Muscarinic Acetylcholine M2 Receptors Regulate Lateral Habenula Neuron Activity and Control Cocaine Seeking Behavior.

The lateral habenula (LHb) balances reward and aversion by opposing activation of brain reward nuclei and is involved in the inhibition of responding for cocaine in a model of impulsive behavior. Previously, we reported that the suppression of cocaine seeking was prevented by LHb inactivation or nonselective antagonism of LHb mAChRs. Here, we investigate mAChR subtypes mediating the effects of endogenous acetylcholine in this model of impulsive drug seeking and define cellular mechanisms in which mAChRs alter LHb neuron activity. Using in vitro electrophysiology, we find that LHb neurons are d

Bilateral Habenula deep brain stimulation for treatment-resistant depression: clinical findings and electrophysiological features.

Deep brain stimulation (DBS) of structures in the brain's reward system is a promising therapeutic option for patients with treatment-resistant depression (TRD). Recently, DBS of the habenula (HB) in the brain's anti-reward system has also been reported to alleviate depressive symptoms in patients with TRD or bipolar disorder (BD). In this pilot open-label prospective study, we explored the safety and clinical effectiveness of HB-DBS treatment in seven patients with TRD or BD. Also, local field potentials (LFPs) were recorded from the patients' left and right HB to explore the power and asymme

Malocclusion Generates Anxiety-Like Behavior Through a Putative Lateral Habenula-Mesencephalic Trigeminal Nucleus Pathway.

Malocclusion is an important risk factor for temporomandibular disorder (TMD), a series of disorders characterized by dysfunction in the orofacial region involving the temporomandibular joint (TMJ) and jaw muscles. We recently showed that experimental unilateral anterior crossbite (UAC) produced masseter hyperactivity through a circuit involving the periodontal proprioception, trigeminal mesencephalic nucleus (Vme), and trigeminal motor nucleus (Vmo). Anxiety is a common complication in patients with TMD. The lateral habenula (LHb) is involved in emotional modulation and has direct projections

Ancestral organisation and asymmetric circuit architecture of the habenula revealed in a basal ray-finned fish.

The habenula is a conserved dorsal diencephalic hub that integrates information related to motivational state, behavioural flexibility, and cognitive control. In many vertebrates, it displays pronounced left-right asymmetry, yet how structural lateralisation is reflected in circuit architecture remains poorly understood, partly because phylogenetically informative lineages remain undersampled. Here, we characterise habenular connectivity in the Senegal bichir (Polypterus senegalus), a member of an early-diverging ray-finned fish lineage with marked habenular asymmetry. Using neuronal tracing,

Habenula as a Marker of Thalamic Asymmetrical Skew and a Landmark for the Vim-Vc Boundary in MRgFUS Thalamotomy.

BACKGROUND: Traditional targeting based on the anterior commissure-posterior commissure (AC-PC) line in MR-guided focused ultrasound (MRgFUS) thalamotomy does not account for individual thalamic asymmetry, potentially leading to suboptimal outcomes or adverse effects. OBJECTIVE: To investigate whether habenular torsion, an anatomical marker of thalamic asymmetry, predicts clinical outcomes and sensory side effects following ventral intermediate nucleus (Vim) thalamotomy. METHODS: We retrospectively analyzed 157 patients (essential tremor: 121; Parkinson's disease: 36) who underwent unilatera

Neuronal activation in the paraventricular thalamus and lateral habenula during activity-based anorexia in mice.

Self-starvation in anorexia nervosa patients ultimately progresses into a state of energy deficit and the body responds with homeostatic changes giving rise to many of the core symptoms. To accelerate progress towards effective therapies, we need to better understand the neurobiological underpinnings of this disease. Hence, we sought to identify brain regions of potential importance for anorexia nervosa by exploring neuronal activity in the activity-based anorexia (ABA) mouse model using Fos-TRAP2 technique. Given the function of many nuclei in the dorsal thalamus that centre around the integr

A visual circuit related to the habenula mediates the prevention of cocaine relapse by bright light treatment.

Despite significant advancements in our understanding of addiction at the neurobiological level, a highly effective extinction procedure for preventing relapse remains elusive. In this study, we report that bright light treatment (BLT) administered during cocaine withdrawal with extinction training prevents cocaine-driven reinstatement by acting through the thalamic-habenular pathway. We found that during cocaine withdrawal, the lateral habenula (LHb) was recruited, and inhibition of the LHb via BLT prevented cocaine-driven reinstatement. We also demonstrated that the effects of BLT were media

Habenula responses to potential and actual loss in major depression: preliminary evidence for lateralized dysfunction.

The habenula has been implicated in predicting negative events and in responding to unexpected negative outcomes. Animal models of depression have supported the hypothesis that perturbations in habenula activity contribute to the pathophysiology of Major Depressive Disorder (MDD), a psychiatric illness characterized by abnormalities in responding to negative feedback and by pessimism in evaluating the likelihood of future events. No research to date, however, has examined human habenula responses to potential and experienced negative outcomes in MDD. In this study, depressed and healthy contro

Habenula as a relay in the descending pathway from nucleus accumbens to periaqueductal grey subserving antinociception.

This study explored the possibility of a relay at habenula for the descending neural pathway of antinociception. The latency of the escape response elicited by radiant heat on the snout of the rabbit was taken as index of nociception. (1) Microinjection of 20 micrograms of morphine into nucleus accumbens resulted in a one-fold increase in nociceptive threshold 20-40 min after the injection. This effect of morphine was markedly attenuated by naloxone or met-enkephalin antiserum administered to the nucleus habenula, suggesting that the release of met-enkephalin in habenula is essential for the a

TMEM16A ablation in cholinergic medial habenula neurons induces early-onset schizophrenia-like phenotypes in mice.

Schizophrenia is a heterogeneous psychiatric disorder that remains inadequately treated with current therapies. Developing appropriate animal models that reflect the broad spectrum of schizophrenia symptoms is crucial for advancing our understanding of the disease and identifying effective treatments. However, existing animal models often have limitations in fully recapitulating the diverse symptomatology observed in humans. Previously, we reported that mice with conditional ablation of TMEM16A (ANO1) in cholinergic neurons of the medial habenula (ANO1 cKO) exhibit behavioral patterns indicati

Post-traumatic stress disorder-induced behavioral modulation by the medial septum-medial habenula neural pathway.

Post-traumatic stress disorder (PTSD) is characterized by anxiety, excessive fear, distress, and weakness as symptoms of a psychiatric disorder. However, the mechanism associated with its symptoms such as anxiety-like behaviors is not well understood. It is aimed to investigate the underlying mechanisms of the medial septum (MS)-medial habenula (MHb) neural circuit modulating the anxiety-like behaviors of PTSD mice through in vivo fiber photometry recording, optogenetics, behavioral testing by open-field and elevated plus maze, fluorescent gold retrograde tracer technology, and viral tracer te

Hyperactive mTORC1 disrupts habenula function and light preference in zebrafish model of Tuberous sclerosis complex.

Mechanistic target of rapamycin complex 1 (mTORC1) is an integration hub for extracellular and intracellular signals necessary for brain development. Hyperactive mTORC1 is found in autism spectrum disorder (ASD) characterized by atypical reactivity to sensory stimuli, among other symptoms. In Tuberous sclerosis complex (TSC) inactivating mutations in the TSC1 or TSC2 genes result in hyperactivation of the mTORC1 pathway and ASD. Here, we show that lack of light preference of the TSC zebrafish model, tsc2 vu242/vu242 is caused by aberrant processing of light stimuli in the left dorsal habenula

Virtual social interaction reveals that the dorsal habenula-IPN pathway is essential for targeting the opponent.

Conspecific social interaction is among the most important brain functions to be investigated. We designed a virtual reality (VR) system that enables real-time teleportation of head-fixed adult zebrafish movements onto their 3D avatars. To facilitate dyadic fight-like behavior in the VR space, we devised a task in which avoidance of periodic mild electric stimulation is achieved by attacking the opponent, and a transient electric shock acts as negative feedback for bite-like events. Moreover, a transgenic fish with an impaired dorsolateral habenula-to-interpeduncular nucleus pathway, known as

Cocaine Blocks Cholinergic Activity in the Medial Habenula Prior to But Not After Induced Preference for the Drug.

Descending projections from the medial habenula potently influence brainstem systems associated with reward and mood. Relatedly, the ventral, cholinergic segment of the nucleus has been linked to nicotine and cocaine addiction. Here we report that cocaine has no effect on baseline firing in the ventral medial habenula but entirely blocks the self-sustained activity initiated by endogenous acetylcholine. This effect was not altered by antagonists to dopamine receptors and thus presumably reflects a direct action on cholinergic receptors. Remarkably, cocaine had no effect on endogenous cholinerg

Static and dynamic functional connectivity of the habenula in late-life depression patient with suicidal ideation.

BACKGROUND: Suicide is one of the most lethal complications of late-life depression (LLD), and habenular dysfunction may be involved in depression-related suicidality and may serve as a potential target for alleviating suicidal ideation. This study aimed to investigate abnormal functional connectivity of the habenula in LLD patients with suicidal ideation. METHODS: One hundred twenty-seven patients with LLD (51 with suicidal ideation (LLD-S) and 76 without suicidal ideation (LLD-NS)) and 75 healthy controls (HCs) were recruited. The static functional connectivity (sFC) and dynamic functional

Habenula activation patterns in a preclinical model of neuropathic pain accompanied by depressive-like behaviour.

Pain and depression are complex disorders that frequently co-occur, resulting in diminished quality of life. The habenula is an epithalamic structure considered to play a pivotal role in the neurocircuitry of both pain and depression. The habenula can be divided into two major areas, the lateral and medial habenula, that can be further subdivided, resulting in 6 main subregions. Here, we investigated habenula activation patterns in a rat model of neuropathic pain with accompanying depressive-like behaviour. Wistar rats received active surgery for the development of neuropathic pain (chronic co

Stimulation of mu opioid, but not GABAergic, receptors of the lateral habenula alters free feeding in rats.

Overconsumption, or eating beyond the point of homeostasis, is a key feature in the development of obesity. Although people are consuming beyond the point of homeostasis, they are not consuming constantly or indefinitely. Thus, there is likely a mechanism that acts to terminate periods of food intake at some point beyond satiation and prior to aversion, or the negative effects of extreme excess (nausea, bloating, etc.). The purpose of the present study was to assess the lateral habenula as a candidate region for such a mechanism, due to its connectivity to midbrain reward circuitry, sensitivit

Shifted pallidal co-release of GABA and glutamate in habenula drives cocaine withdrawal and relapse.

Cocaine withdrawal produces aversive states and vulnerability to relapse, hallmarks of addiction. The lateral habenula (LHb) encodes negative stimuli and contributes to aversive withdrawal symptoms. However, it remains unclear which inputs to LHb promote this and what the consequences are for relapse susceptibility. We report, using rabies-based retrolabeling and optogenetic mapping, that the entopeduncular nucleus (EPN, the mouse equivalent of the globus pallidus interna) projects to an LHb neuronal subset innervating aversion-encoding midbrain GABA neurons. EPN-to-LHb excitatory signaling is

Excitation of lateral habenula neurons as a neural mechanism underlying ethanol-induced conditioned taste aversion.

KEY POINTS: The lateral habenula (LHb) has been implicated in regulation of drug-seeking behaviours through aversion-mediated learning. In this study, we recorded neuronal activity in the LHb of rats during an operant task before and after ethanol-induced conditioned taste aversion (CTA) to saccharin. Ethanol-induced CTA caused significantly higher baseline firing rates in LHb neurons, as well as elevated firing rates in response to cue presentation, lever press and saccharin taste. In a separate cohort of rats, we found that bilateral LHb lesions blocked ethanol-induced CTA. Our results stron

5HT(1B) receptor-mediated pre-synaptic depression of excitatory inputs to the rat lateral habenula.

Accumulating lines of evidence indicate that the lateral habenula (LHb), which reciprocally interacts with raphe nuclei (RN), displays hyperactivity including synaptic potentiation of excitatory inputs to the LHb during a depressed state. Despite the potential importance of glutamatergic excitatory synapses in depression-like behavior, modulation of these LHb synapses by monoamines such as serotonin (5HT) is not fully understood at the cellular and molecular level. Therefore, we used whole cell voltage-clamp recording to examine the molecular mechanisms by which 5HT modulates glutamatergic tra
