Gamma-aminobutyric acid attenuates cortisol-induced damage in human colorectal adenocarcinoma cells via Nrf2 signaling.

Long-term psychological stress is associated with increased intestinal epithelial permeability. In the human central nervous system, gamma-aminobutyric acid (GABA), a non-protein amino acid found in bacteria, plants, and animals, acts as an inhibitory neurotransmitter that controls the cardiovascular system, reduces blood pressure, enhances mood, and encourages sleep. It is still unclear how GABA controls the function of the colon epithelial barrier under long-term stress. This study explored the potential of GABA to ameliorate cortisol-induced damage in human colorectal adenocarcinoma cells (

Ingested Engineered Nanomaterials Affect the Expression of Mucin Genes-An In Vitro-In Vivo Comparison.

The increasing use of engineered nanomaterials (ENM) in food has fueled the development of intestinal in vitro models for toxicity testing. However, ENM effects on intestinal mucus have barely been addressed, although its crucial role for intestinal health is evident. We investigated the effects of ENM on mucin expression and aimed to evaluate the suitability of four in vitro models of increasing complexity compared to a mouse model exposed through feed pellets. We assessed the gene expression of the mucins MUC1, MUC2, MUC5AC, MUC13 and MUC20 and the chemokine interleukin-8 in pre-confluent an

Preclinical evaluation of novel imidazoacridinone derivatives with potent activity against experimental colorectal cancer.

Novel imidazoacridinone derivatives, C1310 and C1311, have been evaluated for their potential to inhibit tumour cell growth in vitro and in vivo. A cell line panel, including seven human and murine colon carcinoma cell lines and three in vivo models, was used. The compounds were found to be potent inhibitors of tumour cell growth with IC50 values ranging between 10 nM and 2 microM in human colon cancer cell lines. Statistically significant tumour growth delay (P < 0.01) was observed after a single intraperitoneal (i.p.) dose of C1311 (100 mg kg-1 body weight) in MAC15A, MAC29 murine and HT29 h

Fenretinide targeting of human colon cancer sphere cells through cell cycle regulation and stress-responsive activities.

Cancer stem cells (CSCs) are considered to be the main cause of chemoresistance and the resultant low survival rate of patients with cancer. N-(4-hydroxyphenyl)-retinamide, known as fenretinide or 4HPR, is a synthetic derivative of all-trans-retinoic acid. It is a promising anticancer agent, has minimal side effects and synergizes with other anticancer agents to reinforce their anticancer efficacy. The present study investigated whether fenretinide eliminated colon sphere cells. HT29 and HCT116 cells incubated in low-serum culture medium were more sensitive to fenretinide treatment than those

The role of Ca2+/calmodulin-dependent protein kinase II in oxaliplatin resistance of colorectal cancer: In vitro and in vivo studies.

BACKGROUND: Oxaliplatin (Oxa) resistance is a major barrier in colorectal cancer (CRC) treatment. The Ca2+/CaM/CaMKII pathway is involved in tumor progression, but its role in chemoresistance remains unclear. METHODS: Oxa-resistant HCT116 and HT29 sublines (HCT116-Oxa and HT29-Oxa) were established through gradual dose escalation. The activation status of the Ca2+/CaM/CaMKII pathway was assessed by measuring intracellular Ca2+ levels and phosphorylation of CaMKII. Functional studies such as CCK-8, clonogenic assays, and flow cytometry-based apoptosis analysis were conducted to assess the impa

Polycystin-1 affects cancer cell behaviour and interacts with mTOR and Jak signalling pathways in cancer cell lines.

Polycystic Kidney Disease (PKD), which is attributable to mutations in the PKD1 and PKD2 genes encoding polycystin-1 (PC1) and polycystin-2 (PC2) respectively, shares common cellular defects with cancer, such as uncontrolled cell proliferation, abnormal differentiation and increased apoptosis. Interestingly, PC1 regulates many signalling pathways including Jak/STAT, mTOR, Wnt, AP-1 and calcineurin-NFAT which are also used by cancer cells for sending signals that will allow them to acquire and maintain malignant phenotypes. Nevertheless, the molecular relationship between polycystins and cancer

Flavokawain C alleviates dextran sulphate sodium-induced colitis through suppressing necroptosis via blocking RIPK1-RIPK3-MLKL axis.

BACKGROUND: Kava (Piper methysticum G. Forst) is a plant native to Pacific Island countries, consumed as a beverage in social and ceremonial contexts, and also used as a traditional medicine for various types of pain and anxiety. Flavokawain C, one of the primary active chalcones isolated from Kava, has been demonstrated to possess antitumor properties. However, its effects and underlying mechanisms in ulcerative colitis (UC) remain unreported. Necroptosis is closely associated with UC pathogenesis, and inhibiting necroptosis represents a potential therapeutic strategy for UC. This study aimed

Cloning and functional expression of a human neuroendocrine vasoactive intestinal peptide receptor.

Vasoactive intestinal peptide (VIP) is a potent mediator of gastrointestinal, nervous, pulmonary, vascular and immune functions. A cDNA was obtained from human HT29 intestinal epithelial cells and found to encode a 457 amino acid, 52 kDa VIP receptor. Transfection of the cDNA into COS-7 cells and 293 cells resulted in expression of specific saturable binding of VIP with a Kd of 0.8 nM, and induction of increases in intracellular cAMP by VIP with an EC50 of 1 nM. The human VIP receptor is homologous to other G protein-coupled receptors of the secretin-parathyroid hormone receptor family. A 2.8

The paradigm-shifting idea and its practice: from traditional abortion Chinese medicine Murraya paniculata to safe and effective cancer metastatic chemopreventives.

Recent large epidemiological studies demonstrated benefit of oral contraceptives in reducing cancer risk, and our analysis also showed molecular and cellular similarities between embryo implantation and CTCs adhesion-invasion to endothelium. We here hypothesize that abortion traditional Chinese medicine (TCM) may serve well for pre-metastatic chemoprevention. To test the hypothesis, we selected the safe and well-known abortifacient TCM Murraya paniculata and identified a most-promising extracted fraction G (containing flavonoids and coumarins) from its many raw ethanol/dichloromethane extracts

Synthesis and anticancer activity of betulin-succinate derivatives in vitro and in the zebrafish xenograft tumor model.

Betulin is a naturally occurring triterpene holding great promise as a lead for developing new antitumor agents. Given that the design, synthesis and anticancer activity of betulin-succinate derivatives remain unexplored, we herein reported the synthesis and cytotoxicity evaluation of such 22 new derivatives against A172, HT29, SW579, and TCA8113 cancer cell lines in vitro using the CCK8 (Cell Counting Kit-8) assay. The results revealed that most compounds showed potent antitumor activity relative to the positive control and betulin. Particularly, compound 21 also exhibited superior activities

Synthesis and Antiproliferative Activity against Cancer Cells of Indole-Aryl-Amide Derivatives.

Indoles constitute a large family of heterocyclic compounds widely occurring in nature which are present in a number of bioactive natural and synthetic compounds, including anticancer agents or atypical opioid agonists. As a result, exponential increases in the development of novel methods for the synthesis of indole-containing compounds have been reported in the literature. A series of indole-aryl amide derivatives 1-7 containing tryptamine or an indolylacetic acid nucleus were designed, synthesized, and evaluated as opioid ligands. These new indole derivatives showed negligible to very low a

A role for tumor necrosis factor and bacterial antigens in the pathogenesis of Crohn's disease-associated fistulae.

BACKGROUND: Intestinal fistulae represent a severe complication of Crohn's disease (CD). The authors have demonstrated that epithelial-to-mesenchymal transition plays a pivotal role in their pathogenesis. High levels of interleukin-13 and tumor necrosis factor (TNF) are detected in myofibroblast-like transitional cells covering the fistula tracts. Here, a functional role was investigated for the transcription factor Ets-1, TNF, and the bacterial wall component (muramyl dipeptide [MDP]) in the pathogenesis of CD-associated fistulae. METHODS: Perianal fistulae from CD patients were analyzed by

H2 blockers decrease gut mucus production and lead to barrier dysfunction in vitro.

BACKGROUND: Pneumonia and other gut-related infectious complications have been associated with the use of histamine 2 (H2) receptor antagonists such as cimetidine in critically ill patients. The mechanism(s) may include acid suppression with resultant effects on the gut flora. Other possibilities include immunologic effects and perturbation of gut barrier function. Recent work has demonstrated the importance of mucus on the gastrointestinal mucosal barrier. We studied the effect of cimetidine on mucus production and mucosal barrier function in vitro. METHODS: HT29-MTX, a mucus-producing intes
