Bacteroides thetaiotaomicron (Bt) degrades dietary glycans via carbohydrate-active enzymes (CAZymes), but their spatial distribution and functional coordination remain unclear. In this study, we characterized the "labor division" during glycan degradation across four bacterial fractions: cytoplasm, outer membrane, extracellular vesicles, and supernatant. All fractions displayed hydrolytic activity. Proteomic analysis identified abundant glycoside hydrolases, mainly GH32 enzymes encoded by PUL22, and their components varied greatly. Untargeted metabolomics identified 45 shared differential meta

The biochemical and structural characteristics of PtLam, a laminarinase from deep-sea Planctomycetota, have been extensively elucidated, unveiling the fundamental molecular mechanisms governing substrate recognition and enzymatic catalysis. PtLam functions as an exo-laminarinase with the ability to sequentially hydrolyze laminarin, cleaving glucose units individually. Notably, PtLam exhibits proficient transglycosylation capabilities, utilizing various sugar alcohols as acceptors, with lyxose, in particular, yielding exclusively transglycosylated products. Structural analysis of both apo-PtLam

In this paper, the enzymatic activity, substrate specificity and antibiofilm feature of bacteriophage dual-function tail proteins are presented. So far, tail tubular proteins A-TTPAgp31 and TTPAgp44-have been considered as structural proteins of Klebsiella pneumoniae bacteriophages KP32 and KP34, respectively. Our results show that TTPAgp31 is able to hydrolyze maltose as well as Red-starch. The activity of 1 µM of the protein was calculated as 47.6 milli-Units/assay relating to the α-amylase activity. It degrades capsular polysaccharides (cPS), slime polysaccharides (sPS) and lipopolysacchari

C-glycosides are natural products with important biological activities but are recalcitrant to degradation. Glycoside 3-oxidases (G3Oxs) are recently identified bacterial flavo-oxidases from the glucose-methanol-coline (GMC) superfamily that catalyze the oxidation of C-glycosides with the concomitant reduction of O2 to H2O2. This oxidation is followed by C-C acid/base-assisted bond cleavage in two-step C-deglycosylation pathways. Soil and gut microorganisms have different oxidative enzymes, but the details of their catalytic mechanisms are largely unknown. Here, we report that PsG3Ox oxidizes

Hydrolytic enzymes play crucial roles in cellular processes, and dysregulation of their activities is implicated in various physiological and pathological conditions. These enzymes cleave substrates such as peptide bonds, phosphodiester bonds, glycosidic bonds, and other esters. Detecting aberrant hydrolase activity is vital for understanding disease mechanisms and developing targeted therapeutic interventions. This study introduces a novel approach to measuring hydrolase activity using giant magnetoresistive (GMR) spin valve sensors. These sensors change resistance in response to magnetic fie

Under appropriate experimental conditions, some glycoside hydrolases can catalyze transglycosylation reactions; a hypothesis associated with this is that the glycosidic linkages formed will be preferentially hydrolyzed under optimal conditions. Therefore, the hydrolytic and transglycosylation activities of isolated membranes from differentiated Caco-2 cells on sucrose, maltose and isomaltulose were evaluated. After the enzymatic reactions, the di- and trisaccharides obtained were identified by gas chromatography coupled to a mass spectrometer. Differentiated Caco-2 cell membranes exerted hydro

The catabolism of cyanogenic glycosides is initiated by cleavage of the carbohydrate moiety by one or more beta-glycosidases, which yields the corresponding alpha-hydroxynitrile. Until recently, the mode by which cyanogenic disaccharides are hydrolysed was largely unclear. Investigation of highly purified beta-glycosidases from plants containing cyanogenic disaccharides has now indicated that these compounds may be degraded via two distinct pathways, depending on the plant species. beta-Glycosidases from Davallia trichomanoides and Vicia angustifolia hydrolysed (R)-vicianin and (R)-amygdalin a

Carbohydrate-active enzymes (CAZymes) catalyze reactions central to glycan degradation, biosynthesis, and remodeling. Although their mechanisms have traditionally been described using a limited set of canonical models, recent studies are revealing a broader and more dynamic mechanistic landscape. We discuss how QM/MM-based enhanced-sampling methods have expanded our understanding of CAZyme catalysis by uncovering substrate-assisted pathways, complex proton-transfer processes, unconventional catalytic residues, and the roles of substrate distortion and enzyme dynamics in shaping reactivity. 1. Polysaccharide depolymerases and glycoside hydrolases involved in the breakdown of plant structural polysaccharides (hemicellulose and pectins) were monitored in three fractions of the liquid phase of horse caecum digesta: acellular fluid (AF), bacteria (B) and protozoa plus bacteria (PB). 2. Both bacteria and protozoa were found to be involved in the decomposition of pectic substances, with two enzymic activities: depolymerase (polygalacturonase, EC 3.2.1.15; and pectin lyase, EC 4.2.2.10) and esterase (pectinesterase, EC 3.1.1.11). The activity of the PB fraction was higher than that of B

Two-component plant defenses such as cyanogenic glucosides are produced by many plant species, but phloem-feeding herbivores have long been thought not to activate these defenses due to their mode of feeding, which causes only minimal tissue damage. Here, however, we report that cyanogenic glycoside defenses from cassava (Manihot esculenta), a major staple crop in Africa, are activated during feeding by a pest insect, the whitefly Bemisia tabaci, and the resulting hydrogen cyanide is detoxified by conversion to beta-cyanoalanine. Additionally, B. tabaci was found to utilize two metabolic mecha

The covalent attachment of pendant groups to the carbons of d-pyranosides can influence their ability to be recognized by proteins that bind natural sugars. These carbon-modified sugars have received only limited attention, and systematic investigations on their ability to act as substrates of glycosyl hydrolases have not been reported. Here, C6-substituted β-d-galactosides are synthesized and evaluated as putative substrates of β-glycosyl hydrolases. The results indicated that the size of the group at C6-methyl vs. di-methyl vs. phenyl-attenuated their ability to act as substrates. The C6-met

Certain strains of Moraxella bovis produce tissue-damaging enzymes which may initiate or potentiate infectious bovine keratoconjunctivitis. Thirteen reference strains of this species were characterized physiologically and screened for production of various enzymes by some conventional biochemical tests and the API ZYM system (Analytab Products, Plainview, N.Y.). All 13 strains were hemolytic. All hydrolyzed Tween 80 and Tween 85 and displayed C4 esterase, C8 esterase-lipase, and C14 lipase activities. All produced phosphoamidase and phosphatase. All were able to hydrolyze casein and gelatin. A

Complications of short bowel syndrome (SBS) include malabsorption and bacterial overgrowth, requiring prolonged dependence on parenteral nutrition (PN). We hypothesized that the intolerance of whole food in some SBS patients might be due to the effect of dietary fiber on the gut microbiome. Shotgun metagenomic sequencing and targeted metabolomics were performed using biospecimens collected from 55 children with SBS and a murine dietary fiber model. Bioinformatic analyses were performed on these datasets as well as from a healthy human dietary intervention study. Compared to healthy controls, t

This study integrated metagenomic and untargeted metabolomics analysis to characterize differences in kombucha communities and metabolites between black tea-sucrose and theaflavin-sucrose systems. Theaflavins selectively enriched yeasts, particularly Meyerozyma and Lodderomyces, and increased the annotated abundance of carbohydrate-metabolism genes, glycoside hydrolases, and glycosyltransferases. Based on these community-level findings, the newly isolated Meyerozyma sp. C3 was employed to establish a simplified single-strain fermentation model to investigate the biotransformation potential of

Siamenoside I (SI), a high-intensity sweetener, is limited in natural abundance. A β-glycosidase (CoExg1) from Candida orthopsilosis L7 (isolated from the Siraitia grosvenorii rhizosphere) exhibits intrinsic sequential regioselectivity, enabling efficient SI production from mogroside V (MGV). Fermentation optimization increased SI yield from 70.34% (7 d) to 84.06% (72 h). CoExg1 rapidly cleaves the C-3 β-1,6 bond (89.35% SI in 20 min), then slowly hydrolyzes the C-24 bond to mogroside IIIE (MIIIE, 12 h)─a time-controlled process requiring no external regulation. Kinetic analysis showed a ∼50-f
