Histopathological analysis of Leri-Weill dyschondrosteosis: disordered growth plate.

Leri-Weill syndrome (LWS) is a dominant (pseudoautosomal) skeletal dysplasia with mesomelic short stature and bilateral Madelung deformity, due to dyschondrosteosis of the distal radius. It results from the loss of one copy of the Short Stature Homeobox Gene (SHOX) from the tip of the short arm of the X or Y chromosome. SHOX molecular testing enabled us to evaluate the histopathology of the radial physis in LWS patients with a documented SHOX abnormality. A widespread disorganisation of physeal anatomy was revealed with disruption of the normal parallel columnar arrangement of chondrocytes. Ta

Epiphyseal growth plate fracture: Salter and Harris type V.

One of the most easily misdiagnosed injuries, which can result in severe consequences in young patients, is a fracture involving the epiphyseal growth plate. The Salter and Harris type V fracture is a relatively uncommon injury resulting from a severe crushing force applied through the epiphysis to an area of the growth plate. It is most likely to occur in the region of the knee or ankle. The following is a case report of a comminuted compression fracture of the epiphysis of the fifth metatarsal head, with complete transverse midshaft fracture of the proximal phalanx of the fifth digit.

A Retinoid Antagonist Attenuates Bone Growth Inhibition Caused by Growth Plate Injury in Mice.

Growth plate injury, which may trigger growth plate fusion or arrest, is a common occurrence in pediatric orthopaedics that can lead to angulation of the limb and limb length discrepancy. Growth plate injuries are currently treated palliatively, with surgical intervention when severe limb length discrepancy or angulation is found during follow-up visits. The cellular changes in the chondrocytes in injured growth plate have not been fully elucidated, and understanding these factors will provide important insights for the development of therapies that promote growth plate repair and prevent grow

Recent research on the growth plate: Impact of inflammatory cytokines on longitudinal bone growth.

Children with inflammatory diseases usually display abnormal growth patterns as well as delayed puberty. This is a result of several factors related to the disease itself, such as malnutrition, hypercortisolism, and elevated levels of pro-inflammatory cytokines. These factors in combination with glucocorticoid treatment contribute to growth retardation during chronic inflammation by systemically affecting the major regulator of growth, the GH/IGF1 axis. However, recent studies have also shown evidence of a direct effect of these factors at the growth plate level. In conditions of chronic infla

Disturbance of Growth in Pediatric Patients Due to Osteomyelitis Caused by Growth Plate Infection.

Osteomyelitis, a severe bone infection, poses a multifaceted challenge to healthcare professionals. While its pathophysiology and treatment have been extensively studied, the impact of osteomyelitis on skeletal growth, particularly in pediatric patients, is an area that warrants attention. This abstract highlights the significance of understanding and managing growth disturbances in osteomyelitis, providing key findings and recommendations for clinicians. Understanding growth disturbance in osteomyelitis is essential because it can lead to lifelong consequences for pediatric patients. The infe

Growth plate cartilage-targeting nanoparticles for pharmacological treatment of hypochondroplasia.

Hypochondroplasia (HCH) is a systemic cartilage disorder caused by gain-of-function mutations in FGFR3, resulting in overactivation of signaling and short stature. Pharmacological therapy remains the primary treatment but provides only partial symptomatic relief, as the avascular and alymphatic nature of growth plate cartilage severely limits drug delivery and accumulation. To address this challenge, we developed CT-CM-NPs, a drug delivery system coated with primary chondrocyte membranes and further modified with the collagen II-binding peptide WYRGRL. This design retains the inherent biologic

Growth Plate Injuries: Advances and Future Directions in Regenerative Medicine.

Growth plate injuries account for up to 30% of pediatric fractures, with a substantial number leading to complications such as bony bar formation that can impair longitudinal growth, resulting in limb length discrepancies, angular deformities, or premature growth arrest. Conventional treatments focus on surgical resection of the bone bridge and interpositional grafting, but these approaches often fail to restore the native architecture or biological function of the physis, and recurrence is common. Recent advances in regenerative medicine offer promising alternatives that move beyond mechanica

Enhanced BMP signalling causes growth plate cartilage dysrepair in rats.

Growth plate cartilage injuries often result in bony repair at the injury site and premature mineralisation at the uninjured region causing bone growth defects, for which underlying mechanisms are unclear. With the prior microarray study showing upregulated bone morphogenetic protein (BMP) signalling during the injury site bony repair and with the known roles of BMP signalling in bone healing and growth plate endochondral ossification, this study used a rat tibial growth plate drill-hole injury model with or without systemic infusion of BMP antagonist noggin to investigate roles of BMP signall

Effect of thiram on avian growth plate chondrocytes in culture.

Thiram is a dithiocarbamate pesticide that causes tibial dyschondroplasia (TD), a growth plate defect, in poultry. Deaths of transitional zone chondrocytes appear to interrupt endochondral bone development leading to the broadening of growth plate. The mechanism of action of thiram on chondrocytes is not well understood. Since proteins play major roles in different aspects of cell's metabolism, growth, and survival, the objective of this study was to find whether thiram produces proteomic changes that could impair the development of chondrocytes. The chondrocytes, isolated from proximal tibial

Growth plate mechanics and mechanobiology. A survey of present understanding.

The longitudinal growth of long bones occurs in growth plates where chondrocytes synthesize cartilage that is subsequently ossified. Altered growth and subsequent deformity resulting from abnormal mechanical loading is often referred to as mechanical modulation of bone growth. This phenomenon has key implications in the progression of infant and juvenile musculoskeletal deformities, such as adolescent idiopathic scoliosis, hyperkyphosis, genu varus/valgus and tibia vara/valga, as well as neuromuscular diseases. Clinical management of these deformities is often directed at modifying the mechani

Iliac crest growth plate analysis in slipped capital femoral epiphysis.

Previous studies have demonstrated numerous histologic and histochemical abnormalities in the involved growth plate of patients with slipped capital femoral epiphysis (SCFE). Whether these changes are primary or secondary with regard to the slip is unknown. We examined an uninvolved site of patients with SCFE and controls, the iliac crest growth plate, by histochemical means for general morphology, proteoglycans, glycoproteins, and collagen fibril orientation. No differences were noted between the two groups studied. This observation suggests that previously reported slipped capital femoral ep

Growth-plate behavior in whole joint replantation in the rabbit.

We studied the behavior of the distal femoral and proximal tibial physes after whole knee-joint replantation in skeletally immature New Zealand White Rabbits. We developed an operative model in which a rabbit's knee was isolated on a vascular pedicle and was then replanted using microvascular technique. In a short-term study (three weeks or less) of ten rabbits, the patency of the vascular repairs and the resumption of metabolic activity of the cells in the growth plates were assessed. A long-term study (ninety days) of eight rabbits was designed to monitor growth after replantation. The resul

Compression injury of the epiphyseal growth plate: fact or fiction?

It has been hypothesized that a longitudinal compression force on a growing long bone may cause partial or complete closure of a physis by crushing the germinal cell layer. In the absence of a fracture of the physis, the roentgenograms at the time of injury would be normal. Thus, this diagnosis can be made only in retrospect. In order to prove such a hypothesis, the following criteria would have to be met: (a) normal findings on roentgenograms taken in at least two planes at the time of injury, (b) no treatment, and (c) subsequent positive roentgenographic findings showing partial or complete

Growth plate extracellular matrix defects and short stature in children.

Many etiological factors causing short stature have already been identified in humans. In the last few years, the advent of new techniques for the detection of chromosomal and molecular abnormalities has made it possible to better identify patients with genetic causes of growth failure. Some of these factors directly affect the development and growth of the skeleton, since they damage the epiphyseal growth plate, where linear growth occurs, influencing chondrogenesis. In particular, defects in genes involved in the organization and function of the growth plate are responsible for several well-

Guided growth using a tension-band plate in Blount's disease.

Our observational study's objective was to determine how effective guided growth with tension-band plates was to correct the deformity in Blount's disease. We reviewed the records of 14 children (18 limbs) with Blount's disease who were treated with tension-band plates as the only surgical intervention at a single institution over eight years. Five children (seven limbs) had infantile Blount's disease with Langenskiöld stage ≤2. Nine children (11 limbs) had late-onset Blount's disease. The mean age at operation was 7.2 years (SD, 3.1, range, 2.9-11.8). The tension-band plate effectively correc

Familial Short Stature-A Novel Phenotype of Growth Plate Collagenopathies.

CONTEXT: Collagens are the most abundant proteins in the human body. In a growth plate, collagen types II, IX, X, and XI are present. Defects in collagen genes cause heterogeneous syndromic disorders frequently associated with short stature. Less is known about oligosymptomatic collagenopathies. OBJECTIVE: This work aims to evaluate the frequency of collagenopathies in familial short stature (FSS) children and to describe their phenotype, including growth hormone (GH) treatment response. METHODS: Eighty-seven FSS children (pretreatment height ≤ -2 SD both in the patient and his or her shorte

[Prevalence of metatarsal growth plate lesions in Swiss fattening bulls].

The prevalence of osteochondral changes in the metatarsal growth plates of fattening bulls was investigated. Plantarodorsal radiographic views of the metatarsal region of 204 hind legs from 102 slaughtered fattening bulls were taken. Radiographic lesions in metatarsal growth plates were confirmed in selected cases using computed tomography (n=14) and histology (n=6). Radiographic criteria for growth plate lesions were axis deviations between the metatarsal bones and the digit, radiolucencies in the growth plate, increased bone density in the metaphysis, irregularities of the growth plate and m

Characterisation of Growth Plate Dynamics in Murine Models of Osteoarthritis.

The purpose of this study was to investigate growth plate dynamics in surgical and loading murine models of osteoarthritis, to understand whether abnormalities in these dynamics are associated with osteoarthritis development. 8-week-old C57BL/6 male mice underwent destabilisation of medial meniscus (DMM) (n = 8) surgery in right knee joints. Contralateral left knee joints had no intervention (controls). In 16-week-old C57BL/6 male mice (n = 6), osteoarthritis was induced using non-invasive mechanical loading of right knee joints with peak force of 11N. Non-loaded left knee joints were internal

Emulsion-free chitosan-genipin microgels for growth plate cartilage regeneration.

The growth plate is a cartilage tissue near the ends of children's long bones and is responsible for bone growth. Injury to the growth plate can result in the formation of a 'bony bar' which can span the growth plate and result in bone growth abnormalities in children. Biomaterials such as chitosan microgels could be a potential treatment for growth plate injuries due to their chondrogenic properties, which can be enhanced through loading with biologics. They are commonly fabricated via an emulsion method, which involves solvent rinses that are cytotoxic. Here, we present a high throughput, no

SHP2 Regulates the Osteogenic Fate of Growth Plate Hypertrophic Chondrocytes.

Transdifferentiation of hypertrophic chondrocytes into bone-forming osteoblasts has been reported, yet the underlying molecular mechanism remains incompletely understood. SHP2 is an ubiquitously expressed cytoplasmic protein tyrosine phosphatase. SHP2 loss-of-function mutations in chondroid cells are linked to metachondromatosis in humans and mice, suggesting a crucial role for SHP2 in the skeleton. However, the specific role of SHP2 in skeletal cells has not been elucidated. To approach this question, we ablated SHP2 in collagen 2α1(Col2α1)-Cre- and collagen 10α1(Col10α1)-Cre-expressing cells
