Tonotopic specialization of MYO7A isoforms in auditory hair cells.

Mutations in Myo7a cause Usher syndrome type 1B and non-syndromic deafness, but the precise function of MYO7A in sensory hair cells remains unclear. Using long-read sequencing, we identify and characterize a novel isoform, MYO7A-N, expressed in auditory hair cells alongside the canonical MYO7A-C. Isoform-specific knock-in mouse models reveal that inner hair cells primarily express MYO7A-C, while outer hair cells express both isoforms in opposing tonotopic gradients. Both isoforms are localized to the upper tip-link insertion site, consistent with a role in the tip link for mechanotransduction.

Pathology and physiology of auditory neuropathy with a novel mutation in the MPZ gene (Tyr145->Ser).

We studied a family with hereditary sensory motor neuropathy and deafness accompanying a missense mutation in the MPZ gene. Pathological examination of the cochlea in one of the family members revealed marked loss of auditory ganglion cells and central and peripheral auditory nerve fibres within the cochlea. The inner hair cells were of normal number with preserved morphology. The outer hair cells were normal in number except for a 30% reduction in just the apical turn. Examination of the sural nerve and the auditory nerve adjacent to the brainstem showed marked loss of fibres with evidence of

Trimethyltin disrupts auditory function and cochlear morphology in pigmented rats.

Trimethyltin (TMT) produces auditory deficits, presumably of cochlear origin, in rats. The present study identified pathological changes in the cochlea following treatment with TMT and correlated them with auditory threshold changes. Thresholds were determined by reflex-modulation audiometry, before and after treatment with TMT or with saline vehicle. Animals were then perfused and their cochleas embedded for examination as block-surface preparations or radial sections. In the first week following treatment, all TMT-treated rats showed threshold shifts of 40 to 60 dB at 40 kHz, and smaller thr

Do Disinfectants Induce Ototoxicity and Cause Structural Damage to the Inner Ear?

BACKGROUND: This study examines the impact of commonly used disinfectants on hearing and the structural integrity of the inner ear. OBJECTIVE: To evaluate the effects of indophor, alcohol, and chlorhexidine on auditory function and cochlear morphology in mice. METHODS: Sixty-four mice were evenly divided into four groups: saline, indophor, alcohol, and chlorhexidine. Each mouse received a 25 μL injection of the designated liquid into the left ear, while the right ear was remained untreated. Auditory brainstem response (ABR) testing was performed at three intervals: before injection, 24 h pos

Human cochlear changes in noise induced hearing loss.

The temporal bone histopathological findings in 14 ears with noise induced hearing loss are presented. The morphological changes consist mainly of hair cell loss, which is more severe in the 9 mm to 13 mm region of the cochlear duct. Within the area of maximum hair cell loss, there is a greater loss of outer hair cells than of inner hair cells. There is a good correlation between the permanent auditory threshold shifts and the spatial location of the sensory lesion according to the anatomical frequency scale.

Revisiting the cochlear and central mechanisms of tinnitus and therapeutic approaches.

This short review aims at revisiting some of the putative mechanisms of tinnitus. Cochlear-type tinnitus is suggested to result from aberrant activity generated before or at the cochlear nerve level. It is proposed that outer hair cells, through their role in regulating the endocochlear potential, can contribute to the enhancement of cochlear spontaneous activity. This hypothesis is attractive as it provides a possible explanation for cochlear tinnitus of different aetiologies, such as tinnitus produced by acute noise trauma, intense low-frequency sounds, middle-ear dysfunction or temporomandi

ATP8B1-TMEM30B Flippase Activity Maintains Stereocilia Lipid Asymmetry Required for Hearing.

Sensory hair cells convert sound-induced vibrations into electrical signals through a process called mechanoelectrical transduction (MET). While the protein components of the MET complex are well studied, increasing evidence indicates that MET channel properties are significantly modulated by the surrounding lipid bilayer. The asymmetric distribution of membrane lipids between the inner and outer membrane leaflets is well established to shape membrane mechanics. The recent discovery that the core MET components TMC1 and TMC2 also act as lipid scramblases suggests a direct role for membrane lip

Variable number of TMC1-dependent mechanotransducer channels underlie tonotopic conductance gradients in the cochlea.

Functional mechanoelectrical transduction (MET) channels of cochlear hair cells require the presence of transmembrane channel-like protein isoforms TMC1 or TMC2. We show that TMCs are required for normal stereociliary bundle development and distinctively influence channel properties. TMC1-dependent channels have larger single-channel conductance and in outer hair cells (OHCs) support a tonotopic apex-to-base conductance gradient. Each MET channel complex exhibits multiple conductance states in ~50 pS increments, basal MET channels having more large-conductance levels. Using mice expressing flu

Fusion of stereocilia on inner hair cells in man and in the rabbit, rat and guinea pig.

The complexity of the problem of sensorineural hearing loss is illustrated by the well-known poor correlation between pure tone thresholds and loss of sensory cells in the organ of Corti and between pure tone thresholds and other auditory functions such as discrimination of speech and thresholds of the intra-aural muscle reflex (MER). The present work demonstrates that two types of inner ear pathology are present in several mammalian species, including man. Both types can be induced by noise. In addition to the earlier well-known loss of outer hair cells, it is shown that abnormal sensory hair

Differential Expression of Bcl-2 in the Cochlea and Auditory Cortex of a Mouse Model of Age-Related Hearing Loss.

Bcl-2, the first gene shown to be involved in apoptosis, is a potent regulator of cell survival and known to have protective effects against a variety of age-related diseases. However, the possible relationship between hearing and Bcl-2 expression in the cochlea or auditory cortex of C57BL/6 mice, a mouse model of age-related hearing loss, is still unknown. Using RT-PCR, immunohistochemistry, and Western blot analysis, our results show that Bcl-2 is strongly expressed in the inner hair cells and spiral ganglion neurons of young mice. In addition, moderate Bcl-2 expression is also detected in t

Myosin-XVa Controls Both Staircase Architecture and Diameter Gradation of Stereocilia Rows in the Auditory Hair Cell Bundles.

Mammalian hair cells develop their mechanosensory bundles through consecutive phases of stereocilia elongation, thickening, and retraction of supernumerary stereocilia. Many molecules involved in stereocilia elongation have been identified, including myosin-XVa. Significantly less is known about molecular mechanisms of stereocilia thickening and retraction. Here, we used scanning electron microscopy (SEM) to quantify postnatal changes in number and diameters of the auditory hair cell stereocilia in shaker-2 mice (Myo15sh2) that lack both "long" and "short" isoforms of myosin-XVa, and in mice l

Editorial: 'auditory neuropathy' and cochlear implantation - myths and facts.

A review of current opinion concerning 'auditory neuropathy' is presented. It is suggested that electrophysiological tests, including electrocochleography, auditory brainstem responses and electrically evoked auditory brainstem responses, together with imaging, can provide information regarding the site of the underlying pathological conditions that may produce the combination of otoacoustic emissions in the absence of auditory brainstem responses in children with hearing loss. It is suggested that in 75% of cases auditory neuropathy can merely be a result of surviving outer hair cells when in

Toluene disrupts outer hair cell morphometry and intracellular calcium homeostasis in cochlear cells of guinea pigs.

The aromatic hydrocarbon, toluene, has been demonstrated to disrupt auditory system function both in occupational epidemiological and in laboratory animal investigations. This agent, along with several other organic solvents, impairs hearing preferentially at middle frequencies-a finding that distinguishes these agents from the traditional high-frequency impairment observed with ototoxic drugs such as aminoglycoside antibiotics and cisplatin. Prior investigations have identified the outer hair cell as a probable target for toluene exposure, although studies designed to evaluate spiral ganglion

Hydroxypropyl-β-cyclodextrin causes massive damage to the developing auditory and vestibular system.

2-hydroxypropyl-β-cyclodextrin (HPβCD), a cholesterol chelator used to treat Niemann-Pick C1 (NPC1) lysosomal storage disease, causes hearing loss in mammals by preferentially destroying outer hair cells. Because cholesterol plays an important role in early neural development, we hypothesized that HPβCD would cause more extensive damage to postnatal cochlear and vestibular structures in than adult rats. This hypothesis was tested by administering HPβCD to adult rats and postnatal day 3 (P3) cochlear and vestibular organ cultures. Adult rats treated with HPβCD developed hearing impairment and o

Auditory neuropathy in children.

Auditory neuropathy is a sensorineural disorder characterized by absent or abnormal auditory brainstem evoked potentials and normal cochlear outer hair cell function. A variety of processes is thought to be involved in its pathophysiology and their influence on hearing may be different. We present here the diagnostic sequence and management of two new cases of auditory neuropathy in breastfeeding children.

An auditory-periphery model of the effects of acoustic trauma on auditory nerve responses.

Acoustic trauma degrades the auditory nerve's tonotopic representation of acoustic stimuli. Recent physiological studies have quantified the degradation in responses to the vowel /E/ and have investigated amplification schemes designed to restore a more correct tonotopic representation than is achieved with conventional hearing aids. However, it is difficult from the data to quantify how much different aspects of the cochlear pathology contribute to the impaired responses. Furthermore, extensive experimental testing of potential hearing aids is infeasible. Here, both of these concerns are addr

Pejvakin, a Candidate Stereociliary Rootlet Protein, Regulates Hair Cell Function in a Cell-Autonomous Manner.

Mutations in the Pejvakin (PJVK) gene are thought to cause auditory neuropathy and hearing loss of cochlear origin by affecting noise-induced peroxisome proliferation in auditory hair cells and neurons. Here we demonstrate that loss of pejvakin in hair cells, but not in neurons, causes profound hearing loss and outer hair cell degeneration in mice. Pejvakin binds to and colocalizes with the rootlet component TRIOBP at the base of stereocilia in injectoporated hair cells, a pattern that is disrupted by deafness-associated PJVK mutations. Hair cells of pejvakin-deficient mice develop normal root

Measures of auditory brain-stem responses, distortion product otoacoustic emissions, hair cell loss, and forward masked tuning curves in the waltzing guinea pig.

Measures of the auditory brain-stem response (ABR), distortion product otoacoustic emission (2f1-f2), hair cell loss, and forward masked tuning curves were obtained from waltzing guinea pigs and their age-matched controls at postnatal day 2, 9, 15, and 30. A mild ABR threshold shift (10-15 dB) is seen by 2 days postnatal and gradually increases to a more severe threshold shift (40-50 dB) by postnatal day 15. Already by 30 days the auditory brain-stem response, being beyond the output of the instrumentation, could not be elicited. The mean distortion product otoacoustic emission (DPOE) amplitud
