AIMS: Polymyxins remain the mainstay antibiotic for the treatment of infections caused by multidrug-resistant bacteria. However, with the increase in the use of polymyxins, the simultaneous rise of polymyxin-resistance cases has been another global threat, necessitating the need for novel therapeutic strategies. This work aimed to evaluate the antibacterial activity of cationic peptoids against polymyxin-resistant bacteria of global priority.
METHODS AND RESULTS: Three paired polymyxin-sensitive/polymyxin-resistant strains were included, along with two clinical isolates and one reference stra

The rise in bacterial resistance to conventional antibiotics has led to the search for alternative antimicrobial agents, such as flavonoids, which are widely distributed in plants and known for their antibacterial properties. This review examines the antibacterial activity of flavonoids against Gram-negative bacteria and identifies gaps in current knowledge. The analysis of available data revealed that several flavonoids, such as liricidin-7-O-hexoside, licoflavone C, and dorsmanins C and F, demonstrated remarkable antibacterial potential. It was also observed that flavonoids exhibit multifact

Antimicrobial resistance poses a severe threat to human health, with colistin serving as a critical medication in clinical trials against multidrug-resistant Gram-negative bacteria. However, the efficacy of colistin is increasingly compromised due to the rise of MCR-positive bacteria worldwide. Here, we reveal a notable metabolic disparity between mcr-positive and -negative bacteria through transcriptome and metabolomics analysis. Specifically, pyridoxal 5'-phosphate (PLP), the active form of vitamin B6, was significantly diminished in mcr-positive bacteria. Conversely, supplementing with PLP

UNLABELLED: Polymyxin is used as a last resort antibiotics for infections caused by multi-drug resistant (MDR) Gram-negative bacteria and is often combined with other antibiotics to improve clinical effectiveness. However, the synergism of colistin and other antibiotics remains obscure. Here, we revealed a notable synergy between colistin and flavomycin, which was traditionally used as an animal growth promoter and has limited activity against Gram-negative bacteria, using checkerboard assay and time-kill curve analyses. The importance of membrane penetration induced by colistin was assessed b

Since the introduction of quinolone and fluoroquinolone antibiotics to treat bacterial infections in the 1960s, there has been a pronounced increase in the number of bacterial species that have developed resistance to fluoroquinolone treatment. In 2017, the World Health Organization established a priority list of the most critical Gram-negative resistant pathogens. These included Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Escherichia coli. In the last three decades, investigations into the mechanisms of fluoroquinolone resistance have revealed that mutations in

The continuous reduction of clinically available antibiotics has made it imperative to exploit more effective antimicrobial therapies, especially for difficult-to-treat Gram-negative pathogens. Herein, it is shown that the combination of an antimicrobial nanozyme with the clinically compatible basic amino acid L-arginine affords a potent treatment for infections with Gram-negative pathogens. In particular, the antimicrobial activity of the antimicrobial nanozyme is dramatically increased by ≈1000-fold after L-arginine stimulation. Specifically, the combination therapy enhances bacterial outer

Genes encoding endolysins were identified and cloned from three different Escherichia coli bacteriophages, 10-24(13), PBEC30, and PBEC56. Putative antimicrobial peptide (AMP)-like C-terminal alpha helix structures with amphipathic natures were predicted from the three endolysins. Each gene was cloned and expressed as hexahistidine-tagged forms, and the products were purified and characterized. The purified endolysins exhibited antibacterial activities against a variety of Gram-negative bacteria including Escherichia coli, Pseudomonas aeruginosa, Acinetobacter baumannii, and Klebsiella pneumoni

Antibody activity to cotton dust bacteria in the nasal secretions of cotton-mill cardroom workers was studied using the ELISA technique. Secretion samples were collected by inserting a roll of filter paper into the nasal cavities of the subjects. The amount of secretion was significantly less among cotton-mill workers than workers in a wood factory. No significant differences were seen between byssinotic and nonbyssinotic cotton-mill workers. No significant differences were observed between cotton and wood workers regarding antibody activity against Pseudomonas syringae and Enterobacter agglom

AIM: We report synthesis of capped gold nanoparticles (C-AuNPs) of ≈20-30 nm by reducing HAuCl4 with flower and leaf extracts of Ocimum tenuiflorum, leaves of Azadirachta indica and Mentha spicata and peel of Citrus sinensis plants.
METHODS: Atomic force microscopy (AFM) and transmission electron microscopy (TEM) determined their size, shape and topographical structures. The C-AuNPs with UV-Vis spectrophotometer produced a maximum absorption within 530-535 nm wavelengths. Their Fourier transform IR stretching frequencies, from 450 to 4000 cm-1, have inferred HAuCl4 reduction to Au.
RESULTS:

The continued rise of antibiotic resistance threatens to undermine the utility of the world's current antibiotic arsenal. This problem is particularly troubling when it comes to Gram-negative pathogens for which there are inherently fewer antibiotics available. To address this challenge, recent attention has been focused on finding compounds capable of disrupting the Gram-negative outer membrane as a means of potentiating otherwise Gram-positive-specific antibiotics. In this regard, agents capable of binding to the lipopolysaccharide (LPS) present in the Gram-negative outer membrane are of par

OBJECTIVES: Differences in management and outcomes of brain abscesses due to gram-positive (GPB) versus gram-negative bacteria (GNB) are not well defined.
METHODS: A retrospective review of adult patients with brain abscesses due to monomicrobial infection from 2009 through 2020 was performed.
RESULTS: A total 177 patients had a monomicrobial brain abscess; 143 (80.8%) caused by GPB and 34 (19.2%) by GNB. Patients with GNB had more history of head/neck surgery than those with GPB (58.8% vs 36.4%; P = 0.02). Pathogens in the GNB group included Pseudomonas aeruginosa (29.4%), Klebsiella spp (2

Physically disrupting microorganism membranes to enable antibiotics to overcome resistance mechanisms that inhibit or excrete antibiotics has great potential for reducing antibiotic doses and rendering resistance mechanisms inert. We demonstrate the synergistic inactivation of a Gram-positive (Staphylococcus aureus) and two Gram-negative (Escherichia coli and Pseudomonas aeruginosa) bacteria by combining 222 30 kV/cm electric pulses (EPs) or 500 20 kV/cm EPs with 300-ns EP duration with various antibiotics with different mechanisms of action is demonstrated. Doses of antibiotics that produced

Antimicrobial resistance causes millions of infections and deaths each year, necessitating new approaches to minimise these effects. Here, we present a direct comparison of crystalline, MoS2 nanosheets (MNS) and amorphous MoS2 nanoflowers (MNF) against Escherichia coli MG1655 (Gram-negative bacteria) and Staphylococcus aureus RN4220 (Gram-positive bacteria). The crystalline MNS sheets of different sizes were obtained using exfoliation of bulk MoS2, while the amorphous MNF was formed using hydrothermal synthesis. Using XPS, the MNF was found to contain MoO3 in addition to MoS2, while the MNS ha

Antimicrobial resistance (AMR) represents one of the most pressing global public health challenges of the 21st century, driven by the widespread inappropriate use of antibiotics and the remarkable adaptive capacity of pathogenic bacteria This narrative review provides an integrated comparative analysis of AMR mechanisms across Gram-positive and Gram-negative bacteria, with particular emphasis on clinically significant ESKAPE pathogens. The four principal resistance mechanisms: enzymatic drug inactivation, reduced drug uptake, target site modification, and active efflux, are examined comparativ

Faropenem is a new oral beta-lactam antibiotic unique from carbapenems and other available beta-lactams. Determinants of the in vitro activity of beta-lactam antibiotics include affinity to penicillin-binding proteins (PBPs) and beta-lactamase stability. In this study, the binding affinity of faropenem to various PBPs and its impact on the morphology of Staphylococcus aureus and Escherichia coli were evaluated. In general, faropenem demonstrated high binding affinity to high-molecular-weight PBPs but low affinity to low-molecular-weight PBPs. In S. aureus and Streptococcus pneumoniae, faropene

Efflux mechanisms that account for resistance to a variety of antimicrobial agents are commonly found in a wide range of bacteria. Two major groups of efflux systems are known, specific exporters and transporters conferring multidrug resistance (MDR). The MDR systems are able to remove antimicrobials of different classes from the bacterial cell and occasionally play a role in the intrinsic resistance of some bacteria to certain antimicrobials. Their genes are commonly located on the bacterial chromosome. In contrast, the genes coding for specific efflux systems are often associated with mobile

INTRODUCTION: The global rise of multidrug-resistant (MDR) Gram-negative bacteria (GNB) has intensified the threat of chronic and hard-to-treat infections, many of which are associated with biofilm formation. These biofilms confer enhanced resistance to antimicrobials and immune responses, posing a major clinical challenge.
AREAS COVERED: This review summarizes the biological mechanisms of biofilm formation in GNB and explores both traditional and novel strategies for their prevention and eradication. The literature search covered peer-reviewed articles from major databases, focusing on thera
