Whole Genome Sequence Analysis of Pig Respiratory Bacterial Pathogens with Elevated Minimum Inhibitory Concentrations for Macrolides.

Macrolides are often used to treat and control bacterial pathogens causing respiratory disease in pigs. This study analyzed the whole genome sequences of one clinical isolate of Actinobacillus pleuropneumoniae, Haemophilus parasuis, Pasteurella multocida, and Bordetella bronchiseptica, all isolated from Australian pigs to identify the mechanism underlying the elevated minimum inhibitory concentrations (MICs) for erythromycin, tilmicosin, or tulathromycin. The H. parasuis assembled genome had a nucleotide transition at position 2059 (A to G) in the six copies of the 23S rRNA gene. This mutation

[Bacteriological diagnosis of Haemophilus pleuropneumonia in swine].

Haemophilus pleuropneumonia (Matthews and Pattison, 1961; Shope, 1964) should now be accepted as the valid name of the pathogen of hemophilus pleuropneumoniae of swine, according to studies and the description by Kilian and co-workers (1978), while the name of Haemophilus parahaemolyticus should be considered a synonym. On top of Haemophilus pleuropneumoniae, Haemophilus parasuis and Haemophilus spec. "minor group" are also important to pneumonia, serositis, and arthritis (Glässer's disease) in swine, whereas Haemophilus suis is hardly any longer isolated. Evidence as to dependence on growth f

Baicalin Alleviate Apoptosis via PKC-MAPK Pathway in Porcine Peritoneal Mesothelial Cells Induced by Glaesserella parasuis.

Glaesserella parasuis (GPS), a causative agent of Glässer's disease, is thought to be the main fatal cause of peritonitis in swine, thus resulting in high mortality and morbidity and significant economic losses to the swine industry. However, the mechanisms of GPS infection-induced apoptosis and possible therapeutic pathway for GPS infection in peritonitis remain unclear. Baicalin has important biological functions during disease treatment, such as antiviral, bacterial inhibition, anti-apoptosis, and anti-inflammatory. However, whether baicalin has anti-apoptotic effects during the process of

Comparative study of the effects of baicalin and probenecid on microRNA expression profiles in porcine aortic vascular endothelial cells infected by Glaesserella parasuis.

BACKGROUND: Glaesserella parasuis elicits severe inflammatory responses and vascular damage, thus resulting in high mortality and morbidity in pigs; consequently, early diagnosis and treatment are critical to controlling economic losses. MicroRNAs (miRNAs) have been demonstrated to be involved in vascular endothelial inflammation. Baicalin is an effective Chinese medicinal herb with anti-microbial, anti-inflammatory, and anti-oxidant activity. Probenecid has activity toward multiple mammalian biological processes. Herein, we compared the effects of baicalin and probenecid on the miRNA expressi

Acute phase protein concentrations in colostrum-deprived pigs immunized with subunit and commercial vaccines against Glässer's disease.

Haemophilus parasuis is the etiological agent of Glässer's disease, which is characterized by fibrinous polyserositis, polyarthritis and meningitis in pigs. This study was focused on the characterization of the acute-phase response after immunization and infection of colostrum-deprived pigs with H. parasuis serovar 5, by measuring serum concentrations of three positive acute-phase proteins (APPs) (pig major acute-phase protein pig, MAP; haptoglobin, HPG; C-reactive protein, CRP) and one negative APP (apolipoprotein A-I, ApoA-I). Six experimental groups were established: a non-immunized but inf

IL-17 signaling pathway in monocytes drives the persistent inflammatory cytokine storm caused by Glaesserella parasuis infection in pigs.

Glaesserella parasuis, the causative agent of Glässer's disease, is a significant swine pathogen characterized by fibrinous polyserositis, meningitis, and arthritis, causing significant economic losses to the global swine industry. In this study, infection of piglets with G. parasuis induced a sustained systemic inflammatory response, marked by persistently elevated mRNA expression levels of pro-inflammatory cytokines (IL-1β, IL-6, IL-8, TNF-α) and acute-phase proteins (SAA, HP) in whole blood. This response was accompanied by severe inflammatory cell infiltration in the lungs, heart, spleen,

Methylome and Transcriptome-Based Integration Analysis Identified Molecular Signatures Associated With Meningitis Induced by Glaesserella parasuis.

Glaesserella parasuis (G. parasuis) can elicit serious inflammatory responses and cause meningitis in piglets. Previous epigenetic studies have indicated that alterations in host DNA methylation may modify the inflammatory response to bacterial infection. However, to date, genome-wide analysis of the DNA methylome during meningitis caused by G. parasuis infection is still lacking. In this study, we employed an unbiased approach using deep sequencing to profile the DNA methylome and transcriptome from G. parasuis infected porcine brain (cerebrum) and integrated the data to identify key differen

Swine Influenza Virus and Association with the Porcine Respiratory Disease Complex in Pig Farms in Southern Brazil.

Despite the putative endemic status of swine influenza A virus (swIAV) infections, data on the occurrence of swine influenza outbreaks are scarce in Brazil. The aim of this study was to detect and subtype swIAVs from six outbreaks of porcine respiratory disease complex (PRDC) in southern Brazil. Nasal swabs were collected from 66 piglets with signs of respiratory disease in six herds. Lung tissue samples were collected from six necropsied animals. Virus detection was performed by PCR screening and confirmed by virus isolation and hemagglutination (HA). Influenza A subtyping was performed by a

Baicalin, Amoxicillin, and Probenecid Provide Protection in Mice Against Glaesserella parasuis Challenge.

Glaesserella parasuis (G. parasuis) causes Glässer's disease and systemic inflammatory responses in the host. The currently available therapies have limited efficacy and fail to achieve a balance between anti-inflammatory and antibacterial effects. In this study, we investigated the effects of baicalin, amoxicillin, and probenecid on blood biochemical parameters, routine blood indicators, survival rate, bacterial burden, and pathological tissue damage in G. parasuis-challenged mice. Treatment with baicalin, amoxicillin, and probenecid significantly modified the blood biochemical parameters and

Antibacterial effects of cinnamaldehyde and hesperitin on resistant Glaesserella parasuis by suppressing QseBC two-component system.

BACKGROUND: Glaesserella parasuis (G. parasuis) is one of the most important porcine pathogens causing Glässer's disease, and QseBC two-component system (TCS) is associated with various behaviors of G. parasuis. Our preliminary tests confirmed that plant-derived compounds cinnamaldehyde (CAL) and hesperitin (HES) exhibited promising antimicrobial activity against G. parasuis. Here, we further investigate the antimicrobial effects of CAL and HES on G. parasuis and the underlying mechanisms. RESULTS: We observed that CAL and HES affected the morphology and physiology of G. parasuis based on the

Characterization of a universal neutralizing monoclonal antibody against Glaesserella parasuis CdtB.

Glaesserella parasuis is the etiological agent of Glässer's disease. Although present as a symbiotic bacterium in the respiratory tract of healthy pigs, G. parasuis invades piglets under stress conditions and causes severe systemic infection characterized by fibrinous polyserositis, polyarthritis, and meningitis, which caused high mortality in weaned and nursery herds. Further, the lack of cross-protection against the various serotypes of G. parasuis is a serious concern for the swine industry. Cytolethal distending toxin (CDT) is essential for the pathogenicity of G. parasuis and is a conserv

Glaesserella parasuis serotype 5 disrupts the swine respiratory epithelial barrier via NBR1-mediated selective autophagic degradation of Claudin-1.

Glaesserella parasuis (G. parasuis) colonizes the upper respiratory tract of pigs and can cause systemic infection through disruption of the epithelial barrier of the respiratory tract, resulting in substantial economic losses to the swine industry worldwide. A previous study revealed that infection with the G. parasuis serotype 5 strain HPS5-SQ breaks through the swine respiratory epithelial barrier by inducing autophagic degradation of the tight junction protein Claudin-1. However, the underlying mechanism remains incompletely understood. In this study, co-immunoprecipitation (Co-IP) results

Development of a one-run real-time PCR detection system for pathogens associated with porcine respiratory diseases.

The etiology of Porcine respiratory disease complex is complicated by infections with multiple pathogens, and multiple infections increase the difficulty in identifying the causal pathogen. In this present study, we developed a detection system of microbes from porcine respiratory by using TaqMan real-time PCR (referred to as Dempo-PCR) to screen a broad range of pathogens associated with porcine respiratory diseases in a single run. We selected 17 porcine respiratory pathogens (Actinobacillus pleuropneumoniae, Boldetella bronchiseptica, Haemophilus parasuis, Pasteurella multocida, Pasteurella

Copper-zinc superoxide dismutase in Haemophilus species.

Copper-zinc superoxide dismutases ([Cu,Zn]-SODs) are ubiquitous in eukaryotes but have rarely been found in prokaryotes. A gene for [Cu,Zn]-SOD (sodC) has recently been cloned from Haemophilus influenzae type b and H. parainfluenzae, so other Haemophilus and related species were screened for the presence of [Cu,Zn]-SODs by visualization of bands of SOD activity in non-denaturing polyacrylamide gels and by gene probing. Strains of H. aphrophilus, H. paraphrophilus, H. haemolyticus, H. paraphrohaemolyticus, some non-typable H. influenzae, H. haemoglobinophilus (canis) and H. parasuis were all fo

Glaesserella parasuis serotype 4 exploits fibronectin via RlpA for tracheal colonization following porcine circovirus type 2 infection.

Porcine circovirus type 2 (PCV2) often causes disease through coinfection with other bacterial pathogens, including Glaesserella parasuis (G. parasuis), which causes high morbidity and mortality, but the role played by PCV2 and bacterial and host factors contributing to this process have not been defined. Bacterial attachment is assumed to occur via specific receptor-ligand interactions between adhesins on the bacterial cell and host proteins adsorbed to the implant surface. Mass spectrometry (MS) analysis of PCV2-infected swine tracheal epithelial cells (STEC) revealed that the expression of
