Beta-defensin 22 is a major component of the mouse sperm glycocalyx.

Surface components of sperm isolated from the cauda epididymides were stabilized by whole sperm fixation for immunization of rabbits. The resulting immunoglobulins (Igs) recognized a single protein of 130 kDa (non-reduced) or 54-57 kDa (reduced) on western blots of cauda sperm. Igs recognized the same 54-57 kDa protein band on whole tissue blots of the corpus and cauda epididymidis and vas deferens. No immunoreactive bands were detected on blots of the prostate, seminal vesicles, testes, caput epididymis, or any of various non-reproductive tissues. Removal of sperm from the vas deferens prior

Tumour acidosis remodels the glycocalyx to control lipid scavenging and ferroptosis.

Aggressive tumours are defined by microenvironmental stress adaptation and metabolic reprogramming. Within this niche, lipid droplet accumulation has emerged as a key strategy to buffer toxic lipids and suppress ferroptosis. Lipid droplet formation can occur via de novo lipogenesis or extracellular lipid-scavenging. However, how tumour cells coordinate these processes remains poorly understood. Here we identify a chondroitin sulfate (CS)-enriched glycocalyx as a hallmark of the acidic microenvironment in glioblastoma and central nervous system metastases. This CS-rich glycocalyx encapsulates t

Nanovaccine Based on Glycocalyx-Mimicking Nanovehicle to Potentiate the Immunogenicity of Neoantigen.

Neoantigens are promising candidates for personalized cancer vaccines and immunotherapies. However, low immunogenicity and insufficient cross-presentation of neoantigens remain a major challenge. Inspired by natural glycocalyx and its important functions in immune response, here we report a glycocalyx-mimicking nanovehicle constructed from (oligo)mannoside-modified acid-sensitive glycopolymers to improve the efficiency of tumor neoantigens. These amphiphilic glycopolymers assembled into nanoparticles could serve as immune activators for dendritic cells maturation. The encapsulation of neoantig

Global Research Trends on Endothelial Glycocalyx in Sepsis: A Bibliometric Analysis.

BACKGROUND: Sepsis remains a major global health challenge, with glycocalyx dysfunction playing an important role in its pathogenesis. Recent research highlights EG degradation as a key contributor to vascular permeability, inflammation, and organ failure. Despite growing interest, a comprehensive bibliometric analysis of global research trends on EG in sepsis is lacking. METHODS: We conducted a bibliometric analysis using data from Web of Science and Scopus (2005-2025). Inclusion criteria were original English-language research articles. Bibliometrix R-package was employed to analyze publica

Updates on the endothelial glycocalyx in heart failure with preserved ejection fraction.

The endothelial glycocalyx (GCX) is a specialized form of extracellular matrix with a dynamic synthesis and degradation balance. It plays a pivotal role in regulating cell signaling, sensing the extracellular environment, and maintaining vascular permeability. In the setting of heart failure (HF), particularly in HF with preserved ejection fraction (HFpEF), systemic endothelial GCX degradation may represent an early pathophysiological mechanism. Indeed, HFpEF results from comorbidities-related proinflammatory states leading to endothelial dysfunction and subsequent myocardial hypertrophy, fibr

The glomerular endothelial glycocalyx as a therapeutic target in proteinuric kidney disease.

Endothelial glycocalyx lines every blood vessel throughout the body, and has key roles in vascular biology, including vascular permeability and inflammation. Accumulated evidence from the past 15 years shows that the glomerular endothelial glycocalyx is a vital component of the glomerular filtration barrier, which limits the filtration of macromolecules such as albumin. However, the contribution of endothelial glycocalyx to the pathogenesis of proteinuria and its potential as a therapeutic target have not been fully explored. Experimental disruption of the glomerular endothelial glycocalyx inc

Enzymatic Modulation of the Pulmonary Glycocalyx Enhances Susceptibility to Streptococcus pneumoniae.

The pulmonary epithelial glycocalyx is rich in glycosaminoglycans such as hyaluronan and heparan sulfate. Despite their presence, the importance of these glycosaminoglycans in bacterial lung infections remains elusive. To address this, we intranasally inoculated mice with Streptococcus pneumoniae in the presence or absence of enzymes targeting pulmonary hyaluronan and heparan sulfate, followed by characterization of subsequent disease pathology, pulmonary inflammation, and lung barrier dysfunction. Enzymatic degradation of hyaluronan and heparan sulfate exacerbated pneumonia in mice, as eviden

The role of the alveolar epithelial glycocalyx in acute respiratory distress syndrome.

The alveolar epithelial glycocalyx is a dense anionic layer of glycosaminoglycans (GAGs) and proteoglycans that lines the apical surface of the alveolar epithelium. In contrast to the pulmonary endothelial glycocalyx, which has well-established roles in vascular homeostasis and septic organ dysfunction, the alveolar epithelial glycocalyx is less understood. Recent preclinical studies demonstrated that the epithelial glycocalyx is degraded in multiple murine models of acute respiratory distress syndrome (ARDS), particularly those that result from inhaled insults (so-called "direct" lung injury)

Effects of therapeutic plasma exchange on the endothelial glycocalyx in septic shock.

BACKGROUND: Disruption of the endothelial glycocalyx (eGC) is observed in septic patients and its injury is associated with multiple-organ failure and inferior outcomes. Besides this biomarker function, increased blood concentrations of shedded eGC constituents might play a mechanistic role in septic organ failure. We hypothesized that therapeutic plasma exchange (TPE) using fresh frozen plasma might influence eGC-related pathology by removing injurious mediators of eGC breakdown while at the time replacing eGC protective factors. METHODS: We enrolled 20 norepinephrine-dependent (NE > 0.4 μg/

Evaluation of the endothelial glycocalyx damage in patients with acute coronary syndrome.

BACKGROUND: Endothelial glycocalyx (EG) is sugar-based cell-bound surface molecules linked to transmembrane proteins observed on the endothelial surface of the vessels. Damage to this structure causes an increase in platelet and leucocyte adhesion and shear stress in the vessel. We hypothesized a possible link between EG damage and acute coronary syndrome (ACS). METHODS: We measured the syndecan-1 levels (a biomarker of EG damage) in 141 patients (99 men) with ACS and compared to those of 45 patients (24 men) with non-coronary chest pain (NCCP) and of 24 (14 men) healthy individuals (CONTROL)

Glycocalyx injury in adults with nephrotic syndrome: Association with endothelial function.

BACKGROUND: Glomerulopathy is a group of diseases that affect mainly young adults. Endothelial dysfunction, atherosclerosis, and increased cardiac mortality can complicate the evolution of such patients. However, there is no study evaluating endothelial glycocalyx in this pathology. METHODS: This cross-sectional study included 49 patients with untreated primary nephrotic syndrome that were otherwise healthy. In addition to routine laboratory measurements, syndecan-1, intercellular adhesion molecule-1 (ICAM-1), and e-selectin were measured. Moreover, flow-mediated dilation (FMD) was used as th

Hypothesis: arterial glycocalyx dysfunction is the first step in the atherothrombotic process.

We present evidence that the 0.5 microm thick gel layer, lining the inner wall of healthy blood vessels, the glycocalyx, is the first line of defence against atherothrombotic disease. All blood vessel linings are coated with this gel, a highly negatively charged structure, rich in anionic sites mostly represented by the sialic acid moieties of glycoproteins and the sulphate and carboxyl groups of heparan-sulphate proteoglycans. Blood flow in arteries is associated with a shear stress at the glycocalyx, which signals the underlying endothelial cells to release nitric oxide (NO), an anti-atherog

Alterations in the intestinal glycocalyx and bacterial flora in response to oral indomethacin.

Nonsteroidal anti-inflammatory drugs (NSAIDs), used extensively in clinical medicine, tend to cause adverse effects in the gastrointestinal tract. Earlier work has shown that oral administration of indomethacin produced oxidative damage in the small intestine and attenuation of the glycocalyx layer of the mucosa. The present study assessed, in greater detail, the alterations produced in the glycocalyx of rat small intestinal mucosa in response to indomethacin, with specific reference to surfactant-like particles (SLP) and brush border membranes (BBM). Changes in gut flora in response to the dr

Enzyme-Resistant Glycocalyx Bionic Coating for Corneal Bandage Lens Surface Engineering.

Advanced implantable medical devices often need to integrate multiple functions onto one surface to meet the complex needs in vivo. The construction of glycocalyx bionic coatings that mimic the natural multifunctional glycocalyx barrier of blood vessels, intestines, and ocular surfaces is an emerging strategy for function integration. However, in practical application scenarios such as corneal bandage lens (CBL) for corneal injury treatment, bio-enzymes from immune cells and bacteria can degrade the natural polymers constructing the glycocalyx bionic coating, such as hyaluronic acid, resulting

Febrile temperature enhances Plasmodium falciparum cytoadhesion by disrupting the endothelial glycocalyx.

Fever, a universal host defense in infection and inflammation, paradoxically contributes to neurological complications in malaria. While febrile temperatures enhance the expression of parasite virulence proteins that mediate vascular adhesion and disease severity, its effects in the endothelium remain elusive. Here we present a 3D fever-on-a-chip model that recapitulates human brain and lung microvessels under febrile conditions. Short febrile episodes at 40 °C, common in treated cerebral malaria patients, rapidly enhanced iRBC and immune cell binding under flow. Mechanistically, we demonstrat

Endothelial glycocalyx in hepatopulmonary syndrome: An indispensable player mediating vascular changes.

Hepatopulmonary syndrome (HPS) is a serious pulmonary vascular complication that causes respiratory insufficiency in patients with chronic liver diseases. HPS is characterized by two central pathogenic features-intrapulmonary vascular dilatation (IPVD) and angiogenesis. Endothelial glycocalyx (eGCX) is a gel-like layer covering the luminal surface of blood vessels which is involved in a variety of physiological and pathophysiological processes including controlling vascular tone and angiogenesis. In terms of lung disorders, it has been well established that eGCX contributes to dysregulated vas

Transmission electron microscopy demonstration of reduced endothelial glycocalyx in severe preeclampsia.

INTRODUCTION: Endothelial glycocalyx is a carbohydrate-rich layer lining the luminal side of blood vessels. Its damage was demonstrated in different groups of critically ill patients. Indirect evidence showed that endothelial glycocalyx degradation might be an important factor in pathophysiology of preeclampsia. The aim of our study was to demonstrate endothelial glycocalyx by transmission electron microscopy and to compare its amount in the omentum vessels of pregnant patients with severe preeclampsia and two control groups. METHODS: Patients with severe preeclampsia who had a cesarean secti
