Ingestion of guar-gum hydrolysate partially restores calcium absorption in the large intestine lowered by suppression of gastric acid secretion in rats.

We examined the effects of feeding guar-gum hydrolysate (GGH), a highly fermentable form of dietary fibre with low viscosity, on Ca absorption in the small and large intestines in rats under conditions in which gastric acid secretion was suppressed by a proton pump inhibitor, omeprazole. We also examined the role of the caecum in influencing these effects. The study was designed in a 2 x 2 x 2 factorial arrangement with two diet (GGH-containing (50 g/kg diet) and GGH-free diets) groups, two injection (omeprazole and vehicle) groups and two operation (sham and caecectomy) groups. Influence of L-glutamine on aspirin-induced gastric lesions and absorptions as well as antipyretic, analgesic and anti-inflammatory effects of aspirin in rats and mice.

L-glutamine markedly suppressed the development of the gastric lesions induced by aspirin in pylorus-ligated rats. In non-ligated normal rats, aspirin was absorbed rapidly after administration and was maintained in the blood in high salicylate concentration thereafter. When aspirin was administered in combination with L-glutamine, the absorption of aspirin was at nearly the same level as when aspirin had been given alone. In pylorus-ligated rats, administration of aspirin was followed by slow increment in blood salicylate concentration. Blood salicylate level in these rats was higher when aspi

The Effects of a Whey Protein and Guar Gum-Containing Preload on Gastric Emptying, Glycaemia, Small Intestinal Absorption and Blood Pressure in Healthy Older Subjects.

A whey protein/guar gum preload reduces postprandial glycaemia in type 2 diabetes through slowing gastric emptying. However, gastric emptying has previously been assessed using a stable isotope breath test technique, which cannot discriminate between slowing of gastric emptying and small intestinal absorption. This preload also may be useful in the management of postprandial hypotension. We evaluated the effects of a whey protein/guar preload on gastric emptying, glucose absorption, glycaemic/insulinaemic and blood pressure (BP) responses to an oral glucose load. Eighteen healthy older partici

Effect of denervation of the pylorus and transection of the duodenum on acetaminophen absorption in rats; possible mechanism for early delayed gastric emptying after pylorus preserving pancreatoduodenectomy.

Early delayed gastric emptying has been reported as a frequent complication following pylorus preserving pancreatoduodenectomy (PPPD). We investigated the effect of division of the pyloric branch of the vagus nerve and/or transection of the duodenum on gastric emptying using the acetaminophen method in rats to speculate the unknown etiology of early delayed gastric emptying after PPPD. Twenty-four male Wistar rats were divided into the following four groups; Group S, sham operation as controls; Group N, disturbance of neuro-vascular supply to the pylorus; Group D, temporary interference of the

Jejunal pouch reconstruction after total gastrectomy is associated with better short-term absorption capacity and quality of life in early-stage gastric cancer patients.

BACKGROUND: No consensus exists regarding the best reconstruction style after total gastrectomy (TG). Roux-en-Y oesophagojejunostomy is a simple option for gastrointestinal tract reconstruction. Recently, jejunal pouch reconstruction has been suggested as an appropriate approach. We compared the postoperative outcomes of the two surgical approaches using a well-characterized cohort of gastric carcinoma patients. METHODS: A total of 60 patients who underwent TG were divided into two groups according to the reconstruction style. Both groups were compared regarding patient characteristics, perio

The effect of pharmacological modification of gastric emptying and mouth-to-caecum transit time on the absorption of sugar probe marker molecules of intestinal permeability in normal man.

The present study examined the hypothesis that altered motility of the gastrointestinal tract affects absorption of probe markers of intestinal permeability. Seven healthy subjects, aged 32-44 years, received saline, 600 micrograms atropine or 10 mg metoclopramide in randomized order at weekly intervals. After 10 min they ingested a test solution containing 5 g lactulose, 5 g mannitol and 2 g 3-O-methyl glucose in 100 ml tap water. The molarity of the solution was 542 mmol l-1 and the dose administered was 80 ml m-2 body surface area. Gastric emptying was measured by ultrasound, mouth-to-caecu

Studies on the protective effect of deglycyrrhinised liquorice against aspirin (ASA) and ASA plus bile acid-induced gastric mucosal damage, and ASA absorption in rats.

By per-oral intubation of the stomach, four groups of rats (n = 22) received ASA 64 mg/kg, alone or with taurodeoxycholic acid (TDC, 5 mM), DGL (2000 mg/kg) or TDC plus DGL. At four hours the severity of bleeding was assessed by a lesion scoring system and expressed as medians and quartiles. In a separate study two groups of rats (n = 7) received ASA (128 mg/kg) or ASA plus DGL (2000 mg/kg) and salicylate levels measured in serial tail blood samples at 20, 40, 60 and 80 minutes. Lesion scores were increased from 6(3;10) with ASA alone to 12(5;16) by TDC (p less than 0.05) and reduced by DGL to

Integrated gastric to intestinal pH-shift dissolution and permeation testing can predict the marked oral absorption enhancement of an itraconazole-carboxymethyl ethylcellulose amorphous solid dispersion.

In this study, we aimed to investigate correlation of oral absorption and dissolution-permeation behaviors carboxymethyl ethylcellulose (CMEC)-based amorphous solid dispersion (ASD) for itraconazole (ITZ). Amorphization of ITZ and formation of the ASDs through ball milling were confirmed by powder X-ray diffraction, thermal and spectroscopic analyses, using hydroxypropyl methylcellulose acetate succinate (HPMCAS) as an additional carrier. ITZ rapidly dissolved in pH 1.2 medium; however, this was suppressed by ASD formation. The dissolution of ITZ in pH 6.8 medium was <0.2 μg/mL, which was impr

In vitro-in silico framework for evaluating intra-subject variability in the oral absorption of BCS class 1 drugs mediated by the gastric interdigestive migrating motor complex.

The intensity of gastric peristaltic motility and the rate of gastric emptying exert a substantial influence on the gastrointestinal absorption of orally administered drugs, particularly on the rate of absorption. The objective of this study was to elucidate how these factors influence drug absorption and to establish a simple framework for predicting the variability in oral drug pharmacokinetics. To minimize inter-subject confounding, this study focused on the intra-subject variability (CVintra) of the maximum plasma concentration (Cmax). Tablets of acetaminophen (AA) and bisoprolol fumarate

An enteric polymer mitigates the effects of gastric pH on oral absorption of poorly soluble weak acid drugs from supersaturable formulations: A case study with dantrolene.

This study demonstrated that an enteric polymer can mitigate the effects of gastric pH on the oral absorption of a poorly water-soluble weak acid drug, dantrolene (DNT). An amorphous solid dispersion (ASD) of DNT with hydroxypropyl methylcellulose (HPMC) acetate succinate (ASD-HPMCAS) was prepared as the enteric released ASD (ER-SF). ASD with HPMC (ASD-HPMC) and DNT sodium salt were also used as immediate-release supersaturable formulations (IR-SFs) with and without water-soluble polymer, respectively. In vivo study with rats and in vitro study with a dissolution/permeation (D/P) system were p

Cocrystal mitigates the effects of elevated gastric pH on oral absorption of weakly basic drugs: a case study with ketoconazole-succinic acid cocrystal on beagle dogs.

It is well documented that weakly basic drugs, such as ketoconazole (KTZ), will exhibit reduced oral absorption and lower bioavailability in elevated gastric pH conditions. Because this occurs frequently in patients taking medicines to reduce stomach acid, it is crucial to identify formulation strategies that ensure adequate drug exposure. The purpose of this work was to evaluate the in vitro and in vivo effects of KTZ cocrystals with acidic coformers in mitigating the negative effects of elevated gastric pH on KTZ dissolution and absorption after oral administration. Gastrointestinal absorption of a new reversible proton pump inhibitor, YJA-20379-8, and its pharmacokinetics after oral administration in acetic acid-induced gastric ulcer in rats.

The absorption of YJA-20379-8 (3-butyryl-4-[5-(R)-(+)-methylbenzylamino]-8-ethoxy-1,7-naphthyrid ine) from various rat gastrointestinal segments was evaluated using in-situ closed-loops. The pharmacokinetics of the drug were also evaluated after oral administration to rats with acetic acid-induced gastric ulcer (AIURs). The concentrations of YJA-20379-8 in the biological samples were analyzed by HPLC. The absorption of YJA-20379-8 from stomach and jejunum was fast, but approximately 50% of the drug was recovered from each segment at 24 h. The total areas under the plasma concentration-time cur

Effects of intragastric administration of L-tryptophan on the glycaemic response to a nutrient drink in men with type 2 diabetes - impacts on gastric emptying, glucoregulatory hormones and glucose absorption.

BACKGROUND: The rate of gastric emptying and glucoregulatory hormones are key determinants of postprandial glycaemia. Intragastric administration of L-tryptophan slows gastric emptying and reduces the glycaemic response to a nutrient drink in lean individuals and those with obesity. We investigated whether tryptophan decreases postprandial glycaemia and slows gastric emptying in type 2 diabetes (T2D). METHODS: Twelve men with T2D (age: 63 ± 2 years, HbA1c: 49.7 ± 2.5 mmol/mol, BMI: 30 ± 1 kg/m2) received, on three separate occasions, 3 g ('Trp-3') or 1.5 g ('Trp-1.5') tryptophan, or control (

Drugs, diseases and altered gastric emptying.

Drugs are usually given orally. They are not absorbed to any extent from the stomach but may be absorbed very rapidly from the small intestine. Thus factors influencing the rate of gastric emptying may alter the rate of absorption of most if not all orally administered drugs. Food, hormones, posture, peritoneal irritation, severe pain, gastric ulcer, diabetes and other metabolic diseases, as well as drugs such as alcohol, anticholinergics, narcotic analgesics, ganglion blocking drugs, antacids and metoclopramide all influence the rate of gastric emptying and they will, in turn, change the rate

A revised Sorensen model: Simulating glycemic and insulinemic response to oral and intra-venous glucose load.

In 1978, Thomas J. Sorensen defended a thesis in chemical engineering at the University of California, Berkeley, where he proposed an extensive model of glucose-insulin control, model which was thereafter widely employed for virtual patient simulation. The original model, and even more so its subsequent implementations by other Authors, presented however a few imprecisions in reporting the correct model equations and parameter values. The goal of the present work is to revise the original Sorensen's model, to clearly summarize its defining equations, to supplement it with a missing gastrio-int
