Effects of Defined Pig Microbiota on Acute Salmonellosis in Gnotobiotic Piglets.

Salmonella enterica subsp. enterica serovar Typhimurium (S. Typhimurium) is a ubiquitous Salmonella serovar that causes enterocolitis in humans, livestock, poultry, and some pets. Gnotobiotic animals are suitable for studying the development of initial colonization in the gastrointestinal tract, host-microbiota cross-talk, and interactions among microbiota strains. To clarify the impact of pre-colonization of the germ-free piglets with defined pig microbiota (DPM) on the subsequent infection with S. Typhimurium LT2 (LT2), we focused on the acute host immune response. Colonization with DPM did

Evaluation of disinfectants and antiseptics to eliminate bacteria from the surface of turkey eggs and hatch gnotobiotic poults.

Bird eggs are in contact with intestinal microbiota at or after oviposition, but are protected from bacterial translocation by a glycoprotein cuticle layer, the shell, and internal membranes. In a preliminary study, turkey eggs were hatched in a germ-free environment. Firmicutes 16S rRNA gene was detected in the cecal microbiota of hatched poults, suggesting that poults may acquire spore-formers by exposure to shell contents during hatching. Generating gnotobiotic poults for research requires elimination of bacteria from the egg's surface without damaging the developing embryo. The ability of

Gut microbiota promote hematopoiesis to control bacterial infection.

The commensal microbiota impacts specific immune cell populations and their functions at peripheral sites, such as gut mucosal tissues. However, it remains unknown whether gut microbiota control immunity through regulation of hematopoiesis at primary immune sites. We reveal that germ-free mice display reduced proportions and differentiation potential of specific myeloid cell progenitors of both yolk sac and bone marrow origin. Homeostatic innate immune defects may lead to impaired early responses to pathogens. Indeed, following systemic infection with Listeria monocytogenes, germ-free and oral

Gnotobiotic transgenic mice reveal that transmission of Helicobacter pylori is facilitated by loss of acid-producing parietal cells in donors and recipients.

Helicobacter pylori is acquired during childhood, but its mode of transmission remains unclear. A genotyped H. pylori isolate (Hp1) that expresses two classes of adhesins was introduced into the stomachs of three types of germ-free FVB/N mice to model factors that may affect spread of H. pylori in humans. Normal mice represented human hosts with normal gastric acid production. Transgenic animals expressing human alpha-1,3/4-fucosyltransferase in their gastric pit cells represented humans with normal acid production and the commonly encountered Lewis(b) histo-blood group receptor for the bacter

Enteric campylobacter infection in gnotobiotic calves and lambs.

Gnotobiotic calves and lambs were infected orally with Campylobacter jejuni, C coli or C hyointestinalis to assess pathogenicity. All animals were successfully colonised and excreted mucoid faeces but showed no other clinical signs. Campylobacters colonised the large intestine better than the small intestine, in which bacterial numbers decreased with time after infection. Campylobacters were found occasionally in the lumen of crypts in close proximity to epithelial cells and included in a mucus-like material. Lesions were mostly in the large intestine in calves whereas in lambs they were prese

Isolation and characterization of a mucin-degrading strain of Peptostreptococcus from rat intestinal tract.

A mucin-degrading microorganism was isolated from the intestinal tract by serial sectioning from the serosal side of the caecum wall from a conventional rat. The ability of degrading the intestinal water-soluble mucin was present both in vivo after monocontamination of germ-free rats and in vitro, when adding the microbe to Mucin medium. The morphology, Gram-positive cocci single or in short chains and the very weak biochemical activities allow us to place this strain in the species Peptostreptococcus micros.

Microbial flora in the digestive tract and action of lactose on mineral metabolism.

Germ-free (GF) and conventional (CV) adult rats were given for 4 weeks a semi-synthetic diet containing 10 p. 100 lactose (L) or no lactose (LO). The axenic state had an unfavourable effect on sodium and potassium absorption. On the contrary, it increased the absorption and retention of calcium, phosphorus and magnesium. The favourable effect of lactose on magnesium metabolism was not different in GF and CV rats. On the contrary, lactose increased the absorption and retention of phosphorus and calcium more in GF than in CV animals. It augmented the amount of calcium present in a soluble, ultra

Histamine and 5-hydroxytryptamine in the intestinal tract of germ-free animals, animals harbouring one microbial species and conventional animals.

Histamine and 5-hydroxytryptamine were determined in the intestinal tract of germfree and conventional rats and mice. Comparable histamine data were collected in Clostridium perfringens mono-associated rats, while 5-hydroxytryptamine determinations were extended to include the chicken. In rats and mice harbouring an intestinal microflora, bacterial formation of histamine occurs mainly in the caecum. Compared to values found in germ-free animals, histamine levels in the wall of the small intestine of the conventional animal tend to be higher, though in the rat the data are not consistent. Mono-

Placental endocrine function is controlled by maternal gut Bifidobacterium in germ-free mice.

BACKGROUND: Recent studies have shown that the maternal gut microbiota can regulate placental growth, particularly the transport region, in association with fetal growth. However, the specific role of certain microorganisms in modulating the hormonal production of the placenta, which is critical for supporting fetal development and maintaining a healthy pregnancy, remains largely unexplored. In this context, the objective of this study is to determine whether the maternal colonisation with the early life gut bacterium Bifidobacterium breve UCC2003 regulates placental endocrine function. Identification of a microbial sub-community from the feral chicken gut that reduces Salmonella colonization and improves gut health in a gnotobiotic chicken model.

A complex microbial community in the gut may prevent the colonization of enteric pathogens such as Salmonella. Some individual or a combination of species in the gut may confer colonization resistance against Salmonella. To gain a better understanding of the colonization resistance against Salmonella enterica, we isolated a library of 1,300 bacterial strains from feral chicken gut microbiota which represented a total of 51 species. Using a co-culture assay, we screened the representative species from this library and identified 30 species that inhibited Salmonella enterica subspecies enterica

Mechanisms of Kwashiorkor-Associated Immune Suppression: Insights From Human, Mouse, and Pig Studies.

Malnutrition refers to inadequate energy and/or nutrient intake. Malnutrition exhibits a bidirectional relationship with infections whereby malnutrition increases risk of infections that further aggravates malnutrition. Severe malnutrition (SM) is the main cause of secondary immune deficiency and mortality among children in developing countries. SM can manifest as marasmus (non-edematous), observed most often (68.6% of all malnutrition cases), kwashiorkor (edematous), detected in 23.8% of cases, and marasmic kwashiorkor, identified in ~7.6% of SM cases. Marasmus and kwashiorkor occur due to ca

Carbohydrate metabolism is essential for the colonization of Streptococcus thermophilus in the digestive tract of gnotobiotic rats.

Streptococcus thermophilus is the archetype of lactose-adapted bacterium and so far, its sugar metabolism has been mainly investigated in vitro. The objective of this work was to study the impact of lactose and lactose permease on S. thermophilus physiology in the gastrointestinal tract (GIT) of gnotobiotic rats. We used rats mono-associated with LMD-9 strain and receiving 4.5% lactose. This model allowed the analysis of colonization curves of LMD-9, its metabolic profile, its production of lactate and its interaction with the colon epithelium. Lactose induced a rapid and high level of S. ther

Modulation of gut wall paf-acether and precursors by intestinal microflora.

Comparison between holoxenic and axenic mice led to the conclusion that the presence of an intestinal microflora produced a decrease in wall paf in conventional mouse caecum, whereas an increase in wall lyso-paf and alkyl-acyl-glycerophosphocholine (A-A-GPC) content was noticed. By contrast, the presence of flora had no significant incidence on wall paf, lyso-paf and A-A-GPC content of conventional mouse jejunum. Thus, the modulation of gut wall phospholipid composition by intestinal microflora is evidenced for the first time.

Pathology of cloacal bursae of gnotobiotic turkeys orally inoculated with Escherichia coli.

Cloacal bursae from three-week-old gnotobiotic turkeys were examined by light and electron microscopy and bacteriologic techniques at 1, 2, 5, 8, 12, and 16 days after oral inoculation of highly virulent (group 1) and weakly virulent (group 2) Escherichia coli. In both groups a significant decrease in follicular volume and increase in interstitial volume were associated with infection. Follicular cortical, follicular medullary and total follicular transectional areas increased with time in inoculated and control turkeys. In group 1, granulocytic inflammation developed in bursae on day 1 and di

Resistance of gnotobiotic nude mice to Listeria infection.

Unlike the acute and short-term course of experimental listeriosis in phenotypically normal (nu/+) mice, Listeria monocytogenes infection in conventionally raised athymic Nude mice results in chronic inflammation with persistence of bacteria and lesions in the internal organs such as liver and spleen. Although it could be shown that germfree Nude mice had no intestinal microbial flora and lacked measurable serum immunoglobulins, neither the course nor the histomorphology of experimental listeriosis differed significantly from that of their conventionally raised littermates. The results indicat

Inoculation of neonatal gnotobiotic dogs with a canine rotavirus.

Neonatal gnotobiotic dogs were inoculated orally with a rotavirus isolated from a pup with fatal diarrhea, and in the gnotobiotic dogs, diarrhea was observed between postinoculation hours (PIH) 20 and 24. The diarrhea persisted through PIH 154, and inoculated pups had clinical signs of dehydration after PIH 24. Negative-contrast electron microscopy of the feces from inoculated pups revealed rotavirus particles from PIH 12 through 154. Using an indirect-fluorescent antibody test, serum rotavirus antibody was detected in inoculated pups by PIH 96. In the duodenum, jejunum, and ileum of inoculate
