Schistosome worms inhabit mammalian mesenteric veins. Their eggs cause chronic inflammation, which progresses to periportal fibrosis in 5 to 30% of cases, increasing portal blood pressure and leading to esophageal varices. Episodes of bleeding cause hepatic necrosis and may ultimately lead to hepatic failure and the death of the patient. Schistosome infections can also cause pulmonary hypertension and heart failure. The mechanisms of fibrogenesis and fibrolysis are beginning to be unraveled, but it remains unclear why disease occurs only in certain subjects, as also observed for other types of

Systemic inflammatory proteins have been reported to be related to inflammatory bowel disease (IBD) in previous observational research. However, their causal links remain obscure. Herein, we performed a Mendelian randomization (MR) analysis to analyze the causality between systemic inflammatory proteins and IBD. Genetic variants related to systemic inflammatory proteins were extracted from a meta-analysis of genome-wide association study (GWAS) data of 8293 European participants. Summary statistics of IBD diverse subtypes were obtained from the international IBD genetic consortium (IIBDGC). We

BACKGROUND/PURPOSE: Although metabolic dysfunction-associated steatotic liver disease (MASLD) has been proposed to replace the diagnosis of non-alcoholic fatty liver disease (NAFLD) with new diagnostic criteria since 2023, the genetic predisposition of MASLD remains to be explored.
METHODS: Participants with data of genome-wide association studies (GWAS) in the Taiwan Biobank database were collected. Patients with missing data, positive for HBsAg, anti-HCV, and alcohol drinking history were excluded. MASLD was defined if having hepatic steatosis on ultrasound, plus at least one of cardiometab

BACKGROUND: Frailty, defined as a phenotype of decreased physiological reserves and diminished ability to respond to stressors, has been linked to the development of chronic diseases. Epidemiological evidence connecting frailty to non-alcoholic fatty liver disease (NAFLD) and cirrhosis risks remain sparse. We aimed to assess the longitudinal associations of frailty with the risks of severe NAFLD and cirrhosis in middle-aged to older adults and further explore the modification role of genetic risk on these associations.
METHODS: This study included a total of 398 386 participants from the UK B

Europace : European pacing, arrhythmias, and cardiac electrophysiology : journal of the working groups on cardiac pacing, arrhythmias, and cardiac cellular electrophysiology of the European Society of Cardiology2017eng
AIMS: Coronary artery disease accounts for the majority of sudden cardiac deaths (SCD) in the older population whereas cardiomyopathies and arrhythmogenic abnormalities predominate in younger SCD victims (<35 years) with a significant genetic component. The elucidation of the pathogenetic cause of death might be relevant for the prevention of further deaths within affected families. Aim of this study was to determine the portion of underlying genetic heart diseases among unexplained putative SCD cases from a large German forensic department.
METHODS AND RESULTS: We included 10 forensic cases

This study aimed to explore the association between dietary fiber intake and the risk of metabolic dysfunction-associated steatotic liver disease (MASLD), as well as liver fat content, while considering genetic predispositions of MASLD, gut microbial abundance, and butyrate levels. This study analyzed data from 190,276 participants in the UK Biobank. Dietary fiber intake was assessed using 24-h dietary recall. MASLD cases were diagnosed through hospital admission records and death registries, and liver fat content was measured via magnetic resonance imaging. The genetic predispositions of MASL

BACKGROUND: Previous studies suggest that outdoor sunlight exposure was associated with a lower risk of Parkinson’s disease (PD). However, the interaction of genetic predisposition and the potential role of lifestyle risk factors in mediating this association remains unclear from prospective evidence. METHODS: A cohort study based on the UK Biobank enrolled participants between 2006 and 2010, with the latest follow-up in November 2022. In the prospective population-based study 375,599 UK adults aged 37–73 years were enrolled. The outdoor light time was assessed using a questionnaire survey to

BACKGROUND: Little is known about the role that combined sleep behaviors play in the association with chronic liver disease (CLD) risk.
METHODS: We included 408,560 participants initially free of CLD from the UK Biobank. A healthy sleep pattern was defined by early chronotype, sleep duration of 7-8 h/day, no insomnia, no snoring, and no excessive daytime sleepiness. Cox regression models were used to examine the association of healthy sleep pattern with incident CLD and their interaction with PNPLA3 genetic risk.
RESULTS: During a median 12.5 years of follow-up, we documented 10,915 incident

Hypertension, cardiovascular disease and kidney failure are associated with persistent hyperglycaemia and the subsequent development of nephropathy in people with diabetes. Diabetic nephropathy is associated with widespread vascular disease affecting both the kidney and the heart from an early stage. However, the risk of diabetic nephropathy in people with type 1 diabetes is strongly genetically determined, as documented in familial transmission studies. The search for the underlying genes has been extensive, using specific hypotheses, sibling linkage studies and genome-wide association studie

BACKGROUND: Metabolic risk factors have been shown to be associated with severe liver disease (SLD) in Chinese populations. However, there is limited evidence on the combined impact of these factors, or the genetic variants associated with SLD.
OBJECTIVES: We examined the associations of combined metabolic risk factors with risks of SLD, both overall and by genetic predisposition to SLD.
METHODS: The study population involved 486,828 participants of the prospective China Kadoorie Biobank aged 30-79 years from 10 diverse areas in China without a history of cancer or liver disease at baseline.

INTRODUCTION: Chronic beryllium disease (CBD) is an occupational lung disease caused by the inhalation of beryllium dust, fumes or metallic salts.
CURRENT DATA: Beryllium affects the lungs via particles deposited in the pulmonary alveoli. These are ingested by alveolar macrophages which act as antigen presenting cells to CD4+ T lymphocytes. T lymphocytes proliferate in response to beryllium antigens and combined with macrophages produce numerous epithelioid granulomas with the release of inflammatory cytokines (IFNgamma, IL-2, TNFalpha and IL6) and growth factors. Beryllium induces macrophage

Recombinant tumor necrosis factor alpha (TNF-alpha) administration significantly delayed the development of lupuslike nephritis in the New Zealand black x New Zealand white (NZB x NZW)F1 and to a lesser extent in the MRL-lpr/lpr model systems. TNF-alpha treatment was effective when treatment was initiated at 2, 3, or 4 months of age but was ineffective if initiated as late as 6.5 months of age. Treatment of (NZB x NZW)F1 mice for 3 months was more effective than treatment continued for 6 months. Anti-TNF-alpha antibodies did not develop in these mice. Flow microfluorometry analysis showed no m

Background and Objectives: Celiac disease (CD) is a multifactorial immune-mediated disorder, triggered by the ingestion of gluten in genetically-predisposed subjects carrying MHC-DQ2 and -DQ8 heterodimers, which are encoded by four HLA-DQ allelic variants, overall. This meta-analysis aims at providing further epidemiological support to the predominant relevance of one specific allele, namely HLA-DQB1*02, in the predisposition and genetic risk of CD. Materials and Methods: We performed a search of MEDLINE/PubMed, Embase, Web of Science, and Scopus, retrieving all publications (case-control stud

The Georgia Cardiovascular Twin Study is a longitudinal study of biobehavioral antecedents of cardiovascular disease in youth and young adults, including around 500 twin pairs with roughly equal numbers of African Americans and European Americans. Focus of study includes the longitudinal change in relative influence of genetic and environmental factors (especially chronic stress) on development of risk factors for cardiovascular disease. Approaches include quantitative genetic modeling of phenotypic twin data as well as the examination of the influence of polymorphic variation in candidate gen

While prior research has indicated a possible association between serum 25 hydroxyvitamin D [25(OH)D] and celiac disease (CD), key questions remain unresolved without clear consensus. To assess association, we perform Mendelian randomization and a systematic review and meta-analysis of existing studies in European ancestry. A systematic review and meta-analysis performed by using both standardized mean difference and a Bayesian framework. Moreover, Mendelian randomization conducted via Generalized Summary-data-based Mendelian randomization (GSMR) and two-sample Mendelian randomization. Data on

BACKGOUND AND AIM: There are some common genetic features between celiac disease (CD) and diabetes mellitus type 1 (DM). However, the genetic risk factors have not been fully clarified for CD and the co-occurrence of CD and DM. KIR (killer immunoglobulin-like receptor) genes regulate the cytolitic activity of NK-cells and T lymphocytes. The aim of this study is to evaluate the contribution of KIR genes, KIR ligands, and combinations of KIR/ KIR ligands on the genetic predisposition to CD and co-occurrence of CD and DM.
MATERIAL AND METHODS: Forty six patients with CD (n = 46), 20 patients wit

BACKGROUND: The rest-activity rhythm amplitude (RARA), as a fundamental human behavior, has been linked to various health conditions. However, its causal relationship with ischemic heart disease (IHD), along with the potential modification by genetic predisposition, remains unclear.
OBJECTIVE: This study aimed to investigate the causal association between RARA and IHD using a triangulation approach that incorporated both observational and Mendelian randomization (MR) analyses, and to determine whether genetic predisposition modifies this relationship.
METHODS: First, a prospective cohort ana

INTRODUCTION: This study was done to further reveal the role of the innate immune system in celiac disease.
METHODS: Dendritic cells were matured from venous blood of patients with active or treated celiac disease and DQ2-DQ8-positive or negative controls. Dendritic cells were treated with a peptic-tryptic digest of gliadin (500 microg/ml) and their activation was analyzed by fluorescent-activated cell sorting analysis, cytokine secretion, and their ability to elicit T cell proliferation.
RESULTS AND DISCUSSION: Gliadin upregulated interleukin (IL)-6, IL-8, and IL-12 (p40) secretion in dendr

BACKGROUND: Depression and chronic pain are the two most important causes of disability (Global Burden of Disease Study 2013). They occur together more frequently than expected and both conditions have been shown to be co-morbid with cardiovascular disease. Although shared socio-demographic risk factors (e.g. gender, deprivation) might explain the co-morbidity of these three conditions, we hypothesised that these three long-term, highly prevalent conditions co-occur and may be due to shared familial risk, and/or genetic factors.
METHODS AND FINDINGS: We employed three different study designs
