Superovulation affects glucosamine trisaccharides in the glycocalyx of the rat endometrium at the time of implantation.

The presence of glucosamine trisaccharides in the glycocalyx of surface epithelial cells of the endometrium of hyperstimulated pregnant rats was compared with that of normal pregnant animals by means of lectin-avidin-ferritin histochemistry. The lectin Phytolacca americana was used. A statistically significant reduction in ferritin particles and hence, glucosamine trisaccharides, was found in the glycocalyx of the hyperstimulated animals when compared with that of the controls. This reduction could contribute to the failure of implantation in hyperstimulated animals.

Autoradiographic localizations of [35S]-sulfate and [3H]-glucosamine in the hamster ciliary epithelium.

The localization of [35S]-sulfate and [3H]-glucosamine in the hamster non-pigmented ciliary epithelium was studied by light and electron microscopic autoradiography. The radioactivity was concentrated in the Golgi complex of the non-pigmented ciliary epithelium 10 mins after injections of [35S]-sulfate or [3H]-glucosamine. Silver grains of both isotopes covered the cytoplasm 60 mins after the administration and were then associated with the basal infoldings. Radioactivity of [3H]-glucosamine was observed in the basal lamina of the non-pigmented ciliary epithelium and the ciliary zonule; howeve

Inhibition of glucosamine synthesis by salicylates, hydrocortisone and two non-ulcerogenic drugs.

The in vitro inhibition of the glucosamine synthetase activity of rat gastric mucosal homogenates by sodium salicylate and phenylbutazone has been previously reported and has been adduced as a possible mechanism for the ulcerogenic effect of these drugs. This inhibition has been confirmed in human gastric and colonic mucosal homogenates and it is also shown by acetylsalicylic acid and by hydrocortisone hemisuccinate. However, paracetamol and gentamicin, neither of which is known to be ulcerogenic, also inhibited the enzyme to a similar degree to the ulcergenic drugs. This refutes glucosamine s

Glucosamine Promotes Autophagy and Attenuates Hepatic Steatosis Via O-GlcNAcylation-Mediated Mechanisms.

Autophagy is a key cellular process regulating lipid turnover and maintaining hepatic homeostasis, and its impairment is closely associated with the pathogenesis of nonalcoholic fatty liver disease (NAFLD). In this study, we examined the effects of glucosamine (GlcN), a hexosamine biosynthetic pathway intermediate, on autophagy and lipid accumulation using both human hepatocellular carcinoma (HepG2) cells and a high-fat diet (HFD)-induced NAFLD mouse model. GlcN treatment led to a dose- and time-dependent increase in the expression of autophagy-related markers LC3 and p62 at both mRNA and prot

Synthesis and anti-inflammatory activities of two new N-acetyl glucosamine derivatives.

N-acetyl glucosamine (NAG) is a natural amino sugar found in various human tissues with previously described anti-inflammatory effects. Various chemical modifications of NAG have been made to promote its biomedical applications. In this study, we synthesized two bi-deoxygenated NAG, BNAG1 and BNAG2 and investigated their anti-inflammatory properties, using an in vivo and in vitro inflammation mouse model induced by lipopolysaccharide (LPS). Among the parent molecule NAG, BNAG1 and BNAG2, BNAG1 showed the highest inhibition against serum levels of IL-6 and TNF α and the leukocyte migration to l

Genetic variation in glucosamine metabolism contributes to the colonization resistance of Bacteroides to Salmonella.

The gut microbiota contributes to variation in susceptibility to enteric infection, but the underlying mechanisms remain poorly defined. Here, we leverage a natural model of disease resistance comparing Salmonella-resistant indigenous Tibetan chickens with susceptible commercial broilers to uncover conserved mechanisms of colonization resistance relevant to human health. We find that enrichment of Bacteroides, particularly B. salanitronis, is associated with resistance, and supplementation with B. salanitronis significantly reduces S. Enteritidis colonization in susceptible hosts. Mechanistica

Effect of Chitosan Degradation Products, Glucosamine and Chitosan Oligosaccharide, on Osteoclastic Differentiation.

Chitosan, a natural cationic polysaccharide derived from crustaceans and shellfish shells, is known for its advantageous biological properties, including biodegradability, biocompatibility, and antibacterial activity. Chitosan and its composite materials are studied for their potential for bone tissue repair. However, the effects of chitosan degradation products, glucosamine (GlcN) and chitosan oligosaccharide (COS), on osteoclasts remain unclear. If these chitosan degradation products promote osteoclastic differentiation, careful consideration is required for the use of chitosan and related m

Brain glycogen serves as a critical glucosamine cache required for protein glycosylation.

Glycosylation defects are a hallmark of many nervous system diseases. However, the molecular and metabolic basis for this pathology is not fully understood. In this study, we found that N-linked protein glycosylation in the brain is metabolically channeled to glucosamine metabolism through glycogenolysis. We discovered that glucosamine is an abundant constituent of brain glycogen, which functions as a glucosamine reservoir for multiple glycoconjugates. We demonstrated the enzymatic incorporation of glucosamine into glycogen by glycogen synthase, and the release by glycogen phosphorylase by bio

Effects of glucosamine supplements on painful temporomandibular joint osteoarthritis: A systematic review.

The purpose of this study was to systematically review the literature for studies that assessed the effects of glucosamine supplements (GS) on pain and maximum mouth opening (MMO) restriction compared to other therapies, placebo or no intervention on painful temporomandibular joint osteoarthritis (TMJ OA). Randomised controlled trials were selected in a two-phase process. Seven electronic databases, in addition to three grey literature databases, were searched. Risk of bias was assessed using the Cochrane Collaboration's tool for assessing risk of bias in randomised trials. Twelve potentially

Glucosamine for the Treatment of Osteoarthritis: The Time Has Come for Higher-Dose Trials.

Although clinical trials with glucosamine in osteoarthritis have yielded mixed results, leading to doubts about its efficacy, the utility of glucosamine for preventing joint destruction and inflammation is well documented in rodent models of arthritis, including models of spontaneous osteoarthritis. The benefit of oral glucosamine in adjuvant arthritis is markedly dose dependent, likely reflecting a modulation of tissue levels of UDP-N-acetylglucosamine that in turn influences mucopolysaccharide synthesis and the extent of protein O-GlcNAcylation. Importantly, the minimal oral dose of glucosam

Hepatotoxicity associated with glucosamine and chondroitin sulfate in patients with chronic liver disease.

Glucosamine and chondroitin sulfate are molecules involved in the formation of articular cartilage and are frequently used for symptom relief in patients with arthrosis. These molecules are well tolerated with scarce secondary effects. Very few cases of possible hepatotoxicity due to these substances have been described. The aim of this paper is to report the frequency of presumed glucosamine hepatotoxicity in patients with liver disease. A questionnaire was given to 151 consecutive patients with chronic liver disease of different etiology (mean age 59 years, 56.9% women) attended in an outpat

The efficacy of glucosamine and chondroitin sulfate in the treatment of osteoarthritis: are these saccharides drugs or nutraceuticals?

This review summarizes recent knowledge on the efficacy of glucosamine (GS) and/or chondroitin sulfate (CS) in the therapy of mild to moderate osteoarthritis (OA). OA, the most common joint disease is a significant source of disability, quality of life impairment and a considerable burden to any health care system. In the Czech Republic, glucosamine sulfate (GS) and chondroitin sulfate (CS) are available both as prescription drugs and as food supplements. Based on available data both are useful in the earlier stages of OA when combined with other modalities such as weight loss and exercises. T

The incorporation of glucosamine into enterobacterial core lipopolysaccharide: two enzymatic steps are required.

The core lipopolysaccharide (LPS) of Klebsiella pneumoniae is characterized by the presence of disaccharide alphaGlcN-(1,4)-alphaGalA attached by an alpha1,3 linkage to l-glycero-d-manno-heptopyranose II (ld-HeppII). Previously it has been shown that the WabH enzyme catalyzes the incorporation of GlcNAc from UDP-GlcNAc to outer core LPS. The presence of GlcNAc instead of GlcN and the lack of UDP-GlcN in bacteria indicate that an additional enzymatic step is required. In this work we identified a new gene (wabN) in the K. pneumoniae core LPS biosynthetic cluster. Chemical and structural analysi
