cod have increased in the last decades. Eastern Baltic cod is transport host for this parasite that has a high affinity for the liver of the fish. The liver is a highly vital organ and damage to the liver tissue can result in reduced functionality of the organ. Previous studies have revealed that cod with high infections loads reveal impaired physiological performance, reduced nutritional condition and show signs of having a liver disease. Yet, little is known about the pathological changes and inflammatory r

Integrative omics-analysis of lipid metabolism regulation by peroxisome proliferator-activated receptor a and b agonists in male Atlantic cod.

Lipid metabolism is essential in maintaining energy homeostasis in multicellular organisms. In vertebrates, the peroxisome proliferator-activated receptors (PPARs, NR1C) regulate the expression of many genes involved in these processes. Atlantic cod (Gadus morhua) is an important fish species in the North Atlantic ecosystem and in human nutrition, with a highly fatty liver. Here we study the involvement of Atlantic cod Ppar a and b subtypes in systemic regulation of lipid metabolism using two model agonists after in vivo exposure. WY-14,643, a specific PPARA ligand in mammals, activated cod Pp

Evidence of predation events by marine mammals at offshore wind farms.

Anthropogenic structures in the marine environment alter the availability and distribution of food for marine animals with potential consequences for individual fitness, population and community dynamics. Knowledge of how marine mammals' behaviour or foraging activities are influence by existing wind farms is essential to predict the effect of future large-scale renewable energy development on local or regional populations. During acoustic tracking of demersal gadoid fish at two offshore wind farms in Scotland, six Atlantic cod were predated by a marine mammal close to or in the vicinity of tu

Effects of hollow glass microspheres on cold-water pelagic copepods and fish.

The highly reflective properties of hollow glass microspheres (HGM) have motivated their application as a reflective shield onto Arctic sea ice to increase albedo and reduce melting. However, to ensure safe application in the environment, their potential environmental impacts must be assessed. While most HGMs will be positively buoyant in seawater and thus remain in the upper part of the water column, small, cracked and broken HGMs will sink. In this study, we conducted a series of experiments to investigate interactions between HGMs and cold-water pelagic copepods (Calanus finmarchicus) and f

Computational Study of Network and Type-I Functional Divergence in Alcohol Dehydrogenase Enzymes Across Species Using Molecular Dynamics Simulation.

Alcohol dehydrogenases (ADHs) are critical enzymes involved in the oxidation of alcohols, contributing to various metabolic pathways across organisms. This study investigates type I functional divergence within three ADH1 families: Saccharomyces cerevisiae (PDB ID: 4W6Z), Gadus morhua (PDB ID: 1CDO), and Homo sapiens (PDB ID: 1HDX). Understanding the molecular evolution and mechanisms underlying functional divergence of ADHs is essential for comprehending their adaptive significance. For this purpose, we performed a computational analysis that included structural characterization of ADHs throu

Anisakid Nematodes as Possible Markers to Trace Fish Products.

In this work a total of 949 fish samples were analysed for the identification of nematode larvae belonging to the Anisakidae family. Biomolecular application for the identification of Anisakidae larvae can be an optimal instrument for the traceability of fish products, described on the Reg. EC 178/2002. Results confirm a correlation between geographical distribution of fishes and presence of specific Anisakid larvae. FAO 37 zone (Mediterranean sea) showed a prevailing distribution of Anisakis pegreffii and a minimal presence of A. simplex s.s. in hybrid form with Anisakis pegreffii. FAO 27 zon

Egg and larval production of Eastern Baltic cod in captivity over one spawning season.

Eastern Baltic cod (Gadus morhua) has experienced a dramatic decline in biomass, distribution, size and condition over recent decades, related to a combination of fishing pressure and environmental stressors. Reduced parental size and condition are known to impact reproductive success, affecting egg quality, fertilization and hatching success and larval survival. However, how these reproductive traits vary throughout the spawning season at the population level remains poorly understood for this stock. To address this knowledge gap, this study investigated egg and larval characteristics of capt
