Acute trauma can lead to life-long changes in susceptibility to psychiatric disease, such as post-traumatic stress disorder (PTSD). Rats given free access to a concentrated glucose solution for 24 h beginning immediately after trauma failed to show stress-related pathology in the learned helplessness model of PTSD and comorbid major depression. We assessed effective dosing and temporal constraints of the glucose intervention in three experiments. We exposed 120 male Sprague-Dawley rats to 100, 1 mA, 3-15 s, inescapable and unpredictable electric tail shocks (over a 110-min period) or simple re

Animal models have been developed to investigate aspects of stress, anxiety, and depression, but our understanding of the circuitry underlying these models remains incomplete. Prior studies of the habenula, a poorly understood nucleus in the dorsal diencephalon, suggest that projections to the medial habenula (MHb) regulate fear and anxiety responses, whereas the lateral habenula (LHb) is involved in the expression of learned helplessness, a model of depression. Tissue-specific deletion of the transcription factor Pou4f1 in the dorsal MHb (dMHb) results in a developmental lesion of this subnuc

One of the most problematic situations for the estimated 60 million working donkeys and mules across the world is within the brick kiln environment. In countries such as Egypt, Nepal, India and Pakistan, donkeys and mules carry dried bricks into small kilns either by pulling carts or carrying loaded packs. The health and welfare problems of this environment are well documented: they may carry excessive loads; work long hours, have difficulties accessing food, water and rest options; and can frequently suffer untreated illnesses including dental problems, lameness and open wounds from poorly fi

When an organism is subjected to stress, gastric ulcers or ulcerations commonly develop but the vulnerability to and amount of pathology induced varies considerably between individuals. The role of psychological factors in determining the occurrence and severity of these ulcerations is amply demonstrated in the studies reviewed here. The present paper (a) gives a brief history of the search for the causes of gastric ulcer, (b) provides a review of our own research which reveals that vulnerability to gastric ulceration is modulated by psychologically meaningful experiences, and (c) offers a mul

Attributional responses made to both good and bad events by male and female Ss (N = 117) differing in degree of clinical depression were examined in light of the reformulated learned helplessness model of depression. Data comparison with control Ss provided support for the model's prediction. Clinically depressed Ss assumed more personal responsibility than control Ss for their life outcomes, especially the negative ones. While significant main effects were demonstrated in the clinical sample for all three variables, the significant two and three-way interaction effects revealed a complex rela

Background: Preconception screening traditionally focuses on physical health indicators and diagnosed psychopathology, potentially overlooking subclinical psychological vulnerabilities that could predict perinatal mental health outcomes. This study examined whether transdiagnostic psychological assessment before pregnancy provides information beyond childhood adversity and current symptom severity. Methods: In this prospective cohort study, 170 healthy nulliparous women attempting natural conception underwent preconception assessment and were followed through postpartum. Preconception measures

BACKGROUND: (R,S)-ketamine has gained attention for its rapid-acting antidepressant actions in patients with treatment-resistant depression. However, widespread use of ketamine is limited by its side effects, abuse potential, and poor oral bioavailability. The ketamine metabolite, (2R,6R)-hydroxynorketamine, exerts rapid antidepressant effects, without ketamine's adverse effects and abuse potential, in rodents.
METHODS: We evaluated the oral bioavailability of (2R,6R)-hydroxynorketamine in three species (mice, rats, and dogs) and also evaluated five candidate prodrug modifications for their c

It is widely accepted that mental stress is an important factor in the development of psychological disorders such as depression. On pre-existing evidence, the so-called green odor may have a relieving and sedative effect on animals exposed to stressful situations. Using two behavioral models of depression, the forced-swim test and learned helplessness paradigm, we investigated whether inhalation of green odor (a 50:50 mixture of trans-2-hexenal and cis-3-hexenol) might alleviate and/or prevent experimentally induced depressive-like states in rats. A 3-min swim every day for 7 days resulted in

Battered wife syndrome is difficult to detect because the women usually do not volunteer the diagnosis. They often present with vague somatic complaints such as headache, lower back pain, abdominal pain, pelvic pain and dyspareunia. Four case histories demonstrate the difficulty in recognizing the cause of these complaints. The diagnosis was often missed because straight-forward, non-threatening, open-ended questions were not asked initially. The family physician's primary role is to identify the syndrome and initiate psychotherapy. Psychotherapy is centred on reversing "learned helplessness"

The implantable cardioverter defibrillator (ICD) may be associated with some degree of psychological maladjustment, especially in patients experiencing high rates of discharge. This article reviews the psychological literature related to ICDs and presents two case examples illustrating common psychological complications related to multiple discharges. A brief, psychological intervention can be implemented to improve patient adjustment. To explain possible processes by which psychological complications may develop in ICD patients, two well-established psychological theories of learning and beha

Generalized anxiety disorder is among the world's most prevalent psychiatric disorders and often manifests as persistent and difficult to control apprehension. Despite its prevalence, there is no integrative, formal model of how anxiety and anxiety disorders arise. Here, we offer a perspective derived from the free energy principle; one that shares similarities with established constructs such as learned helplessness. Our account is simple: anxiety can be formalized as learned uncertainty. A biological system, having had persistent uncertainty in its past, will expect uncertainty in its future

OBJECTIVE: Pain is among the most frequently reported, bothersome, and disabling symptoms described by patients with osteoarthritis, rheumatoid arthritis, fibromyalgia, and other musculoskeletal conditions. This review describes a growing body of literature relating catastrophizing, a set of cognitive and emotional processes encompassing magnification of pain-related stimuli, feelings of helplessness, and a generally pessimistic orientation, to the experience of pain and pain-related sequelae across several rheumatic diseases.
METHODS: We reviewed published articles in which pain-related cata

During early development, environmental challenges set the stage for permanent changes in the functioning of the pituitary-adrenal stress response. Since these data have been reported almost exclusively in single rat strains the role of phenotypic and genotypic factors in shaping the stress response is relatively unknown. This study examined whether the phenotypic/genetic profile of the rat influences the long-term response to challenge after early exposure to stress. Two strains of Sprague-Dawley rats were used in this study: one is a stress-induced animal model of "learned helpless" (LH) beh

It was shown previously that cycloprolylglycine, an endogenous neuropeptide, is a positive AMPA receptor modulator and is able to increase the content of BDNF in neurons. In the present study, using the model of learned helplessness in rats, we showed that cycloprolylglycine at a dose of 1 mg/kg at subchronic intraperitoneal administration reduced the time of animal immobility to passive control values (from 167.6 to 83.6 s) on day 12 of the experiment. This indicates the presence of antidepressant-like activity. The results of the study allow us to consider cycloprolylglycine as a component i

Several empirically supported treatments for depression are currently available with little understanding of either principles or mechanisms that are responsible for their effectiveness. This article reviews existing principles and finds that they contain little mechanism information. A connectionist mechanism used to explain why systematic desensitization and response prevention are effective in treating anxiety disorders is reviewed and generalized to understand why empirically supported treatments of depression work. This mechanism suggests a dissonance induction followed by reduction princ

Newborn neurons in the adult hippocampal dentate gyrus (DG) are hyperexcitable, competitive cells involved in behavioral responses to stress and antidepressants. The role and the mechanisms underlying the competitiveness of the newborn neurons in moderating behavioral responses to stress have not yet been comprehensively investigated. Using FosTRAP2 transgenic mice in the learned helplessness (LH) paradigm, we report that voluntary running, a guideline-supported intervention for depression, induces neurogenesis, and that the newly generated neurons compete with LH-TRAPed neurons, thereby reduc

The total dependence that, from necessity, must be the lot of an intensive care patient can lead to a state of learned helplessness as they recover. In addition, the physical frailty of these patients further confounds their first attempts at independence. It is at this stage that patients need clear information about the road ahead and in a form that they can refer back to as needed when they are discharged to the general wards and then home. As many intensive care patients have little or no memory of the intensive care unit (ICU) afterwards and only gradually come to understand how ill they

Theories on the aetiology of depression in humans are intimately linked to animal research on stressor controllability effects. However, explicit translations of established animal designs are lacking. In two consecutive studies, we developed a translational paradigm to study stressor controllability effects in humans. In the first study, we compared three groups of participants, one exposed to escapable stress, one yoked inescapable stress group, and a control group not exposed to stress. Although group differences indicated successful stress induction, the manipulation failed to differentiat
