Sphincteric mechanism of the main pancreatic duct in the dog. An experimental model of the isolated sphincteric preparation.

This study describes an in vitro experimental model of the sphincter at the lower end of the main pancreatic duct in the dog. This model, employing a drop counter to measure the drop rate, monitors the perfusion rate of Tyrode through the sphincter. Acetylcholine (ACh), employed to establish the sensitivity and viability of the isolated sphincter, produced a contraction of the sphincter which was concentration-response related. Saline, as a control, atropine and hexamethonium did not affect the drop rate. Atropine (20 microgram) partially abolished the contraction produced by 50 microgram of A

Cholinergic regulation of adrenal medullary blood flow.

To determine the relative role of nicotinic and muscarinic mechanisms in splanchnic nerve stimulation (NS)-induced adrenal catecholamine secretion and medullary vasodilation, 12 pentobarbital-anesthetized dogs were subjected to three identical stimulations. The first NS was performed before drug administration and served as a control. The second NS was performed after administration of either the muscarinic antagonist, atropine 0.5 mg/kg (group 1), or the nicotinic antagonist, hexamethonium 20 mg/kg (group 2). The third NS was performed after administration of both drugs. NS in the absence of

Prevention of organophosphate intoxication with presynaptic cholinergic blockers.

In Vivo cardiovascular neuromuscular preparation of rats was used to study the prophylaxis and mechanism of diisopropyl fluorophosphate (DFP) intoxication by presynaptic cholinolytic agents (ganglionic blocking agents), including hexamethonium, trimethaphan and mecamylamine. The lethal action of DFP was partially or completely prevented by pretreatment of rats with hexamethonium (10 mg/kg), trimethaphan (80 mg/kg) and mecamylamine (30 mg/kg). Combined use of these drugs with 2-PAM (100 mg/kg) improved further the prophylaxis of DFP lethality. The mechanism of prophylaxis could be due to the re

Actions of piperidine and dimethylphenylpiperazinium (DMPP) on afferent discharges of the cat's carotid body.

Chemoreceptor discharges were recorded in vivo from fine filaments of carotid sinus nerve; their frequency was used as index of receptor activity. Effects of piperidine on chemoreceptors were studied and compared with those of dimethylphenylpiperazinium (DMPP) on chemoreceptors in adult cats. Intracarotid-arterial injections of piperidine produced a transient increase in chemoreceptor discharges, the threshold dose ranging from 10 to 50 microgram i.a. The excitatory effect of piperidine was not affected by atropine, hexamethonium or GABA. DMPP (0.2-0.5 microgram i.a.) induced a marked increase

The inhibitory action of ketamine on the rat's gastric secretion.

Ketamine exhibited effects on gastric acid secretion total acidity and acid output. Reduction of secretion by ketamine did not appe,r to be significantly different from values obtained with atropine or hexamethonium. In addition to atropine- and hexamethonium-like effects, ketamine also appeared to decrease gastric secretion through a potentiating activity on the sympathetic nervous system. When sympathetic activity was potentiated by cocaine, the antisecretory effect of ketamine appeared to be significantly reduced, compared with that of atropine. Furthermore, after alpha- or alpha- and beta-

Role of vagus nerve in postprandial antropyloric coordination in conscious dogs.

It is generally believed that gastric emptying of solids is regulated by a coordinated motor pattern between the antrum and pylorus. We studied the role of the vagus nerve in mediating postprandial coordination between the antrum and pylorus. Force transducers were implanted on the serosal surface of the body, antrum, pylorus, and duodenum in seven dogs. Dogs were given either a solid or a liquid meal, and gastroduodenal motility was recorded over 10 h. Gastric emptying was evaluated with radiopaque markers mixed with a solid meal. Dogs were treated with hexamethonium, N(G)-nitro-l-arginine me

Effects of erythromycin in the dog upper gastrointestinal tract.

The effects of erythromycin on motor and electrical behavior of the antrum, pylorus, and duodenum were determined in chronically instrumented, awake dogs. Erythromycin infusion resulted in an abrupt, powerful increase in motility. The motility index increased 18-fold in the antrum, 15-fold in the pylorus, and 8-fold in the duodenum. Bradyarrhythmia with a 30% decrease in slow-wave frequency occurred in all animals. Retrograde giant contractions in association with retching and vomiting occurred in 88% of the dogs. Neostigmine was less potent than erythromycin in increasing motility. Hexamethon

Effects of cholinergic antagonists on ghrelin release and expression in the crop, proventriculus, and duodenum of newly hatched chickens.

The aim of this study was to evaluate the effect of the cholinergic receptor antagonists on ghrelin (GHRL) expression and release from parts of the gastrointestinal system in an unique animal model-newly hatched chickens. Ghrelin was released from explants of the crop, proventriculus, and duodenum tissues in vitro. The expression of GHRL, along with that of ghrelin O-acyltransferase (GOAT) and growth hormone secretagogue receptor (GHSR-1a), was also observed in the crop, proventriculus, and duodenum. This is the first report on ghrelin expression, synthesis, and release in the avian crop. The

Central injections of noradrenaline and adrenaline differentially affect plasma free fatty acid and glucose in conscious pigeons (Columba livia).

The possible involvement of central noradrenergic and/or adrenergic circuits in central mechanisms controlling free fatty acids and glucose levels was investigated in conscious pigeons. The effects of intracerebroventricular injections of noradrenaline (80 nmol) or adrenaline (80 nmol) on plasma free fatty acids and glucose concentrations were examined. The possible role of the autonomic nervous system, of sympathetic terminals and of pituitary hormone release in the metabolic responses induced by intracerebroventricular injections of adrenaline and noradrenaline was investigated by systemic p

Nicotinic receptors of frog ganglia resemble pharmacologically those of skeletal muscle.

The actions of ACh antagonists were studied on synaptic currents of autonomic ganglia of the frog. Fast excitatory synaptic currents (ESCs) were recorded from cardiac and paravertebral neurons with the use of the 2-microelectrode voltage-clamp method. The actions of 4 ACh antagonists, tubocurarine, hexamethonium, trimetaphan, and decamethonium were studied. Tubocurarine was effective at reducing the peak amplitude of ESCs (50% inhibition at 3 microM). In contrast, tubocurarine (1-30 microM) reduced the time constant of ESC decay by only 9% compared with controls. Both of these effects of tuboc

Studies on an isolated innervated preparation of guinea-pig trachea.

An isolated preparation of the guinea-pig trachea with intact parasympathetic and sympathetic innervation has been devised. Responses to nerve stimulation were recorded as increases or decreases in intraluminal pressure from the fluid-filled trachea. The preparation maintained a positive resting intraluminal pressure of 3-4 cmH2O. This was unaffected by atropine, hexamethonium or propranolol. Brief pressor responses, which could be completely blocked by atropine or hexamethonium, were obtained by applying short trains of stimuli to the cervical segment of the right vagus. The amplitude of resp

Efficacy of antidotes and their combinations in the treatment of acute carbamate poisoning in rats.

BACKGROUND: Physostigmine and its analogues neostigmine, pyridostigmine and rivastigmine are carbamates nowadays used in many indications, including antidotal effects against antimuscarinic poisonings, reversal of competitive neuromuscular block, myasthenia gravis, Alzheimer's disease and prophylaxis against nerve agent intoxications. Use of these medicinal carbamates, but also of carbamate insecticides, created need for research into the potential and mechanisms of action of several antidotes against carbamate poisonings, including anticholinergics and oximes. AIM: The goal of this experimen

The acute effects of capsaicin on the cardiovascular system.

Arterial blood pressure and heart rate were recorded from male Wistar rats anaesthetized with urethane. Intravenous injection of capsaicin, 1 microgram, produced a reproducible triphasic effect on blood pressure, comprising an initial fall in blood pressure and heart rate, followed by a transient and then a sustained pressor response. The depressor response and bradycardia were abolished by vagal section. The transient pressor response was altered in shape by hexamethonium. Slow intravenous infusion of capsaicin, 50 micrograms over 12 min, produced only a sustained pressor response accompanied

Ganglion blocking activity of cimetidine in the anaesthetized cat.

Cimetidine produced a dose-dependent and reversible inhibition of contractions of the nictitating membrane elicited through stimulation of the preganglionic nerve supplying the superior cervical ganglion in the anaesthetized cat. Postganglionic nerve stimulation resulted in only a slight and variable inhibition of the contractions. Both hexamethonium and cimetidine also produced a dose-dependent and reversible fall in arterial blood pressure. The ganglion blocking activity of cimetidine was much weaker than that of hexamethonium; the ED50 ratio (cimetidine:hexamethonium) calculated from the cu

Effect of sodium propionate on the contractile response of the rat ileum in situ.

Effects of short-chain fatty acids (SCFA) on the contractile response of rat ileum were studied in vivo. The contractile response was estimated by means of changes in the intraluminal pressure under the isometric condition. Intravenous administration of sodium salts of propionate, butyrate, valerate or caproate produced biphasic contractions: an initial phasic contraction and a subsequent tonic contraction. The effect of propionate was studied in detail. A sigmoid dose-response curve was obtained for the phasic contraction. Atropine, hexamethonium and tetrodotoxin inhibited the phasic contract

Effects of adrenergic neuron and ganglion blockers on hemodynamics and plasma catecholamine levels after corticosteroids during hemorrhagic shock in the dog.

The involvement of adrenergic mechanisms in the ability of massive doses of methylprednisolone to potentiate the vasodilatory effects of phenoxybenzamine during controlled hemorrhagic shock was investigated. Dogs were subjected to ligation of the adrenals and, with the exception of the controls, were pretreated with either hexamethonium or bretylium. Despite careful surgery, the adrenal ligation, per se, failed to alter the concentration of plasma catecholamines. Hexamethonium and bretylium, on the other hand, both decreased the levels of plasma catecholamines: noradrenaline to a greater exten

Effects of some beta-adrenoceptor antagonists on orthostatic hypotension in repeatedly cold- (SART-) stressed rats.

Rats stressed by specific alternation of rhythm in temperature (SART) show various symptoms of disautonomia, increased pulse rates, continuous hypotension, and severe orthostatic hypotension (OH) when they are subjected to postural change. The OH symptoms are improved by muscarinic M2-receptor blockers. In the present study, effects of beta-adrenoceptor blocking agents on OH in SART-stressed rats were investigated. Anesthetized rats were restrained on a board in the supine position, and direct blood pressure and ECG were measured automatically using Fluclet Jr.2. Postural change was performed

Influence of acetonylgeraniin, a hydrolyzable tannin from Euphoria longana, on orthostatic hypotension in a rat model.

Acetonylgeraniin, an active principle isolated from the seeds of Euphoria longana Lam. (Sapindaceae), reversed the fall in arterial blood pressure in conscious hypertensive rats (SHRs) with orthostatic hypotension induced by injection of hexamethonium into animals subjected to 90 degrees head-up tilts for 60 seconds. However, acetonylgeraniin failed to affect prazosin-induced orthostatic hypotension. Plasma noradrenaline (NA) and mean blood pressure were elevated dose-dependently by an intravenous injection of acetonyl-geraniin into the rats; this increase in blood pressure was totally abolish

Propagated motor activity in the small intestine of urethane-anaesthetized rats: inhibitory action of sympathetic and capsaicin-sensitive nerves.

1. In the duodenum/small intestine of urethane-anesthetized rats, a series of atropine- and hexamethonium-sensitive phasic contractions activity were recorded by using a balloon. With two balloons, one in the proximal duodenum (B1) and the other at a distal site (B2) a co-ordinated motor activity was observed which migrated from B1 to B2. 2. Spontaneous activity of the duodenum was greater in either adrenalectomized, guanethidine- or capsaicin-pretreated than in control rats. Effects of ganglionic blockade and adrenalectomy on the cardio-respiratory response to cow's urine concoction in rats.

The cardio-respiratory effects of cow's urine concoction (CUC) were studied in anaesthetized normal, hexamethonium-treated and adrenalectomized rats. The results showed an early response characterized by a transient depressor effect followed by a more sustained pressor effect. This was followed by a late depressor response. Hypotension was most marked in the adrenalectomized group. The cardio-respiratory effects of CUC in the normal and hexamethonium-treated groups, were milder than in the adrenalectomized group.
