Neurotherapeutic effects of Ginkgo biloba extract and its terpene trilactone, ginkgolide B, on sciatic crush injury model: A new evidence.

Ginkgo biloba leaves extract (GBE) was subjected to neuroprotective-guided fractionation to produce eleven fractions with different polarities and constituents. The intermediate polar fraction was shown to be terpene trilactones-enriched fraction (TEGBE). Out of this fraction, pure ginkgolide B (G-B) was further purified and identified based on its spectral data. The effects of GBE and TEGBE were evaluated in comparison to that of G-B in the crush sciatic nerve injury rat model. To evaluate the neuroprotective effects, sixty Wistar male rats were randomly allocated into 6 groups: naive, sham,

Ginkgolide A alleviated liver fibrosis by modulating PI3K/AKT/NF-κB signaling to inhibit M1 macrophage polarization and macrophage-HSC crosstalk.

Liver fibrosis (LF) is a progressive pathological process driven by chronic hepatocyte injury and aberrant wound healing, culminating in excessive extracellular matrix (ECM) deposition primarily mediated by activated hepatic stellate cells (HSCs). A dysregulated hepatic immune microenvironment, orchestrated in large part by macrophages, plays a critical role in driving HSCs activation. Ginkgolide A (GA), a bioactive diterpenoid lactone from Ginkgo biloba leaves, possesses potent anti-inflammatory and antioxidant activities. However, its role and underlying mechanism in LF, particularly via mod

Ginkgolide B alleviates traumatic brain injury in mice via inhibiting the HIF-1α/AKT/mTOR signaling pathway and modulating microglial polarization.

Traumatic brain injury (TBI) is commonly accompanied by aberrant microglial activation, uncontrolled neuroinflammation and subsequent neurological dysfunction. Ginkgolide B (GB) exerts prominent neuroprotective and anti-inflammatory activities; however, the precise molecular mechanism underlying its therapeutic efficacy against TBI remains poorly defined. The present study aimed to explore whether GB relieves TBI by modulating the HIF-1α/AKT/mTOR cascade and redirecting microglial polarization. A controlled cortical impact (CCI) model was established in mice followed by intraperitoneal GB admi

Inhibition of platelet-activating factor (PAF)-induced chemotaxis and PAF binding to human eosinophils and neutrophils by the specific ginkgolide-derived PAF antagonist, BN 52021.

We have investigated the effects of BN 52021 (a specific PAF antagonist derived from Ginkgo biloba) on PAF-induced human eosinophil and neutrophil chemotaxis. In response to an optimal concentration of PAF (10(-6) mol/L), the drug was significantly more potent (p less than 0.001) in inhibiting eosinophil as compared to neutrophil locomotion. These inhibitory effects were observed in a dose-dependent manner with a concentration of drug required to produce 50% inhibition of 7.0 (+/- 2.2) X 10(-6) mol/L and 2.3 (+/- 0.2) X 10(-5) mol/L for eosinophils and neutrophils, respectively. Involvement of platelet-activating factor (PAF) in cerebral post-ischemic phase in Mongolian gerbils.

Platelet-activating factor (PAF) is a potent mediator of anaphylaxis and shock. In addition, evidence for PAF participation in gastric, intestinal and heart post-ischemic phase has been recently demonstrated. Ginkgo biloba extracts improve cerebral metabolism and protect brain against hypoxic damage in various models of cerebral ischemia. Potent and specific antagonists of PAF have been found in Ginkgo biloba and termed Ginkgolides: BN 52020, BN 52021, BN 52022, BN 52024. We therefore undertook the investigation of the role of Ginkgolides in cerebral ischemia obtained by bilateral ligature of

Pharmacological studies supporting the therapeutic use of Ginkgo biloba extract for Alzheimer's disease.

The standardized Ginkgo biloba extract EGb 761(definition see editorial) has been shown to produce neuroprotective effects in different in vivo and in vitro models. Since EGb 761 is a complex mixture containing flavonoid glycosides, terpene lactones (non-flavone fraction) and various other constituents, the question arises as to which of these compounds mediates the protective activity of EGb 761. Previous studies have demonstrated that the non-flavone fraction was responsible for the antihypoxic activity of EGb 761. Thus, we examined the neuroprotective and anti-apoptotic ability of the main

Serum Pharmacochemistry-Guided DARTS-MS Profiling Reveals Potential Mechanisms of Caragana jubata Against Hypoxic Pulmonary Hypertension.

Hypoxic pulmonary hypertension (HPH) is a progressive vascular disease characterized by an abnormal increase in pulmonary arterial pressure resulting from pulmonary vasoconstriction and pulmonary vascular remodeling (PVR). Excessive proliferation and migration of pulmonary artery smooth muscle cells (PASMCs) are key drivers of PVR. Caragana jubata (Pall.) Poir. (C. jubata), known as "zuomaoxing" in Tibetan medicine, is traditionally used to treat blood-related disorders. However, the potential preventive and therapeutic effects of C. jubata on HPH remain unclear. Here, we integrated in vivo ph

LC-MS based strategy for chemical profiling and quantification of dispensing granules of Ginkgo biloba seeds.

Ginkgo biloba seeds have been used as a traditional Chinese medicine for hundreds of years to treat diseases such as cervicitis, cough, asthma and other lung diseases. As a novel form, the dispensing granules (GSDG) of Ginkgo biloba seeds have been widely employed in clinic. However, its chemical profiling is not yet clear, which has restricted in-depth research in many fields. In this study, a high performance liquid chromatography coupled with quardrupole time-of-flight mass spectrometry method was used for the component characteration with the help of accurate molecular weights, fragmentati
