The potentiation of barbiturate-induced narcosis by procarbazine.

Procarbazine (MIH), N-isopropyl-alpha-(2-methylhydrazino)-p-toluamide (NSC-77213), a clinically effective antineoplastic agent, induced sleep in mice at its optimally effective dose (400 mg/kg) and prolongs hexobarbital sleeping times. MIH (400 mg/kg) increased the period of sleep following hexobarbital (100 mg/kg) nearly 10-fold. Nonhypnotic doses of MIH also significantly prolonged hexobarbital-induced sleep. Hexobarbital half-life in plasma was prolonged 6 to 7 times by prior treatment with MIH (400 mg/kg). Liver microsomes from mice treated with MIH exhibited decreased metabolism of the fo

[Effect of pyriditol on drug metabolism].

Pyriditol (encephabol, enerobol, pyrithioxin, etc.), a disulfide derivative of pyridoxin, exerts an inhibitory effect on hexobarbital and amphetamine metabolism i vivo and on ethylmorphine N-demethylation in vitro. In the latter case the inhibition proceeds according to the mixed type of action. Pyriditol potentiates the hypnotic action of hexobarbital and barbital as well as the effects of amphetamine stereotypy. The mechanism of the potentiating of hexobarbital and amphetamine effects is of combined character and is conditioned both by the physiological properties of pyriditol and its inhibi

Some aspects of the changes induced by chronic barbital treatments in the male rat.

Oral barbital treatments consisting of average daily doses of 200 mg/kg with durations of 15-50 weeks have been used to induce functional tolerance and physical dependence. Tolerance is usually studied with a hexobarbital threshold while increased excitation has been studied by various means, such as induction of convulsions with pilocarpine or choline. After barbital treatments for 30 weeks brain weights are reduced by approximately 10 per cent. This reduction is not due to changes in body weight or water content of the brain. The decrease is still found 30 days after the end of the barbital

Biotransformation of N-methylcyclobarbital in vivo in rabbit and rat.

Metabolism of N-methylcyclobarbital in the rabbit and rat has been studied in vivo for the purpose of comparison with the C5-methylated analogue, hexobarbital. In the rabbit, the main route of the metabolism of N-methylcyclobarbital is glucuronide formation after hydroxylation at the 3'-position of the parent compound. Dehydrogenation of the 3'-hydroxy product, a major pathway in the metabolism of hexobarbital, was a minor route in the case of N-methylcyclobarbital. In addition, a new type of metabolite, thought to be dihydroxylated products from spectral studies, was isolated. In the rat, the

Effect of perinatal low protein diets on the ontogeny of select hepatic cytochrome p450 enzymes and cytochrome p450 reductase in the rat.

In the present study, we administered two low protein diets (LPDs) to rats during pregnancy and lactation and determined their effect on the ontogeny of select hepatic cytochrome P450 (P450) isoforms in their offspring. The L93 and LM76 LPDs were derived from the American Society of Nutrition recommended AIN93G and a modified version of the AIN76A purified control diets, respectively. The LPDs contained 8% crude protein in the form of casein, whereas the purified control diets contained 19% casein. A regular cereal-based diet (NP) was also included, and, therefore, a total of five groups were

[Use of synthetic steroids, inhibitors of microsomal enzymes, for retention of hexenal in the body and for prolongation of hexenal anesthesia under experimental conditions].

17alpha-hydroxydeoxycorticosterone and its acetate derivative inhibited the activity of rat liver microsomal enzymes. The inhibitory effect of steroids was not accompanied by induction of the enzyme synthesis, which was characteristic for the other known inhibitors of mixed function oxydases (SKF 525-A, DPEA, MPDK). These properties of the steroids suggested their use for prolongation of the effect of drugs, which are inactivated by microsomal enzymes. The suggestion was tested using hexobarbital as an example. The transformation of hexobarbital in vitro by the rat liver microsomal enzymes was

Interaction between probenecid and two lipid-soluble barbiturates in the rat.

The effect of pretreatment with probenecid (200 mg/kg, i.p.) on the sensitivity of the central nervous system (CNS) to thiopental and hexobarbital was investigated with an EEG-threshold method. The threshold dose was significantly decreased by pretreatment with probenecid for thiopental but not for hexobarbital. This was due to an increased penetration of thiopental into the CNS, but for hexobarbital an increase in penetration could also be demonstrated by analysis of brain and serum concentrations after infusion of an equal dose of barbiturate. The concentrations in brain at the EEG-threshold

Enzyme-inhibiting action of hydrocortisone and desoxycorticosterone of mixed function oxidases.

Comparative studies have been carried out on the action of two corticosteroids: the mineralocorticoid desoxycorticosterone and the glucocorticoid hydrocortisone, upon single administration in male albino rats, on the hexobarbital sleeping time and the activity of the mixed function oxidases. Applied in equimolar doses, the two steroids prolong the hexobarbital sleeping time, the effect of desoxycorticosterone being stronger. The potentiation of hexobarbital sleeping time by hydrocortisone differs in degree in animals from the different age groups, being most pronounced in mature animals compar

Barbiturate and amphetamine activity in rats fed a magnesium-deficient diet.

Hexobarbital hypnosis and d-amphetamine toxicity were observed in rats fed a commercially available rat food preparation (lab chow) and in another two groups fed complete nutritionally defined diets with or without magnesium. Neither hexobarbital sleeping time nor hexobarbital blood concentration at awakening were affected after 4, 8 or 12 days of feeding the deficient diet. However, increased sensitivity to d-amphetamine was observed after 11 days of feeding the same magnesium deficient diet to other rats. The LD50 of d-amphetamine in magnesium deficient rats was less than 0.5 that recorded f

Sex-related differences in doxapram-induced alteration of drug action in the rat.

Pretreatment of rats with doxapram (20 mg/kg, i.p.) potentiates the duration of hexobarbital hypnosis and inhibits the rate of hepatic hexobarbital metabolism in female rats but not male rats. These data show that there is a sex-related difference in the ability of doxapram to alter both the drug response and hepatic drug metabolism in rats.

Strain independent elevation of hepatic mono-oxygenase enzymes in female mice.

1. Hepatic microsomal drug metabolizing activity was compared in male and female AL/N, Balb/c, Balb/cJ, CD-1, C57BL/6J, C57BL/10J, DBA/2, and DBA/2J mice. 2. Cytochrome P-450-dependent hexobarbital hydroxylase and aminopyrine N-demethylase activities were sexually dimorphic with apparent maximal velocities consistently higher in females. Hexobarbital-induced sleeping times were greater in males, corresponding to their lower hepatic mono-oxygenase enzyme activities measured in vitro. 3. No murine sex differences were observed for the hydroxylation of aniline, while UDP-glucuronyltransferase act

A preliminary evaluation of drug biotransformation in hepatocytes of genetically defined rat strains.

This study was directed to use the genetically developed isoprenaline-sensitive (S), isoprenaline-resistant (R) and spontaneous hypertensive rats (SHR) as standard diseased animal models for in vitro liver function evaluation of drug biotransformation. Hepatic hexobarbital hydroxylase and glutathione transferase (GST) were evaluated by using hexobarbital and 1-chloro-2,4-dinitrobenzene (CDNB) as substrates, at concentrations of 0.21 mmol/l and 1 mmol/l, respectively. The assay was conducted by using isolated hepatocytes in suspension and hepatocytes in a bioreactor configuration. The data demo

Effect of manganese on hepatic drug metabolism in male and female rats.

Experiments were conducted to examine the effect of manganese on drug response and metabolism in male and female rats. Three days after administration of manganese chloride (5 mg Mn/kg, ip), the duration of hexobarbital hypnosis was prolonged in male but not in female rats. Hepatic microsomal metabolism of aniline, ethylmorphine, and hexobarbital was significantly inhibited in male rats. Metabolism of aniline and ethylmorphine was also inhibited in female rats but to a lesser extent than in males. Hexobarbital metabolism was not inhibited in female rats. Cytochrome P-450 content was depressed

Short-term and prolonged treatment with oral contraceptives and liver function.

Experiments on female rats were performed to observe the effects of progesterone and two synthetic progestens: ethinodiol-diacetate and methylestrenolone on liver function. In acute experiments animals were treated for the duration of one sexual cycle. All three compounds prolonged the hexobarbital biotransformation time. This effect is reversible and normalised 48 h after stopping treatment. The testosterone inducement of microsomal mixed function oxydases remained at the level of controls after methylestrenolone pretreatments and was even higher in the ethinodiol-diacetate and progesterone p

The interaction between electrically induced convulsions and tolerance in the abstinence period after chronic barbital treatments in the rat.

The effect of electrically induced convulsions was tested on the tolerance to hexobarbital after chronic barbital treatment in male rats. In two experiments barbital was given in the drinking water for more than 20 weeks. The dose was around 200 mg/kg/day. Tolerance was tested with an EEG threshold method where hexobarbital is infused intravenously to obtain a criterion of burst suppression. If on the third abstinent day an electrical convulsion was induced 1 h prior to the threshold determination then the hexobarbital thresholds were reduced compared with barbital-treated animals where no con

[The effect of thiamine deficiency on the actions of drugs affecting the central nervous system in rats (author's transl)].

Male Wistar rats, 35-days-old, maintained on a thiamine deficient diet for 30 days showed marked growth inhibition and a heart rate less than 70% of that of control rats. We examined the effect of thiamine deficiency on the action of drugs effecting the central nervous system at this period. In thiamine deficient rats treated with chloral hydrate 200 mg/kg, ketamine 100 mg/kg sodium pentobarbital 50 mg/kg, and hexobarbital 100 mg/kg, the sleeping time increased. Pretreatment with 15 mg/kg of the metabolic enzymes inhibitor, SKF-525A, 30 min prior to the hexobarbital administration resulted in

Sex-related alteration of drug action in carrageenan-induced inflammation in the rat.

Twenty-four hours after treatment with carrageenan the duration of hexobarbital-induced hypnosis was prolonged in males but not females in Wistar rats bearing carrageenan-induced paw edema. Similar treatment with carrageenan led to significant inhibition of the metabolism of hexobarbital by hepatic enzymes obtained from male but not female animals. These data suggest that there is a sex-related difference in the ability of carrageenan to alter drug actions in rats.

The effect of caffeine on barbiturate sleeping time and brain level.

The effect of caffeine and coffee on the sleeping time induced by barbital was tested in rats. It was found that caffeine pretreatment shortened barbital-induced sleeping time and that coffee had an effect in direct proportion to its caffeine content; decaffeinated coffee was without effect. It was hypothesized that the shortening was due to an interaction at the brain level. This hypothesis was tested using hexobarbital which is metabolized by the liver whereas barbital is only negligibly so metabolized. Pretreatment with caffeine was found to shorten the hexobarbital sleeping time even thoug
