Germinal center immune dynamics: challenges for effective vaccination in the elderly.

Follicular (F) and germinal center (GC) immune dynamics are crucial in generating pathogen-specific antibodies with high affinity and neutralizing activity capable of combating infections. GCs are populated by highly differentiated CD4 T-cell and B-cell populations with unique phenotypes, functions, and molecular signatures. Aging, which is associated with a state of "immunosenescence," is characterized by compromised B-cell responses to infections and reduced vaccine efficacy, pointing to altered GC immunoreactivity and function. Therefore, there is a need for novel, improved approaches to st

Escalation of Germinal Center Responses in Chronic Litomosoides sigmodontis Filarial Infection.

T follicular helper (TFH) cells are the primary CD4+ T helper cell subset providing help to B cells for efficient antibody responses in vaccination, allergy, autoimmunity, and infectious diseases. Despite their critical involvement in immunity, TFH cells' specific role in filarial infections remains unclear. Using the rodent filarial model Litomosoides sigmodontis, we examined TFH and germinal center (GC) B cell responses in lung-draining mediastinal lymph nodes (medLNs) over a 110-day infection period in naive and infected wildtype (WT) BALB/c mice, as well as eosinophil-deficient dblGATA mic

The transcription complex p52-ETS1 is essential for germinal center formation.

The NF-κB family comprises five transcription factors (RELA, RELB, C-REL, NF-κB1 (p50) and NF-κB2 (p52)) that form homo- or heterodimers among themselves to regulate gene expression by binding DNA. Here we show that p52 activates transcription without directly binding DNA but as a heterotetrameric complex with ETS1, a transcription factor outside the NF-κB family. By generating a knock-in mouse model (Nfkb2ki/ki) with three mutated residues on p52 required for its interaction with ETS1, but not RELB, we demonstrate that the p52-ETS1 complex regulates the expression of transcription factors OCT

Compromised steady-state germinal center activity with age in nonhuman primates.

Age-related reductions in vaccine-induced B cells in aging indicate that germinal centers (GCs), the anatomical site where the development of humoral responses takes place, may lose efficacy with age. We have investigated the baseline follicular and GC composition in nonhuman primates (NHPs) with respect to their age. There was a marked reduction in follicular area in old animals. We found significantly lower normalized numbers of follicular PD1hi CD4 T (Tfh) and proliferating (Ki67hi ) GC B cells with aging, a profile associated with significantly higher numbers of potential follicular suppre

Chronic psychological stress impairs germinal center response by repressing miR-155.

Germinal centers (GC) are vital to adaptive immunity. BCL6 and miR-155 are implicated in control of GC reaction and lymphomagenesis. FBXO11 causes BCL6 degradation through ubiquitination in B-cell lymphomas. Chronic psychological stress is known to drive immunosuppression. Corticosterone (CORT) is an adrenal hormone expressed in response to stress and can similarly impair immune functions. However, whether GC formation is disrupted by chronic psychological stress and its molecular mechanism remain to be elucidated. To address this issue, we established a GC formation model in vivo, and a GC B

A case of progressively transformed germinal center-type IgG4-related lymphadenopathy.

Progressively transformed germinal centers (PTGC), a lymph node process unfamiliar to most otolaryngologists, is a morphological variant of reactive lymphofollicular hyperplasia of lymph nodes. Immunoglobulin (Ig)G4-related disease (IgG4-RD) is a newly identified condition, characterized by hyper-IgG4-γ-globulinemia and mass-forming or hypertrophic lesions associated with infiltration of IgG4(+) plasma cells in the affected organs. Recently, a case study of PTGC was reported that fulfilled the diagnostic criteria of IgG4-RD (IgG4(+) PTGC) [1]. A 68-year-old male was referred to our hospital wi

Cutting Edge: Redox Signaling Hypersensitivity Distinguishes Human Germinal Center B Cells.

Differences in the quality of BCR signaling control key steps of B cell maturation and differentiation. Endogenously produced H2O2 is thought to fine tune the level of BCR signaling by reversibly inhibiting phosphatases. However, relatively little is known about how B cells at different stages sense and respond to such redox cues. In this study, we used phospho-specific flow cytometry and high-dimensional mass cytometry (CyTOF) to compare BCR signaling responses in mature human tonsillar B cells undergoing germinal center (GC) reactions. GC B cells, in contrast to mature naive B cells, memory

Downregulation of FOXP1 is required during germinal center B-cell function.

B-cell maturation and germinal center (GC) formation are dependent on the interplay between BCL6 and other transcriptional regulators. FOXP1 is a transcription factor that regulates early B-cell development, but whether it plays a role in mature B cells is unknown. Analysis of human tonsillar B-cell subpopulations revealed that FOXP1 shows the opposite expression pattern to BCL6, suggesting that FOXP1 regulates the transition from resting follicular B cell to activated GC B cell. Chromatin immunoprecipitation-on-chip and gene expression assays on B cells indicated that FOXP1 acts as a transcri

Germinal center B cells are dispensable in prion transport and neuroinvasion.

Transmissible spongiform encephalopathies (TSEs) are fatal neurodegenerative diseases of animals and humans. Many TSEs are initiated by prion replication in the lymphoreticular system (LRS). The cellular and molecular prerequisites for prion trafficking within the LRS are not fully understood. Here we have manipulated CD40 and its ligand to investigate whether genetic or pharmacological ablation of germinal center B cells (GCBs), which migrate into and out of germinal centers, influences prion pathogenesis. In contrast to previous reports, no alteration of prion pathogenesis was detected in mi

Nuclear and cytoplasmic AID in extrafollicular and germinal center B cells.

Activation-induced cytidine deaminase (AID) is necessary for immunoglobulin somatic hypermutation (SHM) and class switch recombination (CSR) in T-dependent immune response in germinal centers (GCs). The structural similarity of AID with RNA-editing enzymes and its largely cytoplasmic location have fueled controversial views of its mode of interaction with DNA. We show that AID, a mature B-cell-restricted cytoplasmic antigen, is relocated into the nucleus in 2.5% of CDKN1B(-), CCNB1(-) GC cells. The GC dark zone and the outer zone (OZ), but not the light zone, contain nuclear and cytoplasmic AI

Germinal center B cell apoptosis requires both caspase and cathepsin activity.

Follicular dendritic cells (FDCs) select B cells during germinal center (GC) reactions. The B cells that are able to bind to the FDCs receive a signal that leads to the termination of endonuclease activity in the nuclei of those B cells. This signal must be in addition to the signals transferred through the cross-linkage of the B cell receptors and signals resulting from the interactions of the adhesion molecules lymphocyte function-associated Ag-1 and very late Ag-4 with ICAM-1 and VCAM-1, respectively. In this report, we present evidence that the FDCs silence all apoptotic processes in GC B

Spatial compartmentalization shapes distinct germinal center states during respiratory viral infection.

BACKGROUND: Germinal centers (GCs) typically arise within the lymph node cortex, where follicular dendritic cell (FDC) networks and follicular helper T cells coordinate B-cell expansion, affinity maturation, and durable immunity. Whether proliferating B-cell aggregates in the medulla engage bona fide GC programs or instead represent a distinct GC-like differentiation state remains unresolved. METHODS: In this study, using mediastinal lymph nodes from SARS-CoV-2-infected cynomolgus macaques, we combined immunofluorescence with Xenium spatial transcriptomics to identify LYVE1+ sinus-associated

The ubiquitin system in normal and infected germinal center B cells.

The ubiquitin system is a highly regulated cellular mechanism that plays a central role in germinal center (GC) B cells, essential for protective humoral immunity and the site of gammaherpesvirus latency establishment. GC B cells undergo extraordinary levels of proliferation, DNA damage, and apoptosis prior to differentiating to long-lived memory or plasma cells, and ubiquitin pathways underpin these activities. Oncogenic gammaherpesviruses infect B cells and drive them through a GC activation program. Viral programs exploit ubiquitin biology to establish latency in GC B cells and persist for

Cathepsin S regulates germinal center reactions and anti-tumor humoral responses.

B cells mature into antibody-secreting cells by antigen processing and presentation and undergo productive germinal center (GC) reactions in secondary lymphoid organs. In solid tumors, antigen presentation and local anti-tumor humoral immunity can arise through the formation of GC-like tertiary lymphoid structures (TLS). Here, we show that cathepsin S activity is essential to form mature GC and TLS by regulating the communication of B cells with CD4+ Tfh cells and follicular dendritic cells. The absence of cathepsin S activity impairs B cells' affinity maturation and high-affinity antibody pro

Resolving the cellular profile of germinal center-analogous microstructures in zebrafish.

Teleost fish lack canonical germinal centers (GCs); however, they can mount antigen-specific immune responses, raising longstanding questions about how antibody affinity maturation is organized in cold-blooded vertebrates. Melanomacrophage centers (MMCs) have been proposed as GC analogues, yet their cellular composition and immune organization remain poorly defined. To address this, we isolated autofluorescent MMCs from zebrafish spleens and performed single-cell RNA sequencing to characterize their immune and stromal cell populations at high resolution. Our analysis identified centroblast-lik

Elevated enteric putrescine suppresses differentiation of intestinal germinal center B cells.

The dysregulation of B cell maturation and putrescine metabolism has been implicated in various diseases. However, the causal relationship between them and the underlying mechanisms remain unclear. In this study, we investigated the impact of exogenous putrescine on B cell differentiation in the intestinal microenvironment. Our results demonstrated that administration of exogenous putrescine significantly impaired the proportion of germinal center B (GC B) cells in Peyer's patches (PPs) and lamina propria. Through integration of bulk RNA sequencing and single-cell RNA sequencing (scRNA-seq), w

Accelerated involution of germinal center in palatine tonsils in IgA nephropathy.

BACKGROUND: Altered immunological responses in the palatine tonsils may be involved in the pathogenesis of IgA nephropathy (IgAN). The germinal center serves as the site for antigen-specific humoral immune responses in the palatine tonsils. Germinal center involution is frequently observed in the palatine tonsils of IgAN (IgAN tonsils). However, the pathogenic significance of these characteristic changes remains unclear. This study aimed to investigate the morphological changes in secondary lymphoid follicles in IgAN tonsils and to evaluate the correlation between the morphometric results and

Belatacept inhibit human B cell germinal center development in immunodeficient mice.

The humoral response mediated by alloantibodies directed against donor HLA molecules (DSAs) is one of the main causes of graft loss in kidney transplantation. Understanding the pathophysiology leading to humoral kidney rejection as the development of therapeutic tools is therefore a main objective in the field of solid organ transplantation and necessitate adapted experimental models. Among the immunosuppressive agents used in renal transplantation, belatacept, a fusion protein targeting T costimulatory molecules has shown its ability to prevent more efficiently the secretion of DSA by differe

Type I interferon induces CXCL13 to support ectopic germinal center formation.

Ectopic lymphoid structures form in a wide range of inflammatory conditions, including infection, autoimmune disease, and cancer. In the context of infection, this response can be beneficial for the host: influenza A virus infection-induced pulmonary ectopic germinal centers give rise to more broadly cross-reactive antibody responses, thereby generating cross-strain protection. However, despite the ubiquity of ectopic lymphoid structures and their role in both health and disease, little is known about the mechanisms by which inflammation is able to convert a peripheral tissue into one that res

KLHL6 Is Preferentially Expressed in Germinal Center-Derived B-Cell Lymphomas.

OBJECTIVES: KLHL6 is a recently described BTB-Kelch protein with selective expression in lymphoid tissues and is most strongly expressed in germinal center B cells. METHODS: Using gene expression profiling as well as immunohistochemistry with an anti-KLHL6 monoclonal antibody, we have characterized the expression of this molecule in normal and neoplastic tissues. Protein expression was evaluated in 1,058 hematopoietic neoplasms. RESULTS: Consistent with its discovery as a germinal center marker, KLHL6 was positive mainly in B-cell neoplasms of germinal center derivation, including 95% of fol
