Effect of the R126C mutation on the structure and function of the glucose transporter GLUT1: A molecular dynamics simulation study.

Glucose transporter 1 (GLUT1) is responsible for basal glucose uptake and is expressed in most tissues under normal conditions. GLUT1 mutations can cause early-onset absence epilepsy and myoclonus dystonia syndrome (MDS), with MDS potentially lethal. In this study, the effect of the R126C mutation, which is associated with MDS, on structural stability and substrate transport of GLUT1 was investigated. Various bioinformatics tools were used to predict the stability of GLUT1, revealing that the R126C mutation reduces the structural stability of GLUT1. Molecular dynamics (MD) simulations were use

Cryptosporidium parvum alters glucose transport mechanisms in infected enterocytes.

The parasite Cryptosporidium parvum Tyzzer 1912 destroys parts of the intestinal brush border membrane which is important for the uptake of nutrients like glucose. In this study, glucose transport mechanisms of the host cells (IPEC-J2 cells) infected by C. parvum were investigated. The mRNA expression levels of glucose transporters (GLUT) 1 and 2 and Na+-coupled glucose transporter (SGLT) 1 were compared in infected and uninfected cells over an infection time of 24-96 h by RT-qPCR. Furthermore, the protein expression of SGLT 1 and GLUT 2 was quantified in western blot studies. While the protei

Expression and localization of glucose transporters in rodent submandibular salivary glands.

BACKGROUND/AIM: The submandibular gland is one of the three major salivary glands, producing a mixed secretion; this saliva is hypotonic compared to plasma. It also secretes glucose, but the mechanisms responsible for this process are poorly understood. Our study addressed the question whether glucose transporters are expressed and how are they localized within specific rodent submandibular cells, in order to estimate a possible implication in salivary glucose disposal. METHODS: Immunohistochemistry, RT-qPCR and Western blotting were performed to determine the presence/localization of glucose

Estrogen-Driven Maintenance of GLUT1/GLUT4/SGLT1 under glucose starvation drives energy homeostasis in bovine PMNs.

The negative energy balance (NEB) and fluctuations in estrogen (17β-estradiol, E2) during the perinatal period alter glucose metabolism in bovine polymorphonuclear neutrophils (PMN) by affecting the activity of glucose transporters. In the peripheral blood, glucose uptake by PMNs is primarily dependent on the Glucose transporter type1 (GLUT1), Glucose transporter type4 (GLUT4), and Sodium-glucose cotransporter1 (SGLT1). However, the mechanisms through which E2 regulates energy metabolism in these cells, particularly through the modulation of glucose transporter activity, are currently unclear.

Atomistic Insights into Anomeric and Stereochemical Effects on Glucose Transport by GLUTs.

Although β-glucose is more abundant than α-glucose in aqueous solution, GLUT3 preferentially binds α-glucose due to favorable interactions and conformational complementarity within the protein binding site. This study explores the anomeric preferences of glucose transporters GLUT1 and GLUT3 for α- and β-glucose using classical MD, providing mechanistic insight into previously reported differences in anomer-specific transport rates during net influx, efflux, and exchange flux, as well as asymmetric binding of glucose anomer derivatives. Analysis of hydrogen-bonding frequencies between glucose a

Hypometabolism, Alzheimer's Disease, and Possible Therapeutic Targets: An Overview.

The brain is a highly dynamic organ that requires a constant energy source to function normally. This energy is mostly supplied by glucose, a simple sugar that serves as the brain's principal fuel source. Glucose transport across the blood-brain barrier (BBB) is primarily controlled via sodium-independent facilitated glucose transport, such as by glucose transporter 1 (GLUT1) and 3 (GLUT3). However, other glucose transporters, including GLUT4 and the sodium-dependent transporters SGLT1 and SGLT6, have been reported in vitro and in vivo. When the BBB endothelial layer is crossed, neurons and as

Unlocking the Potential of Sotagliflozin in Diabetes Mellitus targeting SGLT 1 & SGLT 2: A Comprehensive Review.

Sotagliflozin, a novel dual inhibitor of sodium-glucose cotransporters 1 and 2 (SGLT1/2), represents a promising therapeutic advancement for managing diabetes mellitus. By inhibiting SGLT1 in the small intestine and SGLT2 in the kidneys, sotagliflozin uniquely improves glycemic control through reduced postprandial glucose absorption and enhanced urinary glucose excretion. This dual mechanism has shown significant benefits for both type 1 and type 2 diabetes, including reduced insulin requirements, better glycemic control, weight loss, and improved cardiovascular and renal outcomes. Clinical tr

Characterization of glucose transport and glucose transporters in the human choriocarcinoma cell line, BeWo.

In this study we have characterized 2-deoxyglucose (2DG) transport and hexose transporter expression in the human choriocarcinoma cell line, BeWo. 2DG uptake in BeWo cells displayed saturable kinetics (V(max), 29+/-1.5 nmol/min/mg protein;K(m), 1.5+/-0.02 m m) and was significantly inhibited in the presence of 2-deoxyglucose, mannose and 3- O -methyl glucose (all at a competing concentration of 30 m m) by up to 97 per cent, but not by galactose or fructose. Glucose uptake was not Na(+)-dependent, but was inhibited by cytochalasin B (by approx 85 per cent) indicating that hexose uptake was medi

GLUT1 and GLUT3 in brain glucose metabolism: mechanisms, regulation, and implications for metabolic disorders.

Brain energy metabolism primarily depends on glucose, which serves as the primary energy source for neuronal activity. Glucose entry into the brain is mediated by glucose transporters, the major isoforms of which are GLUT1 and GLUT3. GLUT1 is responsible for delivering glucose to the brain parenchyma, while GLUT3, with its high affinity for glucose, ensures glucose uptake by neurons. Growing evidence indicates that disturbed glucose metabolism is closely associated with impaired brain function and the progression of neurological diseases, and regulating these transporters may be a potential th

The effect of folate status on the uptake of physiologically relevant compounds by Caco-2 cells.

The aim of this work was to investigate the effect of folate status on the uptake of several physiologically relevant substances by Caco-2 cells. For this, Caco-2 cells cultured in high-folate conditions (HF) and low-folate conditions (LF) were compared. Growth rates of HF and LF Caco-2 cells were similar. However, proliferation rate of LF cells was greater than that of HF cells during the first 2days of culture and slightly smaller thereafter, viability of LF cells was greater than that of HF cells, and apoptosis index was similar in both cell cultures. We verified that in LF cells, comparati

GLUT4 in the endocrine pancreas--indicating an impact in pancreatic islet cell physiology?

The glucose transporter GLUT4 is well known to facilitate the transport of blood glucose into insulin-sensitive muscle and adipose tissue. In this study, molecular, immunohistochemical, and Western blot investigations revealed evidence that GLUT4 is also located in the mouse, rat, and human endocrine pancreas. In addition, high glucose decreased and insulin elevated the GLUT4 expression in pancreatic α-cells. In contrast, high glucose increased GLUT4 expression, whereas insulin led to a reduced expression level of the glucose transporter in pancreatic β-cells. In vivo experiments showed that i

Sotagliflozin: A Review in Type 1 Diabetes.

Sotagliflozin (Zynquista™) is the first dual inhibitor of sodium-glucose co-transporter-1 and -2 (SGLT1 and 2). In the phase 3, inTANDEM 1-3 trials, adjunctive use of oral sotagliflozin (200 mg or 400 mg once daily) improved glycaemic control and reduced bodyweight and insulin requirements relative to placebo over 24 weeks of treatment in adults whose type 1 diabetes (T1D) was inadequately controlled by insulin therapy. Similar benefits were seen with the drug in patients who were overweight/obese [i.e. body mass index (BMI) ≥ 27 kg/m2] in inTANDEM 1 and 2 (pooled). The benefits of sotaglifloz

Getting sweeter: new evidence for glucose transporters in specific cell types of the airway?

New technologies such as single-cell RNA sequencing (scRNAseq) has enabled identification of the mRNA transcripts expressed by individual cells. This review provides insight from recent scRNAseq studies on the expression of glucose transporters in the epithelial cells of the airway epithelium from trachea to alveolus. The number of studies analyzed was limited, not all reported the full range of glucose transporters and there were differences between cells freshly isolated from the airways and those grown in vitro. Furthermore, glucose transporter mRNA transcripts were expressed at lower level

Peritoneal Expression of SGLT-2, GLUT1, and GLUT3 in Peritoneal Dialysis Patients.

INTRODUCTION: In peritoneal dialysis (PD) patients, the peritoneal membrane is affected by glucose-based solutions used as peritoneal dialysate fluids. This exposure leads to changes of the membrane which may eventually culminate in fibrosis and method failure. In vitro or animal studies demonstrated that glucose transporters are upregulated upon exposure to these solutions. Expression studies of glucose transporters in human peritoneum have not been reported yet. METHODS: Expression of SGLT-2, GLUT1, and GLUT3 in human peritoneal biopsies was analyzed by real-time polymerase chain reaction a
