BACKGROUND: Doubled haploid (DH) lines generated via the wheat × maize system represent an effective technology for accelerating wheat breeding. However, achieving low-cost, large-scale DH production remains a core technical challenge.
RESULTS: In this study, we conducted flowering-synchronization experiments to determine the hybridization window between wheat and maize. Wheat was sown year-round in the field, while maize was planted both in the field and in high tunnels. We also evaluated a novel pollination method, four new nutrition solutions for cut-tiller culture, and a newly designed op

Ionizing radiation causes various types of DNA damage in mammalian cells, leading to tissue reactions and stochastic effects. Different cultured cell lines have been used to study DNA alterations at the molecular level; however, most previous studies have used diploid cells, making it difficult to accurately analyze structural variations (SVs), such as insertions and deletions (indels), in autosomal chromosomes. Here, we report the development of a unique experimental system using HAP1 cells, a human haploid cell line derived from the chronic myeloid leukemia (CML) cell line KBM-7. To compare

Evidence has been sought for a genetically determined predisposition among children with juvenile rheumatoid arthritis (JRA) who are also at particular risk for the development of inflammatory eye disease.45 unrelated Caucasian patients (41 female) with early-onset pauciarticular JRA were human leukocyte antigen (HLA) types. 28 of the study group were found to be HLA-DRw5 compared with 16 of 84 controls (X(2), 24.3, P = <0.001). 9 patients were HLA-DRw8 compared with 4 of 84 controls (X(2), 7.51, P = <0.01). Iritis developed in 24 of the 45 children studied, 17 of whom were typed as HLA-DRw5 w

Intracytoplasmic spermatid injection into oocytes has limited efficiency in cattle, with no offspring generated so far, partly due to ambiguous spermatid identification. This study aimed to develop and validate a method for isolating and characterizing bovine spermatids to improve the efficiency of spermatid intracytoplasmic injection. First, we optimized a protocol for spermatid isolation from bull testis using a discontinuous Percoll gradient and 10 μm mesh cell strainers. Next, we established a stage-specific separation strategy based on DNA content, size, and granularity using flow cytomet

Genetic diversity is essential for populations adapting to environmental-changes. Following genetic bottlenecks, invasive species often have reduced diversity, yet must rapidly adapt to novel environments. This paradox is especially pronounced in parasites, which face repeated transmission bottlenecks and anthropogenic countermeasures. The challenge of maintaining diversity in the short-term intensifies in haplodiploid species, where males inherit and transmit only half of their mother's genome, limiting effective population size. To examine how such species may maintain adaptability, we study

Albinism is a major limitation in androgenesis-based doubled haploid (DH) production in cereals, significantly limiting the efficiency of this powerful breeding tool. Albino plantlets, which do not develop functional chloroplasts, are unable to photosynthesize and thus cannot survive outside of in vitro culture. Despite numerous attempts to optimize androgenesis protocols, albinism remains difficult to eliminate, largely due to its strong genotype dependence. Recent studies have shed light on the molecular and cellular mechanisms underlying this phenomenon, linking it to early plastid differen

Haploid embryonic stem cells (haESCs) are derived from the inner cell mass of the haploid blastocyst, containing only one set of chromosomes. Extensive and accurate chromatin remodelling occurs during haESC derivation, but the intrinsic transcriptome profiles and chromatin structure of haESCs have not been fully explored. We profiled the transcriptomes, nucleosome positioning, and key histone modifications of four mouse haESC lines, and compared these profiles with those of other closely-related stem cell lines, MII oocytes, round spermatids, sperm, and mouse embryonic fibroblasts. haESCs had

BACKGROUND: Generating chromosome-scale haplotype resolved assembly is important for functional studies. However, current de novo assemblers are either haploid assemblers that discard allelic information, or diploid assemblers that can only tackle genomes of low complexity.
RESULTS: Here, Using robust programs, we build a diploid genome assembly pipeline called gcaPDA (gamete cells assisted Phased Diploid Assembler), which exploits haploid gamete cells to assist in resolving haplotypes. We demonstrate the effectiveness of gcaPDA based on simulated HiFi reads of maize genome which is highly he

In a diallel cross analysis using doubled haploids, both additive and additive x additive genetic variances can be estimated. In addition, mean and variance of recombination values can also be estimated. Linkage affects the covariance between half-sibs the most, the covariance between grandparents and grand offspring to an intermediate degree and the covariance between full sibs the least. The covariance of parent and offspring is not affected by linkage.

Three partially inbred substrains of Obese strain chickens were studied for the spontaneous development of autoimmune thyroiditis. The influence of the major histocompatibility complex (B haplotype) was marked in one, less marked but still significant in a second, and barely detectable and transient in a third substrain. These differences in the effect of B haplotype may be due to the overriding action of genes other than those within the B haplotype.

The evidence for a genetic determination of susceptibility to leprosy is reviewed. To test the hypothesis that an HLA (histocompatibility leukocyte antigen)-linked gene is associated with such susceptibility, the association between the distribution of leprosy within a family and the segregation of HLA haplotypes was investigated among 72 families who lived in Karigiri, Tamil Nadu State, South India. A statistically significant association was found for families in which siblings had tuberculoid leprosy and in which neither parent had leprosy. The findings from the data of this study agree wit

Deleterious mutations arise continually in natural populations, yet their full life histories-from origin to elimination-are rarely observed directly. Our understanding of mutational fate in the wild comes from indirect inference rather than from tracking a single mutation through time and space. Haplodiploid systems provide a rare opportunity to observe negative selection in action: because males are haploid, recessive alleles are immediately exposed to selection rather than masked by heterozygosity. Haplodiploidy has long been hypothesized to accelerate the purging of deleterious variation.

Understanding the genetics of adaptation and speciation is critical for a complete picture of how biodiversity is generated and maintained. Heterogeneous genomic differentiation between diverging taxa is commonly documented, with genomic regions of high differentiation interpreted as resulting from differential gene flow, linked selection and reduced recombination rates. Disentangling the roles of each of these non-exclusive processes in shaping genome-wide patterns of divergence is challenging but will enhance our knowledge of the repeatability of genomic landscapes across taxa. Here, we comb

Triploid and haploid conceptions are not viable and are a common occurrence in humans, where they account for 10% of all pregnancy losses. Despite the parent of origin being important in the etiology of the pregnancy, our knowledge of their causes is limited, especially at the point of conception. Using a dataset of 96,660 biopsies and a validation dataset of 44,324 from human blastocyst embryos generated by intracytoplasmic sperm injection, we estimate that 1.1% of human conceptions (n = 1,063) contain extra or missing chromosome sets in zygotes. In our cohort of intracytoplasmic-sperm-inject

A cytogenetic study was undertaken to quantify, by chromosomal karyotyping, the incidence and type of chromosomal abnormalities present in Day-6 in vitro-produced (IVP) porcine embryos. Morphologically normal Day-6 blastocysts (n=318) were fixed and grouped into six classes according to the number of total cells (from < or =20 to 61-70). Of 248 embryos suitable for analysis, 97 (39.1%) displayed chromosomal abnormalities. The abnormalities included haploidy (9.3%), polyploidy (71.1%) and mixoploidy (19.6%). Within polyploid embryos, triploidy and tetraploidy showed the highest incidence (56.5

In a series of 24 patients with chronic T-lymphoid disorders [13 T-prolymphocytic leukaemia (T-PLL) and 11 Sezary syndrome] we have studied (i) chromosome 17p abnormalities and p53 allele deletion by fluorescence in situ hybridization; (ii) mutation in the exons of the p53 gene by direct DNA sequencing; and (iii) p53 protein expression by immunocytochemistry and, in some cases, also by flow cytometry with DO-1, a monoclonal antibody to the p53 protein. The study revealed p53 deletion and accumulation of p53 protein in the absence of mutation in the exons that included the hot-spots and differs

We have previously detected chromosome abnormalities in human embryos whilst identifying the sex for preimplantation diagnosis of X-linked disease. In this study we assess the incidence of these abnormalities, both for sex chromosomes and autosomes 1 and 17, using dual fluorescent in situ hybridization (FISH). Sixty-nine normally fertilized embryos of good morphology at the 6-10 cell stage (day 3 post-insemination) were examined. The embryos were spread whole using HCl and Tween 20 to dissolve the cytoplasm. Thirty-four embryos were analyzed for the sex chromosomes and 35 for autosomes 1 and 1

BACKGROUND: Next-generation sequencing (NGS) is widely used for assessing CDKN2A status in cancers due to its high throughput and efficiency. However, discrepancies between NGS and fluorescence in situ hybridization (FISH)-the gold standard for detecting copy number variations (CNVs)-are occasionally observed. This study aimed to evaluate the accuracy of NGS in detecting CDKN2A deletions by comparing it with FISH.
METHODS: A total of 1118 non-small cell lung cancer (NSCLC) patients who underwent NGS at Fudan University Shanghai Cancer Center (FUSCC) were retrospectively analyzed. Among them,
