Neuropeptide Modulation Enables Biphasic Internetwork Coordination via a Dual-Network Neuron.

Linked rhythmic behaviors, such as respiration/locomotion or swallowing/chewing, often require coordination for proper function. Despite its prevalence, the cellular mechanisms controlling coordination of the underlying neural networks remain undetermined in most systems. We use the stomatogastric nervous system of the crab Cancer borealis to investigate mechanisms of internetwork coordination, due to its small, well-characterized feeding-related networks (gastric mill [chewing, ∼0.1 Hz]; pyloric [filtering food, ∼1 Hz]). Here, we investigate coordination between these networks during the Gly1

Neuroanatomy of Hyalinella punctata: Common patterns and new characters in phylactolaemate bryozoans.

Studies on the bryozoan adult nervous system employing immunocytochemical techniques and confocal laser scanning microscopy are scarce. To gain a better view into the structure and evolution of the nervous system of the Phylactolaemata, the earliest extant branch and sister taxon to the remaining Bryozoa, this work aims to characterize the nervous system of Hyalinella punctata with immunocytochemical techniques and confocal laser scanning microscopy. The cerebral ganglion is located between the anus and the pharynx and contains a lumen. Two ganglionic horns and a circum-oral nerve ring emanate

Slow conductances could underlie intrinsic phase-maintaining properties of isolated lobster (Panulirus interruptus) pyloric neurons.

The rhythmic pyloric network of the lobster stomatogastric system approximately maintains phase (that is, the burst durations and durations between the bursts of its neurons change proportionally) when network cycle period is altered by current injection into the network pacemaker (Hooper, 1997a,b). When isolated from the network and driven by rhythmic hyperpolarizing current pulses, the delay to firing after each pulse of at least one network neuron type [pyloric (PY)] varies in a phase-maintaining manner when cycle period is varied (Hooper, 1998). These variations require PY neurons to have

Central pattern generating neurons simultaneously express fast and slow rhythmic activities in the stomatogastric ganglion.

Neuronal firing patterns can contain different temporal information. It has long been known that the fast pyloric and the slower gastric motor patterns in the stomatogastric ganglion of decapod crustaceans interact. However, the bidirectional influences between the pyloric rhythm and the gastric mill rhythm have not been quantified in detail from preparations that spontaneously express both patterns in vitro. We found regular and stable spontaneous gastric and pyloric activity in 71% of preparations of the isolated stomatogastric nervous system of the lobster, Homarus americanus. The gastric [

Insights into early molluscan neuronal development through studies of transmitter phenotypes in embryonic pond snails.

Pond snails have long been the subject of intense scrutiny by researchers interested in general principles of development and also cellular and molecular neurobiology. Recent work has exploited both these fields of study by examining the ontogeny of the nervous system in these animals. Much of this work has focussed upon the development of specific transmitter phenotypes to provide vignettes of neuronal subpopulations that can be traced from early embryonic life through to adulthood. While such studies have generally confirmed previous explanations of gangliogenesis in gastropods, they have al

Dynamic control of irregular bursting in an identified neuron of an oscillatory circuit.

In the oscillatory circuits known as central pattern generators (CPGs), most synaptic connections are inhibitory. We have assessed the effects of inhibitory synaptic input on the dynamic behavior of a component neuron of the pyloric CPG in the lobster stomatogastric ganglion. Experimental perturbations were applied to the single, lateral pyloric neuron (LP), and the resulting voltage time series were analyzed using an entropy measure obtained from power spectra. When isolated from phasic inhibitory input, LP generates irregular spiking-bursting activity. Each burst begins in a relatively stere

Pyloric motor pattern modification by a newly identified projection neuron in the crab stomatogastric nervous system.

1. We have used multiple, simultaneous intra- and extracellular recordings as well as Lucifer yellow dye-fills to identify modulatory commissural neuron 5 (MCN5) and characterize its effects in the stomatogastric nervous system (STNS) of the crab, Cancer borealis. MCN5 has a soma and neuropilar arborization in the commissural ganglion (CoG; Figs. 1 and 2), and it projects through the inferior esophageal nerve (ion) and stomatogastric nerve (stn) to the stomatogastric ganglion (STG; Figs. 1-3). 2. Within the CoGs, MCN5 receives esophageal rhythm-timed excitation and pyloric rhythm-timed inhibit

Adenylate cyclase activity from Hirudo medicinalis segmental ganglia: modulation by calcium and calmodulin.

An adenylate cyclase activity stimulated by serotonin and calmodulin is present in the segmental ganglia of the leech Hirudo medicinalis. Removal of the endogenous calcium binding protein does not alter the responsiveness of the enzyme to serotonin. The calmodulin antagonist, trifluoperazine, drastically reduces the amine stimulatory effect on both intact and calmodulin-depleted membranes. We suggest that calmodulin-sensitive and serotonin-stimulated adenylate cyclase are, at least functionally, independent.

Neuropeptide degradation produces functional inactivation in the crustacean nervous system.

The pentapeptide proctolin (Proct.; Arg-Tyr-Leu-Pro-Thr) is a modulatory transmitter found throughout the crustacean nervous system. No information is available in this system, however, as to how the actions of this peptide are terminated. To study this issue in the crab Cancer borealis, we incubated exogenous proctolin (10(-5) M) with either the thoracic ganglion (TG) or with conditioned saline (CS) that had been preincubated with the TG. We removed aliquots at standard time points for analysis by reverse-phase high-performance liquid chromatography (HPLC). We found that over time the proctol

Distinct Strategies Regulate Correlated Ion Channel mRNAs and Ionic Currents in Continually versus Episodically Active Neurons.

Relationships among membrane currents allow central pattern generator (CPG) neurons to reliably drive motor programs. We hypothesize that continually active CPG neurons utilize activity-dependent feedback to correlate expression of ion channel genes to balance essential membrane currents. However, episodically activated neurons experience absences of activity-dependent feedback and, thus, presumably employ other strategies to coregulate the balance of ionic currents necessary to generate appropriate output after periods of quiescence. To investigate this, we compared continually active pyloric

Increased robustness and adaptation to simultaneous temperature and elevated extracellular potassium in the pyloric rhythm of the crab, Cancer borealis.

Animals must deal with numerous perturbations, oftentimes concurrently. In this study, we examine the effects of two perturbations, high extracellular potassium and elevated temperature, on the resilience of the pyloric rhythm of the crab, Cancer borealis. At control temperatures (11°C), high potassium saline (2.5× K+) depolarizes the neurons of the stomatogastric ganglion (STG), and the pyloric rhythm becomes quiescent. Over minutes, while remaining depolarized in high potassium, the pyloric network neurons adapt, and resume their spiking and bursting activity. We compared adaptation to high

A classic model animal in the 21st century: recent lessons from the leech nervous system.

The medicinal leech (genus Hirudo) is a classic model animal in systems neuroscience. The leech has been central to many integrative studies that establish how properties of neurons and their interconnections give rise to the functioning of the animal at the behavioral level. Leeches exhibit several discrete behaviors (such as crawling, swimming and feeding) that are each relatively simple. Importantly, these behaviors can all be studied - at least at a basal level - in the isolated nervous system. The leech nervous system is particularly amenable to such studies because of its distributed nat

Natural neural output that produces highly variable locomotory movements.

We recorded fast extensor tibiae motor neuron activity during single-legged treadmill walking in the stick insect, Carausius morosus. We used this activity to stimulate the extensor muscle motor nerve, observed the resulting extensor muscle contractions under isotonic conditions, and quantified these contractions with a variety of measures. Extensor contractions induced in this manner were highly variable, with contraction measures having SDs of 12 to 51%, and ranges of 82 to 275%, when expressed as percentages of the means, an unexpectedly wide range for a locomotory pattern. Sensitivity threshold and response characteristics of infrared detection in the beetle Melanophila acuminata (Coleoptera: Buprestidae).

The minimum detection threshold of the infrared sensitive beetle, Melanophila acuminata, was measured with a helium-neon laser that emitted light at a wavelength of 3.39 microm. Extracellular recordings were taken both at the pit organ responsible for detection and at the interganglionic connectives in the thorax of the beetle. At the pit organ, generator and action potentials from single neurons were measured with a sharpened tungsten electrode. At the connectives that linked the fused second meso-/metathoracic and prothoracic ganglia, compound action potentials were measured with a tungsten

When can neuronal activity-dependent homeostatic plasticity maintain circuit-level properties?

Neural circuits are remarkably robust to perturbations that threaten their function. Activity-dependent homeostatic plasticity (ADHP) is a stabilizing mechanism that supports robustness by tuning neuronal ion conductances to combat chronic over- or under-activity. Its restorative capacity has been demonstrated in the pyloric circuit of the crustacean stomatogastric ganglion, whose neurons must burst in a specific order to coordinate digestive muscles. After disruption by physical and pharmacological manipulations, this circuit reliably recovers not only the activity levels of constituent neuro

Neural control of olfaction and tentacle movements by serotonin and dopamine in terrestrial snail.

We investigated the role of serotonin (5HT) and dopamine (DA) in the regulation of olfactory system function and odor-evoked tentacle movements in the snail Helix. Preparations of the posterior tentacle (including sensory pad, tentacular ganglion and olfactory nerve) or central ganglia with attached posterior tentacles were exposed to cineole odorant and the evoked responses were affected by prior application of 5HT or DA or their precursors 5-hydroxytryptophan (5HTP) and L: -DOPA, respectively. 5HT applications decreased cineole-evoked responses recorded in the olfactory nerve and hyperpolari

Animal-to-animal variability in motor pattern production in adults and during growth.

Which features of network output are well preserved during growth of the nervous system and across different preparations of the same size? To address this issue, we characterized the pyloric rhythms generated by the stomatogastric nervous systems of 99 adult and 12 juvenile lobsters (Homarus americanus). Anatomical studies of single pyloric network neurons and of the whole stomatogastric ganglion (STG) showed that the STG and its neurons grow considerably from juvenile to adult. Despite these changes in size, intracellularly recorded membrane potential waveforms of pyloric network neurons and

[Neurotoxicity of the environmental pollutants: tin (Sn2+) effects on currents induced by acetylcholine in the snail neurons].

In isolated single neurone or in whole ganglia preparation, SnCl2 caused a decrease in the acetylcholine-induced inward current in a dose-dependent way. Threshold concentration was 0.1 mkM and saturated at 5 mkM SnCl2. After 10-min. pre-treatment with SnCl2 the effect was more obvious (by 20%) than after 3-min pre-treatment. Sn(CH3)2 decreased the amplitude of Ach-induced current similarly to the inorganic Sn. The effect of Sn(CH3)2 was irreversible. The findings support earlier data that the agonist-activated channels are important targets for toxic metals. Direct action upon neuronal membran

Allogeneic and xenogeneic grafts in pulmonate gastropod molluscs: fates of neural transplants.

Neural intracerebral allo- and xenografts in pulmonate gastropods demonstrated a variation in the tolerance of neural xenogeneic grafts that was dependent on the phylogenetic distance between the donor and the host. Like allografts, neural congeneric xenografts (Hp/Haa and H1/Haa) of cerebral ganglia (CG) were tolerated and restored growth in juvenile mesocerebrum-deprived (Haa) snails. However, CG neural xenografts between different genera of stylommatophorans (Achatina fulica/Haa) or between genera of different orders (Lymnaea stagnalis: Basommatophora/Haa: Stylommatophora) revealed an inter

Differential modulation of chemical and electrical components of mixed synapses in the lobster stomatogastric ganglion.

1. Two pairs of neurons in the pyloric network of the spiny lobster, Panulirus interruptus, communicate through mixed graded chemical and rectifying electrical synapses. The anterior burster (AB) chemically inhibits and is electrically coupled to the ventricular dilator (VD); the lateral pyloric (LP) and pyloric (PY) neurons show reciprocal chemical inhibition and electrical coupling. We examined the effects of dopamine (DA), serotonin (5HT) and octopamine (Oct) on these mixed synapses to determine the plasticity possible with opposing modes of synaptic interaction. 2. Dopamine increased net i
