The importance of genetic testing for dystonia patients and translational research.

Genetic testing through a variety of methods is a fundamental but underutilized approach for establishing the precise genetic diagnosis in patients with heritable forms of dystonia. Our knowledge of numerous dystonia-related genes, variants that they may contain, associated clinical presentations, and molecular disease mechanism may have significant translational potential for patients with genetically confirmed dystonia or their family members. Importantly, genetic testing permits the assembly of patient cohorts pertinent for dystonia-related research and developing therapeutics. Here we revi

[Advance in research on the genetic etiology of spontaneous intracerebral hemorrhage].

Spontaneous intracerebral hemorrhage (SICH) is a form of brain parenchymal hemorrhage caused by a variety of non-traumatic reasons, resulting in cerebral artery, veins or capillaries rupture. The etiology of SICH is variable, with hypertensive intracerebral hemorrhage being the most common, accounting for 60% ~ 81% of all cases. Cerebral amyloid angiopathy, drug use related hemorrhage, Moyamoya disease are also important causes of SICH. Previous studies showed that genetic factors play an important role in the pathogenesis of SICH. Here the genetic mechanisms of SICH and classification of its

INFRAVEC: research capacity for the implementation of genetic control of mosquitoes.

Mosquitoes represent a major and global cause of human suffering due to the diseases they transmit. These include parasitic diseases, i.e. malaria and filariasis, and viral infections such as dengue, encephalitis, and yellow fever. The threat of mosquito-borne diseases is not limited to tropical and subtropical regions of the world. Trade and climate changes have opened new niches to tropical vectors in temperate areas of the world, thus putting previously unaffected regions at risk of disease transmission. The most notable example is the spread of Aedes species, particularly the Asian tiger m

[Genetics of anxiety disorders. Current clinical and molecular research].

The authors give an overview of the present state of knowledge on the genetics of anxiety disorders. According to ICD-10 or DSM III/IV classification, anxiety disorders comprise panic disorder, generalized anxiety, phobias, and post-traumatic stress disorder. In the context of the conceptual change from psychodynamic anxiety neuroses to complex, multifactorial anxiety disorders, a summary of biological hypotheses of the pathogenesis of anxiety derived from pharmacotherapy, challenge tests, and animal model disorders is provided. The relevant findings from clinical genetic studies (twin and fam

Of replications and refutations: the status of Alzheimer's disease genetic research.

Alzheimer's disease (AD) is a genetically complex and heterogeneous disorder. To date, mutations in three genes (APP, PSEN1, PSEN2) have been described to cause familial early-onset AD. In addition, a common polymorphism in the gene encoding apolipoprotein E (APOE) has been associated with the more common late-onset form of the disease. However, many studies have shown that genetic factors other than APOE play an important role in late-onset AD. Along these lines, a recent report predicted the existence of at least four additional late-onset AD genes, one of which was estimated to have a much

[Development of genetic research in ophthalmology with special reference to Switzerland].

The relationship between ophthalmology and genetics has always been a very active one, and existed even before Mendel's laws of inheritance were known. The pedigree method in particular yielded satisfactory results even to the first researchers. Later, Helmholtz's invention of the ophthalmoscope (in 1851) and the rediscovery of Mendel's laws of heredity (in 1900) made major contributions to the progress of human genetics in ophthalmology. Credit is due to J. F. Horner of Zurich (1831-1886) for having first established the X-chromosomal inheritance of colorblindness. Subsequently, the developme

The Danish Lymphoid Cancer Research (DALY-CARE): Genetic Cohort Profile.

PURPOSE: The Danish Lymphoid Cancer Research (DALY-CARE) Genetic Cohort was established to support research into how genetic factors influence clinical outcomes in lymphoid cancers (LCs), including disease progression, treatment response, toxicity, and survival. Individual-level genetic data were combined with detailed clinical information from national health registers, hospital-based electronic health records (EHR), laboratory data, and pathology reports. The cohort enables large-scale studies of genetic susceptibility, disease course, and therapy-related outcomes in LCs and provides a platf

Global Perspectives on Returning Genetic Research Results in Parkinson Disease.

BACKGROUND AND OBJECTIVES: In the era of precision medicine, genetic test results have become increasingly relevant in the care of patients with Parkinson disease (PD). While large research consortia are performing widespread research genetic testing to accelerate discoveries, debate continues about whether, and to what extent, the results should be returned to patients. Ethically, it is imperative to keep participants informed, especially when findings are potentially actionable. However, research testing may not hold the same standards required from clinical diagnostic laboratories and hold

Genetic animal modeling for idiopathic scoliosis research: history and considerations.

PURPOSE: Idiopathic scoliosis (IS) is defined as a structural lateral spinal curvature ≥ 10° in otherwise healthy children and is the most common pediatric spinal deformity. IS is known to have a strong genetic component; however, the underlying etiology is still largely unknown. Animal models have been used historically to both understand and develop treatments for human disease, including within the context of IS. This intended audience for this review is clinicians in the fields of musculoskeletal surgery and research. METHODS: In this review article, we synthesize current literature of ge

IL-6 polymorphisms: a useful genetic tool for inflammation research?

In 1998, we described a novel polymorphism in the promoter (G>C, rs1800795) of the IL-6 (IL6) gene. The common allele, G, exhibited higher transcriptional activity in gene reporter assays and was associated with higher serum IL-6 levels in a small cohort of healthy subjects. We explored the ethnic distribution of these alleles and found significant differences among people of mixed European descent, Africans, and Gujarati Asians. Disease association was established in a cohort of 92 children of mixed European descent from the United Kingdom with systemic juvenile idiopathic arthritis (sJIA), w

Defining nicotine dependence for genetic research: evidence from Australian twins.

BACKGROUND: Whether current criteria used to define nicotine dependence are informative for genetic research is an important empirical question. The authors used items of the DSM-IV and of the Heaviness of Smoking Index to characterize the nicotine dependence phenotype and to identify salient symptoms in a genetically informative community sample of Australian young adult female and male twins. METHOD: Phenotypic and genetic factor analyses were performed on nine dependence symptoms (the seven DSM-IV substance dependence criteria and the two Heaviness of Smoking Index (HSI) items derived from

Recent Advances and Future Directions in Alzheimer's Disease Genetic Research.

Alzheimer's disease (AD) is a complex neurodegenerative condition which, despite its high prevalence and socioeconomic impact on the world, has an etiology that remains poorly understood. The genetic causes of AD are complex and have been continuously studied for decades. They range from rare pathogenic, highly penetrant mutations in early-onset (EOAD) forms, which account for 5% of the cases to multiple-risk alleles across different genes in late-onset (LOAD) forms. Monogenic causes of EOAD allocate within APP, PSEN1, and PSEN2 genes in 10-15% of cases. The most significant risk factor in LOA

[Advances in genetic research on Non-syndromic congenital joint synostosis].

Congenital joint synostosis (CJS) is a functional impairment resulting from failure in joint morphogenesis during embryonic development. Clinically, it may be classified as syndromic (sCJS) and non-syndromic (nsCJS) disorders. Common sCJS include chromosomal disorders such as Klinefelter syndrome and single-gene disorders like Apert/Pfeiffer/Crouzon syndromes, Holt-Oram syndrome, Ehlers-Danlos syndrome, and Radial-ulnar synostosis with thrombocytopenia, presenting with multiple system/organ anomalies. By contrast, nsCJS manifest with only joint abnormalities, affecting one or multiple joints.

Canadian Research Ethics Board Leadership Attitudes to the Return of Genetic Research Results to Individuals and Their Families.

Genomic research may uncover results that have direct actionable benefit to the individual. An emerging debate is the degree to which researchers may have responsibility to offer results to the biological relatives of the research participant. In a companion study to one carried out in the United States, we describe the attitudes of Canadian Research Ethics Board (REB) chairs to this issue and their opinions as to the role of the REB in developing related policy.

Research participants and stakeholders' views on feedback of genetic research findings: a qualitative study of the H3Africa Kidney Disease Research Network in Ghana.

BACKGROUND: There is evidence that variants of the Apolipoprotein L1 gene in Africans and people of African descent increase the risk of developing chronic kidney disease (CKD). A study conducted by the H3Africa Kidney Disease Research Network showed that 28.2% of the Ghanaian study population carried 2 APOL1 high-risk variants. Relatives of patients with CKD may be at increased risk of developing CKD. Researchers of this project are faced with an ethical dilemma of whether, study participants and their relatives should be informed about their risk of developing CKD. The aim of this study was

dbGVOSCC: a comprehensive database of genetic variation for systems genetics research on oral squamous cell carcinoma.

INTRODUCTION: Oral squamous cell carcinoma (OSCC) is a highly aggressive malignancy of the oral epithelium, marked by a high rate of lymph node metastasis and a profound negative impact on patients' quality of life. Despite its severity, no routine screening program currently exists for OSCC. To address the genetic heterogeneity underlying OSCC, we have developed a database of genetic variation in oral squamous cell carcinoma (dbGVOSCC; http://www.sysbio.org.cn/dbGVOSCC/). METHODS: OSCC literature (1991-2024) was queried from PubMed and screened manually and via PubTator, following predefined
