Unveiling the responses of Alexandrium pacificum to phosphorus utilization by physiological and transcriptomic analysis.

Phosphorus (P) is an essential macronutrient impacting bloom formation of marine dinoflagellates. The dinoflagellate Alexandrium pacificum is a cosmopolitan species known to frequently cause dense blooms in estuarine and coastal waters worldwide, while the physiological and molecular responses of A. pacificum to P utilization are still not well understood. Herein, the growth, P utilization, toxin production and transcriptomes of A. pacificum grown under P-deficient, inorganic P-replete, and organic P-replete conditions were compared. The results indicated that P-deficient adversely affected th

Natural attenuation and engineering-based removal of marine algal toxins: current status and future perspectives.

The global surge in harmful algal blooms (HABs), amplified by ocean warming and eutrophication, has intensified ecological and public health risks through the production of potent marine algal toxins. Given their ubiquitous occurrence, environmental persistence, and adverse effects on marine ecosystems and coastal economies, increasing efforts have been devoted to mitigating toxic HABs and associated toxin pollution. This review presents a systematic, integrated analysis of natural attenuation processes and engineering-based removal technologies for marine algal toxins, addressing a critical g…

Storm-induced coastward expansion of Margalefidinium polykrikoides bloom in Chesapeake Bay.

An unusual coastward expansion of the toxic dinoflagellate species Margalefidinium polykrikoides was observed in 2020 summer after a tropical storm passing Chesapeake Bay. Such coastward expansion was only recorded in 2007. A newly developed coupled Lagrangian particle tracking and harmful algal bloom model driven by environmental variables was used to investigate the underlying mechanisms and successfully reproduced the expansion patterns. Persistent pre-storm southerly winds favored the delivery of bloom source water originated inside the bay to the coast. Storm-induced strong upwelling of d…

Slow vertical floating velocity and rapid buoyancy loss promote subsurface distribution of Raphidiopsis raciborskii: Insights from a vertical migration model.

Raphidiopsis raciborskii (Wołoszyńska) Aguilera et al., a potentially toxic filamentous cyanobacterium, typically forms subsurface aggregations, rather than surface blooms. This contrasts with many other problematic cyanobacteria and makes early detection and management more difficult. We hypothesize that this subsurface distribution arises from the interaction between light-driven buoyancy regulation and trait-dependent factors related to sinking. To test this hypothesis, we quantified changes in cellular density and relative gas vesicle volume under different light irradiances and incorporat

Analysis tool for swimming marine microorganisms using acoustic tweezer.

Phytoplankton, as primary producers through photosynthesis, are fundamental to marine ecosystems. However, their rapid proliferation in coastal regions has increasingly led to harmful algal blooms (HABs), commonly known as red tides, which pose significant environmental, economic, and health risks. A deeper understanding of phytoplankton dynamics, particularly their movement patterns such as diel vertical migration (DVM), is essential for effective monitoring and prevention of HABs. Traditional measurement techniques, including acoustic Doppler current profiling (ADCP) and acoustic backscatter…

The allelopathic efficacy of aqueous extracts derived from Pyropia haitanensis on five species of bloom-forming microalgae: An environmentally viable approach to control harmful microalgal blooms.

Various methods have been used to control harmful algal blooms (HAB), including chemical and physical strategies, but their long-term ecological harm remains. As key ecosystem components, macroalgae like Pyropia haitanensis offer potential; we explore the antialgal effects of its water-soluble extracts on microalgal blooms. Initially, the inhibitory effects of water-soluble extracts from the residual biomass following methanol and ethanol extraction of dried P. haitanensis were evaluated against five bloom-forming microalgal species for 120 h. In comparison with methanol, the powder residue su…

Spatial and temporal characterization of cyanobacteria blooms in the Mississippi Sound and their relationship to the Bonnet Carré Spillway openings.

During the spring and summer of 2019, an unprecedented cyanobacterial harmful algal bloom (cyanoHAB) was responsible for beach advisories on 25 beaches along the Mississippi Sound for over 3 months. Due to the preceding heavy rainfall and flooding within the Mississippi River watershed, for the first time in history, the Bonnet Carré Spillway (BCS) opened twice in one year during 2019. The coastal cyanoHAB coincided with the second BCS opening. The main objectives of this study were: (1) to investigate the potential for using the National Aeronautics and Space Administration (NASA) ocean color…

Use of biosensors for the detection of marine toxins.

Increasing occurrences of harmful algal blooms (HABs) in the ocean are a major concern for countries around the globe, and with strong links between HABs and climate change and eutrophication, the occurrences are only set to increase. Of particular concern with regard to HABs is the presence of toxin-producing algae. Six major marine biotoxin groups are associated with HABs. Ingestion of such toxins via contaminated shellfish, fish, or other potential vectors, can lead to intoxication syndromes with moderate to severe symptoms, including death in extreme cases. There are also major economic im…

Spatial distribution of prokaryotes in a harmful dinoflagellate bloom near the Huangqi Peninsula, Fujian, China.

In recent decades, increasingly frequent harmful algal blooms (HABs) have severely impacted the coastal aquaculture across China, but the mechanisms underlying their initiation and termination remain unclear. Emerging evidence suggests that planktonic prokaryotes potentially play a critical role in HAB dynamics. Using 16S rDNA high-throughput sequencing, this study investigated the spatial variations in planktonic prokaryotic communities during a mixed Prorocentrum and Karenia bloom in Lianjiang coastal waters in late May 2024. The results revealed that the bloom significantly reduced bacteria…

Ecological insights from field and laboratory studies of Akashiwo sanguinea.

Blooms of the dinoflagellate Akashiwo sanguinea occur in coastal ecosystems worldwide and can have significant ecological consequences. Along the central California coast, long-term oceanographic and harmful algal bloom observations indicate that A. sanguinea has exhibited seasonal blooms since approximately 2017, coinciding with negative upwelling anomalies and warm, stratified conditions characteristic of late summer and early fall. In laboratory experiments, three A. sanguinea strains isolated from different sites and/or environmental conditions along the central California coast exhibited …

A modified stratification index method to assess reservoir water quality trends.

Harmful algal blooms (HABs) have been a harm to reservoir health for decades and it is believed that as climate changes and temperatures rise, frequency and severity of HABs are likely to increase as well. Understanding the relationships between physical factors in a reservoir and bloom trends could be key for keeping rising blooms at bay. A modified stability index based on temperature dependent water density at multiple depths was adapted into a code to process high frequency reservoir monitoring data. Metrics and visual tools were developed to use this stability index to objectively analyze…

Symptom frequency and exposure to a cyanobacteria bloom in Florida.

This investigation was undertaken to characterize health effects associated with a major bloom of blue-green algae due to the proliferation Microcystis aeruginosa that occurred in Florida in 2018. Cyanobacteria produce multiple toxins, including the potent hepatotoxic microcystins (MCs), that have been reported to cause illness in exposed persons worldwide. Widespread exposure to toxins released by blue-green algae during the 2018 bloom was shown by the presence of MCs in the nasal passages of 95 percent of the individuals studied previously in south Florida (Schaefer et al., 2020). The curren

Physiological and transcriptomic responses of Heterosigma akashiwo to nitrogen deficiency and urea utilization.

Nitrogen (N) is an essential nutrient for phytoplankton growth. The availability of ambient N and its chemical forms are critically important for the formation of harmful algal blooms (HABs). Although previous studies have explored the transcriptomic responses of Heterosigma akashiwo to N conditions, these analyses were largely descriptive and lacked in-depth characterization of the molecular mechanisms underlying urea utilization and N deficiency responses. To address this gap, we conducted a comprehensive physiological and transcriptomic investigation of this globally distributed HAB species…

Acclimation temperature influences phage susceptibility in a toxin-producing strain of Microcystis aeruginosa.

Microcystis aeruginosa is a freshwater cyanobacterium known for its role in the formation of harmful algal blooms. However, less well characterized are the host-phage interactions that occur in these toxic blooms. Here, we describe the relationship between a microcystin-producing strain of M. aeruginosa, NIES-298, and a lytic bacteriophage Ma-LMM01. In this study, we explored how host pre-acclimation to different temperature conditions influenced sensitivity to subsequent infection by Ma-LMM01. We focused on 19°C, a cooler temperature often associated with microcystin-producing blooms in sprin…

Contrasting growth phase-dependent microbiota dynamics between dinoflagellates and diatoms.

Bacteria associated with phytoplankton can profoundly influence host physiology and bloom dynamics. Yet it remains poorly understood whether the microbiota of dinoflagellates is restructured across host growth phases, and how such dynamics differ from those of other phytoplankton. Here, we tracked the microbiota of three harmful algal bloom (HAB)-forming dinoflagellates (Alexandrium catenella, A. pacificum, Gymnodinium catenatum) and three diatoms (Cylindrotheca closterium, Pseudo-nitzschia pungens, Coscinodiscus granii), each represented by two strains of distinct spatiotemporal origin and sa…

Toxic Characteristics and Action Mode of the Mixotrophic Dinoflagellate Akashiwo sanguinea on Co-Occurring Phytoplankton and Zooplankton.

The mixotrophic dinoflagellate Akashiwo sanguinea frequently forms harmful algal blooms around the world and has caused massive deaths of shellfish, finfish and birds, yet its toxic mechanism is still unclear. In this study, toxic effects of A. sanguinea on co-culturing phytoplankton and zooplankton were investigated. The results showed that sonicated cultures of A. sanguinea JX13 and JX14, isolated from the Pearl River Estuary, had a significant lethal effect on the rotifer Brachionus plicatilis, with the highest mortality rate of 80%. The highest inhibition rates of A. sanguinea cultures JX1…

An optimized CaO2-functionalized alginate bead for simultaneous and efficient removal of phosphorous and harmful cyanobacteria.

Simultaneous removal of phosphorus (P) and algae is important to mitigate eutrophication, however, it is rather challenging in remediation of harmful algal blooms (HABs)-contaminated water. In this study, a wet alginate bead functionalized by CaO2 particle formed layer by layer was prepared with an in-situ method and optimized to remove phosphorous and inhibit algae growth. The stable H2O2 release with a concentration level of 0.06 mM was observed for a period of 26 d. The content of peroxy groups (-O-O-) in the optimal bead was 0.44 mmol·g-1 through permanganate-based titration study. For sol…
