Functional Characterization of Two PLP-Dependent Enzymes Involved in Capsular Polysaccharide Biosynthesis from Campylobacter jejuni.

Campylobacter jejuni is a Gram-negative, pathogenic bacterium that causes campylobacteriosis, a form of gastroenteritis. C. jejuni is the most frequent cause of food-borne illness in the world, surpassing Salmonella and E. coli. Coating the surface of C. jejuni is a layer of sugar molecules known as the capsular polysaccharide that, in C. jejuni NCTC 11168, is composed of a repeating unit of d-glycero-l-gluco-heptose, d-glucuronic acid, d-N-acetyl-galactosamine, and d-ribose. The d-glucuronic acid moiety is further amidated with either serinol or ethanolamine. It is unknown how these modificat…

Xenobiotic nuclear receptor-mediated regulation of UDP-glucuronosyl-transferases.

UDP-glucuronosyltransferases (UGTs) belong to the Phase II drug metabolizing enzymes. UGTs mediate the transfer of glucuronic acid, from UDP glucuronic acid to predominantly hydrophobic xeno- and endobiotic chemicals, thus facilitating their detoxification and excretion. Deficiency in the expression and/or activity of UGTs may lead to genetic and acquired diseases such as Crigler-Najjar syndrome and jaundice. UGT genes are classified into UGT1A and UGT2B subfamily, and each subfamily and each isoform shows tissue-specific distribution pattern. The underlying mechanisms for this tissue specific…

Sex differences in salicylic acid metabolism in Nigerian subjects.

The urinary metabolites of a single dose (1 g) of salicylic acid were investigated in black Nigerian subjects (78 females 44 males). Qualitatively, the major metabolites were the glycine and glucuronic acid conjugates. Quantitatively, there was a statistically significant difference in the level of these metabolites between female and male subjects (P less than 0.001) (using Student's t-test). The results of the present study compared with earlier published data show a statistically significant quantitative difference between black Nigerians and Caucasians (P less than 0.001). The results sugg…

The UDP-glucuronosyltransferases of the blood-brain barrier: their role in drug metabolism and detoxication.

UDP-glucuronosyltransferases (UGTs) form a multigenic family of membrane-bound enzymes expressed in various tissues, including brain. They catalyze the formation of β-D-glucuronides from structurally unrelated substances (drugs, other xenobiotics, as well as endogenous compounds) by the linkage of glucuronic acid from the high energy donor, UDP-α-D-glucuronic acid. In brain, UGTs actively participate to the overall protection of the tissue against the intrusion of potentially harmful lipophilic substances that are metabolized as hydrophilic glucuronides. These metabolites are generally inactiv…

Covalent binding of zomepirac glucuronide to proteins: evidence for a Schiff base mechanism.

Carboxylic acids can undergo irreversible binding to proteins via their acyl glucuronide metabolites. Such binding occurs in humans and in vitro for bilirubin and the anti-inflammatory drugs zomepirac and tolmetin and may have toxicological significance. The mechanism of the binding is uncertain, but is thought to involve nucleophilic displacement of glucuronic acid by free cysteine thiols, tyrosine, or lysine residues of the protein. We now present evidence, using zomepirac glucuronide, for an alternative mechanism where irreversible binding occurs by isomerization of the glucuronide and form…

Possible genetic influence on conjugate formation in salicylic acid metabolism.

Subjects (7 males and 7 females) were dosed with salicylic acid (1 g) and hourly urinary samples were analyzed for its metabolites. The results obtained showed that the female subjects had higher capacity for salicylurate formation than the male (P less than or equal to 0.025). The urinary hourly excretion ratio of salicylurate and salicylglucuronic acid was about or greater than 1 while in the male this ratio is less than 0.50. A comparison of this ratio between female and male showed a highly significant difference (P less than or equal to 0.001). The high capacity of glucuronic acid pathway…

A sensitive procedure for the diagnosis of N-acetyl-galactosamine-6-sulfate sulfatase deficiency in classical Morquio's disease.

The trisaccharide 6-sulfo-N-acetylgalactosamine-glucuronic acid-6-sulfo-N-acetyl-[1-3H]galactosaminitol was used as a substrate for the determination of N-acetylgalactosamine-6-sulfate sulfatase activity. The amount of liberated sulfate was measured indirectly by separating monosulfated reaction products from the substrate on Dowex 1 X 2 microcolumns in a simple two step procedure. Fibroblast homogenates from patients with various genotypes, except classical Morquio's disease, released 410 +/- 90 pmol sulfate/h/mg cell protein. The enzyme exhibited a pH optimum of pH 4.8 and a KM of about 1 X …

(S)oxazepam glucuronidation is inhibited by ketoprofen and other substrates of UGT2B7.

1,4-Benzodiazepine anxiolytics such as diazepam and halazepam are converted in vivo to oxazepam, an active metabolite with a hydroxyl group at the asymmetric C3 position. D-glucuronic acid couples with the C3 hydroxyl group of oxazepam to form pharmacologically inactive diastereomeric glucuronide conjugates. Conjugation with glucuronic acid is catalysed by the microsomal UDP-glucuronosyltransferase (UGT) enzyme system, which includes an undetermined number of isozymes. Although 1,4-benzodiazepines are ultimately cleared as oxazepam glucuronide, little is known about the particular UGT isozyme(…

Identification of Acidic Triterpenoid Saponins from Silene vulgaris Using a Methylation-Based Isolation and LC-MS Analysis Strategy.

The Silene genus plants in the Caryophyllaceae family are a large genus with over 850 species. It has been known that Silene genus plants contain triterpenoid saponins. These saponins have glucuronic acid in the sugar chain and are difficult to separate in chromatography. In this study, a strategy was developed to clarify the distribution of triterpenoid saponins in whole plants of Silene vulgaris by isolation of neutralized saponins using methylation reactions, which were used as standard substances for LC-MS analysis, and elucidating their characteristic MS and MS/MS fragment patterns. The n…

Determination of trichloroethylene metabolites in rat liver homogenate using headspace gas chromatography.

An headspace gas chromatographic (HSGC) method for determination of trichloroethylene metabolites in rat liver homogenates is described. These metabolites are chloral hydrate (CH), trichloroethanol (TCE), trichloroacetic acid (TCA) and the glucuronic acid conjugate of trichloroethanol (TCE-beta-glucuronide). The method is based on selective thermal conversion of CH and TCA into chloroform, which is determined together with trichloroethanol by HSGC using electron-capture detection. TCE-beta-glucuronide was determined as the difference between free TCE and total TCE after enzymatic hydrolysis wi…

Evidence for rapid uptake of D-galacturonic acid in the yeast Saccharomyces cerevisiae by a channel-type transport system.

D-Galacturonic acid is a major component of pectins but cannot be metabolized by Saccharomyces cerevisiae. It is assumed not to be taken up. … The uptake was not inhibited by any compound tested as candidate competitive inhibitor, including D-glucuronic acid, which was also transported. The characteristics of D-galacturonic acid uptake are consistent with involvement of a channel-type system, probably encoded by multiple genes.
