Spinal cord compression and hindlimb paresis in cats with mucopolysaccharidosis VI.

Six cats with mucopolysaccharidosis VI had hindlimb paresis and other clinical signs associated with compression of the thoracolumbar spinal cord. In 5 cats, the neurologic abnormality progressed over 2 to 4 weeks to loss of thoracolumbar spinal cord function. In 1 cat, the hindlimb paresis remained stable for 18 months. In the cats with progressive worsening of hindlimb function, the abnormality was caused by compression of the spinal cord from proliferation of bony tissue in the thoracolumbar region. In all affected cats, the compression occurred from T12 to L2. In 1 cat, an attempt to relie

Cutaneous and muscular rasodilation in the canine hindlimb evoked by central stimulation.

Using stereotaxic procedures, we electrically stimulated specific sites in the hypothalamus and midbrain of anesthetized dogs pretreated with guanethidine and atropine methonitrate. A tract in which stimulation caused noncholinergic dilator responses in the hindlimbs was identified. The course of this trace was different from that subserving cholinergic vasodilation in the hindlimb musculature. In a number of experiments we studied the proportional distribution of blood flow to leg and paw. Responses restricted to the paw were regarded as occurring mainly in cutaneous vessels; those restricted

Rat Hindlimb Below-Knee Amputation Model and Immediate Targeted Muscle Reinnervation.

Amputation-related pain reduces patient quality of life, contributes to opioid use, and can reduce functional rehabilitation and prosthesis use. Preclinical models that reliably produce post-amputation pain while preserving mobility are essential for studying the biological mechanisms underlying these outcomes. Here, we present a detailed, reproducible, rodent surgical protocol using Sprague-Dawley rats to model hindlimb below-knee amputation. The procedure involves isolating and transecting the sciatic nerve branches, including the tibial, common peroneal, and sural nerves, producing sustaine

Pelvic axial rotation in hindlimb lame horses changes after diagnostic analgesia.

BACKGROUND: Visual assessment of mild to moderate hindlimb lameness has low sensitivity and specificity, potentially leading to flawed diagnostic procedures. Pelvic axial rotation (roll) makes the assessment more complex but is also a potential source of information. OBJECTIVES: To describe pelvic roll patterns in horses with unilateral hindlimb lameness before and after diagnostic analgesia. STUDY DESIGN: Retrospective study. METHODS: Optical motion capture data from 20 horses with unilateral hindlimb lameness before and after successful local analgesia (assessed visually and objectively)

Brown adipose tissue transplantation ameliorates hindlimb ischemic damage in diabetic mice.

Peripheral arterial disease (PAD) is a common complication associated with diabetes, which can lead to foot ischemia. The condition is often accompanied by infection and necrosis, ultimately leading to diabetic foot ulcers and the risk of amputation. Brown adipose tissue (BAT) and its secreted cytokines play an essential role in the regulation of glucose homeostasis, the modulation of inflammatory responses, and vascular endothelial cell proliferation. The transplantation of BAT into ischemic regions may offer therapeutic benefits in alleviating the symptoms associated with PAD. A diabetic mou

Hindlimb unloading induces time-dependent disruption of testicular histology in mice.

Mechanical unloading of the body in the hindlimb unloaded (HU) mice induces pathology in multiple organs, but the effects on testes are poorly characterized. We investigated the histology and Raman spectroscopy of the mouse testes following HU condition. We divided male, c57BL/6j mice into ground-based controls or experimental groups for two and four weeks of HU. The testes tissues were dissected after euthanasia to investigate histological and Raman spectroscopic analysis. We found an HU-induced atrophy of testes irrespective of the time duration (p < 0.05). Histological analysis revealed tha

Pulsed electromagnetic field improves postnatal neovascularization in response to hindlimb ischemia.

Pulsed electromagnetic fields (PEMF) have been shown to promote proliferation and regeneration in the damaged tissue. Here, we examined whether PEMF therapy improved postnatal neovascularization using murine model of hindlimb ischemia, and the underlying cellular/molecular mechanisms were further investigated. Hindlimb ischemia was induced by unilateral femoral artery resection using 6-8 week-old male C57BL6 mice. Then, mice were exposed to extracorporeal PEMF therapy (4 cycles, 8min/cycle, 30 ± 3 Hz, 5 mT) every day until day 14. Our data demonstrated that PEMF therapy significantly accelerat

Rats exhibit aldosterone-dependent sodium appetite during 24 h hindlimb unloading.

Hindlimb unloading (HU) is an animal model of microgravity and bed rest. In these studies, we examined the role of ingestive behaviours in regulating body fluid balance during 24 h HU. In the first experiment, all rats were given distilled water to drink while two groups were also given access to a sodium chloride solution (0.9% or 1.8%). Water and saline intakes were measured before, during and after 24 h of HU. Rats reduced water intake during 24 h HU in all conditions. During HU, rats increased their intakes of both saline solutions (0.9% NaCl (n= 11): control 7.8 +/- 3 ml; HU 18.2 +/- 4 ml

Platelet activating factor-induced microvascular permeability increases in the cat hindlimb.

Changes in microvascular permeability induced by platelet activating factor (PAF) were measured in the isolated, perfused cat hindlimb preparation, and compared to the effect produced by another inflammatory mediator, histamine. Permeability was assessed from changes in the protein reflection coefficient, as measured from changes in hematocrit and protein concentration resulting from microvascular fluid filtration. The findings were 1) PAF produces transient increases in permeability similar to histamine, but PAF is approximately 30 times as potent; 2) the permeability changes induced by 76 nM

Hindlimb proportions, allometry, and biomechanics in Old World monkeys (primates, Cercopithecidae).

The traditional focus on morphological rather than mechanical units has obscured some significant functional differences in the hindlimbs of primates. This paper examines the allometric and biomechanical basis for some distinctive proportional differences among pairs of morphological units in the hindlimb, and especially the foot, of cercopithecid primates. Five major conclusions are reached. First, many hindlimb dimensions scale allometrically with body mass to maintain mechanical similarity within taxonomic and locomotor groups. Therefore, the majority of traditional indices which describe t

The effect of treadmill speed and gradient on equine hindlimb muscle activity.

REASONS FOR PERFORMING STUDY: Locomotion requires successful negotiation of different terrains, but we currently know little of how the musculoskeletal system adapts to cope with positive and negative slopes. OBJECTIVE: To compare the effects of treadmill speed and gradient on equine hindlimb muscle mean electromyographic (EMG) intensity. METHODS: Surface EMG recorded the activity of gluteus medius (GM), biceps femoris (BF), vastus lateralis (VL), gastrocnemius lateralis (GL) and extensor digitorum longus (EDL) in 6 horses at walk (1.4-1.6 m/s) and trot (2.6-3 m/s) on 3 different treadmill g

Sclerotomes in the thoracic and lumbar spine, pelvis, and hindlimb bones of rats.

UNLABELLED: Pain in lumbar radiculopathy shows a segmental distribution in muscles and bones, requiring knowledge of myotomes and sclerotomes for diagnosis of the involved nerve roots. The rostrocaudal coordinate in sensory space was examined for 49 reference sites placed on the periostium in the spine and hindlimb bones of rats to clarify the sclerotomes. Neurotracer 1,1'-dioctadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI) was applied to reference sites. DiI-labeled neurons were counted from the T9 through S3 dorsal root ganglia. The coordinate was calculated for each referenc

Differentiation between fore- and hindlimb bones and locomotor behaviour in primates.

Primate appendicular limb bones were measured on the cross-sectional geometry at the mid-length of the humerus and femur and on the external dimensions of long bones of the same individuals. Cross sections were directly measured by means of computer tomography or direct sectioning. The morphometry of bones and locomotor behaviour is discussed from the viewpoint of the functional differentiation between the fore- and hindlimbs. The primate group which daily adopted a relatively terrestrial locomotor type demonstrates robust forelimb bones compared with the group which adopted a fully arboreal l

Segmental regulation pattern of body surface temperature in the rat hindlimb.

Body surface zones or 'thermatomes', whose temperature is regulated by a single spinal segment, were investigated by thermography in the rat hindlimb. First, the spatial relation between the dermatome delineated by dye extravasation and the corresponding thermatome was investigated in rats pretreated with intravenous application of Evans blue. Electrical stimulation of the spinal nerves and sympathetic trunk segments at L3 and L5 induced a distal dominant temperature decrease. In contrast, Evans blue extravasation appeared in the medial (in L3) and lateral (in L5) paw only by electrical stimul

Blockade of morphine-induced hindlimb myoclonic seizures in mice by ketamine.

Morphine administration can lead to a variety of side-effects, including myoclonus. In an animal model, high morphine doses given intrathecally elicit hindlimb myoclonic seizures which are not influenced by traditional opioid receptor antagonists, such as naloxone. Ketamine prevents this seizure-like activity in a dose-dependent manner. The response is stereoselective, with S-ketamine far more potent than R-ketamine. A competitive NMDA antagonist, NPC17742, also prevents the seizures, although less potently than ketamine. Dextromethorphan has limited activity in this model, while haloperidol a

An electromyographic analysis of hindlimb function in Alligator during terrestrial locomotion.

The neuromuscular control of the hindlimb of American alligators (Alligator mississippiensis) walking on a treadmill was analyzed using simultaneous electromyography (EMG) and cineradiography. EMG and kinematic data were integrated with myological information to discern the interplay of muscles mediating hip and knee movement during the high walk. Twelve muscles, subdivided into 23 individual heads, cross the hip joint of Alligator. Activity patterns of 12 heads of 11 hip muscles and one knee muscle were recorded and quantified. An additional five heads from four muscles were recorded in singl

Effect on arterial pressure of rhythmically contracting the hindlimb muscles of cats.

Although static contraction of the hindlimb muscles of anesthetized cats is known to reflexly increase arterial pressure and heart rate, the cardiovascular effects of rhythmic contractions of these muscles is unclear. To help clarify this issue, we determined, in chloralose-anesthetized cats, the effects on arterial pressure and heart rate of rhythmically contracting the hindlimb muscles at a frequency of 5 Hz. In addition, we determined the effect of rhythmic contractions on the impulse activity of group III and IV muscle afferents whose activation is known to increase cardiovascular function

Hydrogen ion concentration and oxygen uptake in an isolated canine hindlimb.

Oxygen utilization (VO2) and lactate production by an isolated perfused canine hindlimb was evaluated at various hydrogen ion concentrations. A membrane lung perfusion system was established such that blood flow and temperature could be fixed at normal levels. Oxygen, nitrogen, and carbon dioxide (CO2) gas flows to the membrane lung were independently regulated to provide a fixed arterial oxygen content (CaO2). By changing CO2 flow, the pH of the arterial blood was varied between 6.9 and 7.6 at 10-min intervals. The mean O2 delivery (CaO2 X blood flow) was between 16.3 ML O2/min and 20.5 ml O2

An optogenetic mouse model of hindlimb spasticity after spinal cord injury.

Spasticity is a common comorbidity of spinal cord injury (SCI), disrupting motor function and resulting in significant discomfort. While elements of post-SCI spasticity can be assessed using pre-clinical SCI models, the robust measurement of spasticity severity can be difficult due to its periodic and spontaneous appearance. Electrical stimulation of sensory afferents can elicit spasticity-associated motor responses, such as spasms; however, placing surface electrodes on the hindlimbs of awake animals can induce stress or encumbrance that could influence the expression of behaviour. Therefore,
